2.A new cyclic diarylheptanoid from the bark of Myrica rubra.

To study the chemical constituents from the bark of Myrica rubra, fourteen compounds were isolated from the methanolic extract using various chromatographic techniques, including silica gel, Sephadex LH-20 and preparative HPLC. Their structures were identified on the basis of chemical properties and spectroscopic data, as 3, 5-dimethoxy-4-hydroxymyricanol (1), myricanol (2), myricanone (3), myricanol 11-sulfate (4), myricitrin (5), quercetin (6), quercetin-3-rhamnoside (7), tamarixol (8), uvaol (9), ursolic acid (10), taraxerol (11), myricadiol (12), β-sitosterol (13) and β-daucosterol (14). Among them, compound 1 is a new compound, named as 3, 5-dimethoxy-4-hydroxymyricanol, compounds 8, 9 were isolated from the genus Myrica for the first time.

3.Effect of Oxcarbazepine on multidrug resistance associated protein 1 expression in hippocampus of kainic acid-induced seizure in rats during development period rats

Objective To observe the expressions of multidrug resistance-associated protein (MRP) 1 in the hippocampus of the rat models with chronic epilepsy,and to explore whether antiepileptic drug oxcarbazepine(OXC) affects the expression of MRP1.Methods One hundred rats of 14 days old were randomly divided into model group(n =52) and the control group(n =48).The model group received intraperitoneal injection of kainic acid(KA) 1 mg/kg (0.5 g/L) to induce seizures,and the control group rats were injected the same dose of 9 g/L chloride.According to Lado standard classification of seizures,the young rats whose seizure degree was beyond 5 and became status epilepticus after intraperitoneal injection used as successful seizure models if they caught the spontaneous seizures after 2 weeks.When spontaneous seizures were developed,48 surviving KA rats were divided into KA group and KA + OXC group.Rats in the control group were divided into NS group and NS + OXC group.After spontaneous recurrent seizures,the treatment groups began to take drugs.Each group started to be sacrificed from the beginning of drug perfusion at different times and they were divided into the 4th week group,the 6th week group and the 8th week group.The expression of MRP1 in hippocampus(CA3 area) was detected by immunohistochemistry methods.Results 1.Epileptic performance:all rats injected with NS had no epileptic performance.In the chronic epilepsy,Ⅰ-Ⅴ Racine grade of spontaneous recurrent seizures were found in all rats with KA.The rats in KA group had higher epilepsy seizure frequency than those in KA + OXC group (P < 0.05).2.The rats in NS group and NS + OXC group had fewer MRP1 positive cells,and there was no significant difference at all time points between NS group and NS + OXC group (P > 0.05).The trend of expression in KA group and KA + OXC group gradually increased.The MRP1 positive cells in KA + OXC group compared with those in the KA group was significantly different in the 8th week(P <0.05),while there was no significant difference in the 4th week and the 6th week(all P >0.05).And there was a significant difference in the positive cells in KA group compared with those of KA group(P <0.05).The expression of MRP1 positive cells in KA + OXC group increased significantly compared with those in NA + OXC group(all P < 0.05).Conclusions MRP1 plays an important role in resistance mechanisms of refractory epilepsy,while the expression of MRP1 can be induced by using OXC in a long term.

5.Evaluation of the liver reserve using lidocaine test on experimental liver injuries in rats.

OBJECTIVESTo study the effectiveness of the lidocaine test in evaluating the liver reserve of rats with experimental liver injury in different phases.
METHODS40 healthy male Wistar rats were divided randomly into experimental and control groups. Rats of the experimental group received subcutaneous CCl4 in oil injection, and rats of the control group received saline injections. Monoethylglycinexylidide (MEGX) test, common hepatic function tests and histological examination of the livers were performed on all the rats.
RESULTSWith the development of the severity in liver injury, the concentrations of the serum MEGX in lidocaine test decreased gradually, which were consistent with liver histological changes. However, the results from the common liver function tests were all abnormal in the experimental group and were not consistent with the liver histological changes.
CONCLUSIONThe results obtained from the MEGX test are more agreeable to liver histological changes than those from common liver function tests. The results from the MEGX test can represent liver histological changes concisely.

