1.Treatment Features and Clinical Thought of Pro. WU Xuesu Treating Subclinical Thyroid Inflammation

[Objective] To summarize treatment ideas for subclinical thyroid inflammation by Professor WU Xuesu.[Methods] By learning from Professor WU Xuesu, studying relevant literature cases;collecting and reporting the case of her clinical characteristics for the treatment of subclinical thyroid inflammation, analyzed and summarized treatment experience. [Results] It is argued that the root of subclinical thyroid inflammation is turbid-phlegm, nitrogen stasis cardiac knoting in the anterior portion, it should be treated by resolving phlegm mainly,and secondary promoting Qi activating blood. [Conclusion] The treatment ideas of Professor WU Xuesu for subclinical thyroid inflammation has a good curative effect.
2.New progress in the study of protein kinase C(PKC)inhibitors

Protein kinase C (PKC) is a group of phospholipid-dependent serine/threonine protein kinases, which togeth-er with protein kinase A (PKA) and protein kinase G (PKG) constitute a superfamily of serine/threonine protein kinase AGC. PKC includes classic PKC, novel PKC, atypical PKC and some members of kinase associated with PKC (PRK). PKC is wide-ly distributed in mammalian tissues and cells, which plays an important biological role in growth and metabolism, prolifera-tion and differentiation of cells. A series of studies have demonstrated that variations of multiple cells, occurrence and devel-opment of diseases are related to the abnormal expression of PKC. Therefore, designing and searching for efficient PKC inhib-itors have very important implications for synthesis of many kinds of effective drugs and treatment of a variety of clinical dis-eases including cancer, cardiovascular, and hypertension, et al. In recent years, the research on PKC inhibitors has become the focus of domestic and foreign research. A large number of literatures have reported many kinds of effective PKC inhibi-tors, and analyzed their function site, mechanism, clinical trial data and so on. The discovery of these PKC inhibitors has im-portant implications for structural analysis of PKC and the treatment of diseases. So in this paper, the efficient PKC inhibi-tors are summarized.
3.Changes in the myocardial and liver tissues of exercise-induced fatigue rats after acupuncturing different acupoints

BACKGROUND:According to the theory of Traditional Chinese Medicine, stimulating certain acupoints of the body after exercise may improve its antioxidant capacity, but different acupoints have different effects. OBJECTIVE:To compare the effect of different acupuncture combinations on the antioxidant capacity of myocardial and liver tissues in rats after exhausted swimming exercise, and screen out the optimal acupoint combination for exercise-induced fatigue. METHODS:Seventy Sprague-Dawley male rats were divided into five groups: control group (n=10), model group (n=15),Sanyinjiao +Zusanli group (n=15),Shenshu +Zusanli group (n=15),Dazhui+Zusanli group (n=15). Except the control group, rats in the other four groups were subject to exhausted swimming. In the three acupuncture groups, rats were given acupuncture at corresponding acupint combinations at 15 minutes after exercise. RESULTS AND CONCLUSION:Compared with the control group, the activities of superoxide dismutase and glutathione peroxidase in the myocardial and liver tissues were significantly reduced in the model group, while the malonaldehyde content in cardiac muscle was significantly increased. Compared with the model group, the activities of superoxide dismutase and glutathione peroxidase in the cardiac muscle were significantly increased inSanyinjiao +Zusanli group, the superoxide dismutase in liver tissue was significantly increased inShenshu+Zusanli group, the malonaldehyde content in the myocardial tissue was significantly decreased in Sanyinjiao +Zusanli group and Dazhui +Zusanligroup, the malonaldehyde content in the liver tissue was significantly decreased in Sanyinjiao +Zusanligroup. Different acupuncture combinations have different effects on the activities of superoxide dismutase and glutathione peroxidase, as wel as the malonaldehyde content in the myocardial and liver tissues of fatigued rats. Acupunture could increase the antioxidant ability in cardiac muscle of rats and the best combination might beDazhui+Zusanli; acupoint combinations ofShenshu +Zusanli andSanyinjiao +Zusanli are specific for liver tissue.
4.Mechanisms and reversal strategies of tumor multidrug resistance

Tumor multidrug resistance (MDR) is the leading cause of clinical chemotherapy failure and death.Researches show that the occurrence of MDR is related to P-glycoprotein,multidrug resistance-associated proteins,breast cancer resistance-associated proteins,lung resistance-related proteins and other factors.Now the reversal strategies of MDR include chemical drugs reversal,gene reversal,immune reversal,traditional Chinese medicine reversal and drug-loaded nano-system reversal,which make some progress and help to improve the effect of cancer chemotherapy.
6.Mining gene expression profi le in def icient cold syndrome with characteristics functional modules

Objective:To identify the deficient cold syndrome relevant genes and functional modules,based on gene expression profiles and gene functional knowledge to study the molecular mechanism of deficient cold syndrome.Methods:Screening the differential expression genes with Array Tools,and testing the classificational results to the samples.The Gene Ontology Tool was used for annotating characteristics functional gene,selecting functional modules enriched with differentially expressed genes;and then identifing the feature modules and analyzing their association with the deficient cold syndrome.Results:The accuracy rate of classification samples was 93% with 25 different gene,the accuracy rate was 87%-100% with 11 functional modules.Conclusion:These features function modules had some relevance to the occurrence of deficient cold syndrome.It was more favorable interpretation of the disease by studying functional modules based on gene expression profiles.
7.Effects of leptin on proliferation and differentiation of human preadipocyte

