1.Synergistic lethal effects of cetuximab combined with chemotherapy and/or radiotherapy in laryngeal squamous carcinoma cells.

OBJECTIVE:
To determine the sensitivity of cetuximab induced apoptosis in laryngeal squamous carcinoma cells Hep-2, and to evaluate the synergistic killing effects and regulation mechanism of cetuximab alone or cetuximab in combination with chemotherapeutic agents (cisplatin) or radiation means on Hep-2 cells.
METHOD:
To investigate the cytotoxicities of cetuximab, cisplatin and radiation, cell counting kit-8 (CCK-8) assay was used for the detection of cell growth inhibition ratio, and fluorescence activated cell sorter FACS for the apoptotic rate and cell cycle distribution.
RESULT:
Cetuximab had inhibitive effect on Hep-2 cells within a certain range of concentration in a time- and dose-dependence manner. The inhibition concentration 50% (IC50) of cetuximab on Hep-2 cells for 24 h was 1 036.84 microg/ml. For application of cisplatin and radiation, the apoptotic rate of Hep-2 cell was higher by combining with cetuximab than their single or combined administration. Moreover, the cell cycle arrested at G0/G1 phase.
CONCLUSION
Laryngeal cancer Hep-2 cells was sensitive to the cetuximab induced apoptosis. Cetuximab combined with cisplatin and/or radiation can increase the antiproliferative effects on Hep-2 cells. These findings suggest the synergistic combination of cetuximab and cytotoxic agents was sequence depended.

2.Biocompatibility of chitosan carrier with rabbit corneal endothelium

Background There are a lot of studies about the carrier of corneal endothelial transplantation,but the best carrier has not been defined.Objective This study was to investigate the biocompatibility of chitosan carrier with rabbit corneal endothelium in vivo.Methods Fresh eye-balls were obtained from 10 New Zealand white rabbits.Rabbit corneal endothelial cells (CECs) were isolated and cultured on chitosan carrier in vitro.The morphology and density of rabbits CECs were observed every day,and the expressions of fibronectin (FN),collagen-1 (Coil-I) and Zonula occludens 1 (ZO-1) were detected by immunoinfluorescence.The morphology and ultrastructure of CECs were observed under the scanning and transmission electron microscope.Chitosan carrier with CECs was implanted into the anterior chamber of the left eyes in ten healthy New Zealand white rabbits,and only paracentesis of anterior chamber was performed in the right eyes as controls.The inflammation of ocular anterior segment was examined under the slit lamp microscope,and corneal thickness was measured 1 week,4 and 8 weeks after operation.Corneal endothelium cell density and morphology were examined under the corneal endothelial microscope at postoperative 2 weeks.Corneal samples were collected for the regular histopathological examination to observe the inflammatory reaction at postoperative 1 month and 3 months.Paired t test was used for statistical analyses between the control group (left eyes) and the experimental group (right eyes).The use and care of the animals followed the Statement of ARVO.Results CECs formed an intact monolayer of cells with the uniform shape and size on the chitosan membrane after incubated for 5 days.The cells reached confluence of 90% 7 days after cultured with the 40% hexagon cells.Under the scanning electron nicroscope,rabbit CECs showed the round or polygon in the shape with the microvillus on the cell surface.The cells connected closely by desmosome.The processes,pseudopodiums and microvillus on the cellular surface,vacuole in the cytoplasm,expanded endoplasmic reticulum with ribosome and abundant chromatin were exhibited under the transmission electron microscope.The immunofluorescence examination revealed the positive expressions of FN,Coll-Ⅰ and ZO-1 in the CECs on the chitosan carrier.In the in vivo experiment,the exudation in the anterior chamber and corneal edema were seen under the slit lamp microscope 3 days after implantation of chitosan carrier with CECs.However,the inflammation was gone 14 days after operation.The differences of the corneal thickness were no significant between the experimental group and the control group 1 week and 4,8 weeks after operation (t =1.377,P=0.265;t =1.795,P=0.165 ; t =0.390,P =0.760).In addition,no significant differences were found in the CECs density and the hexagon cells rate between the two groups(P =0.365,0.062).The histopathological examination showed that the inflammatory cells around the chitosan membrane were disappeared 3 months after operation and showed a good corneal structure.Conclusions Chitosan carrier has a good biocompatibility with rabbit CECs and anterior chamber,and it may be a potentially good carrier for CECs transplantation.
3.Relationship between hypertriglyceridemic-waist phenotype and clustering of cardiovascular risk factors in adolescents

