1.Exploration of Deoxyschizandrin Metabolic Processes in Vivo Based on Blood Microdialysis

Objective To establish an HPLC method for evaluating deoxyschizandrin in rat plasma; To explore deoxyschizandrin metabolic processes in blood.Methods The blood microdialysis probe was implanted into the right atrial enlargement through right jugular vein to detect the concentration of deoxyschizandrin in blood. Chromatographic conditions for detecting of dialysis fluid were as follows: mobile phase was methanol-acetonitrile- water (40:35:25); flow rate was 1 mL/min; column temperature was 30℃; UV detection wavelength was 254 nm. The data obtained after being calculated by the recovery rate of the probe in vivo were dealt with DAS 2.0 software. Pharmacokinetic parameters were calculated, and the concentration-time related curves were plotted.Results The deoxyschizandrin curve was linear (r=0.999 5) within the range of 2–500μg/mL. The RSD of intra-day pecision and inter-day pecision were both <3%. The stability of the solution was good in 12 hours at roomtemperature. ke was (0.364±0.047)/h; t1/2 was (1.637±0.302)h; Tmax was (0.612±0.194)h; Cmax was (165.92±28.830)μg/mL; AUC0-t was (240.793±25.540)μg?h/mL; AUC0-∞ was (282.710±30.727)μg?h/mL.Conclusion The method is convenient to perform, and the retention time of HPLC is short. This method is suitable for detecting the content of deoxyschizandrin in dialysis fluid, which can also be used to investigate the metabolic processes of deoxyschizandrin in the rat blood.
2.Impact of conflict psychological stress on rats'sleep time and sleep phrase

ObjectiveTo discuss the impact of conflict psychological stress on rats'sleep time and sleep phrase.Methods20 Wistar male rats were randomly divided into vehicle group and model group,10 in each group.The improved experiment of Vogel drinking water conflict was used to establish the model of conflict psychological stress in rats and monitoring the changes of rats'weight and concentration of corticosterone (CORT) in serum after a week.After one week,the 12 h EEG and EMG of rats were traced by the polygraph in order to clarify the effect of conflict psychological stress on rats′ sleep time and sleep phrase.ResultsAfter one week,compared with vehicle group the weight of model group decreased significantly ( t =2.87,P < 0.05 ),the concentration of CORT in serum raised significantly( t=5.19,P < 0.05 ).A week later,compared the vehicle group,total sleep time( TST),light sleep,slow wave sleep(SWS) and rapid eye movement sleep (REM) time in model group rats ( (337.8 ±84.2)min,( 169 ±49.2) min,( 136.9 ±40.8)min,(33.1 ± 10.1 ) min for vehicle group; ( 155.4 ±16.7 ) min,( 111.6 ± 16.4 ) min,( 30.6 ± 4.9 ) min,( 12.6 ± 3.3 ) min for model group) were significantly reduced ( t =6.75,2.43,5.51,5.78,P < 0.05 ),and the proportion of lightsleep increased ( (49.6 ± 3.4 ) %,( 71.7 ±5.4)% ) ( t =6.90,P<0.05),the proportion of SWS decreased( (40.3 ± 10.9)%,( 19.8 ±3.2)% ) ( t=6.74,P < 0.05 ) ;but no significant change in the proportion of REM ( t =1.57,P> 0.05 ).Rats,sleep-wake rhythm figure indicated that the rats'sleep duration reduced and the number of awakening increased ; sleep tended to be fragmentation.ConclusionsConflict psychological stress lead to hypothalamus-pituitary-adrenal axis hyperactivity,weight loss,and the rhythm of sleep and sleep phase change in rats.
3.Advances in nano-hydroxyapatite and its composite

Nano-hydroxyapatite has been widely used as reconstructive and prosthetic material for osseous tissue,owing to its excellent biocompatibility and tissue bioactivity.But the poor mechanical property of hydroxyapatite restricts its further application.In order to enhance the comprehensive performance of the material,many researches have been dedicated to the synthesization of the composite materials.This article reviews the main preparation methods of nano-hydroxyapatite and the advancement in research of its composite.The directions in this research area are described as well.
4.Traditional Chinese medicine for cancer pain.

Pain is one of the common symptoms of cancer which seriously affects the quality of life of the patients. Cancer pain is mainly treated with the three-step method, biological therapy or nerve block therapy based on antitumor therapy. However, up to 50 percent of patients with cancer-related pain do not receive adequate pain relief, affecting their physical and psychological well-being, and leading to a lower quality of life for the patient after conventional treatment. Clinical observation suggests that traditional Chinese medicine may alleviate cancer-related pain either by oral administration, topical administration, acupuncture or other means with continuing non-addictive and non-drug-resistant qualities. However, scientific evaluation of the efficacy of herbs in the treatment of pain is insufficient; the underlying mechanisms are unclear and, safety and toxicity remain a concern.
5.Collagen/silk fibroin nerve conduits used for repairing peripheral nerve defect:application and development

