1.Influence of TNF-? gene polymorphisms on TNF-? production and disease

Tumor necrosis factor ?(TNF ?) is a potent proinflammatory cytokine. In human, TNF ? gene is located within the highly polymorphic major histocompatibility complex(MHC) region on chromosome 6p21.3. TNF gene cluster contains many polymorphisms including microsatellites and single nucleotide polymorphisms(SNPs).Many of these polymorphisms were found to be in linkage disequilibrium with HLA class Ⅰand Ⅱ alleles. Some of the TNF ? gene polymorphisms were found to influence TNF ? production in vitro , for example the 308SNP. Many studies have shown that this SNP and others within the TNF ? gene associate with different inflammatory conditions. Whether this phenomenon is due to the direct influence if the SNP in question and/or due to linkage disequilibrium with other polymorphisms within the TNF ? gene or the HLA system is still controversial.
2.Diagnostic Value of Galactography and Mammoscopy for Mammary Ductal Diseases:An Analysis of 100 Cases

Objective To improve the diagnostic rate of mammary ductal disease by galactography and mammoscopy.Methods 100 cases of the galactographic and mommoscopy materials were retrospectively analyzed.Results Tumorous diseases including intraductal papilloma,intraductal papillary carcinoma and ductal carcinoma,they accounted for 14%,intraductal papilloma and intraductal papillary carcinoma were the most in this group(12%).Non-tumorous diseases including mammary ductal ectasia with chronicmastitis,plasma cell mastitis,mammary cyst,lobular hyperplasia and cystic hyperplasia,they accounted for 71%,mammary ductal ectasia was the most in this group(42%).Conclusion The galactography and mammoscopy are very valuable in the diagnosis and differentiating diagnosis of mammary ductal diseases.
3.Study on recombinant chicken ? interferon production in yeast Pichia pastoris and detection for its antiviral activity

Objective:To obtain eukaryotic expressing protein of chicken interferon ? (ChIFN-?) and research its anti-virus activity.Methods: Chicken interferon ? mature protein gene was cloned and amplified by reverse transcripition-polymerase chain reaction(RT-PCR) from the total mRNA in the lymphocyte of chicken blood stimulated with ConA for 4~10 hours.The gene was inserted into the expression vector pPICZa-A,which had been cleaved by EcoR I and Xba I.The recombinant vector was linearized by Sac I and transferred into yeast Pichia pastoris,strain X33,anti-virus activity of the recombinant cytokine was detected by the classical experiment cell pathological effect inhibition assay.Results:The result showed that the preparation of recombinant interferon had higher anti-virus activity(10?48 U/ml).Conclusion: The recombinant chicken interferon ? with anti-virus bioactivity has been obtained.
4.Molecular Characterization of the Duck Enteritis Virus UL4 Gene

Duck enteritis virus (DEV) is a herpesvirus that causes an acute, contagious and fatal disease. In the present article, the DEV UL4 gene was cloned and sequenced from a vaccine virus. A degenerate oligonucleotide primer for the consensus site of herpesvirus UL3 gene and a specific primer located in UL5 were used in the polymerase chain reaction (PCR) to amplify a DNA product 2 086 bp in size. DNA sequence analysis revealed that a 714 bp open reading frame (ORF) of DEV encoding a 237 amino acid polypeptide is homologous to the family of herpesvirus UL4 proteins and therefore has been characterized as a DEV UL4 gene. Alignment of the DEV UL4 protein sequence with those of other alphaherpesviruses showed that 10 amino acid residues are completely conserved. Phylogenetic tree analysis showed that the seventeen alphaherpesviruses viruses analyzed were classified into four large groups, and the duck enteritis virus branched separately, closely related to the Mardiviruses group comprising Gallid herpesvirus 2 (GaHV-2), Gallid herpesvirus 3 (GaHV-3) and Meleagrid herpesvirus 1 (MeHV-1). The present study showed that the evolutionary relationship of the UL4 protein could be used for classification of alphaherpesviruses.
5.The combination of flow cytometry and automated cell image analyzer in determining the nature of ascites and pleural effusion nature

