1.Effects of 8-bromo-7-methoxychrysin and sorafenib on apoptosis of liver cancer stem-like cells

BACKGROUND:We tried to combine 8-bromo-7-methoxychrysin and sorafenib in order to offset the tolerance of hepatocelular cancer stem cels to sorafenib, thereby comprehensively improving the therapeutic efficacy on hepatocelular carcinoma. OBJECTIVE:To observe the effects of 8-bromo-7-methoxychrysin, sorafenib and their combination on apoptosis of liver cancer stem-like cels SMMC-7721, and to analyze their mechanisms. METHODS:SMMC-7721 cels were treated with 8-bromo-7-methoxychrysin, sorafenib alone and their combination for 24 hours. Then, flow cytometry was used to detect cel apoptosis and western blot assay was used to determine nuclear factor-κB protein expression. RESULTS AND CONCLUSION:Compared with 8-bromo-7-methoxychrysin group and sorafenib group, the apoptotic rates of SMMC-7721 cels were significantly enhanced after treatment with the combination of 8-bromo-7-methoxychrysin group and sorafenib (2.5, 5, 10, 50 μmol/L), and meanwhile, the protein expression of nuclear factor-κB was down-regulated significantly. These findings indicate that the combined therapy of 8-bromo-7-methoxychrysin group and sorafenib can enhance the apoptosis of SMMC-7721 cels, which may be associated with down-regulation of nuclear factor-κB protein.

Insulinoma is a rare tumor derives from pancreatic islet B cells,the cause of etiology is unknown with insidious onset.Its clinical manifestations are of diversification,the high misdiagnosis rate with the low cure rate owing to the complex of pancreatic anatomy and the lack of effective localization diagnosis and differential diagnosis method for benign and malignant lesions,the treatment of insulinoma is still one of challenge in clinical application considering that some patients unable to tolerate surgery or unwilling surgery.In recent years there have been some new treatments except surgery,this review will introduce the some treatment methods of insulinoma for the moment.

Objective To assess the mutagenicity of polycyclic aromatic hydrocarbons(PAHs) extracted from the atmospheric particulates collected from different aerodynamic sizes,seasonal variation and the sampled sites,and to detect the effect of these factors on the mutagenic activity of PAHs.Methods PAHs were extracted from the atmospheric particulates(PM10 and PM2.5) collected from two different sites,business area and industrial area(summer and winter respectively).The mutagenicity of these PAHs samples was evaluated by the Salmonella mutagenicity assay(Ames assay),using Salmonella typhimurium strains TA98 and TA100 with and without metabolic activation(S9).Each sample was assayed in three doses(125,250 and 500?g per plate).Results All samples induced the number of revertants to be higher measured with TA98 with and without S9 mix.The level of specific mutagenic activity measured with strain TA98 was higher than TA100.Metabolic activation with the S9 fraction increased the mutagenicity of the industrial area samples studied for the strain TA98.All groups showed a good dose-response relationship of mutagenicity(P

