1.Sdudy on Subchronic Toxicity of Fipronil in Rats

Objective To study the subchronic toxicity of fipronil in rats and to protect and improve the environment and human health. Methods 80 SD rats were randomly divided into four groups,20 in each groups (10 males and 10 females). The rats were treated with fipronil by gavage,in doses of 25,10,4,0 mg/kg,once a day,10 ml/(kg?d),for 90 consecutive days. The test method was according to GB15670—1995. Results The acute toxicity symptom of rats in high dose showed that cachexia,dull,hair floppy. No acute toxicity symptoms were found in the moderate and low dose groups. No significant difference was found in the serum and urine routine test indexes. Liver index increased significantly in the high dose(25 mg/kg)and moderate dose(10 mg/kg) exposed rats. The activity of aspartateaminotransferase (AST) in the serum increased only in male rats. No significant changes in indicators were found in the low exposed rats. The histopathologic examination showed that in the high and moderate dose groups,in the liver,spot and focus necrosis,the hepatocytes around central veins became bigger and cloudy swelling,some hepatocytes presented ballooning degeneration. No obvious pathological change was seen in the low dose group. Conclusion Fipronil has a toxicity to the liver,based on 90 day oral toxic test in rats,no-observed adverse effect level (NOAEL) is 4 mg/(kg?d).

3.Correlation between upper gastrointestinal adenomas and colorectal neoplasia: a controlled case study

ObjectiveTo evaluate the risk of colorectal neoplasia among patients with gastric or duodenal adenoma.MethodsWe retrospectively reviewed 39 patients with gastric or duodenal adenomas between 2007 and 2011 as the study group.A total of 78 patients without upper gastrointestinal adenomas who underwent colonoscopy were included as a control group.Colonoscopic findings were compared between the two groups.ResultsPositive finding rate of colorectal neoplasia in study group was 51.3% (20/39),with adenomas in 38.5% (15/39) and cancer in 12.8% (5/39),which were all significantly higher than those of the control group,14.1% (11/78) ( P < 0.001 ),12.8% ( 10/78 ) ( P < 0.05 ) and 1.3% ( 1/78 ) ( P < 0.05),respectively.The results were similar when upper digestive tract was further divided into stomach and duodenum(50.0% vs.12.5% and 52.2% vs.15.2% ).ConclusionPatients with gastric or duodenal adenomas are at a significantly higher risk for colorectal neoplasia.Colonoscopy should be recommended for these patients.
4.Clinical information analysis of neurological emergency:a report of 1979 cases

For this cross-sectional study , the clinical data of 1979 patients at our emergency department from October 2012 to June 2013 were collected and analyzed.The relevant data included gender , age,visit time,complaints,signs and diagnosis.The ratio of female:male was 1 to 0.71.The age groups of 50-59 and 70-79 years were the most.The peak of visit time was 8-10 am accounting for 21.26%.The common complaints included dizziness ,headache and paralysis.The most frequent emergency diseases were acute cerebral vascular accident , dizziness or vertigo due to various diseases , headache and peripheral neuropathy.Ischemis stroke was the top one disease in elders.Conclusion The middle-aged and elderly are the main populations at our emergency department.And cerebrovascular disease is a common disease.

Objective To investigate the in vitro effect of wild-type PTEN gene transfection on the growth of human colon cancer cell lines. Methods With liposome transduction technique,the retrovirus vector pBp-PTEN,which contains wild-type PTEN gene segment and pBabe-puro,was transfected into Lovo,one of the PTEN-deficient human colorectal carcinoma cell lines.After identification by Western blotting,cell growth,cell cycle and apoptosis before and after transfection were studied. 6.Screening of DOC-2 interactive proteins and identification of the interaction between DOC-2 and TGF?Ⅲ receptor

