1.Optimization of the Formulation of Alprazolam Sustained-release Tablet and Study on Its Release in Vitro Characteristics

OBJECTIVE:To optimize the formulation of alprazolam sustained-release tablet and investigate its in vitro release profile.METHODS:Orthogonal experiment was carried out to optimize the formulation of alprazolam sustained-release tablet with the matching percentages of hydroxypropyl methyl cellulose(HPMC),ethyl cellulose and lactose and the specification of the HPMC as factors and with the in vitro release rate as index.The in vitro release rate of the optimized preparation was investigated as well.RESULTS:The optimal formulation of alprazolam sustained-release tablet as follows:the matching percentages of HPMC,ethyl cellulose and lactose were 30%,4% and 15%,respectively,and the specification of the HPMC was K4M.The alprazolam sustained-release tablet prepared in this optimized formulation achieved a sustained release of 24 h with its release profile in line with the Higuchi equation.CONCLUSION:The optimized alprazolam sustained-release tablet is reasonable in formulation and characterized by sustained release.
2.Content Determination of Psoralen in Psoralea-pericarpium Juglates Liniment by HPLC

OBJECTIVE:To establish a method for the content determination of psoralen in Psoralea-pericarpium juglates lini-ment. METHODS:HPLC was performed on the column of Shim-pack VP-ODS C18 with mobile phase of methanol-water(60∶40, V/V)at a flow rate of 0.8 ml/min,detection wavelength was 246 nm,column temperature was 25 ℃,and injection volume was 20μl. RESULTS:The linear range of psoralen was 2.49-39.90 μg/ml;RSDs of precision,stability and reproducibility tests were lower than 2.00%;recovery was 97.23%-100.58%(RSD=1.17%,n=6). CONCLUSIONS:The method is fast,simple,accurate and re-liable,and can be used for the content determination of psoralen in Psoralea-pericarpium juglates liniment.
3.Determination of the Content of Meropenem and Observation of the Stability of Meropenem in Mixing With Infusion Fluids

OBJECTIVE:To establish a HPLC method for determining the content of Meropenem and to observe the stability of Meropenem in mixing with 5%GS,GNS and NS.METHODS:The conditions of HPLC were:column,Nova-Pak C18;mobile phase,acetonitrile-0.05mol KH2PO4(4∶96);flow rate,1.0ml/min;detection wavelength,300nm.After Meropenem mixing with 5%GS,GNS and NS,the changes of external apperance were observed in room temperature,the content was detected with HPLC and the pH and insoluble particles were determined.RESULTS:The calibration curve was in good linearity in the range of 12.5~250?g/ml(r=0.9 999)and the recovery was 99.7% with RSD=1.1%.CONCLUSION:This detection method is effective,rapid and accurate.Meropenem is stable in infusion fluids for 4 hr.
4.Preparation Technology of Psoralea corylifolia-Juglans regia Tincture

OBJECIVE:To study the preparation technology of Psoralea corylifolia-Juglans regia tincture.METHODS: Taking Psoralea corylifolia and Juglans regia as raw material,which were extracted respectively with ethanol and mixed after having been condensed,then added with surface-active agent to prepare the compound preparation Psoralea corylifolia-Juglans regia tincture.RESULTS: The tincture was bright and uniformly sable in color.CONCLUSION: The preformulation and the preparation technology are both reasonable and the tincture is stable in quality.
5.A comparative study on two liver suspension methods in laparoscopic radical gastrectomy

Objective:To compare liver adhesive suspension method and V-shaped liver suspension of anterior abdominal wall in laparoscopic radical gastrectomy.Methods:This study retrospectively evaluated 115 patients who underwent laparoscopic radical gastrectomy from Jan 2018 to Dec 2019 at Shanxi Provincial People′s Hospital, 59 cases were under liver adhesive suspension (study group) and 56 cases under V-shaped liver suspension of anterior abdominal wall (control group). Intraoperative and postoperative conditions were observed and compared between the two groups.Results:All the 115 cases successfully completed surgery. The liver suspension time in study group was (2.4±0.5)min, which was significantly shorter than that of the control group((12.5±2.6) min)( t=-28.100, P<0.01). The total operation time in was (284±69) min and (307±78) min, respectively, ( t=-1.656, P=0.10). There were no statistically significant differences between the two groups in preoperative ALT, AST, total bilirubin, intraoperative incidence of hepatic lacerations and subcapsular hematoma, postoperative total bilirubin level, AST on the day 7 postoperatively, tumor location, TNM staging, surgical method, postoperative duration of hospital stay, and VAS score ( P>0.05). Conclusions:Both methods of liver suspension are safe and feasible in laparoscopic surgery for gastric cancer.Compared with V-shaped liver suspension of anterior abdominal wall, liver adhesive suspension has the advantages of less invasive, safe, and time-saving.
6.Metastatic status of lymph nodes in patients of distal gastric carcinoma

