1.Application of text mining in drug target discovery

Co-citations of foreign and domestic highly cited papers on drug target discovery were analyzed by clustering analysis using BICOMB2.01 and gCLUTO.Semantic analysis of the titles and abstracts in these highly cited papers and their important source literature showed that general trend, theoretical foundation, main methods and principal resources are the major hotspots of text mining in drug target discovery.
2.Bibliometric analysis of domestic Nursing Information System from 2009 to 2013

Objective To analyze the trend of nursing information system(Nursing Information System,NIS) and contemporary situation,so as to provide a reference for the further development of NIS in China.Methods Based on China Academic Journal Network Publishing Database,Wanfang database and VIP Chinese academic periodical database,retrieval published NIS literature from 2009 to 2013 in China.Seven aspects were analyzed:time,areal distribution,cooperative rate,authors,institutions,periodicals and keywords.Results There were 438 articles from 128 published journals,47.9% papers were in the statistical source journals.Research content mainly involved clinical nursing and nursing management.Conclusions NIS research has achieved great development in the past 5 years,but the researches should be strengthened in breadth and depth.
3.Histologic changes of arteries after hydraulic dilation

Objective To study the histologic changes of arteries dilation under different hydraulic pressure. Methods Right carotid arteries were dilated by using hydraulic method with different pressures (40, 80, 120 kPa) normal saline. The endothelial cells, internal elastic membrane (IEM), smooth muscle (SM) and adventitia of the arteries were studied under microscope. The arterial calibre, thickness of tunica intima and smooth muscle were measured under TJTY-300 automatic medical photograph analyser. Results 1) The calibre of the dilated arteries increased. 2) When the pressure was 40 kPa, there were mild injuries of endothelial cells, internal elastic membrane and smooth muscle; all the injuries recovered to normal 1 week later. 3) The degree of injury of the individual layers of the arteries gradually aggravated as the pressure raised up to over 80 kPa. The repair also required long periods of time and was found imperfect. The proliferation of fibres of connective tissues took place one week later. Conclusion 1) Blood flow could be increased after hydraulic dilation of the artery, and the strength of washing away the small thrombosis over the site of anastomosis could be increased. 2) For safety, the hydraulic pressure of arterial dilation should be better kept within 40 kPa.
4.Preparation of EUS-guided radiofrequency probe and test of its functions

Objective To prepare a radiofrequency (RF) ablation probe guided by endoscopic ultrasonography (EUS) and to evaluate its efficacy and safety. Methods A modified 19-gauge needle ( Olympus) was used, which was electrically insulated with shrink tubing, with the tip of lcm naked. The insulation was tested, and the current signal was stable. Three rabbits were anesthetized with 8% chloral hydrate ( 3 ml/kg) intraperitoneally, fixed on the dissecting table. A standard neutral pad was applied to the thigh of the rabbit to complete the electrical circuit, both the pad and the needle electrode connected with RF generator. After anesthesia was ready, the needle was advanced through the gastric wall into the liver. Ablations were performed three times in the same zone. Extent and tissue lesion were measured after ablation. Results Ablation could be successfully performed by the needle electrode, with brown lesions surrounded by normal hepatic tissue. The mean diameter of the ablated zone in the liver was 1.0 cm× 1.2 cm. After ablation,specimens along the passage were subjected to NADH staining, and no lesions were found. HE and NADH staining showed no viable cells in the central ablation area. Conclusion With the advance of the technology and selection of materials, an EUS-guided needle electrode can be made. This preliminary animal trial demonstrates that radiofrequency ablation can be performed effectively and safely by using this EUS-guided needle electrode.
5.Preliminary outcomes of the modified first dorsal metacarpal artery island flap for soft tissue defects of the thumb

