Objective To approach curative effect of using DLI +IL-2 as immunobiotherapy after Mixed-HSCT in acute myelogenous leukemia.MethodAfter times of chemotherapy,8 cases of patients with acute myelogenous leukemia received Mixed-HSCT,then were treated with DLI +IL-2 for 2-7 times.Observed clinical effect for 1 to 5 years.Result DFS in 8 cases of patients with acute myelogenous leukemia received Mixed-HSCT and treated with DLI +IL-2 for 2-7 times were 62.5%.There were no GVHD.Conclusion Immuno-biotherapy with DLI +IL-2 after Mixed-HSCT in patients of acute myelogenous leukemia may be a method to increase DFS efficiently.

3.Effects of paeoniflorin on the proliferation of bone marrow mesenchymal stem cells

BACKGROUND:Studies have shown that paeoniflorin functions as replenishing blood and treatment of autoimmune diseases, and bone marrow mesenchymal stem cels also play an important role in the body’s blood and immune function. However, paeoniflorin effects on bone marrow mesenchymal stem cel proliferation and cytokine secretion and expression are rarely reported. OBJECTIVE:To investigate the effect of paeoniflorin on proliferation of bone marrow mesenchymal stem cels and the expression of interleukin-6. METHODS:Human bone marrow mesenchymal stem cels were separated and culturedin vitro by density gradient centrifugation combined with attachment method. The biological characteristics of bone marrow mesenchymal stem cels were identified by flow cytometry and osteogenic/adipogenic induction. The proliferation of bone marrow mesenchymal stem cels under different concentrations of paeoniflorin was detected by MTT method. The mRNA expression and secretion of interleukin-6 in the supernatant of bone marrow mesenchymal stem cels were detected by RT-PCR and ELISA, respectively. RESULTS AND CONCLUSION: Bone marrow mesenchymal stem cels were isolated successfuly and had osteogenic and adipogenic differentiation potential. Compared with the controlgroup, 2 μmol/L and 10 μmol/L paeoniflorin significantly promoted the proliferation of bone marrow mesenchymal stem cels. 10 μmol/L paeoniflorin could significantly decrease the proportion of bone marrow mesenchymal stem cels in G0/G1 phase and increase this proportion in S phase. Compared with the control group, the experimental group could significantly increase the secretion and mRNA expression of interleukin-6 (P < 0.01). It is concluded that paeoniflorin at certain concentrations can obviously promote the proliferation of bone marrow mesenchymal stem cels, and increase the expression and secretion of interleukin-6.
4.The clinical value of 18 F-FDG PET/CT for the patients with ascites of unknown causes

Objective To assess the clinical value of 18 F-FDG PET/CT for the patients with ascites of unknown causes.Methods A total of 106 patients with ascites undergoing 18 F-FDG PET/CT examination was retrospectively studied and 18F-FDG PET/CT imaging was interpreted by two ways.(1)PET/CT(localization):Diagnostic results of PET/CT depended on the degree of 18 F-FDG uptake in peritoneum with CT was trecoted as anatomical localization,and high 18F-FDG uptake in peritoneum was regarded as malignancy.(2)PET/CT(mor-phology):Morphological changes of peritoneum on CT imaging and the degree of 18 F-FDG uptake on PET ima-ging were simultaneously considered when interpreting the diagnostic results of PET/CT.Results In the differen-tial diagnosis of ascites of unknown cause,the sensitivity,specificity and accuracy of PET/CT( localization) were 90.2%,71.1%and 82.1%,respectively.These values of PET/CT(morphology)were 95.1%,93.3% and 94. 3%,and 85.2%of malignant ascites were found primary lesion.The specificity and accuracy of PET/CT( mor-phology)were higher than those of PET/CT(localization)(P=0.002,P<0.001).Statistical difference was not found in both sensitivities.Conclusion PET/CT( morphology) had higher clinical value than PET/CT( localiza-tion) in the differential diagnosis of ascites with unknown cause,and had important significance in detecting pri-mary cause of malignant ascites.
5.Valsartan Inhibits Myocardial Apoptosis by Down-regulating Myocardial X-box Binding Protein 1 Expression in Experimental Diabetic Cardiomyopathy Rat’s Model

