1.Expression of cyclooxygenase-2 and inducible nitric oxide synthase in cervical carcinomas and its significance

Purpose:To investigate the relation of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) to tumor progression of cervical carcinoma. Methods:The expression and localization of COX-2 and iNOS protein in the 25 patients with cervical carcinomas were determined by immunohistochemical and the gene expression of COX-2 and iNOS were examined by reverse-transcription polymerase chain reaction ( RT-PCR). Results:Immunohistochemical staining for COX-2 and iNOS expression was strongly positive in 15 of 25 (60 %) and 20 of 25 (80 %) cases,respectively. The protein of BRCC36 is a kind of enzyme specifically hydrolyzing K 63-linked poly-ubiquitin chain and widely found in a variety of eukaryotic cells .BRCC36 recognizes diverse substrate proteins , and participates in various kinds of pathophysiological responses such as DNA damage repair , cell signal transduction and cell cycle control .It plays an important role in the process of cancer , angiogenesis and cardiac injury .This review discusses the progress in the inves-tigation on BRCC36 protein to provide the necessary information for searching new therapeutic targets of many diseases .
3.Classification and treatment of periprosthetic femoral fractures after hip arthroplasty

With the increasing number of hip arthroplasty and the extension of implant survival,the risk of periprosthetic femoral fractures are gradually growing.The technical challenge of the surgical management of periprosthetic femoral fractures leads to poor functional outcome,implant survival and patient satisfaction compared to primary hip arthroplasty.The risk factors of periprosthetic femoral fractures involve both of the bone quality of the patient and the surgical techniques of the primary surgery,including age and gender of the patient,osteoporosis,previous surgical history and the type of the components.In order to avoid the occurrence of periprosthetic fractures,precautions including the assessment of the patient status and bone quality as well as the selection of proper prosthesis and surgical procedures should be taken before the primary surgery.During the primary surgery,more attention should be paid to prevent the damage of host bone and make sure the correct placement of the implants.The aseptic loosening should be detected and treated as early as possible in the regular post-operative follow up.The diagnosis of periprosthetic femoral fracture mainly bases on the detailed history,symptoms,signs and serial X-rays after operation.The most extensively used classification system of the periprosthetic femoral fracture is the Vancouver classification.For the treatment of different types of fractures,conservative therapy is recommended for most Vancouver type A fractures.The surgical management is the best choice for most Vancouver type B fractures.Whether the stem revision is necessary or not depends on the stability the component.The treatment of Vancouver type C fractures should be in accordance with the principles of regular femur fractures.However,more attention should be paid to choose proper device according to the existence of the femoral component in the proximal part of the fracture.Through the analysis of the pathogenesis and risk factors of periprosthetic femoral fractures and the discussion of the diagnosis,classification and treatment principles,we expect to provide a standard treatment protocol for the periprosthetic femoral fractures following hip arthroplasty.
4.Pathological Observation of Therapeutic Action of Kuijieling Decoction on Ulcerative Colitis Rats

Objective To observe the therapeutic effect of Kuijieling Decoction on experimental ulcerative colitis rats.Methods Ulcerative colitis rat model was induced by enteroclysis with trinitro-benzene-sulfonic acid(TNBS).Three dosages(18.3 g/kg,9.2 g/kg,4.6 g/kg)of Kuijieling Decoction were used to treat the model rats.After treatment,the serum of rats were obtained to detect the level of IL-1? by enzyme-linked immunosorbent assay.Then the rats were killed for the observation of the colonic pathologic changes by naked eye and for the estimation of the damage scores of colon mucosa.Finally,the colon tissue was fixed to make pathological section and the damage index of colon mucosa was observed under microscope.Results The tissue damage scores of high-and moderate-dosage groups of Kuijieling Decoction were(2.41? 0.79)and(2.83? 0.71)respectively,and were significantly lower than that of model control group(3.75? 0.75;P

