1.Determination simultaneously of gentiopicroside and geniposide in Longdan Xiegan Pill by HPLC

AIM:To establish a HPLC method for determining simultaneously gentiopicroside and geniposide in Longdan Xiegan Pill(Radix et rhizoma Gentianae,Fructus Gardeniae,Radix Scutellariae,etc.).METHODS:C_ 18 column(5 ?m,4.6 mm?250 mm)was used.The mobile phase was methanol-water(23∶77).The detecting wavelength and the temperature of column were at 238 nm and at 40 ℃ respectively.RESULTS:The linear detection range of gentiopicroside was 3.03-30.30 ?g/mL(r=1.000 0),and the average recovery was 98.43%(RSD=1.08%).The linear detection range of geniposide was 9.76-97.60 ?g/mL(r=0.999 9),and the average recovery was 99.95%(RSD=0.91%).CONCLUSION:Two ingredients in Longdan Xiegan Pill could be determined simultaneously by this method which is convenient,rapid and accurate.It can be used for quality control in the production of Longdan Xiegan Pill.
2.Decoction for Longdan Xiegan Tang vs. Longdan Xiegan Pills in Equivalence

OBJECTIVE:To investigate the equivalence of preparations of traditional Chinese medicines with decoction and pill as the representative dosage forms so as to lay a foundation for the evaluation of equivalence.METHODS:Taking the contents of gentiopicroside and geniposide and water soluble extractive as indexes to investigate the equivalence between decoction for Longdan xiegan tang and Longdan xiegan pills.RESULTS:Evaluated by the indexes of contents of gentiopicroside and geniposide,the current daily oral dosage of the decoction was 6.39 times superior to the pill;taking water soluble extractive as index,the decoction was 4.02 times superior to the pill.CONCLUSION:To be equivalent to the decoction,the current daily oral dosage of the pill should be increased appropriately.The degree of increase should be based on the final results of pharmacological and clinical studies.
3.Development and verification of a RP-HPLC method for determination of residual trifluoroacetic acid in human interferon alpha 2b stock solution

ObjectiveTo develop and verify a reversed-phase high performance liquid chromatography(RP-HPLC)method for the determination of residual amount of trifluoroacetic acid(TFA)in human interferon alpha 2b(hIFNα2b)stock solution,and preliminarily apply the method.MethodsThe conditions of liquid chromatography were as follows:ZORBAX300SB-C18 column(4. 6 mm × 250 mm,5 μm);mobile phase:0. 08% phosphoric acid(pH 3. 0)+ 5. 0% methanol solution;detection time:6 min;flow rate:0. 8 mL/min;column temperature:35 ℃;UV wavelength:210 nm;injection volume:20 μL;isocratic elution. The system adaptability,linearity,specificity,accuracy,precision,limit of quantitation(LOQ),limit of detection(LOD)and durability of the method were verified. The TFA residues in three batches of hIFNα2b stock solution samples were determined by the developed method and composite column Comixsil ACRP(100 mm × 4. 6 mm,5 μm).ResultsThe linearity of TFA was good in the range of 10-300 μg/mL(R2= 0. 998 4). The average spike recovery rate was 101. 8% and RSD was 2. 68%. The RSDs of repeatability and intermediate precision verification were 0. 46% and0. 45%,respectively. The LOQ was 10 μg/mL,and the LOD was 2 μg/mL. The slight fluctuation of pH of organic solvent,buffer salt and temperature of column under the specified conditions had no obvious influence on the detection results,and the peak area was affected when the detection wavelength changed. In addition,TFA residues were not detected in three batches of hIFNα2b stock solution by two methods.ConclusionThe developed RP-HPLC method has good system adaptability,specificity,accuracy,precision and durability,and can be used for the determination of TFA residues in hIFNα2b stock solution.
4.Exploration of Deoxyschizandrin Metabolic Processes in Vivo Based on Blood Microdialysis

Objective To establish an HPLC method for evaluating deoxyschizandrin in rat plasma; To explore deoxyschizandrin metabolic processes in blood.Methods The blood microdialysis probe was implanted into the right atrial enlargement through right jugular vein to detect the concentration of deoxyschizandrin in blood. Chromatographic conditions for detecting of dialysis fluid were as follows: mobile phase was methanol-acetonitrile- water (40:35:25); flow rate was 1 mL/min; column temperature was 30℃; UV detection wavelength was 254 nm. The data obtained after being calculated by the recovery rate of the probe in vivo were dealt with DAS 2.0 software. Pharmacokinetic parameters were calculated, and the concentration-time related curves were plotted.Results The deoxyschizandrin curve was linear (r=0.999 5) within the range of 2–500μg/mL. The RSD of intra-day pecision and inter-day pecision were both <3%. The stability of the solution was good in 12 hours at roomtemperature. ke was (0.364±0.047)/h; t1/2 was (1.637±0.302)h; Tmax was (0.612±0.194)h; Cmax was (165.92±28.830)μg/mL; AUC0-t was (240.793±25.540)μg?h/mL; AUC0-∞ was (282.710±30.727)μg?h/mL.Conclusion The method is convenient to perform, and the retention time of HPLC is short. This method is suitable for detecting the content of deoxyschizandrin in dialysis fluid, which can also be used to investigate the metabolic processes of deoxyschizandrin in the rat blood.
5.EFFECTS OF STATIC EXERCISE AND DYNAMIC EXERCISE ON SYSTOLIC TIME INTERVALS (STI) IN CHILDREN