Objective To observe the effects of leptin on the proliferation and differentiation of human preadipocyte in vitro, and to explore the possible mechanism of the generation of obesity regulated by leptin. Methods The human preadipocytes were isolated from human subcutaneous adipose tissue of abdomen and cultured in vitro. The effects of leptin (0-1 000 ng/ml) on the proliferation, lipid accumulation and the mRNA expression of PPAR-γ2 and C/EBP-α, which are the differentiation and transcription factor of human preadipocyte, were analyzed by the methods of MTT, cell counting, extracting stained intracytoplasmic lipid with oil red O and RT-PCR. Results Leptin (1 000 ng/ml) could stimulate the proliferation, lipid accumulation and the mRNA expression of PPAR-γ2 and C/EBP-α (P<0. 05). There were not obvious effects on the proliferation and lipid accumulation in the groups of lower (10 ng/ml) and common (100 ng/ml) concentration (P>0. 05). Conclusion Leptin in higher concentration can stimulate the proliferation and differentiation of preadipocye in vitro, which indicates that leptin may regulate the generation of obesity through acting on the proliferation and differentiation of preadipocyte at the pathologic state of leptin resistance and high leptin concentration in serum.
8.In vitro amplification and ultrastructure of dendritic cells from mouse bone marrow

BACKGROUND: Denddtic cells (DCs) constitute the dominant population of antigen presenting cells (APCs) by possessing potent ability to initiate T cell immunity. The ultrastructure study of DCs is less reported. OBJECTIVE: To investigate the ultrastructure of DCs from mice bone marrow at different maturation stages, and the morphology of DCs between CD40 ligation and tumor necrosis factor-alpha (TNF-α) stimulation in vitro. METHODS: Mice myeloid DCs were generated from bone marrow in vitro using granulocyte-macrophage colony-stimulating factor (GM-CSF)and interleukin-4 (IL-4). Immature DCs were loaded with apoptotic tumor cells (AP-DC), and AP-DC was then stimulated with CD40L-CHO cells and TNF-α for 48 hours, respectively. DCs were routinely sectioned, and ultrastructure was observed under transmission electron microscope. RESULTS AND CONCLUSION: Immature DCs showed a few short and blunt cytoplasmic processes, there were specific morphology lysosomes that liked earphone in some cells; DCs engulfing the apoptotic bodies were observed; sub-cellular structures between CD40 ligation and TNF-α stimulated DCs were different, the former had typical morphology of mature DCs which exhibited many dendritic protrusions, however, some DCs displayed apoptosis and autophagy after TNF-α stimulation. In a conclusion, CD40 ligation plays an essential role in myeloid DCs differentiation and maturation, TNF-α can mediate apoptosis and autophaqy of DCs.
9.QuEChERS-Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry for Determination of 2,4,6-Trinitrophenol, 2,4,6-trinitroresorcinate and 2,4,6-Trinitrophloroglucinol in Soil

A simple, rapid sensitive and accurate ultra performance liquid chromatography tandem mass spectrometric ( UPLC-MS/MS ) method was developed for the simultaneous determination of 2 , 4 , 6-trinitrophenol, trinitroresorcinate, trinitrophloroglucinol residues in soil. The sample was pretreated by using the modified QuEChERS ( quick, easy, cheap, effective, rigged, and safe ) method that completed the extraction and clean-up steps in one procedure. In this none buffer QuEChERS method, samples were extracted with 1% formic acid ﹢ acetonitrile, cleaned up with primary graphitized carbon black ( GCB) and C18 sorbent, then centrifuged and filtrated before detection. The pretreatment method was simple, rapid and effective and can meet the detection requirements. The UPLC-MS/MS method was performed on Waters Accucore PFP (150 mm× 2. 1 mm, 2. 6 μm) and the column temperature was 30 ℃, the gradient elution with acetonitrile and ammonium acetate as the mobile phase and the flow rate was 0. 3 mL/min. The negative electrospray ionization ( ESI-) source under the multiple reaction monitoring ( MRM ) mode and external standard method were used for quantification. The results showed that the correlation coefficients up to 0. 9942 were obtained across a concentration range of 0. 005-5. 0 mg/L. The limits of detection ( LOD) ranged from 0 . 002 to 0 . 005 mg/kg ( S/N=3 ) . The method was validated with soil samples spiked at three fortification levels (0. 01, 0. 1 and 1 mg/kg) and recoveries were in the range of 79. 3%-94. 8% with relative standard deviations (RSD) of 3. 1%-6. 6%.
10.The wrap-round la flap with part toenail combined with the second simepulp flap for repair of total avulsion of fingers

Objective To explore the method for repair of total avulsion of 2~4 fingers and to find the best procedure. Methods The wrap-round flap with part toenail combind with the dorsal pedical flap to repair soft tissue defect of the dorsal of finger and part of the palmar of finger, the second simepulp flap to repair soft tissue defect of the palmar of finger,transfer of two tissues with one pedicle to repair total avulsion of 2~4 fingers. Results All of 9 composite transplants in 6 cases were survived,the received areas were primary healing in 8 finger,1 case repaired by the bilateral wrap-round flaps and second simepulp flaps for index and middle finger,the distal phalanx of middle finger necrosis because of dislocation of DIP,3 months later,injury healed after the necrosis of phalanx excised,but the nail and the nail bed were dead.Except 1 case which the grafted skin necrotized in the donor area of the big toe,the rest had primary healing.All cases were followed up 6~14 months,the activity of joints were: MP 0-70,PIP 0-40.The repaired pulps had excellent appearance,the nail was fine,1 finger's mail was atrophy and 1 finger's nail was defect.The donor area had good fountion without pain and edema,the grafted skin had normal color and the activity of joints were nomal. Conclusion The wrap-round flap with part toenail combined with the second simepulp flap is a good approach in treatment of the skin avulsion injury in the finger.
