Objective To explore the effects of an acute exercise on the contractile protein degradation and 26S proteasome activity in skeletal muscle. Methods Thirty-six male Sprague-Dawley rats were randomly divided into 6 groups, each group consisted of 6 rats. They were killed respectively at rest, immediately after half an hour of exercise(running on the treadmill at 25m/min,5% grade),and 1 hour,2h and 6h after one hour exercise. The 3-methylhistidine(3-MH)content,26S proteasome activity and C2 subunit mRNA expression in rat gastrocnemius muscle were detected.Resuits(1)3-MH content increased immediately and 6 hours after exercise, and reached its peak level immediately after half an hour of exercise(P<0.01).(2)26S proteasome activity increased immediately after half an hour of exercise and 6 hours after exercise(P<0.05).(3)Expression of 26S proteasome C2 subunit mRNA Was lower than baseline during and after exercise, but increased sharply 6 hours after exercise(P<0.01).Conclusion The increase in 26S proteasome activity regulated by subunits gene after exercise could enhance skeletal muscle contractile protein degradation

OBJECTIVE:To explore the clinical efficacy of 131I in the treatment of hyperthyroidism.METHODS:131I therapy for 784 cases of hyperthyroidism was analyzed retrospectively. The dose of 131I(?Ci) was calculated with formula:?Ci=the weight of thyroid(g)?80-120(?Ci?g-1)/the highest absorbed iodine rate(%).Clinical efficacy was evaluated according to the results.RESULTS:394 cases were relieved completely after once therapy,523 cases after twice therapies and 547 cases after three times of therapies.577 patients were visited at random and 94.80% of them were cured.Early-stage toxic reaction had been found in 46 cases with incidence of 7.97%.Hyperthyroidism crisis had not been developed.Temporary hypothyroidism had been found in 68 cases with incidence of 11.79%,while permanent hypothyroidism had been found in 146 cases with incidence of 25.30%.CONCLUSION:131I has an obvious effect on hyperthyroidism with one disadvantage of high incidence of hyperthyroidism.However,131I is an effective therapy which is safe, economical, simple and convenient.

[Summary] To discuss blood glucose control in patients with type 2 diabetes undergoing insulin therapy.A randomized controlled trial was conducted in type 2 diabetics who were taking insulin and who had an HbA1C level ≥ 8%,with a 6-month follow-up period.70 diabetic patients were randomly and equally divided into control group who received standard care, and intervention group who received an individualized personalized medical treatment with health education. The main outcome measures were change of HbA1C,diabetes and medication knowledge, adherence to medications, family blood glucose monitoring, and insulin injection techniques.Questionnaire was used to evaluate the outcomes before and after the intervention. The medication and diabetes knowledge, medication adherence, the correct way for home blood glucose monitoring were significantly improved in intervention group, while remained unchanged in the control group. After 6 months,HbA1C values were significantly reduced in the intervention group while remained unchanged in the controls. The quality of life also significantly improved in the intervention group.

OBJECTIVETo prepare a pH fluorescence probe based on styrylcyanine dyes for live cell imaging.
METHODSThe Probe 1 was prepared by reaction of 4-pyridinecarboxaldehyde with 1,1,2-trimethylbenz[e]indole. The influence of pH on the fluorescent properties was examined, and the cell viability was examined using cell counting kit-8. The Probe 1 was used as a pH fluorescence probe in living cell.
RESULTSProbe 1 emitted green fluorescence under neutral and basic conditions but orange fluorescence under acid condition. Probe 1 selectively stained the cytoplasmic regions of living cells without significantly affecting the cell viability.
CONCLUSIONThe pH-sensitive fluorescent probe prepared based on styrylcyanine possesses good ability of cell membrane permeation for live cell fluorescent imaging.

OBJECTIVE:
To investigate the expression of apoptosis related protein cellular Fas associated death domain like interleukin 1 converting enzyme inhibitory protein (c-FLIP) in peripheral blood mononuclear cells (PBMCs) of patients with rheumatoid arthritis and its relation with extrinsic apoptotic pathway.
METHODS:
Sixty patients with rheumatoid arthritis were collected from Zhangzhou Affiliated Hospital of Fujian Medical University during January 2014 and June 2015, including 22 patients with low activities (DAS28<3.2), 20 patients with middle activities (3.2 ≤ DAS28 ≤ 5.1), and 18 patients with high activities (DAS28>5.1). And 25 healthy controls were also collected. The mRNA and protein expression levels of c-FLIP and the extrinsic apoptotic pathway related proteins Fas-associated protein with death domain (FADD), caspase-8 in PBMCs were detected by real-time RT-PCR and Western blot, respectively. Correlations between c-FLIP and FADD, caspase-8 in PBMCs were analyzed by pearson test.
RESULTS:
mRNA expression levels of c-FLIP, FADD and caspase-8 in PBMCs of patients with rheumatoid arthritis were all higher than those of healthy controls (all <0.05). mRNA expression levels of FADD and caspase-8 in patients with middle activities were significantly higher than those in patients with low activities (all <0.05), but the mRNA expression level of c-FLIP was not significantly higher than that in patients with low activities. mRNA expression level of c-FLIP in patients with high activities was higher than those in patients with middle or low activities (all <0.05), while the mRNA expression level of caspase-8 was lower than those in patients with middle or low activities (all <0.05). mRNA expression level of FADD in patients with high activities was higher than those in patients with low activities (<0.05). Pearson analysis showed that there was a positive correlation between c-FLIP and FADD mRNA expression (=0.323, <0.05), and negative correlation between c-FLIP and caspase-8 mRNA expression (-1.104, <0.05). The protein expression levels of c-FLIP and FADD in patients with middle activities were significantly higher than those in control group and patients with low or high activities (<0.05 or 0.01). The protein expression levels of caspase-8 in patients with middle and high activities were significantly higher than those in control group and patients with low activities (<0.05 or <0.01), and the protein expression level of caspase-8 in patients with high activities was higher than that in patients with middle activities (<0.05).
CONCLUSIONS
c-FLIP may be involved in the extrinsic apoptotic pathway in rheumatoid arthritis, and can provide reference for the evaluation of disease activities.

OBJECTIVETo identify the MPL L391-V392ins12 spliceosome and analyze its frequencies in patients with myeloproliferative neoplasms (MPN).
METHODSMPL aberrant spliceosome was identified through reverse transcription polymerase chain reaction (RT-PCR)combined with cloning sequencing. The mutation of this spliceosome in 248 MPN patients and 200 normal people was determined by allele-specific polymerase chain reaction (AS-PCR).
RESULTSA novel aberrant spliceosome of MPL gene (MPL L391-V392ins12)was identified, i.e. 36 bp intron was retained between exon7 and exon8, and there were 12 amino acids (EGLKLLPADIPV)inserted. MPL L391-V392ins12 mutation was detected in 19 (7.66%)of the 248 patients with MPN, including 1 (1.92%) of 52 patients with PV, 14 (9.66%) of 145 with ET, and 4 (7.84%) of 51 with PMF. And the mutation was not detected in the group of 200 normal people.
CONCLUSIONMPL L391-V392ins12 spliceosome is an aberrant spliceosome present in the MPN. It can be detected in PV, ET and PMF, and more frequently in ET and PMF. This mutation may play an important role in the process of MPN.
