Objective To investigate the regulative effect of the extract of Radix Ranunculi Ternati,Radix Sophorae Flavescenti,Prunella vulgaris L.and Stellera chamaejasme L.on cellular immunity induced by multiple drugs resistant bacillus tuberculosis(MDR-TB) from pneumoconiosis patients complicated with tuberculosis in rats.Methods MDR-TB model in rats was induced by MDR-TB.Normal control group were feed by standard feed,model group were irrigated by normal saline,and the other groups were respectively feed by the extract of Chinese herbal medicines.The content of IFN-γ,IL-4,IL-10 and IL-12 were examined by ELISA.RT-PCR was used to detect the mRNA levels of them.Results The content of IFN-γof the four extracts of Chinese herbal medicines were (2.01 ±0.73 ),( 1.92±0.56),( 1.98 ±0.67 ) and (1.94±0.59) pg/ml,IL-4 were (6.01±1.46),(6.12±1.35),(6.47±1.46) and (6.15±1.44) pg/ml,IL-10 were (12.09±3.07),( 12.45±4.01 ),( 12.13±3.43) and (12.54±3.78) pg/ml,IL-12 were (2.99±0.89),(2.75±0.84),(3.02±0.86) and (2.89±0.75) pg/ml.Compared to the model group,they resulted in significant in serum IFN-γ,IL-12,IL-4 and IL-10 (P＜0.05 or P＜0.01 ),the mRNA levels of them were significantly (P＜0.05 or P＜0.01 ).Conclusion The four extracts of Chinese herbal medicines can enhance the cellar immunological function in rats from up-regulation of the level of genetic transcription,accordingly provide the theory base of healing of pneumoconiosis patients complicated with tuberculosis with them.

Objective Primitive neuroectodermal tumor ( PNET) is a small round cell tumor occurring mostly in children or young adults and categorized into the Ewing sarcoma family of tumors, the purpose of the study was to investigate the clinical features, treatment and prognosis of Children PNET. Methods A retrospective study was performed on clinical data of 13 children with PNET hospitalized in our hospital from March 2010 to Octorber 2014.In order to analyze the clinical effects and prognostic results, statistical analysis was made on their clinical manifestations, CT and ultrasonic results, therapeutic schemes, postoperative pathology and immu-nohistochemical staining results, along with telephone and outpaitient follow-up of average 19 months. Results Of the 13 patients, 4 cases were central nervous system PNET(CNS-PNET) and 9 cases were peripheral PNET(pPNET).The clinical manifestations of the former were headache, vomiting, convolsion, movement disorder or decreased muscle strength, while the latter mainly showed mas-ses and abdominal distention.3 cases with metastasis to lung, lymph nodes and bones respectively showed masses on surface or in cavi-ty in imaging.Homer-Wright pseudorosette textures were found in 12 patients.Immunohistochemical results showed 11 cases with CD99(+), 9 cases with VIM(+) and 8 cases Syn( +).Among 11 patients underwent chemotherapy, 2 cases were lost to follow-up, 7 cases were in stable condition without occurrence and the other 2 cases recurred 4-6 months after chemotherapy.3 of 4 cases with CNS-PNET survived with the sequelae of decreased muscle strength, convolsion and movement disorder.1 case with CNS-PNET and 1 case with pPNET died 7 months and 16 months after diagnosis respectively. Conclusion PNET in children is a very highly malignant tumor with low long-term survival rate.Immunohistochemi-cal examination including CD99 and VIM is of important diagnostic value and CNS-PNET is prone to sequelae.

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a promising anti-cancer agent. However, emergence of drug resistance limits its potential use. Plumbagin is a natural quinonoid compound isolated from plant. In this study, induced apoptosis effect of the combined treatment with plumbagin and TRAIL on human melanoma A375 cell line was examined and possible mechanism was investigated. The cells were divided into four groups: control group, plumbagin group (plumbagin, 5 or 10 mumol/L), TRAIL group (TRAIL, 30 ng/mL) and plumbagin+TRAIL group (combined treatment group). The apoptosis, and the expression of DR4 and DR5 were detected by flow cytometry. The activities of caspase-8 and caspase-3 were determined by colorimetric assay. The results showed that the apoptosis rate was 8.3% in TRAIL group, 10.35%-16.94% in plumbagin group and 52.39%-65.39% in combined treatment group, respectively, with the difference being significant between combined treatment group and plumbagin or TRAIL group (P<0.05 for each). Moreover, plumbagin alone could markedly up-regulate DR5 mRNA and protein expression, and slightly increase DR4 mRNA and protein expression. Treatment of human melanoma A375 cells with plumbagin resulted in the activation of Caspase-3, but not Caspase-8. These results suggest that plumbagin might be useful for TRAIL-based treatment for melanoma.

