1.Advances in the study of regulation of novel organic cation transporter-2.

Novel organic cation transporter-2 (OCTN2), a member of the organic cation transporter family, may transport carnitine and multiple organic cationic drugs. Thus OCTN2 possesses substantial roles in physiology and pharmacology. A number of researches have proven that many factors can regulate the expression and/or function of OCTN2 via different pathways, and then may affect the homeostasis and disposition of drugs. This paper reviews recent progresses in this field.

3.CLONIG AND CHARACTERIZATION OF A NEW ICE NUCLEATION ACTIVE GENE FROM ICE NUCLEATION ACTIVE BACTERIUM ERWINIA ANANAS 110

Using the chromosomal DNA of an ice nucleation active bacterium Erwinia ananas 110 as template, an ice nucleation active (ina) gene was amplified by PCR with Taq plusI DNA polymerase. After sequencing and compared with reported ina genes, the cloned gene was identified as a new ina gene and was registered in GenBank at the accession number of AF387802. The new ina gene, named as iceA, has 3921 bp for its coding region, which encodes 1306 amino acids consisting of repetitive segment (R-domain, 1104aa), which is flanked by N-and C-terminal sequences, with 161 aa and 41aa, respectively.
4.Therapeutic Observation of Acupuncture plus Medication for Cough Variant Asthma

Objective To observe the clinical efficacy of acupuncture plus oral administration of self-made Chinese herbal decoction in treating cough variant asthma (CVA).Method Sixty-two CVA patients were randomized into a treatment group of 32 cases and a control group of 30 cases.The treatment group was intervened by acupuncture plus oral administration of self-made Chinese herbal decoction,and the control group was intervened by Montelukast sodium tablets.Before and after 3 courses of treatment,the score of cough,eosinophil (EOS) count,pulmonary function tests [peak expiratory flow (PEF),forced vital capacity (FVC),and forced expiratory volume in 1 second (FEV1)] were observed.The clinical efficacies were compared between the two groups.Result After the treatment,the score of cough,EOS count,and pulmonary function tests (PEF,FVC,and FEV1) were changed significantly in both groups (P<0.05).The score of cough,EOS level and pulmonary function tests in the treatment group were significantly different from those in the control group after the treatment (P<0.05,P<0.01).The total effective rate was 93.8% in the treatment group versus 80.0% in the control group,and the difference was statistically significant (P<0.05).Conclusion Acupuncture plus oral administration of self-made Chinese herbal decoction is an effective method in treating CVA.
5.Community characters of Lophatherum gracile in Yongchuan district of Chongqing.

To understand Lophatherum gracile plant community's structural characteristics, a survey of community structure and species diversity was conducted through quadrat sampling in Yongchuan district of Chongqing. The results showed that there were 386 species vascular plants, belonging to 117 families and 229 genera. Based on habitat, community structure and species composition, L. gracile were found in three community types: Pinus massoniana community, banboo community, shurb community. Vertical structure was composed of three layers, including tree layer, shrub layer and herb layer. Species in shrub layer was the richness. P. massoniana is the only dominant species of the community, it can not regenerate naturally, the shrub layer has a greater effect on the community of L. gracile in the future. In addition, the banboo community and shurb community is not stable because of human's activity. Therefore, the community characters of L. gracile should be taken care of conservation when the resources are utilized.

6.Establishment of an N-2a cell line stably expressing mouse galanin and the effect of over-expressed galanin on the proliferation and apoptosis of N-2a cell.

OBJECTIVETo construct an N-2a cell line stably expressing PcDNA 3.1-platelet derived growth factor-galanin (GAL) and explore the effect of over-expressed GAL on proliferation and apoptosis of N-2a cell in vitro.
METHODSThe vector containing the target gene was transfected into N-2a cells by liposome, and cell clones stably over-expressing GAL was obtained via G418 screening. GAL mRNA and protein levels were determined by reverse transcriotion-polymerase chain reaction (RT-PCR) and Western blot. The proliferation of N-2a cells was detected by MTT.The cell cycle and apoptosis were detected by flow cytometry.
RESULTSRT-PCT and Western blot indicated that GAL genes were highly expressed in the transfected N-2a cells (i.e.GAL-N-2a). As shown by MTT, the proliferation of the N-2a cells transfected with PcDNA 3.1-PDGF-GAL was significantly slower than the control group(P<0.05). Compared with the non-transfected cells in the control group, the N-2a cells with endogenously overexpressed GAL were arrested at the G0/G1 phases, and the over-expressed GAL protein significantly induced the N-2a cell apoptosis in a concentration-dependent fashion.
CONCLUSIONEukaryotic expression vector PcDNA 3.1-PDGF-GAL can encode the expression of GAL in N-2a cells. Aslo, it can inhibit cell proliferation and promote the cell apoptosis.

