1.Effects of progranulin on proliferation and senescence in gastric cancer cells

Background and purpose:Progranulin (PGRN) is a novel growth factor that plays an important role in the tumorigenicity, tumor cell migration and cell cycle. Its expression in many malignant tumor cells is high. It is not only involved in tumor cell growth, but also closely related with the occurrence and evolution of tumor. This study was to investigate the expression of PGRN in gastric cancer and the effects on proliferation and senescence in gastric cancer cell line BGC823. Methods:Immunohistochemical method was used to detect the expression of PGRN in gastric cancer tissues and adjacent normal tissues; Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect the expression of PGRN in PGRN-siRNA BGC823 cells;MTT method, cell colony formation and cell senescence experiments were used to explore the effects of PGRN on proliferation and senescence in BGC823 cell. Results:PGRN protein levels were high in gastric cancer tissues;Knocking down the PGRN gene in BGC823 decreased the proliferation and clonogenic capacity, cloning efifciency in PGRN-siRNA group was (25.3±3.1)%, in the control group was (72.1±5.7)%, and in the normal cells was (80.3±4.0)%, there was no signiifcant difference between normal group and control group, but there were signiifcant differences among PGRN-siRNA group and the other two groups (P<0.05);Knocking down the PGRN gene in BGC823 cells could promote cell senescence. The positive rate of aging in PGRN-siRNA group was (27.6±2.1)%, in the control group was (3.2±1.3)%, and in the normal group was (1.9±1.2)%, there was no signiifcant difference between normal group and control group. But there were signiifcant differences among PGRN-siRNA group and the other two groups (P<0.05). Conclusion:PGRN can be used as a new marker for gastric cancer, and provide new ideas to the treatment of gastric cancer.
2.Motion signal extraction method based on sEMG energy Gauss distribution characteristics.

Motion segment and extraction from continuous signals is the premise of surface electromyography (sEMG) analysis. For the problem that sEMG energy threshold required repeated manual testing to set, this the paper established a this mathematical model of continuous actions based on Gaussian sEMG energy curve, in which the energy threshold was set according to the distribution of Gaussian signal section, and differentiated the action signals from no-action signals combined with energy comparison method. The experiment results showed the method can achieve continuous repetitive action segmentation, and compared with manual segmentation of the same signal, has a higher degree of coincidence.

3.The expression of protease-activated receptors in colon mucosa tissue of patients with diarrhea-predominant irritable bowel syndrome and its significance

Objective To investigate the expression of protease-activated receptors (PARs) in colon mucosal tissue of patients with diarrhea-predominant irritable bowel syndrome (D-IBS). Methods Colon mucosa tissues were obtained from 28 D-IBS patients and 18 normal controls by colonoscopy.The expression of PARs was detected using Western blotting and real-time PCR. Data were analysed by SPSS 18. 0 softwave, and comparisons between groups were done using Mann-Whitney U test.Results There was no difference in expression of PAR1 between D-IBS patients and normal controls (P=0. 300). The expressions of PAR2 and PAR4 were higher in D-IBS patients than those in normal controls (0.99±0.67 vs 0.63±0.38, P=0.038 and 0.37±0. 14 vs 0.25±0. 11, P=0.013,respectively). The results of real-time PCR were in accordance with those of Western blotting.Conclusion The increased expressions of PAR2 and PAR4 in colon mucosa of D-IBS patients indicate that these two PAR receptors may be involved in the process of D-IBS.
4.Primary Pulmonary Cryptococcosis:A Retrospective Analysis of Sixteen Cases

OBJECTIVE To investigate the clinical pathologic features and therapeutic tools of pulmonary cryptococcosis.METHODS The clinical data about 16 cases of pulmonary cryptococcosis which were diagnosed by histopathologic examinations were reviewed.The survey data recorded over a 15-year period,from 1982 to 2007,were summarized.We analyzed their clinical situations,radiographic manifestations,final diagnosis and therapeutic tools.RESULTS The majority of the patients were middle-aged males.The health condition of the most of the patients was good before infection.six months to five years after surgery and antifungal theragy,no relapse,dissemination and death were observed.CONCLUSIONS The majority of primary pulmonary cryptococcosis patients have not underlying diseases,and their radiography manifestations show single or multiple nodular shadows,tumor shadows and infiltrative shadows.The limited pathological change can be excised and applied with the antifungal drugs.Fluconazole is the first-choice drug for curing pulmonary cryptococcosis.
5.Removing the interference of daratumumab on transfusion compatibility testing and transfusion efficacy comparison

【Objective】 To explore the feasibility of blood transfusion compatibility testing for multiple myeloma(MM) patients treated with anti-CD38 monoclonal antibody daratumumab (DARA) after DARA-Fab fragment blocking, and to evaluate the transfusion efficacy by comparing with dithiothreitol(DTT) method. 【Methods】 After DARA was prepared into DARA-Fab fragments using PierceFab preparation kit, the neutralization effects of different volumes (5, 10, 15, 30 μL) on screening cells and panel cells were confirmed. DARA-Fab fragments and screening cells with specific antigens and corresponding monoclonal antibody reagents were used as the experimental group and the control group with the same volume of saline for incubating and centrifugin.Twenty MM patients treated with DARA were selected for cross-matching with DARA-Fab and DTT respectively, and the laboratory indexes before and after transfusion were statistically analyzed, and the two blood matching methods were compared. 【Results】 After incubating and centrifuging, the results of DARA-Fab fragments(15, 30 μL) with screening cells and serum mixed with DARA were negative, while those of DARA-Fab(5, 10 μL) were positive. 15μL DARA-Fab treated antibody identification cells (2, 3, 4, 5, 7, 9, 11) were negative, antibody identification cells (1, 6, 8, 10, 12) were negative after 30 μL DARA-Fab fragments treatment; the results of MNS, Duffy, Kidd, Kell, Lewis, Rh blood group system of the experimental group were consistent with those of the control group; the hemoglobin (Hb) (g/L) of 20 patients after infusion of RBC (73.90±1.90) was significantly higher than that before transfusion (63.60±1.58), P<0.01. There was no significant difference in total bilirubin(TBil)(μmol/L)(16.25±3.54 vs 17.87±3.57), direct bilirubin(DBIL)(μmol/L)(6.31±2.32 vs 7.10±2.80)and indirect bilirubin(I-Bil)(9.94±1.38 vs 10.77±1.22) before and after infusion(P>0.05).And no statistical difference was noticed in Hb (10.75±1.04 vs 10.30±0.98), TBil (3.31±1.47 vs 3.31±0.55), DBIL(2.76±1.24 vs 2.60±0.83), and I-Bil(1.97±0.40 vs 2.82±0.53) between the DTT treatment method and the DARA Fab fragment treatment before and after transfusion(P>0.05). 【Conclusion】 DARA-Fab can remove the interference of RBC on cross matching by blocking CD38 antigen. This method has no effect on the antigens of common RBC blood group systems, and shows significant blood transfusion efficacy as that of DTT method.
