Objective To investigate the role of intestinal fatty acid binding protein (IFABP) in early diagnosis of acute traumatic intestinal rupture. Methods The patients with suspected acute traumatic intestinal rupture admitted in our emergency department from July 2010 to June 2011 were involved in the study.Their blood samples were taken on admission,1,2,3,4,6,8,12,16,24 and 48 hours after admission. All the patients were given closely medical observation and therapy, and were followed up in aspects of their clinical signs and imageology according to the present diagnosis and treatment routine.Surgical procedures would be carried out as soon as the diagnosis of intestinal rupture was confirmed and the duration between the admission and the final diagnosis was recorded. All the blood samples were determined for the IFABP concentration by means of ELISA.According to the final diagnosis results, the patients were divided into the intestinal rupture group and non-intestinal rupture group. The changes of IFABP concentration and its concentration difference between the two groups at different time points were compared. Results The study involved 33 patients, including 11 patients with confirmed intestinal rupture (intestinal rapture group) and 22 without intestinal rupture (non-intestinal rupture group). The average duration from hospitalization to the final diagnosis in the intestinal rapture group was (7.0 ±2.0) hours. At all the given time points, the IFABP concentration in the intestinal rupture group was significantly higher than that in the non-intestinal rupture group (P < 0.05 ).The IFABP concentration in the intestinal rupture group was ascended on admission, reached the peak one hour later and maintained the level till the surgery, while the IFABP concentration was relatively stable in the non-intestinal rupture group within 24 hours after admission. Conclusion IFABP is the index for early diagnosis of acute traumatic intestinal rupture.

Acta Pharmaceutica Sinica 2003;38(2):129-132
AIMTo establish separation methods of five sulfonamides by using capillary high performance liquid chromatography(mu-HPLC) and electrochromatography. The effect of mobile phase varies such as methanol content, pH, buffer solution concentration and voltage on their chromatographic behavior and electroosmesis flow was investigated. Capillary electrochromatography (CEC) was compared with mu-HPLC at the same condition.
METHODSStationary phase was ODS, mobile phase was methanol and 2 mmol.L-1 H3PO4 buffer solution (pH 3.0-7.0), voltage was 0- -15 kV, flow rate was 10 microL.min-1, pressure was approximately 70 MPa and UV detection wavelength was 254 nm.
RESULTSSeparations on base line have been respectively accomplished for five sulfonamides by mu-HPLC with mobile phase of methanol-2 mmol.L-1 H3PO4 buffer solution (30:70) at pH 5.0 in 67 min, and CEC with the same mobile phase at -5 kV voltage in 25 min.
CONCLUSIONElectroosmesis flow of CEC decreased with the increase in methanol content, buffer solution concentration, increased with the increase in voltage and increase slightly with the increase in pH of mobile phase. Retention values (k) of solutes to be examined decreased with increasing methanol content of mobile phase in mu-HPLC and CEC. Retention values (k) of solutes increased slightly with increasing buffer solution concentration, decreased with increasing voltage in CEC. Trimethoprim(TMP) decreased obviously with increasing voltage in CEC. The effect of pH of mobile phase on retention values (k) was more complex. Five sulfonamides were separated at the same mobile phase condition by mu-HPLC and CEC. And separation speed of CEC was much faster than that of mu-HPLC. CEC was very fit for rapid separation of sulfonamides.

By reverse transcription-polymerase chain reaction (RT-PCR), an open reading frame of pathogenesis-related protein 1 (PR1) was isolated from Panax notoginseng and named as PnPR1. Molecular and bioinformatic analyses of PnPR1 revealed that an open reading frame of 501 bp was predicted to encode a 166-amino acid protein with a deduced molecular mass of 18.1 kD. Homology analysis showed that the deduced amino acid sequence of PR1 protein of Panax notoginseng had a high similarity with other higher plants had the same conservative structure domain of cysteine-rich secretory protein (CAP). The recombinant expressed plasmid pET28a(+)-PnPR1 was expressed in Escherichia coli BL21. The expression conditions were optimized by induction at different times, different temperatures, different IPTG concentrations and different giving times. The optimum expression condition was 0.4 mmol.L-1 IPTG at 28 degrees C for 20 h. The successful expression of PnPR1 provides some basis for protein purification and preparation of the monoclonal antibody.

Objective To study the inhibitory effect of verapamil and nicardipine on human scar fibroblast in serum-free culture and to compare the effectness of the two drugs. Methods We used MTT method to detect the effect of two drugs on human scar fibroblast proliferation:adding verapamil and nicardipine with different concentrations in the culture of fibroblasts which were in logarithmic growth phase (150,100,50,10,0μmol/L).After 24,72,and 120 h,we used MTT method to detect the cell proliferation, and converted the absorbance into growth inhibitory ratio. Results Verapamil and nicardipine showed the definite inhibition on the hypertrophic scar fibroblast (HSFB) and keloid fibroblast (KDFB) which were cultured in vitro. There was some difference in the action feature. In the earlier period, the effect of verapamil was powerful than that of nicardipine. With time, the effect did not reinforce. When fibroblast had been cultured for three to five days, the inhibition became weak. But nicardipine showed lasting inhibition on fibroblast proliferation. Conclusion Combination of verapamil with nicardipine may be a valuable method in the treatment of scar.

Journal of Zhejiang University. Medical sciences 2012;41(5):586-591
Recombinant adeno-associated virus (rAAV) has been widely used as vector for gene therapy. However, the effectiveness of gene therapy based on rAAV needs to be further improved. Enhancement of the transduction efficiency is one of the most important fields for rAAV-based gene therapy. Recent results have showed that the ubiquitin-proteasome system plays an important role in the trafficking of rAAV vector in cytoplasm, and regulation of its function may significantly improve the transduction efficiency of rAAV vector in various types of cells and tissues.
