1.Angiogenesis,Neurogenesis and Neuroprotection of Vascular Endothelial Growth Factor in Cerebral Ischemia (review)
Chinese Journal of Rehabilitation Theory and Practice 2007;13(3):208-211
Vascular endothelial growth factor (VEGF) is angiogenic and neurotrophic factor. It is now known that VEGF also plays an importmant role in neurogenesis and neuroprotection in addition to angiogenesis. VEGF and VEGF receptor increase in the ischemic brain, promote vascular endothelial cell proliferation and decrease infarct volume. VEGF plays neuroprotective and neurotrophic roles by the modulation of the phosphatidylinositol3-kinase (PI3K)/Akt/nuclear factor-κB and MAPK/ERK signaling pathways. VEGF reduces ischemic neuronal apoptosis, promotes neural stem cells proliferation, migration and differentiation and improves nerve functions. In the other hands, VEGF increases permeability of blood vessel which may aggravate cerebral edema early.

Objective:To investigate the effects of comprehensive intervention on the quality of life of middle school students. Methods:Eight classes were extracted with striated cluster sampling method from the first grade of two key junior middle schools in city and countryside; four classes (two classes of the urban and rural) were selected randomly as the experimental group and another four classes (two classes of the urban and rural) as the control group. The experimental group received mental-social intervention for three years. No intervention for the control group. The effect was evaluated with the subjective quality of life of children, and a questionnaire on mental health of middle school students. Results:The scores of life quality, cognitive component and emotion component of experimental group were higher than that of control:(172.6?12.8/145.4?17.4,98.7?10.8/85.0?13.3,73.9?4.3/60.4?7.7; t=20.83, 13.20, 25.29 respectively;the average P

Objective: To explore the effect of comprehensive intervention to correct parental unhealthy rearing behavior on middle-school students. Methods: Eight freshman classes were randomly extracted from two key junior middle schools, four classes as the experimental groups, the other four as the control group. EMBU was applied to measure the difference before and after intervention of both groups. The comprehensive intervention was used, mainly the individual family mental health education and group family education. Results: After intervention, there were significant improvement in the parents' emotional warmth and understanding factors of experimental group(53.0?10.6/49.3?10.1, 55.7?10.9/50.4?10.5,t= 4.24 or 5.80,P

Objective:To express the GPI-anchored CD55 and recombinant transmembrane form CD55 molecules on mouse fibroblasts cell line NIH3T3, and compare their inhibitory function of complement lysis. Methods:In previous study, had constructed the transmembrane form CD55 (CD55-TM) cDNA by linking the extracellular portion of CD55 to the transmembrane and cytoplasmic domains of MCP, and then subcloned into retroviral vector pLXSN. In this experiment, had transfected recombinant CD55-TM and GPI anchored CD55 into PA317 packaging cell to generate stable virus-producing cell lines. And then, mouse fibroblasts cell line NIH3T3 was infected with the virus containing CD55, recombinant CD55-TM or pLXSN alone. The expression of these molecules on NIH3T3 cells was detected by FACS analysis. Complement killing assay was carried out using MTT colorimetric method. Results:FACS analysis showed that both CD55 and its TM version were expressed stably on NIH3T3 cells. Both kinds of CD55 molecules can efficiently protect the NIH3T3 cells from complement mediated lysis and there is no significant difference between them. Conclusion:These data showed that both GPI-anchored CD55 and its TM version can normally expressed on NIH3T3 cells and both can protect the NIH3T3 cells from human complement mediated lysis. These results confirmed the feasibility of TM CD55 based gene therapy could be used for PNH, and also provided an excellent model for the study of signal transduction mechanisms mediated by GPI-anchored protein.

Objective To evaluate effects of pulmonary surfactant (PS) containing ketamine on respiratory failure induced with lung-lavage in rats Methods Twenty Wistar rats were anesthetized with intraperitoneal pentobarbital sodium and were ventilated with the peak inspiratory pressure(PIP) being 1 47 kPa and E:I at 1:1 following intubation through tracheotomy In all rats whole double lung lavage were performed with normal saline (37℃, 40 ml?kg -1 ) 8 10 times after PIP and PEEP were adjusted to 1 96 kPa and 0 49 kPa respectively The rats were randomly divided into A and B groups after PaO 2 decreased to less than 12 kPa In group A (n=10): PS (25mg, 0 5ml) was injected intratracheally to lungs; in group B (n=10): PS (25mg, 0 5ml) containing ketamine 2 5 mg was injected At 120th min after PS injection, PIP and PEEP were gradually adjusted to 1 47 kPa and 0 kpa (ZPEEP), respectively Results PaO 2 in both groups decreased significantly from 59 3 kPa to 10 3 kPa after lung lavage(P0 05 vs pre-lavage) All rats in group B and 4 of 10 rats in group A survived at the end of experiment Conclusions Intratracheal administration with PS containing ketamine not only reverses hypoxemia , but also maintains the respiratory function under low airway pressure without PEEP after respiratory failure induced with lung lavage,

Objective To investigate the effects of different methods of volume therapy on the inflammatory response in patients undergoing liver cancer resection. Methods Forty ASA Ⅰ or Ⅱ patients, aged 40-60 yr,with body mass index 20-25 kg/m2,undergoing liver cancer resection, were randomly divided into 2 groups ( n =20 each):routine fluid replacement group (group Ⅰ ) and goal-directed fluid replacement group (group Ⅱ ).The fluid replacement regime in group Ⅰ =compensatory volume expansion (CVE) + physiological requirements + cumulative loss + confinued loss + the third space losses.CVE was replaced with lactated Ringer's (LR) solution 5 mg/kg before anesthesia induction. The physiological requirements and cumulative loss were replaced with LR solution according to the principle of 4-2-1.The continued loss equal to the intraoperative blood loss was replaced with the equal volume of 6% hydroxyethyl s tarch ( HES 130/0.4).The 3rd space losses were replaced with LR solution 5 ml·kg-1 ·h-1.In group Ⅱ,CVE was replaced with LR sol6ution as in group Ⅰ.LR solution was infused after anesthesia induction at 5 ml·kg-1 ·h-1.6% HES was infused to maintain left ventricular ejection time (LVETc) between 350-400 ms.When 350 ms < LVETc < 400 ms and the amplitude of stroke volume ( SV ) increased by > 10%,6% HES was infused continuously until the amplitude of SV increased by ≤ 10%.Blood samples were taken before anesthesia induction and at the end of operation for measurement of serum TNF-α,IL-2,IL-4,IL-6 and IL-8 concentrations. The adverse cardiovascular reactions were recorded. Results Compared with group Ⅰ,the serum TNF-α,IL-6,IL-8 concentrations were significantly decreased, the serum IL-2 and IL-4 concentrations were significantly increase, and the incidence of hypotension and tachycardia was significantly decreased in group Ⅱ ( P < 0.05).No adverse cardiovascular reactions were found in both groups. Conclusion LVETc and SV-guided volume therapy can maintain the blood volume and inhibit the inflammatory response and is suitable for the patients undergoing liver cancer resection.