Objective: To study the relationship between myocardial X-box binding protein 1 (XBP1) expression and myocardial apoptosis in experimental diabetic cardiomyopathy (DCM) rat’s model and to clarify the mechanism of valsartan inhibiting myocardial apoptosis. Methods: A total of 50 Wistar rats were divided into 2 groups: Control group, the rats received intraperitoneal citrate buffer at 65mg/kg,n=10 and Streptozotocin group, the rats received intraperitoneal streptozotocin at 65mg/kg,n=40, all animals were treated for 7 days. DCM model was established in 37 rats (fasting blood glucose ≥ 16.7mmole/L) and they were further divided into 2 groups: DCM group, the rats received intragastric normal saline,n=20 and DCM + valsartan group, the rats received intragastric valsartan at 30mg/kg·day,n=17. The rats were treated for 16 weeks. The body weight, tail blood pressure, glucose and cardiac function were compared among 3 groups. Myocardial apoptosis was detected by TUNEL staining, RNA and protein expressions of myocardial cytochrome C, cleaved caspase 3, glucose regulation protein 78 (GRP78) and XBP1-s were examined by immunolfuorescence, real time RT-PCR and Western blot analysis. Results: Compared with Control group, DCM group showed disordered cardiac structure, more collagen content and myocardial apoptosis,P<0.05; increased RNA and protein expressions of GRP78, XBP1-s, cleaved caspase 3 and cytochrome C,P<0.05. Compared with DCM group, DCM + valsartan group had rather regularly arranged myocardiocytes, less interstitial ifbrosis and myocardial apoptosis,P<0.05; decreased RNA and protein expressions of GRP78, XBP1-s, cleaved caspase 3 and cytochrome C,P<0.05. Conclusion: Valsartan may inhibit myocardial XBP1 activation and therefore, reduce the myocardial apoptosis in experimental DCM rat’s model.
6.A noninvasive diagnostic model of liver fibrosis using serum markers in primary biliary cirrhosis

Objective To verify and assess diagnostic value of noninvasive diagnostic model of liver fibrosis in primary biliary cirrhosis (PBC) based on conventional laboratory markers.Methods Seventythree patients with PBC diagnosed by liver biopsy between January 2003 and June 2011 in Beijing Friendship Hospital,Capital Medical University were recruited in this study.Correlation analysis and logistic regression analysis between the conventional laboratory markers and histology stages were assessed.A liver fibrosis diagnostic model was established based upon aforementioned biomarkers and verified by its sensitivity and specificity for predicting the liver fibrosis.Results The predictive model ( H index) consisting of five conventional laboratory markers,i.e.,platelet count,serum cholinesterase,albumin,HDL-C and prothrombin time activity,could predict advanced fibrosis ( stages Ⅲ-Ⅳ ) with an AUCROC of 0.861.The sensitivity of predicting the absence of advanced fibrosis using H index < - 2.20 was 96.6% and the specificity of predicting the presence of advanced fibrosis using H index > 0.41 was 93.2%.Conclusion The established noninvasive diagnostic model consisting of five laboratory markers could accurately distinguish pathological changes of early stage PBC ( stages Ⅰ - Ⅱ ) from advanced stage PBC ( stages Ⅲ-Ⅳ).
7.Effects of interferon-γ on expression of adhesion molecules in human umbilical cord mesenchymal stromal cells.

This study was purposed to investigate the effects of interferon (IFN)-γ on expression of adhesion molecules in mesenchymal stromal cells derived from human umbilical cord tissue (UC-MSC). The UC-MSC were isolated from human umbilical cord by tissue culture. The expressions of specific markers on UC-MSC were detected by flow cytometry in the physiological condition. The adipogenic and osteogenic induction of UC-MSC was detected by alizarin and Oil red O staining. UC-MSC were exposed to IFN-γ (100, 1 000, 10 000 U/ml) for 24 h, the expressions of CD54, CD58, CD44, CD49d, CD62p, CD62L, CD102 and CD106 on cell surface were detected using flow cytometry. The results showed that in physiological condition, UC-MSC extremely low expressed CD102, CD106, CD62P, CD62L, while the expression of CD54 was relatively high (41.58 ± 0.83)%. When stimulated by IFN-γ, the expression of CD102, CD106, CD62P, CD62L increased slightly, but still low (< 5%), while CD54 and CD58 upregulated concentration-dependently up to (59.66 ± 1.36)% and (43.96 ± 0.62)% respectively. The expression of CD49d upregulated to (51.33 ± 0.74)% when UC-MSC exposed to IFN-γ 100 U/ml. CD44 increased to (73.22 ± 1.93)% when UC-MSC exposed to IFN-γ 1 000 U/ml. It is concluded that IFN-γ can elevate significantly the expression of CD54, CD49d, CD44 and CD58, but has no significant effect on CD102, CD106, CD62P and CD62L expression on the surface of UC-MSC.

