1.Influence of beryllium ions on genotype of Porphyromonas gingivalis fimbriae gene cluster

BACKGROUND:In oral warm and moisture circumstance, al oy which contains Be is easily to be eroded to release Be2+. But there is stil no research focusing on beryl ium influence on genotype of Porphyromonas gingivalis fimbriae gene cluster. OBJECTIVE:To investigate Be 2+effect on genotype of Porphyromonas gingivalis fimbriae gene cluster. METHODS:The revived Porphyromonas gingivalis was resuscitated for 48 hours in the anaerobic culture medium with different concentration of Be 2+(10×10-6, 5×10-6, 1.25×10-6). Through PCR amplification and sequencing, we investigated the effects of Be2+RESULTS AND CONCLUSION:(1) When Be on genotypes of Porphyromonas gingivalis fimbriae gene cluster. 2+concentration was 5×10-6, we found the peak of 217 and 257 sites on DNA sequence expressing G/A overlap peak, different from G single peak of the other groups, suggesting the suspicious bases changes, part of the single base G mutated into A. (2) On al concentrations, we found a base group composed of seven A bases was inserted into the 101 site of DNA sequence. Up to now, there is no direct contacts of the mutations occurring to Be2+concentration. Changes of gene may lead to the shifting of the reading frame, the abnormal synthesis of proteins, the change of Porphyromonas gingivalis fimA gene toxicity, and lastly the unbalance of the micro-ecological environment.

3.Correlation between Toxoplasma infection and occurrence of children with cerebral palsy

ObjectiveTo investigate whether the Toxoplasma infection is the risk factor of occurrence of children cerebral palsy.Methods236 children with cerebral palsy and 428 normal children were included. Children were laminated to 4 groups by age:-3 months,-6 months,-9 months,-12 months. Final diagnosis was made by one or more finding following: plasma Toxoplasma DNA(TOX-DNA), plasma Toxoplasma circulating antigen(TOX-CAG), plasma Toxoplasma IgM(TOX-IgM), exclusion other congenital infected diseases.ResultsThe OR and 95%CI were 10.13,3.74-27.42 in -3 months,7.71,2.45-24.28 in -6 months,8.84,1.85-42.31 in-9 months(P<0.01 respectively), and 4.30, 0.83-22.28(P>0.05)in -12 months. ConclusionThere is correlation between the Toxoplasma infection and the children cerebral palsy; the earlier Toxoplasma infection, the greater the risk of cerebral palsy. Congenital Toxoplasma infection may be one of important factors in children cerebral palsy.
4.Progress in Noninvasive Assessment of Liver Fibrosis in Patients with Chronic Hepatitis B

Chronic hepatitis B(CHB)is one of the most commom cause of liver fibrosis. Accurate assessment of liver fibrosis is essential for the strategy of treatment and judgement of prognosis . Liver biopsy is the gold standard for staging fibrosis,but it is invasive with high cost,low reproducibility and poor acceptance by patients. Therefore,it is urgent to explore a noninvasive modality for the assessment of liver fibrosis. Recent evidence highlights that elastographic techniques, biochemical markers and the diagnostic model consisted of several serum markers have the potential for the diagnosis of liver fibrosis. This article reviewed the progress in noninvasive assessment of liver fibrosis in patients with CHB.
5.Effect of Genciclovir on Cytomegalovirus Infection of Children with Cerebral Palsy

ObjectiveTo investigate the therapeutic effect of genciclovir on cytomegalovirus (CMV) infection of children with cerebral palsy (CP).Methods36 CP children with CMV infection were randomly divided into treatment group and control group with 18 cases in each group. Children of treatment group were treated by genciclovir with 5 mg/kg i.v. drip q l2 h for 14 days at inductive phase and 7.5 mg/kg i.v. drip qod for 6 weeks at maintain phase. Cases of control group were treated by acyclovir with 5 mg/kg i.v. drip q 8 h for 14 days at inductive phase and 7.5 mg/kg i.v. drip q l2 h for 6 weeks at maintain phase. CMV-DNA contents in urine of two groups were measured before and after treatment.ResultsCMV-DNA contents in urine of two groups were not different before treatment (P>0.05). For control group, CMV-DNA contents were also not different before and after treatment. For treatment group, CMV-DNA contents after treatment significantly decreased compared with pre-treatment and that of control group after treatment (P<0.05).ConclusionGenciclovir is effect and safety for CMV infection of CP children.