Oyecave:To observe the influence of nociceptin/orphanin FQ(N/OFO)on cerebral infarction volume and somatosellsOry evoked potential(SEP)in focal cerebral ischemia in rats.Methods:Forty one SD rats were randomly alloomed into middle artery occlusion(MCAO)sham-operation(n=5),isehemic(n=8),N/OFQ 10μg(n=7),N/OFQ 1 μg(n=7),N/OFQ0.1 μg(n=7),and artificiai cerebrospinal fluid(ACSF)(n=7)groups.A model of middle cerebral artery occlusion(MCAO)in rats was induced using intraluminal suture method.Reperfusion was performed 2 hours after MCAO.One hour after MCAO,N/OFQ 10 μg,N/OFQ 1 μg,N/OFQ O. 1 μg,and the same volume of ACSF were injected intraventricularly in the N/OFQ 10 μg,N/OFQ 1 μg,N/OFQ 0. 1 μg,and ACSF groups,respectively. The cerebral infarction volurne was detected 24 hours after reperfusion,and SEP was recorded. Results:1he amplitude of SEP P1 decreased in the sham-operation group. There was no significant change in P1 peak latencies.There were no significant differences hetween the N/OFQ 0. 1 μg group and the ACSF group in SEP amplitudes,P1 peak lantecies and cerebral infarction volume. As compared with the ACSF group,the SEP amplitudes were further decreased in the N/OFQ 1 μg and N/OFQ 10 μg groups,but there were no significant change in P1 peak lantecies. One hour after reperfusion,the SEP amplitude in the ACSF group almost returned to the level of preischemia,the recovery slowed down in the N/OFQ 1 μg group,and it still did not recovered 3 hours after reperfusion in the N/OFQ 10 μg group. The dose of N/OFQ and SEP response showed dose-effect relationship,The higher the dose,the deeper the SEP depression and the slower the recovery. At 24 hours after reperfusion,the cerebral infarction vlumes in the shamoperation,ACSF,N/OFQ 0. 1 μg,N/OFQ 1 μg,and N/OFQ 10 μg groups were 0 mm3,24.180 ±4.088 mm3,23.090±4.523 mm3,35.304 ± 6. 824 mm3,and 40. 806±6. 716 mm3,respectively. There was no significant difference between N/OFQ 0. 1 and ACSF groups. There were significant differences between N/OFQ 1 μg and 10 μg groups and ACSF group (all P < 0.01 ). Conclusions:Intracerebroventricular injection of N/OFQ in the early stage of cerebral ischemia decreases the SEP amplitude,prolongs the time of recovery,and increases cerebral infarction volume,which shoves that it may aggravate cerebral ischemic injury.
5.Study on the relationship between liver cirrhosis ascites and plasma sodium

Objective To compare sodium supplement and sodium restriction to the effect of the extinction and prognosis of liver cirrhosis ascites, and research the relationship between live cirrhosis ascites and plasma sodium.Methods 119 liver cirrhosis patients were randomly divided into 2 groups :60 cases of sodium supplement and 59 cases of sodium restriction. 60 cases of sodium supplement was kept low-salt diet and intravenous sodium chloride supplement(3 ~5 g/d) ,59 cases of sodium restriction was only kept low-salt diet. Plasma sodium,plasma chloride,urine sodium, urine sodium chloride were detected before treatment, and 6 days, 10 days after treatment respectively. And the urine quantity,extinction of ascites and prognosis were compared. Results There were significant differences on the increase of plasma sodium,plasma chloride,urine sodium,urine chloride and urine quantity after treatment in two groups. Extinction time of ascites was shorter in group of sodium supplement. The morbidity and fatality rate of hepatic encephalopathy, and hepatorenal syndrome in the group of sodium supplement were lower than that in the group of sodium restriction. Conclusion Sodium supplement should be adapted when using diuretic agent to help the extinction of ascites, and to improve recovery.
6.Exogenous VEGF165 regulates skeletal myoblasts transplantation induced Th17/Treg imbalance

Objective To investigate whether Th17/Treg imbalance existed,and whether VEGF165 attenuated the imbalance in allogeneic skeletal myoblasts transplantation(allo-SMT) for acute myocardial infarction (AMI).Methods There were 60 C57BL/6 male mice,3 to 4 weeks old,weighting 0.016-0.020 kg.On the day 1,2,4,and 7 after allo-SMT,the percentages of Th17 and Treg cells,and the ratios of Th17/Treg cells were analyzed by FCM in AMI group,AMI-S group(alloSMT) and AMI-Ⅴ group(with VEGF165 treatment).Subsequently,related proinflammatory and regulatory cytokines and key transcription factors ROR-γt mRNA and Foxp3 mRNA expression were examined by Bio-plex and RT-PCR respectively.Results On the day 1,2,4,and 7 after allo-SMT,the percentage of Treg,related cytokines concentrations and transcript factor Foxp3 mRNA in AMI-S group were lower than those in AMI group,while the AMI-Ⅴ group were higher than those in AMI groups.However,the percentage of Th17 cells,related cytokines concentrations,and ROR-γt mRNA in AMI-S group were higher than those in AMI group; but the AMI-Ⅴ group were lower than those in AMI group.Apparently,compared with AMI group,the ratios of Th17/Treg significantly ascended in AMI-S group and decended in AMI-Ⅴ group.Conclusion Th17/Treg imbalance participated in the formation and development of inflammation and immune response after allo-SMT.However,transfected VEGF165 could relieve the severity of Th17/Treg imbalance.
7.Application of ultrasound biomicroscopy on skin assessment in normal adults and psoriasis patients