Seventy-four men aged 20-50 years with normal glucose tolerance were divided into abdominal obese group(n=36),simple obese group(n=16),and normal body weight group(n=22).One-hour postload plasma glucose(1hPG)and carotid intima-media thickness(IMT)were higher in abdominal obese group than those in simple obese group and control group(all P<0.01).IMT was positively correlated with 1hPG(P<0.05).In multiple regression analysis,waist circumference and triglycerides were independent predictors for IMT.

OBJECTIVE:To establish a method for simultaneous determination of the contents of Cu,Pb,As,Cr,Cd,Ba, Mn,Sb and Hg in Ganmaoling granule. METHODS:ICP-MS was conducted. The sample was handled by nitric acid microwave di-gestion system,the mass concnetration of 9 metal elements were determined by ICP-MS with Ge,In,Bi as internal standard and standard substance of test element standards. RF power was 1 100 W,sampling depth was 8 mm,carrier gas was argon(Ar)with high purity and flow rate was 1.0 L/min. RESULTS:All 9 elements had good linear range between mass concentration and ion peaks (r=0.999 1-0.999 9),detection limits were in the range of 0.3-6.0μg/L;RSDs of precision,stability and reproducibility tests were no more than 6.0%;average recoveries were in the range of 80.0%-113.5%,and RSDs were in 1.0%-4.5%. CONCLUSIONS:The method is simple,rapid and accurate,and can be used for the determination of 9 metal elements in Ganmaoling granule.

Objective A method for quantitative determination of 16 kinds of organochlorine pesticide (OCPs) in aquatic prod-ucts by gas chromatography (GC)was established .Methods The sample was extracted by acetonitrile ,purified by Carb/NH2 ,and then determined by GC .Results The linear relations of 16 kinds of OCPs were good at 0 .05 - 1 .00 mg/L(r> 0 .998) .The limit of detection of OCPs was in the range of 0 .04 - 0 .31 μg /kg(S/N = 3) .When the standard levels were 50 ,100 ,200 μg /kg ,the recovery rates were 72 .6% - 115 .2% ,the relative standard deviations were 0 .6% - 7 .5% .Conclusion The method established in this stud-y is applied to the determination of 16 kinds of OCPs in real samples with satisfactory results .

Forty-eight men with normal glucose tolerance were divided into fatty liver disease ( NAFLD, n =23 ) and non-NAFLD (n =25 ) groups. The blood glucose excursion was evaluated by continuous glucose monitoring system. The results showed that the mean amplitude of glucose excursion[MAGE, (2. 17± 1.13 vs 1.45±0. 42 )mmol/L]and standard deviation of blood glucose[SDBG, (0. 88 ±0. 45 vs 0. 61 ±0. 21 ) mmol/L]were significantly higher in NAFLD group than in non-NAFLD group( both P＜0. 05 ). MAGE and SDBG were positively correlated with body mass index, waist circumference, and the increased value of plasma glucose 0. 5 h after glucose loading( △G30,all P＜0. 05 ). In multiple regression analysis, △G30, waist circumference, and age were significant independent predictors for MAGE( P＜0. 05 or P＜0. 01 ). △G30 and waist circumference were significant independent predictors for SDBG( P＜0. 05 or P＜0. 01 ).