6. Clinicopathologic and molecular features of myoepithelial tumors of salivary glands

Objective:
To investigate the clinicopathological, and molecular characteristics of myoepithelial tumors (MTs) of salivary glands.
Methods:
A total of 37 MTs cases including 13 malignant epithelial tumors (MMTs) and 24 benign epithelial tumors (BMTs) of salivary glands were identified from the archives of the Department of Pathology, General Hospital of Eastern Theater Command, dating from 2006 to 2016. Clinical features, histological patterns, immunohistochemical characteristics and status of EWSR1 gene rearrangement by fluorescence in situ hybridization (FISH) analysis were reviewed in all cases.
Results:
Clinically, 37 MTs cases mainly occurred in the parotid glands, when most of the patients presented with painless masses. Of the 13 MMTs cases, male to female ratio was 7∶6, and the median age was 62 years old. Of the 24 BMTs cases, male to female ratio was 5∶7, and the median age was 54 years old. Immunohistochemically, 37 MTs cases were positive for CKpan, and at least one myoepithelial marker. Twenty six of 37 MTs cases were analyzable for the EWSR1 gene break by FISH. Based on the previous evaluation criterion, the EWSR1 translocation was detected in 4 cases of 11 MMTs, and 4 cases of 15 BMTs. According to the main histological composition of tumor cells, 4 EWSR1-positive MMTs covered 2 clear-cell cases and 2 epithelioid-cell cases, when 4 EWSR1-positive BMTs covered 2 clear-cell cases, 1 plasmacytoid-cell case, and 1 spindle-cell case.
Conclusions
Males and females are affected equally. MTs express immunoreactivity for CKpan, and at least one myoepithelial marker. The EWSR1 rearrangement is present in a subset of MTs, with variable morphological characteristics, and has no statistical significance on clinical behavior.
7. Detection of epidermal growth factor receptor gene mutations in different types of non-small cell lung cancer by droplet digital PCR and amplification refractory mutation system

Objective:
To compare amplification refractory mutation system(ARMS) and droplet digital PCR (ddPCR) in the detection of epidermal growth factor receptor (EGFR) gene mutations in patients with non-small cell lung cancer (NSCLC), and to investigate the clinical value of ddPCR.
Methods:
A total of 79 specimens of NSCLC, including 22 cases of cell block, 18 cases of surgical specimens, 12 cases of biopsy specimens and 27 cases of plasma samples, were analyzed for the mutation status of EGFR gene by ARMS and droplet digital PCR method.
Results:
In 18 cases of surgical specimens and 12 cases of biopsy specimens, the detection results by the two methods were identical with positive rates of 9/18 and 5/12, respectively. In 22 cases of effusion cell blocks, ARMS detected 19-del and L858R of EGFR gene in two cases, in which droplet digital PCR detected 19-del+ T790M mutations in one case and L858R+ T790M mutation in another. L858R mutation was detected by droplet digital PCR in one case but ARMS assay was negative. The remaining 19 cases were consistent by the two methods. In blood samples, the positive rate was 33.3%(9/27) by ARMS and 37.0%(10/27) by droplet digital PCR. Two cases showed L858R and 19-del+ T790M mutation by droplet digital PCR but ARMS assay detected only 19-del. The remaining 25 cases were consistent by the two methods.
Conclusion
Droplet digital PCR method is more sensitive and accurate than ARMS for the detection of EGFR mutations in pleural fluid and blood samples, can be used in clinical test.
8.Expression of microRNA-15b in endometrial carcinoma and its effect on proliferation of endometrial carcinoma cells