ObjectiveTo investigate the features of normal and psoriasis skin on ultrasound biomicroscopy (UBM) and explore the method of thickness measurement.MethodsUsing 50 MHz ultrasound probe of biological microscope, ultrasonographic observation and ultrasonic thickness measurement were conducted in 90 normal adults and 40 psoriasis patients. Innormal patients, ultrasound evaluations were performed at 10 different parts of the body skin.ResultsOn sonogram, the normal skin showed a “sandwich” structure with two parallel hyperechoic bands and the middle isoechoic dots and short liens. The sonograms of the psoriasis skin showed obviously thickened epidermis and dermis, disordered internal structure and clear boundary from adjacent normal skin. The range of the epidermis’ thickness measurement was between the medial forearm (0.12±0.03) mm and the palm (0.29±0.15) mm. The range of dermal thickness measurement was between the back hand (1.18±0.32) mm and parasternal (1.55±0.21) mm. Psoriasis skin was thicker than the uninvolved skin (P<0.001). And the dermis’ thickness of uninvolved skin in psoriasis patients was thicker than that of the normal adults (P<0.001).ConclusionNormal adult’s epidermis, dermis and skin appendages can be shown clearly using 50 MHz ultrasound biomicroscopy. And ultrasound biomicroscopy canaccurately measure the thickness of dermis and epidermis, which provides the basis for the diagnosis and treatment of psoriasis.
8.Perioperative risk factors for postoperative pancreatic fistula in patients with gastric cancer

Objective To explore perioperative risk factors for postoperative pancreatic fistu?la. Methods Six hundred and forty?three cases patients who underwent radical gastrectomy for gastric cancer from January 2010 to June 2015 in the NO. 309 Hospital of Chinese People’ s Liberation Army were selected and divided into Postoperative Pancreatic Fistula( POPF) group with 53 cases and Non?Postoperative Pancreatic Fis?tula( NPOPF) group with 590 cases. Preoperative,intraoperative and postoperative data were analyzed by statisti?cal analysis of two groups. Results POPF rate was 8. 24%(53/643). There were no significanct differences in terms of gender,age,preoperation with disease,preoperative serum albumin,TNM stage,abdominal surgery,sur?gical procedure,lymph node dissection,digestive tract reconstruction,surgical approach,blood loss and length of time between the POPF group and NPOP group(P>0. 05). The level of BMI of POPF group was higher than that of NPOPF group,the difference was statistically significant((25. 63±2. 54) kg/m2 vs. (21. 11±2. 44) kg/m2,t=2. 245,P=0. 025). The number of lymph node dissection in POPF group was (32. 25±5. 82),in NPOPF group was (27. 06±6. 79),there was significant difference between the two groups(t=2. 093,P=0. 037). The operation time in POPF group was (242. 50±52. 30) min,in NPOPF group was (229. 51±59. 21) min,the difference was statistically significant between the two groups( t=2. 398,P=0. 017) . The serum CRP of 1 d in POPF group was (85. 72±12. 05) mg/L,in NPOPF group was (76. 41±12. 52) mg/L,and there was significant difference between the two groups( t=1. 979,P=0. 048) . The serum albumin of 2?4 d after operation in POPF group was (26. 0±5. 9) g/L,in NPOPF group was (32. 6±6. 8) g/L,the difference between the two groups was statistically significant(t=-10. 185,P=0. 000). The drainage fluid amylase of 1 and 2 d after operation in POPF group was (2094+1444) U/L,in NPOPF group was (746+486) U/L,and there was significant difference be?tween the two groups( t=10. 400,P=0. 000) . Logistic regression analysis results showed that body mass index ( BMI) ,lymph node dissection number,time of operation,postoperative 1,2 d drainage fluid amylase and serum albumin of 2?4 d after operation were postoperative occurred pancreatic fistula risk factors( OR=1. 972,183. 6, 2. 004,0. 150,9. 809,P>0. 05). Conclusion BMI,number of dissected lymph node,duration of surgery,serum albumin of 2?4 d after operation and postoperative 1,2 d drainage fluid amylase have important clinical values for predicted POPF.
9.Impact of surgical resection on stage Ⅱ-ⅢA small-cell lung carcinoma patients ’ prognosis