7.Clinical observation on fibrin glue technique in pterygium surgery under surface anesthesia

AIM:To compare the efficiency of fibrin glue to suture technique in pterygium surgery performed with limbal autograft under different methods of anesthesia.
METHODS: A prospective randomised clinical trial was carried out in 60 eyes of 55 patients operated for primary nasal pterygium, which were divided into two groups randomly: experimental group ( 30 eyes in 27 patients ) was under surface anesthesia ( oxybuprocaine ) and control group ( 30 eyes in 28 patients ) was under local anesthesia ( 20g/L lidocaine ). Autologous limbal graft taken from the superotemporal limbus was used to cover the sclera by a fibrin tissue adhesive after pterygium excision. Patients were followed up at least for 6mo. Time of operation, matching degree of graft and VAS score were mainly observed and recorded.
RESULTS: In experimental group the average surgery time was shorter (P=0. 008) and matching degree of graft ( 93%) was better than control group ( 83%) , the differences had statistical significance(P<0.05).
CONCLUSION: The surface anesthesia is enough when using fibrin glue for graft fixation in pterygium surgery, which will shorten surgery time and get better matching degree of graft.
8.New progress on three-dimensional movement measurement analysis of human spine.

Spinal biomechanics, especially the range of spine motion,has close connection with spinal surgery. The change of the range of motion (ROM) is an important indicator of diseases and injuries of spine, and the essential evaluating standards of effect of surgeries and therapies to spine. The analysis of ROM can be dated to the time of the invention of X-ray and even that before it. With the development of science and technology as well as the optimization of various types of calculation methods, diverse measuring methods have emerged, from imaging methods to non-imaging methods, from two-dimensional to three-dimensional, from measuring directly on the X-ray films to calculating automatically by computer. Analysis of ROM has made great progress, but there are some older methods cannot meet the needs of the times and disappear, some classical methods such as X-ray still have vitality. Combining different methods, three dimensions and more vivo spine research are the trend of analysis of ROM. And more and more researchers began to focus on vivo spine research. In this paper, the advantages and disadvantages of the methods utilized recently are presented through viewing recent literatures, providing reference and help for the movement analysis of spine.

9.Identification of Human Lung Cancer-related Epitopes Using Phage display Peptide Library

Objectives: The current study was designed to identify mimic polypeptide epitopes of lung cancer-related antigen WLA-Ag1 through screening random peptide library which was on display in phages, and provide useful information for further study of the characterization of WLA-Ag1 antigen and the potential chance for developing new diagnostic or therapeutic methods for lung cancer. Methods: Through affinity enrichment and immunoscreening of phage-displayed fifteen-peptide libraries with a monoclonal antibody named WLA2C4 against WLA-Ag1, the enriched phages was obtained and plated on agar plates. Some positive clones picked at random were confirmed by ELISA using WLA2C4 monoclonal antibody and DNA sequencing. The homologous comparison of amino acid sequences of positive clones was conducted through searching the protein sequence databank on Internet. The digestion analyses of amino acid sequences were also conducted by a software. Results: Twelve positive clones have been found with a consensual DNA sequence, that is, 5′ -GCT GGT TGG ATT ACT TTT CAT CGT CGT CAT CAT GAT CGT GTT CTT-3′ . Amino acid sequence was AGWITPHRRHHDRVL, with three trypsinase-digestible sites. This coincided with our previous finding that WLA-Ag1 can be digested by trypsinase. Conclusions: The amino acid sequence found in this study might be the epitope of WLA-Ag1. Screening of bioactive peptides from random peptide libraries using monoclonal antibodies as the ligands is an effective method to identify the epitopes of cancer-related antigens.
10.Modeling and experimental study on frequency-domain electricity properties of biological materials.

Frequency-domain electricity properties of four objects, including bullfrog skin, bullfrog muscle, triply distilled water and 0.9% NaCl, were tested in the range of 100Hz-10MHz using home-made electrode and measuring system. The experimental results showed that the resistance of 0.9% NaCl decreased dramatically, that the amplitude frequency characteristics of bullfrog's muscle and skin were similar, but that of triply distilled water did not change significantly. The frequency dependence of 0.9% NaCl showed that the electrode had great influence on the measuring system, so a new equivalent circuit model based on the electrode system was needed. These findings suggest that the new five-parameter equivalent circuit model, which embodies considerations on the interaction between electrodes and tissues, is a reasonable equivalent circuit for studying the electrical characteristics of biological materials.
