Objectives: To study the prolonging of mandible by distr ac tion osteogenesis (DO) after severe trauma in rabbits. Methods:O steotomy was conducted in 16 adult rabbits. To simulate severe trauma, periosteum was removed and inferior alveolar vessels were destroyed on left side. The rabb its were randomly divided into 4 groups with 4 in each group and internal distra ctor was applied in each rabbit. In group A1 and A2 the latency time was 1 day,i n group B1 and B2 that was 7 days.In group A1 and B1 the distraction velocity wa s 0.5 mm/d,in group A2 and B2 that was 1.0 mm/d.The distraction length was 10 mm and retention period was 4 weeks. Results:4 weeks after operati on on right side complete new bone formation was found in the distraction area. On left side that was observed in group B1 and B2,but not in A1 and A2.Nonunion was found in 1,4,0 and 0 cases in group A1,A2,B1 and B2 respectively. The bending strength (10 4 N/m 2) of normal bone was 95,that in group A1,B1,B2 was 25,85 and 60 respectively. Conclusion:DO method can be used even after the blood supply of the mandibles has been destroyed, but the latency period sh ould be longer.

This study was designed to explore the treatment effect of imperatorin(Imp)on chronic endometritis(CE)and the possible mechanism.A rat model of CE was established by injecting phenol gel into the ovary and was grouped into CE group,Imp low dose group(ImpL group,5 mg?kg-1?d-1),Imp high dose group(ImppH group,10 mg?kg-1?d-1),and ImpH+lipopolysaccharide(LPS,10 mg/kg)groups,while sham-treated rats were grouped as control group(CT group).Uterine organ index of all groups were measured;hematoxylin eosin(HE)staining was applied to observe the pathological changes of uterine tissue;immunohistochemistry was applied to detect the expression of vascular endothelial growth factor(VEGF)in the endometrium;ELISA was used to measure the levels of serum IL-6 and TNF-α;RT-qPCR and Western blot were applied to verify the expression of IL-1β,nuclear factor-κB(NF-κB),NOD-like receptor protein 3(NLRP3),and matrix metalloproteinase(MMP)-9 in uterine tissue.Compared with the CT group, the uterus and vagina of the CE group were swollen and congested, and the expression of VEGF in the endometrium was decreased(P<0.05),the levels of IL-6 and TNF-α in serum and the expressions of IL-1β,p-NF-κB/NF-κB,NLRP3,and MMP-9 in uterine tissue were increased(P<0.05).Imp could reverse these pathological changes mentioned above in CE model, while LPS was able to partially suppress the improvement effect of ImpH on CE rats.In conclusion,Imp may have a certain therapeutic effect on CE by inhibiting the activation of NLRP3 inflammatory pathway.

The eukaryotic genome is folded into higher-order conformation accompanied with constrained dynamics for coordinated genome functions. However, the molecular machinery underlying these hierarchically organized three-dimensional (3D) chromatin architecture and dynamics remains poorly understood. Here by combining imaging and sequencing, we studied the role of lamin B1 in chromatin architecture and dynamics. We found that lamin B1 depletion leads to detachment of lamina-associated domains (LADs) from the nuclear periphery accompanied with global chromatin redistribution and decompaction. Consequently, the inter-chromosomal as well as inter-compartment interactions are increased, but the structure of topologically associating domains (TADs) is not affected. Using live-cell genomic loci tracking, we further proved that depletion of lamin B1 leads to increased chromatin dynamics, owing to chromatin decompaction and redistribution toward nucleoplasm. Taken together, our data suggest that lamin B1 and chromatin interactions at the nuclear periphery promote LAD maintenance, chromatin compaction, genomic compartmentalization into chromosome territories and A/B compartments and confine chromatin dynamics, supporting their crucial roles in chromatin higher-order structure and chromatin dynamics.