8.Electrophysiological effects of hydrogen sulfide on guinea pig papillary muscles in vitro.

The cardiac electrophysiological effects of hydrogen sulfide (H2S) were examined in guinea pig papillary muscles in vitro using intracellular microelectrode technique. The results obtained were as follows: (1) the duration of action potential (APD) in the normal papillary muscles was decreased by NaHS (H(2)S donor, 50, 100, 200 micromol/L) in a concentration-dependent manner; (2) in partially depolarized papillary muscles, 100 micromol/L NaHS not only reduced APD, but also decreased the amplitude of action potential (APA), overshoot (OS) and maximal velocity of depolarization at phase 0 (V(max)); (3) pretreatment with ATP-sensitive K(+) (K(ATP)) channel blocker glibenclamide (20 micromol/L) partially blocked the effects of NaHS (100 micromol/L); (4) pretreatment with L-type Ca(2+) channel agonist Bay K8644 (0.5 micromol/L) also partially blocked the effects of NaHS (100 micromol/L); (5) pretreatment with Ca(2+)-free Krebs-Henseleit solution containing glibenclamide (20 micromol/L) completely blocked the effects of NaHS (100 micromol/L); (6) APD in the normal papillary muscles was increased by DL-propargylglycine (PPG, an inhibitor of cystathionine gamma-lyase, 200 micromol/L). All these results suggest that the electrophysiological effects of H(2)S on papillary muscles in our study are due to an increase in potassium efflux through the opening of K(ATP) channels and a decrease in calcium influx. Endogenous H(2)S may act as an important regulator in electrophysiological characters in papillary muscles.

9.Hydrogen sulfide facilitates carotid sinus baroreceptor activity in anesthetized male rats.

BACKGROUNDIt has been reported that hydrogen sulfide (H(2)S) could relax vascular smooth muscle by direct activation of K(ATP) channels and hyperpolarization of the membrane potential. Recently, our study has shown that H(2)S facilitated carotid baroreflex. This study was conducted to investigate the effect of H(2)S on carotid baroreceptor activity (CBA).
METHODSThe functional curve of carotid baroreceptor (FCCB) was constructed and the functional parameters of carotid baroreceptor were measured by recording sinus nerve afferent discharge in anesthetized male rats with perfused isolated carotid sinus.
RESULTSH(2)S (derived from NaHS) 25, 50 and 100 micromol/L facilitated CBA, which shifted FCCB to the left and upward. There was a marked increase in peak slope (PS) and peak integral value of carotid sinus nerve charge (PIV) in a concentration-dependent manner. Pretreatment with glibenclamide (20 micromol/L), a K(ATP) channel blocker, the above effects of H(2)S on CBA were abolished. Pretreatment with Bay K8644 (an agonist of calcium channels, 500 nmol/L) eliminated the role of H(2)S on CBA. An inhibitor of cystathionine gamma-lyase (CSE), DL-propargylglycine (PPG, 200 micromol/L) inhibited CBA in male rats and shifted FCCB to the right and downward.
CONCLUSIONSOur results suggest that exogenous H(2)S exerts a facilitatory role on isolated CBA through opening K(ATP) channels and further closing the calcium channels in vascular smooth muscle. Endogenous H(2)S may activate CBA in vivo.

10.Effect of resveratrol on baroreceptor activity of carotid sinus in anesthetized male rats.

This study is to evaluate the effect of resveratrol on carotid baroreceptor activity (CBA). Resveratrol (30, 60 and 120 micromol x L(-1)) inhibited CBA, which shifted FCCB to the right and downward. There was a marked decrease in peak slope (PS) and peak integral value (PIV) of carotid sinus nerve charge in a concentration-dependent manner. Pretreatment with N(omega)-nitro-L-arginine methyl ester (L-NAME, 100 micromol x L(-1)), an inhibitor of nitric oxide synthase (NOS), eliminated the inhibitory effect of resveratrol. Pretreatment with Bay K8644 (an agonist of L-type calcium channel, 500 nmol x L(-1)) abolished the effect of resveratrol on CBA. A potent inhibitor of tyrosine phosphatase (sodium orthovanadate, 1 mmol x L(-1)) did not influence the effect of resveratrol on CBA. Resveratrol inhibits carotid baroreceptor activity, which may be mediated by the locally released NO and decreased calcium influx. Several studies have showed a cardioprotective effect of resveratrol, with the penetrating study of resveratrol, it may show a potential value in the clinical treatment of cardiovascular disease as an alternative medicine.