Objective To investigate lymph node metastases in distal gastric cancer and its clinical significance. Methods From June 2006 to December 2007, 129 distal gastric cancer patients underwent radical gastrectomy with lymphadenectomy. Dissected lymph nodes were collected in groups, and histopathological studies were performed to detect lymph node metastasis. The relationship between lymph node metastasis and tumor parameters such as diameter, location, infiltrating depth, histological category,Borrmann typing was evaluated. Results Lymph node metastases was found in 80 out of 129 patients (62%). A total of 3295 lymph nodes were harvested with an average of 25.54 lymph nodes per patient,among those 889 lymph nodes were identified with metastasis. The metastasis rate was 18.60%, 48. 84%,37. 98%, 38. 76%, 44. 19%, 31.01%, 10. 85%, 14. 73%, 4. 65%, 1.55% and 0. 78% respectively in No. 1, No. 3, No. 4d, No. 5, No. 6, No. 7, No. 8a, No. 9, No. 11 p, No. 12a, No. 14v lymph node group.No. 3 and No. 6 group nodes were moat frequently invaded by metastasis. Conclusion This study provides the regular pattern of lymph node metastasis in distal gastric carcinoma patients undergoing radical gastrectomy and perigastric lymphadectomy which helps to guide lymphadectomy in terms of less trauma and favorite prognosis.
7.The prevalence, awareness, treatment and control rates of diabetes among residents in Fangshan District

Objective:
To investigate the prevalence, awareness, treatment and control rates of diabetes among residents in Fangshan District, Bejing Municipality, so as to provide reference for strengthening the prevention and control of diabetes.
Methods:
The residents aged 18 to 79 years were selected in Fangshan District using multistage stratified cluster sampling method from March to June 2023. Demographic information, lifestyle, body mass index, fasting blood glucose, and blood lipid were collected through questionnaire surveys, physical examinations and laboratory tests. The prevalence, awareness, treatment and control rates of diabetes among residents were analyzed and their influencing factors were identified using a multivariable logistic regression model.
Results:
A total of 4 438 residents were enrolled, including 2 367 males (53.33%) and 2 071 females (46.67%), and had a mean age of (44.99±14.90) years. The prevalence, awareness, treatment and control rates of diabetes were 15.75%, 57.37%, 56.22% and 22.90%, respectively. Multivariable logistic regression analysis showed that age (aged 30 to 39 years, OR=1.624, 95%CI: 1.011-2.609; aged 40 to 49 years, OR=3.469, 95%CI: 2.163-5.561; aged 50 to 59 years, OR=4.202, 95%CI: 2.595-6.803; aged 60 to 69 years, OR=7.229, 95%CI: 4.406-11.862; aged 70 to 79 years, OR=7.929, 95%CI: 4.665-13.479), obesity (OR=1.368, 95%CI: 1.071-1.748), hypertension (OR=1.943, 95%CI: 1.601-2.359) and dyslipidemia (OR=2.754, 95%CI: 2.287-3.317) were the factors affecting the prevalence of diabetes. Sex (women, OR=1.529, 95%CI: 1.019-2.293), age (aged 40 to 49 years, OR=4.971, 95%CI: 1.681-14.698; aged 50 to 59 years, OR=6.070, 95%CI: 2.045-18.020; aged 60 to 69 years, OR=6.902, 95%CI: 2.287-20.832; aged 70 to 79 years, OR=6.954, 95%CI: 2.191-22.070) and hypertension (OR=1.906, 95%CI: 1.337-2.718) were the factors affecting the awareness of diabetes. Age (aged 40 to 49 years, OR=4.699, 95%CI: 1.594-13.855; aged 50 to 59 years, OR=5.990, 95%CI: 2.023-17.732; aged 60 to 69 years, OR=7.134, 95%CI: 2.370-21.470; aged 70 to 79 years, OR=6.639, 95%CI: 2.102-20.964) and hypertension (OR=1.857, 95%CI: 1.304-2.645) were the factors affecting the treatment of diabetes. Age (aged 40 to 49 years, OR=2.948, 95%CI: 1.349-6.438; aged 60 to 69 years, OR=2.344, 95%CI: 1.049-5.234) was the factor affecting the control of diabetes.
Conclusions
The prevalence rate of diabetes among residents in Fangshan District was high, and the improvements of awareness rate, treatment rate and control rate were not obvious. Health education and management of diabetes should be strengthened for elderly, hypertensive and dyslipidemic residents.
8.In vivo quantification of mandibular bone remodeling and vascular changes in a Wistar rat model: A novel HR-MRI and micro-CT fusion technique