Objective To explore the clinical value of flow cytometry( FCM) and DNA automated cell image analyzer ( AICM) in determine the character of ascites and pleural effusion.Methods This was a cross-sectional study.203 ascites and pleural effusionsamples were random selected from PLA hospital inpatients between August 2013 to June 2014 .The DNA content of sediment cells were detectedthrough the FCM and AICM respectively benign and malignant disease were differentiated according the counts and proportion of aneuploid cells.The sensitivity, specificitywere calculated byROC curves.Results The sensitivity, specificity and accuracy of flow cytometry cell in detectingtumor cells were 78.6%,80.0% and 79.2%%, while the sensitivity, specificity and accuracy of image analyzer were 83.5%,78.6% and 81. 3%respectively.When FCM and AICMwere combined ,the sensitivity, specificity and accuracyincreased to 92.2%, 86.3% and 89.6%.Conclusions Compared toconventional cytology test, the sensitivity and specificity were significantly high when the two methods were combined .Therefore, the combination method can be used to assist in clinical identification of the nature of ascites and pleural effusion and to help the diagnosis of disease.
6.Construction and biological characteristic for the recombinant modified vaccinia virus ankara co-expressing modified GP5 and M protein of porcine reproductive and respiratory syndrome virus.

Modified ORF5 (MORF5) and ORF6 gene of PRRSV were cloned into two multiple cloning sites of MVA transfer vector pLR-gpt to construct the recombinant plasmid pLR-MORF5/ORF6. Homologous recombination between pLR-MORF5/ORF6 and the wtMVA on BHK-21 cell line was mediated with liposome by infecting the cell with 0.01 MOI wtMVA two hours before transfecting the recombinant plasmid into the cell. When the cytopathic effect (CPE) was obvious, virus was collected from the cell plate and the recombinant virus was selected with drug selecting medium (2% MXHAT). After 12 cycles of selection, rMVA with a selection marker Eco gpt was obtained and named as rMVAgpt-MGP5/M. By infecting BHK-Cre expressing Cre recombinant enzyme, the Eco gpt marker in rMVAgpt-MGP5/M was deleted and this rMVA was named as rMVA-MGP5/M. The insertion of MORF5 and ORF6 into the MVA genome was confirmed with PCR analysis and the expression of MGP5 and M protein was identified with Western blot and IFA. Through biological study on the recombinant MVA, no obvious difference was observed between rMVA-MGP5/M and the wtMVA regarding to the CPE and growth curve. The recombinant MVA constructed in this study could coexpress the modified GP5 and M protein and the expressed product had good immunocompetence. Furthermore, the insertion of the MORF5 and ORF6 into MVA genome had no obvious effect on the replication and biological characteristics of this virus.

7.Histopathology of the temporal artery in young ischemic stroke patients with elevated serum immunoglobu-lin E

Objective Ischemic stroke with elevated serum immunoglobulin E ( IgE) in some young patients is regarded as cerebral vasculitis clinically though without sufficient pathological evidence .This study was to investigate the characteristics of vascular lesions in these patients by temporal artery biopsy . Methods We performed histopathologic examinations on the temporal arteries of 32 young ischemic stroke patients with unknown etiology , 16 with normal and the other 16 with elevated serum IgE .We observed inflammatory cells infiltration and mast cells by HE staining and toluidine blue stai-ning respectively and determined the expressions of matrix metalloproteinase -9 (MMP-9), monocyte chemotaxis protein -1 (MCP-1) and serum IgE by immunohistochemistry . Results Compared with the patients with normal IgE , those of the elevated IgE group showed a significantly higher rate of inflammatory cells infiltration (12.5%vs 62.5%, P<0.01), with 1 case of focal necrosis and fi-brinous exudation in the adventitia in the latter group .The average optical density ( OD) of monocyte chemotaxis protein-1 ( MCP-1) in the temporal artery was also dramatically higher in the elevated IgE group than in the normal controls ([9.25 ±5.79] ×10 -5 vs [4.41 ±2.87] ×10 -5, P<0.01).The average OD of matrix metalloproteinase 9 (MMP-9) and intima-media thickness were both increased in the elevated IgE group ([32.79 ±21.38] ×10 -4 and [0.25 ±0.06] mm) but showed no statistically significant differ-ence from those in the normal IgE group ([25.23 ±12.78] ×10 -4 and [0.22 ±0.06] mm) (both P>0.05).Nor was any signifi-cant difference observed in the number of the mast cells between the normal and elevated IgE groups (2.8 ±1.5 vs 3.6 ±2.3, P>0.05). Conclusion The infiltration and necrosis of inflammatory cells and fibrin exudation in the temporal artery of the young pa-tient with elevated serum IgE are likely to be the manifestations of vasculitis , and MCP-1 may play a role in the pathogenesis of the disease.
8.Liposoluble constituent of ethylacetate extract of Holotrichia diomphalia larvae

