1.A systematic review of the mechanisms of acupuncture for myocardial ischemia on central nervous system

Based on the systematic review on acupuncture improving the mechanism of central nervous system in animal model of myocardial ischemia in recent years, we analyzed the effects of acupuncture on medulla oblongata rVLM zone, hippocampus, hypothalamus, spinal cord segment and automalic to the mechanism of positively regulating myocardial ischemic, which provided a new way for the clinical treatment of myocardial ischemia and cardiovascular diseases.
2.Measurement of recombinant human glucagon-like peptide-1(7-36) concentration by ELISA kit methods

AIM: To qualitatively detect the concentration of recombinant human glucagon-like peptide-1(7-36)[rhGLP-1(7-36)] in human blood.METHODS: RhGLP-1(7-36) concentrations of standard and blood samples were detected by ELISA kits.RESULTS: The parameters of specificity,sensitivity,detection limits,reproducibiltiy and recovery can meet the requirements for pharmacokinetic studies.CONCLUSION: The concentration measuring method of rhGLP-1(7-36) in blood samples using ELISA kit produced by LINCO company can meet the detection methods of biological production,which can be used as a measuring method of rhGLP-1(7-36) concentration and for its pharmacokinetic studies.
3.Application and Proposals of X-ray Field Cabin in Earthquake Relief in Wenchuan

Field cabin,a new generation of our military medical support system,was used in the Wenchuan Earthquake relief for the first time. The ordonnance and features of X-ray field cabin were also laid out. In Mianyang city,within 86 days,there were 710 wounded persons were checked up,and the rate of high quality photo was up to 85%. The rational ordonnance,steady performance,easily deployed and withdrawn features of the X-ray field cabin ensure it adapted to long-distance medical support and fulfill its mission effectively. However,some disadvantages of it are also found in the application,and innovating proposal is further introduced.
4.Detection of occult HBV infection and probably window period infection among single NAT reactive blood donors

Objective To analyze hepatitis B virus(HBV) infection stage in single nucleic acid test(NAT)reactive blood donors.Methods Blood donor samples were screened routinely for HBV DNA by using transcription-mediated amplification(TMA) NAT and quantitative polymerase chain reaction(PCR).Then serum markers of HBV were also detected.The HBV infection stage was analyzed.Results Among the 225 single NAT reactive samples,78(34.67%) were identified to be reactive for HBV DNA by TMA NAT discrimination test and/or PCR test,of which 63(82.89%) were occult HBV infection(OBI),13(17.11%) were probably window period infection(pWP),and 2 cases could not be classified for infection stage.Among the OBI samples,49 samples(77.78%) were with HBV DNA concentration less than 20 IU/mL,whereas,there were only 4 samples(30.77%) in pWP samples.The 225 samples were classified into three groups according to the S/CO of NAT, including 1-<6 group,6-<10 group and 10-17 group, the confirmed HBV DNA positive rates of which were 13.11%,13.64% and 47.18%,and the positive rate of 10-17 group was higher than 1-<6 group and 6-<10 group(P<0.05).In all 63 OBI samples,there were 8(12.70%),3(4.76%) and 52(82.54%) samples were classified into S/CO 1-<6,6-<10 and 10-17,respectively.All of the 13 pWP samples were with NAT S/CO of 10-17.Conclusion Part of single NAT reactive blood donors could be with HBV infection,of which OBI might be popular than pWP, with very low concentration of HBV DNA.Deferral of single NAT reactive blood donors could reduce transfusion-transmitted HBV infection.
5.Effect of bag-1 expression on the occurrence of breast cancer and its 5-year survival rate