Objective To investigate the expression of microRNA-15b (miR-15b) in endometrial carcinoma and its effect on proliferation of endometrial carcinoma cells.Methods Eighty-seven cases of endometrial cancer tissues were selected as the research objects from patients with endometrial cancer undergoing endometrial cancer staging surgery in Xinxiang Central Hospital from February 2013 to February 2017.Another 45 cases of normal endometrial tissues from patients with uterine fibroids undergoing line hysterectomy were collected as the control.The expressions of miR-15b in endometrial carcinoma tissues and normal endometrial tissues were detected by real-time quantitative polymerase chain reaction(PCR) and relationship between the miR-15b expression and clinicopathological feature was analyzed.The HEC-1A cells were cultured and were randomly divided into miR-15b mimics group,control sequence group and blank control group.The expression of miR-15b in HEC-1A cells was detected by real-time quantitative PCR.Tetramethylazolazole blue (MTT) assay was used to detect the cell proliferation.Flow cytometry was used to detect the apoptotic rate and cell cycle.Results The relative expression levels of miR-15b in endometrial carcinoma tissues and normal endometrial tissues were 1.32 ±0.13 and 2.49 ±0.16,respectively.The relative expression level of miR-15b in endometrial carcinoma tissues was lower than that in the normal endometrial tissues (t =46.184,P <0.05).The relative expression level of miR-15b in endometrial carcinoma was correlated with International federation of gynecology and obstetrics staging,myometrial invasion and lymph node metastasis (P < 0.05),but it was not correlated with age,histological type,differentiated degree,in filtrating lymph vessel and blood vessel or not.Compared with the blank control group and the control sequence group,the relative expression level of miR-15b in miR-15b mimics group was significantly increased (P < 0.05),the cell proliferation abilities at cultured 24,48,72 and 96 h were decreased (P < 0.05),while the apoptosis rate increased (P <0.05),the proportion of G0 + G1 phase was significantly increased,while the proportion of G2 + M phase was decreased (P <0.05).There was no significant difference in the relative expression level of miR-156,the cell proliferation ability at cultured 12,24,48,72,96 h,the apoptosis rate,the proportion of G0 + G1,S and G2 + M phase between the blank control group and the control sequence group(P <0.05).Conelusion miR-15b is low expression in endometrial carcinoma tissue.Up-regulation of miR-15b expression in endometrial carcinoma cells can inhibit cell proliferation,it may be related to block cell cycle to promote cell apoptosis.
9.Standard-dose of idarubicin in combination with continuous infusion of cytarabine as induction therapy in patients with acute myeloid leukaemia.

The objective of this study was to investigate the efficacy and toxicity of standard-dose idarubicin in combination with continuous infusion of cytarabine as induction therapy in patients with acute myeloid leukemia (AML). A total of 38 AML patients were enrolled, including 30 new diagnosed patients, 8 relapsed and refractory patients. Cytogenetic analysis was performed in all patients, 15 patients had cytogenetic aberrations including 4 complex abnormalities. All patients were treated with standard-dose idarubicin [12 mg/(m(2).d), days 1 to 3] and continuous infusion of cytarabine [100 mg/(m(2).d), days 1 to 7]. The results showed that after one course of induction therapy, the overall response rate was 89.5% (34/38), and 32 out of 38 (84.2%) patients achieved complete remission (CR), including 27 of 30 (90.0%) new diagnosed AML patients, 5 (62.5%) refractory and relapsed AML patients, all 4 patients with complex cytogenetic aberrations achieved cytogenetic CR. Out of 6 relapsed patients 2 showed as extramedullary relapse, 4 showed as bone marrow relapse. The median survival duration was > 22 months and median disease-free survival time was > 16 months. Myelosuppression and infections due to neutropenia were the most frequent adverse effects, severe nonhematologic toxicity and the early death were not observed in the patients. It is concluded that standard-dose of idarubicin combined with continuous infusions of cytarabine as the induction therapy is highly effective and well tolerated approach in patients with AML, this regimen provides an opportune moment for hematopoietic stem cell transplantation.