Objective To explore the survival rate and early-term effects of sensory reconstruction of the modified first dorsal metacarpal artery (FDMCA) flap in treatment of thumb distal soft tissue defect.Methods From March 2004 to October 2007, 65 patients with soft tissue defects of the thumb underwent the FDMCA flap in our department. Forty-nine cases with complete data were included in the study. There were 37 males and 12 females, with an average age of 32 years (ranged, 18-65 years). The conventional surgical methods were used in 18 cases while the improved surgical methods were in 31 cases. The distal necrosis, the vascular crisis rate and the sensory recovery were recorded. Results The mean follow-up period was 2 years (ranged, 20 months to 3 years). Forty-six flaps survived completely. Vascular crisis occurred in five cases harvested by the conventional methods. Eventually, three of them had a partial distal necrosis.Improved FDMCA flap survived well in all 31 patients. Flaps in these patients with nerve anastomosis restored satisfied sense and all sense were located in the thumb. Eighteen cases of nerve transfer group had different levels of mixed feelings (33%) or ectopic sensory (62%). Sense was located in the thumb completely in 1 case. Conclusion The modified FDMCA flap significantly increased the survival rate. Neurorrhaphy could make the sense of the flap locate in the thumb; however it had not been proved that its impact on the recovery of two-point discrimination and stereognosis.
6.DETERMINATION OF FLAVONOIDS IN HAWTHORN FRUITS AND THEIR ANTIOXIDATION EFFECT

Objective: To extract and determine the flavonoids of hawthorn fruits and their antioxidative effect. Method: The ultrasonic method was used for extracting flavonoids. The content of flavonoids was determined by spectrophotometry, and the antioxidation effect was determined by flow-injection chemiluminescence.. Results and Conclution: The recovery rate was 96%~105% and the coefficient of variation was 0.14 % by spectrophotometry. Hawthorn fruits had high antioxidative effect, showing dose-response relation.
7.Some leukocyte differentiation antigens express in side population cells in leukemia K562 cell line

BACKGROUND:To study the phenotypes of side population cells in leukemia is important for understanding the heterogeneity and origin of tumor cells, molecular markers and targeted therapy.
OBJECTIVE:To identify whether the human chronic myeloid leukemia cellline-K562 contains side population cells or not, and to further observe the differences in expressions of leukocyte differentiation antigens from side population cellsubset and non-side population cells subset.
METHODS:Flow cytometry was used to detect whether there were side population cells in the K562 celllines. Then, the expression of CD34+, CD34+CD38-, CD34+CD38+, HLA-DR+cells in the side population subsets and non-side population subsets.
RESULTS AND CONCLUSION:Flow cytometry results showed that the K562 cellline contained side population cells, and the proportion of side population cells was much lower. The side population cells accounted for (2.7±0.5)%of viable cells in K562. The expressions of CD34+cells and CD34+CD38-cells in the side population subset were significantly higher than those in the non-side population subsets. The expressions of CD34+CD38+cells and HLA-DR+cells in the side population subset and non-side population subset did not have a significant difference. Heterogeneity was found in the differentiation antigen expression between the side population subset and non-side population subset.
8.Effect of flipped class model based on SPOC in the course ofMedical Nursingteaching

Objective To explore the application of flipped class model based on SPOC in the course of internal medical nursing. Methods Randomly selected from Shanxi University of Medical grade 2014, 206 undergraduate students as the research object. The control group (104) and experimental group (102) .The experimental group used flipped class model based on Small Private Online Course (SPOC) teaching method;the control group used the traditional teaching mode. After the end of the course, compared two groups nursing students′self-regulated learning ability and critical thinking ability whether there are differences by autonomous learning ability scale and Chinese version of critical thinking disposition inventory (CTDI-CV). Results The experimental group students′theory assessment results, critical thinking ability and self-regulated learning ability were (75.68 ± 9.32), (303.04 ± 16.54), (112.39 ± 11.10) higher than the control group (69.43 ± 11.41), (288.23 ± 15.84), (102.58 ± 9.31), the difference was statistically significant (t = 2.837,3.451,3.512, P<0.05). Conclusions The flipped class model based on SPOC teaching method was better than traditional teaching mode, it improved the students' theory assessment results and self-regulated learning ability and critical thinking ability.
9.Detection of genomic abnormalities in chronic lymphocytic leukemia by fluorescence in situ hybridization