Purpose:
This study was performed to introduce an in vivo hybrid multimodality technique involving the coregistration of micro-computed tomography (micro-CT) and high-resolution magnetic resonance imaging (HR-MRI) to concomitantly visualize and quantify mineralization and vascularization at follow-up in a rat model.
Materials and Methods:
Three adult female rats were randomly assigned as test subjects, with 1 rat serving as a control subject. For 20 weeks, the test rats received a weekly intravenous injection of 30 μg/kg zoledronic acid, and the control rat was administered a similar dose of normal saline. Bilateral extraction of the lower first and second molarswas performed after 10 weeks. All rats were scanned once every 4 weeks with both micro-CT and HR-MRI. Micro-CT and HR-MRI images were registered and fused in the same 3-dimensional region to quantify blood flow velocity and trabecular bone thickness at T0 (baseline), T4 (4 weeks), T8 (8 weeks), T12 (12 weeks), T16 (16 weeks), and T20 (20 weeks). Histological assessment was the gold standard with which the findings were compared.
Results:
The histomorphometric images at T20 aligned with the HR-MRI findings, with both test and control rats demonstrating reduced trabecular bone vasculature and blood vessel density. The micro-CT findings were also consistent with the histomorphometric changes, which revealed that the test rats had thicker trabecular bone and smaller marrow spaces than the control rat.
Conclusion
The combination of micro-CT and HR-MRI may be considered a powerful non-invasive novel technique for the longitudinal quantification of localized mineralization and vascularization.
9.Millimeter wave exposure induces apoptosis in human melanoma A375 cells .

OBJECTIVE:
To investigate the effects of millimeter wave (MMW) exposure on apoptosis of human melanoma A375 cells and explore the mechanisms.
METHODS:
Through electromagnetic field calculation we simulated MMW exposure in cells and calculated the specific absorption rate (SAR). The optimal irradiation parameters were determined according to the uniformity and intensity of the SAR. A375 cells were then exposed to MMV for 15, 30, 60, or 90 min, with or without pretreatment with the caspase-3 inhibitor AC-DEVD-fmk (10 μmol/L) for 1 h at 90 min before the exposure. CCK-8 assay was used to assess the changes in the viability and Annexin-V/ PI staining was used to detect the apoptosis of the cells following the exposures; Western blotting was used to detect the expression of caspase-3 in the cells.
RESULTS:
The results of electromagnetic field calculation showed that for optimal MMV exposure, the incident field needed to be perpendicular to the bottom of the plastic Petri dish with the antenna placed below the dish. CCk-8 assay showed that MMW exposure significantly inhibited the cell viability in a time-dependent manner ( < 0.05); exposures for 15, 30, 60, and 90 min all resulted in significantly increased apoptosis of the cells ( < 0.05). The cells with MMW exposure showed significantly increased expression of caspase-3. The inhibitory effect of MMW on the cell viability was antagonized significantly by pretreatment of the cells with AC-DEVD-fmk ( < 0.05), which increased the cell viability rate from (36.7±0.09)% to (59.8±0.06)% ( < 0.05).
CONCLUSIONS
35.2 GHz millimeter wave irradiation induces apoptosis in A375 cells by activating the caspase-3 protein.