BACKGROUND:Peripheral nerve defect due to limb dysfunction has always been the difficulty faced by the medical profession. Ideal materials and processing technology for constructing a tissue engineering scaffold targeting peripheral nerve repair are stil in research stage. OBJECTIVE:To review the research progress in peripheral nerve repair using col agen/silk fibroin nerve conduits. METHODS:In this paper, the first author retrieved the PubMed and CNKI from 2003 to 2016 to search articles regarding methods of constructing artificial nerve scaffolds and selection of raw materials. Data from these articles were col ected, summarized and analyzed. RESULTS AND CONCLUSION:Forty-six articles were included for final analysis. Col agen and its degradation products trigger no inflammatory response in the host because of high biocompatibility and biodegradability. However, its use is largely limited by its rapid degradation and poor physical performance. Silk fibroin has a high flexibility and biocompatibility, and exhibits a slow degradation in vivo. As a rapid prototyping technique, three-dimensional printing can print various forms of scaffolds within a short time, characterized as high-quality pore structure and large-scale production. Given these, the col agen/silk fibroin nerve conduit prepared using the three-dimensional printing technology can maintain the biocompatibility and even improve the mechanical properties of the raw materials. Until now, more investigations on nerve repair using col agen or silk fibroin have been done, and we have never stopped improving the production process of these scaffolds. Therefore, the col agen/silk fibroin scaffold prepared using the three-dimensional printing technology is expected to become the main candidate for the repair of peripheral nerve defects.
6.Effect of lyophilized powder of Sini powder on 5-HT content and 5-HT(1A) receptor in drosophila heads.

OBJECTIVETo unveil the mechanism of sleep-improving effect of lyophilized powder of Sini powder on drosophila.
METHODDrosophila melanogaster whose strain were wild Canton S, unmated, were collected daring 12 h after their emergencing, then they were anaesthetized by small flow of CO2 and divided into different tubes due to their sex. Both male and female flies were distributed randomly into three groups as blank, control and administration, 32 flies each group. The content of 5-HT in the brain was estimated was estimated with ELISA and had a comparison of the expression of 5-HT(1A) receptor in brain, using real-time quantitative PCR.
RESULTConcentration of 5-HT in brain was increased in administration group, and had a significant difference to control group. The expression of 5-HT(1A) receptor was also up-regulated after administrated of Sini powder.
CONCLUSIONSleep-improving effect of Sini powder on drosophila has a contribution to the increasing of 5-HT and 5-HT(1A) receptor in brain.

7.Review of experimental model of bone defect in situ healing under mechanical loading

Tissue engineering composites for bone defect treatment is currently a hot spot of the research field.Mechanical loading on tissue engineering composites is important to bone formation and related research has attracted more and more attention.In the field of tissue engineering research,the establishment of a standardized animal model is the basis of experimental research for exploring the effects of different mechanical loading on in situ repair of bone defect and its action mechanism.This paper reviews the approaches used to establish experimental model of bone defect under mechanical loading such as animal selection,defect preparation,fixation and loading mode in order to provide a reference for related research.
8.Optimization of the Extraction Technology of Garlic Oil by Box-Behnken Response Surface Method

OBJECTIVE:To optimize the extraction technology of garlic oil. METHODS:Using extraction rate of garlic oil as index,based on single factor test,Box-Behnken response surface method was used to optimize conditions of steam distillation method for the extraction of garlic as fermentation time,solid to liquid ratio,fermentation temperature and the verification test were made for the optimized technology. RESULTS:The optimal extraction technology was as follows as fermentation time of 4.5 h,solid to liquid ratio of 1:7,fermentation temperature of 55 ℃. The average extraction rate of garlic oil in verification test was 0.32%(RSD=1.43%,n=3);the relative error between the measured value and predicted value was 0.06%. CONCLUSIONS:Box-Behnken response surface method is simple,reasonable and feasible to optimize the extraction technology of garlic oil,which can provide a scientific basis for industrial production.
9.Gut microbiota and type 2 diabetes

Recently, gut microbiota is found to be associated with human health and diseases. Environmental and genetic factors both play pivotal roles in the pathogenesis of type 2 diabetes, in which the gut microbiota is regarded as an important environmental factor. In addition, gut microbiota may become a novel therapeutic approach for type 2 diabetes. Present review focused on the association between the alterations of gut microbiome and type 2 diabetes, hyperglycemia, and insulin resistance;also summarized the molecular mechanisms underlying microbe-host interactions in the context of diabetes, potential contribution of the gut microbiota to the pharmacological or surgical treatment of type 2 diabetes, and the interaction of human genetics with gut microbiome. Based on the understanding of gut microbiota-host interactions, probiotic and fecal microbiota transplantation will be potential novel therapeutic approaches for type 2 diabetes.
10.Experimental Study of Vascular Tissue Engineering

Objective To obtain the experimental data of vascular tissue engineering.MethodsThe vascular endothelial cells (VEC) and vascular smooth muscle cells (VSMCs) were acquired and cultured, and then seeded on vascular tissue engineering materials. The porous gelatin-chitosan scaffold with VSMCs was subcutaneously implanted, followed by the observation of the cell growth ten days later.ResultsThe two kinds of cells were successfully cultured and their morpholoical and immunohistochemical characteristics were consistent with vascular endothelial and VSMCs respectively. The VSMCs could grow extensively on the scaffold after the in vivo implantation. The scaffold were wrapped by the fibrous tissue ten days later after the in vitro implantation of VSMCs. The seed cells grew in the scaffold, and the vessel cavity seen in the center of the scaffold, was quite different from the normal vessel structure.ConclusionIt is feasible to implant the VSMCs with fibrin gels into the living body. The vessels reconstructed, though different from the normal structure, is similar to the embryo of the vessels.