Objective To investigate the value of panel fluorescence in situ hybridization (panel FISH)for detection of genomic aberrations in chronic lymphocytic leukemia(CLL). Methods Five types of fluorescein-labelled DNA probes including five sequence specific probes D13S25 for 13q14. 3, RB1, p53, ATM (11 q23)and centromeric probe for chromosome (CSP12) were used to perform fluorescence in situ hybridization assays in 17 patients with CLL. Its results were compared with that obtain by conventional cytogenetic (CC)examination. Results In 17 patients with CLL, CC examination showed that only one case (1/17) was found to have chromosomal abnormality that was simultaneous trisomies 3,8 and 18, whereas panel FISH assay showed that 10 cases (10/17) were found to have genomic aberrations including deletion of D13S25 in 4 cases,deletion of ATM in 2 cases,deletion of p53 in 1 case,deletion of D13S25 combined RB1 in 1 case and 1 case with a variety of abnormalities. Conclusions Panel FISH is a useful method for detection of genomic aberration in CLL If it is combined with CC,it can obviously enhance the detection rate of chromosomal abnormalities in CLL.
10.Effects of miR-145 on the proliferation, apoptosis and cell cycle of a human keratinocyte cell line HaCaT

Objective To investigate the regulatory effects of miR-145 on the proliferation,cell cycle and apoptosis of a human keratinocyte cell line HaCaT.Methods miR-145 mimics and negative control (NC) mimics were chemically synthesized and then transiently transfected into HaCaT cells respectively.After additional culture for different durations,real-time PCR was performed to determine the expression level of miR-145,MTS assay to estimate cell proliferation,and flow cytometry to detect cell apoptosis and cycle.Luciferase assay,real-time PCR and Western blot were conducted to determine whether NRAS was the target gene of miR-145.Results The miR-145 expression level in miR-145 mimic-transfected cells increased by 85.00 ± 1.21 folds compared with NC mimic-transfected cells (t =115.90,P < 0.0001).The transfection with miR-145 mimics significantly inhibited the proliferation of HaCaT cells (F =8.76,P =0.008),and the inhibitory effect significantly varied with the duration (24-96 hours) of culture after transfection,with no interaction effect between the transfection with miR-145 mimics and culture duation (F =1.21,P =0.18).Compared with NC mimic-transfected cells,those transfected with miR-145 mimics showed a significant increase in the proportion of early apoptotic cells (18.9% ± 4.1% vs.4.3% ± 1.2%,t =7.126,P < 0.01),late apoptotic cells (9.3% ± 2.3% vs.3.6% ± 1.6%,t =12.38,P < 0.01),G1-phase cells (85.83% ± 5.2% vs.62.08% ± 6.23%,t =11.78,P =0.007),but a significant decrease in the percentage of G2-phase cells (6.26% ± 1.2% vs.19.36% ± 3.45%,t =7.610,P =0.017) and S-phase cells (7.91% ± 1.3% vs.18.56% ± 5.23%,t =7.230,P=0.019).As luciferase assay showed,luciferase activity was significantly lower in HaCaT cells cotransfected with miR-145 mimics and a recombinant luciferase reporter vector psi-CHECK2-NRAS-wild carrying the wild-type 3'UTR of NRAS than in those cotransfected with NC mimics and the vector psi-CHECK2-NRAS-wild (t =11.09,P =0.008),but similar between cells cotransfected with miR-145 mimics and a recombinant luciferase reporter vector psi-CHECK2-NRAS-mut carrying the mutant-type 3'UTR of NRAS and those cotransfected with NC mimics and the vector psi-CHECK2-NRAS-mut (P > 0.05).Real-time PCR and Western blot revealed that the overexpression of miR-145 mimics had no significant effect on NRAS mRNA expression (P > 0.05),but significantly inhibited NRAS protein expression (1.52 ± 0.07 vs.0.20 ± 0.02,t =28.43,P< 0.01).Conclusion miR-145 might inhibit proliferation and promote apoptosis of HaCaT cells by influencing cell cycle via NRAS.