Objective To investigate the impact of surgical resection on patients with Ⅱ-Ⅲ A stage small cell lung carcinoma (SCLC). Method Data of 61 in-patients who diagnosed as Ⅱ-ⅢA stage SCLC from Jan 1st 2009 to Feb 1st 2014 were analyzed. 23 patients underwent surgical resections were enrolled as the treatment group, while 38 patients without surgical resection were enrolled as the control group. Disease progression was confirmed by monthly examination. The grouping is balanced by propensity score match. The progression-free survival (PFS) time and overall survival(OS) were analyzed with Kaplan-Meier survival method and Cox regression is applied to analyze the covariates. Fisher's exact test was applied to compare one-year survival rate and two-year survival rate. Results The PFS and OS of the treatment group is longer than that of the control group (P < 0.05). Both one-year and two-year survival rates of the treatment group outnumber those of the control group (P < 0.05). Cox regression indicates that surgical resection is an independent prognostic factor (P < 0.05). Conclusion Surgical resection on tolerable patients with stage Ⅱ-Ⅲ A small-cell lung carcinoma is effective on improving the progression-free survival time,one-year and two-year survival rates,and also shows a propensity of a higher overall survival time.
10.Comparison of preoperative PET-CT and pathological analyses in diagnosis of pulmonary tumors

Objective To investigate the consistency in 18F-deoxyglucose positron emission tomography (18F-FDG PET-CT) examination and histopathological analyses in the diagnoses of resectable lung tumors. Methods Retrospective reviews over the clinical data of lung tumor patients by preoperative PET-CT diagnosis and postoperative histopathological diagnosis were conducted to investigate the effects of the two diagnostic methods in terms of lung tumor properties , mediastinal lymph node metastasis , and pulmonary hilar lymph node metastasis. Results The diagnoses by preoperative PET-CT was consistent in differentiation of non-malignancy and malignancy of pathologic lung tumors by 87.3%, at a medium level (κ = 0.401, P < 0.001). McNemar test showed P = 0.508, indicating the two diagnostic methods were insignificantly different in the diagnosis of pulmonary tumors. The preoperative PET-CT was consistent in the diagnosis of the metastasis of pathologic mediastinal lymph node by 85.9%, at a medium level (κ = 0.697, P < 0.001). McNemar test showed P =0.754, indicating no significant difference between the diagnostic methods. The preoperative PET-CT was consistent with postoperative pathological examinations in the differentiations of the metastasis of pulmonary and hilar lymph node by 77.4%, at a medium level (κ=0.523, P < 0.001). McNemar test showed P = 0.454, indicating the two diagnostic methods were no significantly different. Conclusion Preoperative PET-CT and histopathologic examinations may be consistent in lung tumor diagnosis , which provides a basis for a certain significance in the surgical options.