【Objective】To observe the effect of Kuijieling Decoction(KD) on NF-?B p65 protein expression in colonic mucosa and serum tumor necrosis factor ?(TNF-?) content of ulcerative colitis rats.【Methods】Sixty SD rats were randomized into 6 groups: normal group,model group,salicylazosulfapyridine(SASP,0.5g?kg-1?d-1) group and three KD(18.3,9.2 and 4.6 g?kg-1?d-1 respectively) groups.Except the normal group,the rats received enteroclysis of trinitro-benzene-sulfonic acid(TNBS) to induce ulcerative colitis models.Serum content of TNF-? was detected by enzyme-linked immunosorbent assay(ELISA).Then the rats were executed to make tissue section.The tissue sections were stained by immunohistochemistry and the cells with positive expression of NF-?B p65 protein were counted under microscope.【Results】The serum level of TNF-? and the cell number of positive expression of NF-?B p65 protein in high-and moderate-dose KD groups were lower than those in the model group(P

[Objective] To establish a determination method for isolated gastrointestinal intra-cavity perfusion pressure and to explore its application in pharmacological experiment. [Methods] Determination method for isolated gastrointestinal intra-cavity perfusion pressure was established according to motility of the isolated rat gastric muscle strip and water-sac perfusion manometry and ileum peristalsis of Guinea pigs. Intra-cavity pressure of isolated gastrointestinal canal of rat and guinea pig was observed. [Results] Gastric intra-cavity pressure was markedly increased as added with various doses of acetylcholine (1.25, 2.5, 3.25, 5, 6.25 and 7.5 ?g/mL respectively) in nutrient solution. The ileum intra-cavity pressure was also increased as added with various doses of histamine (0.125, 0.25, and 0.5 ?g/mL respectively) . Achievements of the 30-year research work on Pi-Wei Doctrine were summarized. They are: (1) ratio of salivin activity and excretion rate of D-xylose in patients with spleen deficiency are different from those in the normal subjects;(2) the principle of syndrome differentiation and treatment is embodied in the methodology of herbal medicine pharmacology for complex prescription; (3) the above findings are further confirmed by the research in molecular and cellular level which indicates that TCM syndromes and pharmacological mechanisms of herbal medicines have their material basis and the regulatory effect of complex prescription on TCM syndromes is actually the pharmacological mechanism of herbal medicines. It is suggested that the research of herbal medicine should be based on the guidance of traditional Chinese medicinal theory and carried out in clinical practice.

[Objective] To observe the regulatory effect of astragalosides (AST) on the binding capacity of gastrin receptor in gastric parietal cells (GPC) of rats with spleen deficiency. [Methods] SD rats were given gastric gavage of Radix et Rhizoma Rhei (Dahuang) decoction 10 g/kg to induce spleen deficiency. Gastric parietal cells were isolated and purified by Lewin gastric sac method and Percoll density gradient separation method. 2 mL suspension of GPC from rat models was incubated in vitro with AST 20 ?L at the concentrations of 10, 20 and 30 g/L respectively. Suspension of GPC from rat models incubated with phosphate buffered saline (PBS) served as the model group and suspension of GPC from the normal rats incubated with PBS as the normal group. The binding capacity of gastrin receptor in GPC was detected by radioligand receptor assay. Objective To provide a express plasmid carrier platform that can be in common use for preparation of RNase-resistant RNA standards and controls. Methods A cDNA fragment of MS 2 phage RNA genome, which encodes coat protein and maturase protein, and expression vector pET28b DNA are ligated together with T 4 DNA ligase after digested with HindⅢ and EcoR Ⅰ restriction nucleases. Then, a new expression plasmid carrier pI NCCL is contructed。 The prokaryotic expression was carried out by transform pI NCCL into BL21-DE3 E。Coli. Results A new expression plasmid carrier pI NCCL is contructed successfully. RNase-resistant virus-like particles were obtained after prokaryotic expression of pI NCCL. Conclusion The expression plasmid carrier pI NCCL contructed and prokaryotic expression system can be used as a express plasmid carrier platform that can be in common use for preparation of RNase-resistant RNA standards and controls.