Objective To analyze the chemical components of the liposoluble constituent of Holotrichia diomphalia lar-vae .Methods Essential liposoluble compounds were extracted from H .diomphalia through ethylacetate .GC-MS analysis was used to identify its main components .Results 33 peaks were separated and 33 compounds were identified from ethylacetate-ex-tracted H .diomphalia .The main compounds were pentadecylic acid ,Z-11-Hexadecenoic acid ,hexadecanoic acid ,heptade-canoic acid ,oleic acid and octadecanoic acid .Conclusion This work will contribute to the study of medicinal animals and con-firmation of active compounds in H .diomphalia .
9.Obesity and male reproductive health: An update

Obesity has become a growing epidemic that seriously affects the quality of human life. Obesity not only adds the burden to the body but may have negative impacts on various body systems and organs, such as the cardiovascular and metabolic systems. In recent years, some studies have shown that obesity also negatively affects the reproductive system and influences male reproductive health. Male reproductive health is not only related to the normalization of male reproductive function but also plays a role in the ability to conceive healthy offspring. This article summarizes research progress on the effects of obesity on male reproductive health, including endocrine hormone disruption, reproductive organ structural damage, increased oxidative stress, and reproductive cell epigenetic alterations, providing insights for a deeper understanding of the influence of obesity on male reproduction and potential treatments.
10.The effect of butylphthalide injection combined with interventional therapy on patients with acute anterior circulation large vessel occlusive stroke

Objective To investigate the effect of butylphthalide and sodium chloride injection on patients who received endovascular treatment for acute anterior circulation large vessel occlusive stroke.Methods A total of 173 patients were identified from February 2015 to December 2017 in the Department of Neurology of Jingling Hospital in this retrospective observational study.Propensity score-matching analysis was performed to balance differences in baseline characteristics between patients who received butylphthalide injection (butylphthalide group) and those who did not (control group).The modified Rankin Scale scores at 90 days were compared between the butylphthalide and control groups.Results A total of 144 patients who received endovascular treatment for acute anterior circulation large vessel occlusive stroke were finally analyzed,54 cases in the butylphthalide group and 90 cases in the control group.The proportion of good functional outcome at 90 days in the butylphthalide group was higher than that in the control group (63.0% (34/54) vs 44.4% (40/90);x2=4.633,P=0.031).Thirty-six pairs were matched successfully by the propensity score matching,36 patients in the butylphthalide group and 36 in the control group.There was no statistically significant difference in the 90-day functional outcome between the two groups (66.7% (24/36) vs 44.4% (16/36);x2=3.600,P=0.058).One hundred and fifteen patients were recanalized,47 cases in the butylphthalide group and 68 cases in the control group,and after the propensity score matching,30 pairs were analyzed.The proportion of good functional outcome at 90 days in the butylphthalide group was higher than that in the control group (73.3% (22/30) vs 46.7% (14/30);x2=4.444,P=0.035).Conclusion After propensity score-matching,butylphthalide and sodium chloride injection could improve 90-day functional outcome in patients with acute anterior circulation large vessel occlusive stroke and obtained recanalization by endovascular treatment while could not before propensity score-matching.