Objective To explore the differences of the connectivity strength,the clustering coefficient and the local efficiency of the nodes in the brain structural networks in the depression and healthy subjects and then analyze the mode of the local connections of the brain regions and their local efficiency of the transmission of information and their relationship with the severity of the disease in the depression.Methods The Diffusion Tensor Imaging data were obtained from 24 depression patients and 25 healthy controls.The whole cerebral cortex was parcellated into 90 regions by the anatomical label map.Tractography was performed in the whole cerebral cortex of each subject to reconstruct white matter tracts of the brain using the fiber assignment by continuous tracking (FACT) algorithm.And then the brain structural networks were constructed using the complex network theory.The local topological properties of the brain structural networks of the depression and healthy were examined by two sample t-test.Results The local efficiency of the nodes of the networks in depression decreased significantly (the left middle frontal gyrus (orbital part):0.64±0.30,the left hippocampus:0.57±0.07,the right parahippocampal gyrus:0.50±0.15) compared with the healthy (0.88±0.10,0.64±0.06,0.66±0.13 respectively,P=0.00098,0.00039,0.00017,survived critical FDR threshold for multiple comparisons) ; and the clustering coefficient of the nodes of the networks in depression (the left middle frontal gyrus (orbital part):0.14±0.07) decreased significantly when compared with the healthy (0.22±0.06,P=0.000030,survived critical FDR threshold for multiple comparisons).Significant negative correlation was found between the local efficiency of the left middle frontal gyrus (orbital part) and the total scores of HAMD-17 in the depression (r=-0.48,P=0.02).Conclusion The degree of the localization and the local efficiency of the information transmission of the frontal lobe are decreased.The local efficiency in the information transmission in the hippocampal is also decreased.And the local efficiency of the frontal lobe is negatively related with the severity of the disease in the depression.

Objective To explore the differences of the degree and distribution of hub regions of the brain structural networks between the schizophrenia and healthy and then analysis the importance of brain regions in the information transmission in across the whole brain.Methods The diffusion tensor imaging data were obtained from 22 schizophrenia patients and 24 healthy controls.The whole cerebral cortex was parcellated into 90 regions by the anatonical label map.Tractography was performed in the whole cerebral cortex of each subject to reconstruct white matter tracts of the brain using the fiber assignment by continuous tracking (FACT) algorithm.And then the brain structural binary networks were constructed using the complex network theory.The average degree of the network and the degree of the nodes in the network between the brain structural networks of both groups were examined by two sample t-tests.Results The average degree of the brain structural network in the patient group (7.82±0.56) decreased significantly when compared with the control group (8.17 ±0.56; P=0.04).The degree of the nodes in the brain structural network of the patient group (the left hippocampus:11.41 ± 1.84; the left parahippocampal gyrus:6.41± 1.33 ; the left calcarine fissure:11.50±2.97 ; the left fusiform gyrus:8.27± 1.83) decreased significantly when compared with the control group (14.43±2.26; 8.54±2.15; 14.79±2.80; 10.25± 1.36; all P＜0.01,survived critical FDR threshold for multiple comparisons).And the distribution of the hub regions in the temporal and occipital lobes of the patient group was difference from that of the control group.Conclusion The importance of the hippocampus and the brain regions in the occipital lobe is decrease for the information transmission across the whole brain.The damage of the topological properties of these brain regions maybe related to the disorder of the transmission and integration of information in the brain of the schizophrenia.

Objective To explore the differences of the global efficiency of the brain structural networks between the male paranoid schizophrenia and male healthy and its relationship with the psychotic symptoms of the schizophrenia.Methods The diffusion tensor imaging data were obtained from 27 male paranoid schizophrenia patients and 28 male healthy controls.The whole cerebral cortex was parcellated into 90 regions by the anatomical label map.Tractography was performed in the whole cerebral cortex of each subject to reconstruct white matter tracts of the brain using fiber assignment by continuous tracking(FACT) algorithm.And then the brain structural binary networks were constructed using the complex network theory.The average global efficiency of the brain network and the global efficiency of the nodes of both groups were examined by two sample t-test and its relationship with the psychotic symptoms in the male paranoid schizophrenia was explored by the correlation analysis.Results Compared with control group,the average global efficiency of the brain network of the patients decreased significantly (7.87±0.56,8.17±0.56,P=0.005),and the global efficiency of the nodes in the brain network of the patient decreased significantly in the left superior frontal gyrus (orbital part) (P=0.00025),the left superior parietal gyrus (P=0.00011),the left cuneus (P=0.00012) and the left putamen (P=0.00032),all survived FDR correction.Significant negative correlation was found between the global efficiency of the left putamen and the total scores (r=-0.43,P=0.03),the positive scores (r=-0.41,P=0.03) and the cognitive scores (r=-0.40,P=0.04) of PANSS.Conclusion The decreased global efficiency of the left frontal,parietal and occipital lobes and the subcortical structures lead to the occurrence of schizophrenia.And the reduced efficiency of the subcortical structures is associated with the positive symptoms and the abnormal cognitive function of the patients.