The purpose of this study was to investigate STI changes resulting from static and dynamic exercises in boys, 9-10 years old, so as to reveal the effects of physical work of different types and loads on the heart function.Our research showed that the effects of static load and dynamic load on STI in children were different. After dynamic exercise all items of STI changed significantly and PEP/LVET decreased significantly. No PEP change was found after static exercises (50% and 80% max load). Static exercise of 50% max load resulted in no IVCT change, but static exercise of 80% max load resulted in very significant change of IVCT, and the longest recovery period was found afterwards. Thus, 80% static exercise would result in a heavy, transient heart load in children.Our research also showed that the changes of HR?systolic blood pressure?LVET after dynamic load and static load were significantly different indicating that oxygen consumption in heart muscles was higher during dynamic exercise and the effect of dynamic exercise on heart was stronger.
6.Clincial investigation of functional neck dissection in the treatment of multiple cervical tuberculous lymphadenopathy

Objective To compare the clinical efficacy of traditional excision of involved lymph nodes and functional neck dissection in the treatment of patients with multiple cervical tuberculous lymphadenopathy(MCTLP).Methods From Jan 2006 to May 2008,76 patients with MCTLP who presented with focal liquefaction or sinus formation after ineffective conservative therapy were randomly divided into study group(functional neck dissection) and control group.Data regarding the 3-month recurrence rate,postoperative neck function,the mean hospitalization days and medical costs of the 2 groups were compared.Results There was significant difference in relapse rate 3 months postoperatively between study group(1 case,2.56%) and control group(7 cases,16.22%)(
7.On "Pill,Is Sluggish"

OBJECTIVE: To provide reference for research & development and application of pill preparation. METHODS: Our understanding on the statement "Pill, Is Sluggish" was come up with, and the pertinetn literature and research results were analyzed and summarized. RESULTS: The statement "Pill, Is Sluggish" is not only the description of the traditional pill's dosage-form characteristic, but also contains the requirement on administration method. It is a relative description. There is also difference in speed of taking effect between different kinds of pills. Its origin contains the factor of dosage as well as the factor of dosage-form; It does not apply to some new type pills. CONCLUSION: The connotation of "Pill, Is Sluggish" is extremely rich, which should be viewed relatively, comprehensively and developmentally so as to continuously enrich and develop the dosage form theory of Chinese materia medica and make pills serve better for the clinic.

9.The relationships between CTLA-4 gene exon 1 A/G~(49) polymorphism and type 1diabetes mellitus

Objective To investigate the association of cytotoxic T lymphocyte associated antigen 4(CTLA 4) gene polymorphism with type 1 diabetes in Chinese Han population.Methods The A/G phenotype at position 49 of the CTLA 4 gene exon 1 was determined by polymerase chain reaction restriction fragment length polymorphism(PCR RFLP)method in 33 typical type 1 diabetes patients,57 latent autoimmune diabetes in adults(LADA) patients and 84 healthy control subjects of Chinese Han.Results The frequency of the CTLA 4/G 49 phenotype was significantly higher in type 1 diabetes patients than in control subjects(55.6% vs 36.9%, respectively, P =0.0005),but there was no significant difference between typical type 1 DM and LADA groups. Neither the presence nor the absence of G 49 allele influenced the occurrence of islet autoantibody(ICA) and glutamate decarboxylase antibody(GADAb).Conclusion The polymorphism of CTLA 4 gene exon1 confers susceptibility to type 1 diabetes. This association is independent of ICA and GADAb.
10.Application of radiofrequency vaporization in arthroscopic treatment of knee disorders

Objective To investigate effects of radiofrequency vaporization in arthroscopic treatment of knee disorders.Methods Arthroscopic surgery was conducted in 126 consecutive cases of various knee disorders by using the Arthrocare 2000 System,includng 72 cases of meniscus injury,20 cases of synovitis,15 cases of osteoarthritis,8 cases of patella subluxation,6 cases of synovial syndrome,and 5 cases of anterior cruciate ligament rupture.Results All the patients were followed postoperatively.Knee function evaluation with the Neer Scores showed excellent outcomes in 82 cases,good in 35 cases,and fair in 9 cases,the rate of excellent-or-good outcomes being 92.9%.Conclusions Application of radiofrequency vaporization under arthroscopy for knee joint disorders has advantages of simple and accurate performance,effective hemostasis,minimal invasion,and quick recovery of joint functions.
