1.Study of anti-inflammatory function of human bea defensin 2 on rats' acute rhinosinusitis

OBJECTIVE To study the antiinflammatory function of human beta defensin 2(hβD-2) on acute rhinosinusitis in rats,in order to provide a new therapy for acute rhinosinusitis.METHODS Acute rhinosinusitis model were established on SD rats before and after the transfection of plasmid,the rats in experimental group were dropped with recombinant hβD-2 plasmid mixture in nose,while with empty plasmid mixture in control group.Immunohistochemistry method was used to prove the transfection results,nasal mucosa were hematoxylineosin stained to compare the pathological difference of nasal mucosa,nasal lavage fliud was collected and cultured to compare the colony number of the bacteria.RESULTS The expression of hβD-2 was confirmed by immunohistochemistry method,which mainly distributed in mucosal epithelium and gland,pathological results showed that the inflammation of nasal mucosa in experimental group was significantly relieved than that in control group.The number of Staphylococcus auresus colony number was significantly decreased in experimental group,while there was no significantly change in the control group.CONCLUSION Recombinant hβD-2 plasmid can be successfully transfected into the nasal mucosa of rats and expressed effectively.The anti-inflammatory ability of nasal mucosa was increased after the transfection,which is expected to provide a new treatment approach for acute rhinosinusitis.
2.Construction of a competency evaluation indicator system for infection prevention and control nurses in Operating Rooms

Objective:To construct a competency evaluation indicator system for infection prevention and control nurses in Operating Rooms (hereinafter referred to as "IPC") and provide an objective basis for the management of IPC nurses.Methods:From June to November 2022, an initial competency evaluation indicator system for IPC nurses was developed through literature review and semi-structured interviews. The Delphi method was employed to consult 20 experts from 11 provinces and municipalities across the country. Analytical Hierarchy Process (AHP) and mean distribution method were applied to quantify and determine the weight of each level of indicators within the system.Results:Nineteen experts were finally included, with two rounds of questionnaire recovery rates of 95.00% (19/20) and 100.00% (19/19), respectively. The authority coefficients of the experts were 0.858 and 0.861, familiarity coefficients were 0.850 and 0.853, and coefficients of judgment basis were 0.865 and 0.868, respectively. The Kendall's W coefficient of concordance for the two rounds of inquiries were 0.139 and 0.202 ( P<0.05), respectively. The final IPC nurse competency evaluation indicator system included six primary indicators, 22 secondary indicators, and 66 tertiary indicators. Conclusions:The constructed IPC nurse competency evaluation indicator system is scientific, reasonable, objective, and comprehensive, providing a valuable reference for the capability training, assessment, entry standards, and qualification certification of IPC nurses.
3.Analysis of Differences of Antidepressant Effect of Yueju Pill between Balb/c Mice and C57BL/6J Mice

This paper aimed at comparing the differences of antidepressant effects of Yueju Pill between the Balb/c mice and C57 BL/6 J mice and analyzing the effects on synaptic transmission in mice prefrontal cortex. Healthy adult Balb/c mice and C57 BL/6 J mice were randomly divided into control group and Yueju Pill group. Control group was treated with saline, Yueju Pill group was treated with single dose of Yueju Pill (13.5 g·kg-1) . The forced swimming test (FST) was measured 30 min after administration. The field excitatory postsynaptic potential (fEPSP) and long-term potentiation (LTP) of mice prefrontal cortex were detected by the electrophysiological experiment. In FST, the immobility time of Balb/c mice in Yueju Pill group was significantly lower than that in control group (P < 0.01), and the immobility time of C57 BL/6 J mice showed no remarkable difference between Yueju Pill group and control group (P> 0.05) . In electrophysiological experiment, the percentage of slope of fEPSP in Balb/c mice in Yueju Pill group was significantly higher than that in control group (P < 0.01), while there was no significantly difference in C57 BL/6 J mice between Yueju Pill group and control group (P> 0.05) . The LTP of Balb/c mice in Yueju Pill group was significantly increased than that in control group (P < 0.01), while there was no significantly difference in LTP of C57 BL/6 J mice between Yueju Pill group and control group (P> 0.05) . Yueju Pill may display rapid antidepressant effect via increasing fEPSP and LTP in prefrontal cortex of Balb/c mice and then enhancing the synaptic transmission.

With the gradual improvement of medical informatization and the vigorous development of medical and health big data,the exploration and practice of real -world research are becoming more and more mature ,and real -world data have become an important source of evidence for post marketing re -evaluation of drugs . As an important high -quality real -world medical data , electronic medical record data is an indispensable data source for post marketing re -evaluation of drugs . Most of the existing guidelines and norms of real -world research are designed from the perspective of prospective research ,and do not propose specific measures and methods in the implementation of retrospective research ,especially for the operation suggestions on the technical level of using conventionally collected electronic medical record data . In combination with the operational process framework formulated by the existing guidelines and norms ,this paper creatively adds the operating procedures for dataE- validation,data integration ,data verification ,and throughout quality control ,data management and storage of retrospectiveelectronic medical record data ,and describes the data analysis methods and key points involved in carrying out multi -center retrospective real -world research using electronic medical record data ,taking the post marketing safety research of drugs as an example. Finally,the full process operation procedure applicable to the use of multi -center retrospective electronic medical record data is established .
5.Construction of prediction model for early screening in male patients with gastric cancer based on cell -free DNA methylation and machine learning

Objective :
To construct a cell-free DNA ( cfDNA) methylation model for early screening in male pa- tients with gastric cancer by using novel cfDNA methylation detection technology.
Methods :
Methylation informa- tion of the whole genome of gastric cancer patients were detected by cell-free methylated DNA immunoprecipitation and highthroughput sequencing ( cfMeDIP-seq ) technology and locate gastrogenic cfDNA. Then bioinformation methods were used to extract specific methylation labels which could distinguish GC patients and establish diagnosis model by random forest algorithm. Related validation clinical researches were also conducted.
Results :
63 most sig- nificant DMR were selected to construct the cfDNA methylation model based on GC samples and normal control samples,the goal sensitivity was above 85 percent while the goal specificity was above 95% .The sensitivity and specificity of the validation set were 98. 7% and 99. 0% while the area under curve(AUC) was 0. 999.
Conclusion
The cfDNA methylation model constructed in this study has good performance in predicting GC.