BACKGROUND: Bag-1 is a multifunctional and anti-apoptotic gene. Its anti-apoptotic ability is enhanced when binding to bcl-2 to form a complex.Now it is considered as a predictive biomarker for the early diagnosis of breast cancer. However, whether it is useful in the assessment of the prognosis of breast cancer is still elusive.OBJECTIVE: To explore the expression of bag-1 in breast cancer and its role for prognosis.DESIGN: A controlled study with breast cancer, benign breast tumor and normal breast tissues as subjects.SETTING: The Vascular and Endocrine Surgery Department of Xijing Hospital of the Fourth Military Medical University of Chinese PLA.MATERIALS:Totally 100 breast cancer specimens were obtained form May 1995 to May 2000. Ten benign breast tumor and 10 normal breast tissues were used as control. All the specimens were paraffin-embedded and came from the Pathological Department of Xijing Hospital Affiliated to Fourth Military Medical University of Chinese PLA.METHODS: Immunohostochemical strept avidin-biotin complex(SABC) method was adopted to detect bag-1 expression in these specimens.pression levels of bag-1.RESULTS: The positive expression rate of bag-1 in breast cancer (85%) was significantly different form those of benign breast tumor (10%) and normal breast (10%) (χ2= 29.98, P = 0.00). While the positive expression rates in breast cancer of different stages (stage Ⅰ, stage Ⅱ and stage Ⅲ ) were 88%, 82% and 88%, respectively, which has no significant difference (χ2 = 0. 61, P = 0.75) . In duct carcinoma, lobular carcinoma and special carcinoma, bag-1 positive expression rate was 86%, 85% and 80%,which was also no significantly different (χ2 =0.16, P =0.95). In the 94followed patients, the 5-year survival rate of positive bag-1 expression was 79% and that of negative bag-1 expression was just 9%. The difference was significant (χ2 = 0. 07, P = 0.04).CONCLUSION: High bag-1 expression exists in breast cancer and its level is not associated with the clinical stages or pathological types of the cancer.Therefore, bag-1 may be used as a predictive marker for the prognosis of breast cancer.
6.Functional expression of voltage-gated sodium channels Nav1.5 in human breast cancer cell line MDA-MB-231.

Voltage-gated sodium channels (VGSCs) are known to be involved in the initiation and progression of many malignancies, and the different subtypes of VGSCs play important roles in the metastasis cascade of many tumors. This study investigated the functional expression of Nav1.5 and its effect on invasion behavior of human breast cancer cell line MDA-MB-231. The mRNA and protein expression of Nav1.5 was detected by real time PCR, Western Blot and immunofluorescence. The effects of Nav1.5 on cell proliferation, migration and invasion were respectively assessed by MTT and Transwell. The effects of Nav1.5 on the secretion of matrix metalloproteases (MMPs) by MDA-MB-231 were analyzed by RT-PCR. The over-expressed Nav1.5 was present on the membrane of MDA-MB-231 cells. The invasion ability in vitro and the MMP-9 mRNA expression were respectively decreased to (47.82+/-0.53)% and (43.97+/-0.64)% (P<0.05) respectively in MDA-MB-231 cells treated with VGSCs specific inhibitor tetrodotoxin (TTX) by blocking Nav1.5 activity. It was concluded that Nav1.5 functional expression potentiated the invasive behavior of human breast cancer cell line MDA-MB-231 by increasing the secretion of MMP-9.
7.Research progress of RNA m 6A modification in malignant tumor and its mechanism of radiotherapy

N 6-methyladenosine (m 6A) is the most abundant RNA base modification in mammals, especially in eukaryotic messenger RNA (mRNA). N 6-methyladenosine modification can regulate RNA splicing, translocation, stability and ultimately affect protein synthesis. m 6A modification is catalyzed by RNA writers, reduced by erasers and also be recognized by readers. Abnormal changes ofm 6A levels are closely related to tumor occurrence and development, including proliferation, growth, invasion and metastasis. In the process of tumor radiotherapy, m 6A modification affects the efficacy of radiotherapy by affecting DNA damage, tumor stem cell generation and tumor cell radiation sensitivity. This article reviews the role of m 6A-modified epigenetic regulation in malignant tumors and the research progress of its mechanism in tumor radiotherapy, in order to provide new ideas for the development of clinical tumor molecular targeted therapies and radiosensitizers.
8.Avascular necrosis of the metacarpal head: a case report and literature review

