3.Expression and potential application of histone deacetylase in prostate cancer

ObjectiveTo investigate the expression and activity of histone deacetylase (HDAC) in prostate cancer.Methodshe pathological samples of 37 cases of PCa were collected. The mean age of the patients was 73 (53 - 88 ) years, the preoperative t-PSA was 81.69 ( 3.13 - 2000 ) ng/ml, Gleason score: 13 cases were ≤7, 24 cases were >7. Twenty-seven cases of BPH were set as controls. The mean age of the BPH patients was 69 (52 - 84) years, the preoperative t-PSA was 10.93 ( 1.11 - 55.07 ) ng/ml.Western blotting and colorimetric Assay kits were used to determine the HDAC expression and activity. The difference of HDAC activity in benign prostatic hyperplasia and prostate cancer was statistically analyzed.The correlation of the HDAC expression level and values of PSA and Gleason score was also assessed.ResultsHDACs were over-expressed in most cases of prostate cancer, the expression rates were HDAC1 :57%, HDAC2: 68%, HDAC3: 84% and HDAC4: 73%, respectively. The HDAC activity (P <0.05)was significantly different between the prostate cancer and benign prostatic hyperplasia groups. The expression level of HDAC did not correlate with the values of PSA and Gleason score.ConclusionsHDAC was highly expressed and strongly active in prostate cancer. The results suggest that HDAC might be a potential target for the management of prostate cancer patients.
4.Levels of 24 hour movement and associations with childhood obesity in Chinese school students

Objective:
To analyze relationships between levels of 24 hour movement (physical activity, screen time and sleep) and childhood obesity in students aged 7-18 years.
Methods:
This study used data from students aged 7-18 years in the 2015 China health and nutrition survey, and assessed levels of physical activity, screen time and sleep based on the Canadian 24 hour Movement Guidelines for Children and Youth. Associations between levels of 24 hour movement and childhood obesity were assessed using multivariable Logistic regression models.
Results:
The overall rate of overweight and obesity was 21.56%. Proportions of students meeting the guidelines for physical activity, screen time, sleep and three behaviors combined were 24.84%, 38.69%, 57.08%, and 6.77%, respectively. After adjusting for age, sex, location of residence and nationality, students meeting the sleep guideline were less likely to be overweight and obese than those who did not(OR=0.73), students meeting both sleep and screen time guidelines had a lower risk in overweight and obesity than those met neither guidelines(OR=0.58)(P<0.05). However, other behaviors and behavior combinations showed no significant associations with overweight and obesity.
Conclusion
Only a small proportion of students met all three 24 hour movement guidelines. Ensuring adequate sleep and limiting excessive screen time may be an important strategy for childhood obesity prevention and management.

8.Study on protective effect of cyproheptadine on non-alcoholic fatty liver disease induced by dexamethasone in rats

Objective To investigate the effects of cyproheptadine on dexamethasone ( DEX ) -induced non -alcoholic fatty liver disease ( NAFLD ) in rats.Methods SD rats were divided randomly into 5 groups: normal group , model group with high , low dosage ( 2.5 , 1.0 mg· kg-1 ) DEX, high, low dosage (5.0 mg · kg -1 ) cyproheptadine treatment groups.Two weeks later after administration , all rats were killed.Body weight and liver weight of rats were measured.Level of serum liver function and lipoid index, glucose (GLU), free fatty acids ( FFA ) , fasting blood insulin ( FINS ) , adrenocorticotropic hormone ( ACTH) and corticosterone ( CORT) were detected.Rat primary hepato-cytes were isolated and treated with different concentration of DEX and cyproheptadine.The fatty droplets in cells were observed by oil red O staining under electron microscope.GLU in culture supernatant , intrace-llular trigly-ceride ( TG) and FFA levels were detected.Results DEX can cause disorders of glucose and lipid metabolism in rats.The level of alanine transarninase (ALT), aspartate aminotransferas (AST), blood fat, fasting blood glucose and insulin were higher than that of normal group.The hypothalamic pituitary adrenal ( HPA) axis was excessive ac-tivated and the level of ACTH and CORT were increased.While cyproheptadine can reduce the reactivity of HPA axis , improve glucose and lipid metabolism in rats.Similar results were obtained in the rat hepatocyte model tests.Conclusion Cyproheptadine can improve NAFLD induced by DEX in rats through the intervention of the HPA axis.
9.Study on impulsivity and cognitive function in patients with manic episodes of bipolar Ⅰ disorder