Background and purpose:DOC-2 serves as one of the tumor suppressing genes in human ovarian cancer and plays a role in the process of cell growth and differentiation.This study was to investigate the role of DOC-2 in the TGF? signal pathway and verification of the interaction between DOC-2 and TGF?Ⅲ receptor.Methods:The bait vector was constructed by inserting the PID domain of DOC-2(nDOC-2)into yeast express vector pGBKT7.pGBKT7-nDOC2 was transformed into the yeast AH109 and confirmed to be expressed.After the human foetus brain cDNA library had been transformed,the positive clones was screened by both nutrition defect medium and X-?-gal.The putative positive clones were sequenced and analyzed to get the DOC-2 interactive proteins.Furthermore,after the DOC-2 cDNA and TGF?Ⅲ receptor cDNA had been co-transfected into the human ovarian cancer cell line HO-8910 together,the interaction between DOC-2 and TGF?Ⅲ receptor was investigated by immunoprecipitation and Western blot.Results:21 putative positive clones were picked after being screened and sequenced.Three of them were identified as Homo sapiens partial mRNA for betaglycan(TBR Ⅲ gene),homo sapiens protocadherin gamma subfamily C3(PCDHGC3)and APLP1(amyloid beta precursor-like protein 1).The analysis by immunoprecipitation and Western blot showed that the interaction between DOC-2 and TGF?Ⅲ receptor could form protein complex.Conclusions:The three encoding proteins might participate in the DOC-2 signal pathway.DOC-2 might play as an essential role in the TGF?signal pathway by interacting with TGF?Ⅲ receptor.

8.Effects of Hedan Tablets on the Levels of Blood Lipid,Inflammatory Factors and Oxidative Stress in Coro-nary Heart Disease Patients with Type 2 Diabetes Mellitus

OBJECTIVE:To explore the effects and safety of Hedan tablets on the levels of blood lipid,inflammatory factors and oxidative stress in coronary heart disease(CHD)patients with type 2 diabetes mellitus(DM2). METHODS:A total of 118 pa-tients diagnosed as CHD with DM2 were collected from our hospital during Jul. 2013-Nov. 2015,and then divided into observation group(63 cases)and control group(55 cases)according to random number table. Control group received conventional treatment such as controlling blood glucose,regulating blood lipid,vasodilator,anticoagulant. Observation group additionally received He-dan tablets 1.46 g before meal,tid,on the basis of control group. Both groups received treatment for 6 months. The levels of blood lipid (TC,TG,LDL-C,HDL-C),serum inflammatory factors (TNF-α ,IL-10,IL-6,hs-CRP) and oxidative stress indexes (MDA,SOD,NO)before and after treatment as well as the occurrence of ADR were compared in 2 groups. RESULTS:Before treatment,there was no statistical significance in levels of blood lipid,serum inflammatory factors or oxidative stress indexes be-tween 2 groups(P>0.05). After treatment,the serum levels of TC,TG,LDL-C,TNF-α,IL-6,hs-CRP and MDA in 2 groups were decreased significantly,while those of IL-10,SOD and NO were increased significantly;the observation group was signifi-cantly better than the control group,with statistical significance(P<0.05). There was no statistical significance in the incidence of ADR between 2 groups(P>0.05). CONCLUSIONS:Hedan tablets can effectively improve the level of blood lipid,reduce the lev-el of inflammatory factors,relieve inflammatory injury,reduce the level of oxygen free radical,strengthen antioxidant capacity and lighten oxidative stress injury with good safety.
9.Preparation and preservation of hypoxia UW solution.

In order to explore the method to prepare hypoxia UW solution and the stability and preservation of hypoxia UW solution, UW solution was purged by argon or air for 15 min or 60 at a flow rate of 0.8 or 2 L/min, and the oxygen partial pressure of UW solution was detected. The hypoxia UW solution was exposed to the air or sealed up to preserve by using different methods, and the changes of oxygen partial pressure was tested. The results showed that oxygen partial presure of 50 mL UW solution, purged by argon for 15 min at a flow rate of 2 L/min, was declined from 242+/-6 mmHg to 83+/-10 mmHg. After exposure to the air, oxygen partial pressure of hypoxia UW solution was gradually increased to 160+/-7 mmHg at 48 h. After sealed up by the centrifuge tube and plastic bad filled with argon, oxygen partial pressure of hypoxia UW solution was stable, about 88+/-13 mmHg at 72 h. It was concluded that oxygen of UW solution could be purged by argon efficiently. Sealed up by the centrifuge tube and plastic bag filled with argon, oxygen partial pressure of UW solution could be stabilized.