Osteonecrosis is the most common disorder in femoral head and scaphoid. However, avascular necrosis (AVN) in metacarpal head is a rare disease. In the present study, a 14-year-old male patient complained of right-hand pain with a limited range of motion for one month. Physical examination showed that the active flexion was 70° and the extension was limited to 30° of the metacarpophalangeal (MCP) joints. The grip strength decreased to 60% of the contralateral hand. X-rays demonstrated flattening and sclerotic changes in the long finger and ring finger of metacarpal head. CT scanning indicated cystic, osteochondral defects and sclerotic changes in metacarpal head. Imaging examination further confirmed the diagnosis of AVN in the long metacarpal finger and ring finger. After conservative treatments including splint immobilization, non-steroidal anti-inflammatory drugs (NSAIDs) and physiotherapy, the local pain symptoms of the MCP joint gradually disappeared. The range of motion of MCP joint returned to normal with the grip strength score as 105% of the contralateral hand. MRI confirmed excellent remodeling and regeneration in the metacarpal head at two years later. The clinical characteristics, diagnosis and treatments of AVN of metacarpal head were reviewed. Although radiograph examination is commonly used, early-stage osteonecrosis of the metacarpal head should be confirmed by MRI. Given the rarity of this disorder, there is no consensus on the treatments. Metacarpal necrosis is the more common disorder in adolescent patients with a history of trauma. Considering the potential of bone remodeling, juveniles with metacarpal head necrosis could recover by conservative treatments.
9.Effect of Balance Training on Cerebral Cortex and Balance Function of Mouse

Objective To observe the effect of balance training on cerebral cortex and balance function of mouse.Methods Forty-five C57BL/6J female mice were randomly divided into the control group, two-week training group and four-week training group with 15 animals in each group. An new model of balance function training was copied. The balance function, brain index, and the content of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in the mouse cerebral cortex were tested after training.Results The total time passing balance beam of the four-week training group significantly shorten compared with the control group and two-week training group ( P<0.01). Compared with the control group, the brain index of two-week training group increased ( P<0.05), and four-week training group also increased significantly ( P<0.01). Compared with the control group, the SOD activity of cerebral cortex significantly increased (P<0.01), and MDA decreased ( P<0.05) in the four-week training group.Conclusion This new balance training can improve balance function, increase the brain index and decrease lipid peroxidation level in the cerebral cortex of the mice.
10.Expression of voltagE-gated sodium channel SCN5A/Nav1.5 in human ovarian cancer and its significance

Objective:The different subtypes of voltagE-gated sodium channels (VGSCs) are known to correlate with the migration of many malignant cancers. This study was to investigate the significance of functional expression of SCN5A/Nav1.5 in human ovarian cancer and its effects on migration capability of ovarian cancer cells in vitro. Methods: Sodium indicator SBFI and immunofluorescence method were used to detect the distribution of intracellular Na+. Real-time PCR, Western blotting, and immunohistochemistry were used to detect the mRNA and protein expression of SCN5A/Nav1.5. The effect of specific voltagE-gated sodium channels inhibitor tetrodotoxin (TTX) on cell viability was measured by CCK-8 kit. The migration and invasion of ovarian cancer cell lines SKOV-3 were tested by Transwell chamber assay. Results:SCN5A/Nav1.5 were over-expressed in ovarian cancer cell lines SKOV-3 and ovarian cancer specimens at mRNA and protein levels. TTX 30 μmol/L inhibited the intracellular Na+ concentration by (41.51±0.41)%. TTX also suppressed the invasion and migration capacities of SKOV-3 cells by (33.80±1.6)% and (43.60±2.9)%, respectively. The difference was significant (P<0.05). Conclusion:SCN5A/Nav1.5 is involved in the metastasis progression of ovarian cancer in vitro and plays an important role in the initiation and progression of ovarian cancer. It may become a target for ovarian cancer therapy.