8.Effect of M8046 on expression of COX-2/PGE2 in spinal cord and DRG in rats with neuropathic pain.

OBJECTIVETo investigate the effects of glucocorticoid receptor antagonist-M8046 on the behavior and the cyclooxygenase-2/prostaglandin E2( COX-2/PGE2) expression in spinal cord dorsal horn and dorsal root ganglia (DRG) in chronic constrictive injury (CCI) rats.
METHODSOne hundred and forty-four male SD rats were randomly divided into 4 groups, 36 rats in each group: Sham operation group (Sham), chronic constrictive group (CCI), M8046 treated group (M8046) and solvent controlled group (Sc). M8046 3 mg/(kg x d) intraperitoneal injection was given after operation in group M8046. Paw thennal withdrawal (PTWL) and paw mechanical withdrawal threshold (PMWT) of rats were measured on 2 pre-operative and 1, 3, 7, 10, 14 post-operative days. The spinal cord and L15 DRG of the operated side was removed at 3, 7, 14 days after surgery. The change of COX-2 and PGE2 expression was determined by immunohistochemical staining and ELISA separately.
RESULTSPTWL and PMWT in CCI group were significantly lower than those in Sham group on every post-operative day (P < 0.05). PTWL and PMWT in M8046 group were significantly higher than those in CCI group on 7, 10, 14 post-operative day (P < 0.05). In spinal dorsal horn, the level of COX-2 and PGE2 expression in CCI group was significantly higher than that in Sham group (P < 0.05). M8046 could significantly attenuate the activation of COX-2 and PGE2 induced by CCI (P < 0.05). The expression of COX-2 and PGE2 in DRG was similar to that in spinal dorsal horn.
CONCLUSIONThe effects of M8046 ameliorate the CCI-induced neuropathic pain may be related to attenuate the expression of COX-2 and PGE2 in spinal cord and DRG.

10.Diagnosis and treatment of penile verrucous carcinoma: a report of 4 cases.

OBJECTIVETo explore the diagnosis and treatment of penile verrucous carcinoma.
METHODSThe clinical and pathological data of 4 patients with penile verrucous carcinoma were analyzed.
RESULTSThe patients ranged in age from 42 to 76 years (average 52). All the tumors showed exophytic papillary lesions, the biggest being 2.1 to 5.8 cm in diameter. The lesions were confined to the glans penis in two cases and invaded the shafts in the other 2 (1 accompanied by syphilis). One patient, whose tumor was small (1.4 cm in diameter) and confined to the glans penis, underwent glandectomy. One with a larger tumor confined to the glans penis and the other 2 with the shafts involved underwent partial penectomy, including the one accompanied by syphilis, who underwent the operation after treated by Benzathine benzylpenicillin. Histopathological examination of the specimens showed that the tumor cells were mostly well-differentiated and the surgical margins were tumor free in all the 4 cases. HE stain was performed in all the specimens. Microscopic examination revealed papillomatosis and hyperkeratosis of the epithelium, with bulbous projections into the lamina propria consisting of well-differentiated squamous epithelial cells. Marked invasion of the stroma by lymphocytes was noted. Follow-up ranged from 3 to 7 years (average 4.6), revealing no recurrence. The result of the rapid plasma regain (RPR) test was negative but that of the Treponema pallidum passive-particle-agglutination (TPPA) test remained positive in the blood of the patient accompanied by syphilis after treatment.
CONCLUSIONVerrucous carcinoma of the penis is characterized by low malignant potential and locally aggressive nature. It seldom develops metastasis to regional lymphonodes or distant areas. Glandectomy or partial penectomy can be chosen for its treatment, with favorable prognosis.