Objective To explore the role of impulsivity in manic episodes of bipolar I disorder and cognitive impairment. Methods Sixty-one patients with bipolar I manic-episode ( study group) and 43 healthy volunteers (control group) were included in the study,and the cognitive function and impulsivity of the subjects were assessed using the MATRICS consensus cognitive battery (MCCB) and BIS-11 impulsivity scale (BIS-11). The differences in impulsivity and cognitive function between the study group and control group were compared. Partial correlation analysis was used to analyze the correlation between impulsivity and cognitive function in patients with bipolar I manic episode. Results ( 1) The scores of several cognitive function in study group,were significantly lower than those in healthy control group including information pro-cessing speed,attention alertness,word learning,visual learning,working memory,reasoning and problem sol-ving (all P<0. 01). (2)The total score,motor factor score and cognitive factor score of BIS-11 impulse scale in study group were significantly higher than those in control group (58. 39±15. 77 vs 48. 02±11. 16,62. 09± 19. 01 vs 44. 24±21. 09,56. 97±16. 57 vs 50. 06±13. 87,all P<0. 05). Increased overall scores on the bis-11 impulse inventory may be a risk factor for bipolar I episodes( OR=1. 204,95% CI=1. 032-1. 404). (3) In study group,the total score of BIS-11 was negatively correlated with the speed of information processing, working memory,word learning,reasoning and problem solving,and the total score of MCCB(r=-0. 417,-0. 360,-0. 294,-0. 348,-0. 348,P<0. 05). The score of unplanned factor was negatively correlated with the speed of information processing,word learning,the total score of MCCB(r=-0. 397,-0. 302,-0. 358,P<0. 05). The score of cognitive factor was negatively correlated with the speed of information processing,work-ing memory,word learning,reasoning and problem solving,and the total score of MCCB(r=-0. 327,-0. 351,-0. 374,-0. 391,-0. 463,P<0. 05). The score of motor factor was negatively correlated with working memo-ry and the total score of MCCB(r=0. 370,r=0. 389,P<0. 05). Conclusion High impulsivity is a risk factor for manic episodes of bipolar I disorder and may be associated with cognitive impairment in patients with ma-nic episodes of bipolar I disorder.
10.Acute effects of particulate matter on FEV1 and respiratory symptoms of college students in winter.

OBJECTIVETo evaluate the acute effects of indoor and outdoor particulate matter on lung function and respiratory symptoms of college students in winter.
METHODSA panel of 37 college students aged 19-21 in Wuhan were included and the investigation was carried out from 12/23/2009 to 01/05/2010. Daily morning/evening forced expiratory volume in one second (FEV1) and respiratory symptoms (cough, phlegm and runny) were measured and reported by subjects, respectively. Meanwhile, daily data of indoors and outdoors PM10, PM2.5, temperature, and relative humidity were collected. Generalized Estimating Equations (GEEs) were used to estimate the association between particulate matters exposure and respiratory symptoms and FEV1).
RESULTSAverage daily concentrations of indoor, outdoor PM2.5 during the study period were (110.6 ± 42.3), (143.5 ± 51.2) µg/m³, (148.2 ± 43.2) and (239.1 ± 71.3) µg/m³ for indoor, outdoor PM10, respectively. Outdoor exposure to PM2.5 on lag 0 had little effect on evening FEV1). However, researchers observed a significant lagging effect (lag 1 d) and accumulative effect (lag 0-1 d and lag 0-2 d). A rise in the PM2.5 concentration of 10 µg/m³ resulted in the change of evening FEV1) of -0.28% (95% CI: -0.52%, -0.03%), -0.45% (95% CI: -0.81%, -0.08%) and -0.63% (95% CI: -1.15%, -0.11%), respectively. For respiratory symptoms of coughing up phlegm, 10 mg/m³ increase of indoor/outdoor PM2.5 concentration were associated with odds ratio of 1.18 (95% CI: 1.02-1.36) and 1.08 (95% CI: 0.97-1.19), respectively. Moreover, risk of coughing up phlegm increased with lagging time, and accumulative time of indoor and outdoor PM2.5 and PM10.
CONCLUSIONOur study suggested that short-term exposure to particulate matters in winter was significantly associated with acute changes of respiratory symptoms and FEV1) of college students in Wuhan. Moreover, the effects of particulate matters tended to be larger as the increase of lagged and accumulative days.