Objective The aim of the study was to explore the relationship between hypertriglyceridemic-waist(HTWC) phenotype and clustering of cardiovascular risk factors in adolescents. Methods A cross-sectional study was conducted in 3 136 Han adolescents aged 13-17 years in Qinhuangdao. The HTWC phenotype was defined as serum triglyceride concentrations ≥1. 47 mmol/ L and waist circumference ≥90 th percentile for age and sex. Results The prevalence of HTWC phenotype was 3. 3% . The prevalence of clustering of cardiovascular risk factors was 0. 4%in adolescents with normal waist circumference and serum triglyceride and 14. 7% in adolescents with HTWC phenotype. After adjustment for sex, age, body mass index, and family history of diabetes and hypertension, the prevalence of clustering of cardiovascular risk factors in adolescents with HTWC phenotype was 9. 286 fold(95% CI 3. 161-27. 282, P<0. 01) of that in adolescents with normal waist circumference and triglyceride. Conclusion There was a significant correlation between HTWC phenotype and clustering of cardiovascular risk factors, suggesting that this phenotype may be applied as a simple marker for identifying adolescents with clustering of cardiovascular risk factors.
4.Long-pulse Nd:YAG 1064-nm laser treatment for onychomycosis.

BACKGROUNDRecent research shows that lasers can inhibit fungal growth and that Nd:YAG 1064-nm lasers can penetrate as deep as the lower nail plate. The aim of this study was to observe the effect of a long-pulse Nd:YAG 1064-nm laser on 154 nails of 33 patients with clinically and mycologically proven onychomycosis.
METHODSThirty-three patients with 154 nails affected by onychomycosis were randomly assigned to two groups, with the 154 nails divided into three sub-groups (II degree, III degree, and IV degree) according to the Scoring Clinical Index of Onychomycosis. The 15 patients (78 nails) in group 1 were given eight sessions with a one-week interval, and the 18 patients (76 nails) in group 2 were given four sessions with a one-week interval.
RESULTSIn group 1, the effective rates at 8 weeks, 16 weeks, and 24 weeks were 63%, 62%, and 51%, respectively, and the effective rates in group 2 were 68%, 67%, and 53% respectively. The treatment effect was not significantly different between any sub-group pair (P > 0.05).
CONCLUSIONSLong pulse Nd:YAG 1064-nm laser was effective for onychomycosis. It is a simple and effective method without significant complications or side effects and is expected to become an alternative or replacement therapy for onychomycosis.

6.Mediating effect of achievement motivation between perfectionism and procrastination behavior of hematology nurses

Objective:To explore the mediating effect of achievement motivation between perfectionism and procrastination behavior of hematology nurses.Methods:Using the stratified sampling method, a total of 280 hematology nurses from 14 Class Ⅲ Grade A hospitals in Henan Province were selected as the research objects from April 2022 to April 2023. A survey was conducted on hematology nurses using general information questionnaire, the Achievement Motive Scale (AMS), the Chinese Frost Multidimensional Perfectionism Scale (CFMPS) and General Procrastination Scale (GPS). Pearson correlation analysis was used to test the correlation between achievement motivation, perfectionism and procrastination behavior. AMOS 26.0 software was used to establish a structural equation model and verify the mediating effect.Results:A total of 280 questionnaires were distributed in this study, and 271 valid questionnaires were collected, with an effective response rate of 96.79%. Among 271 hematology nurses with AMS, the score for pursuing success was (51.95±10.38), and the score for avoiding failure was (53.11±8.91). The total score of CFMPS was (83.79±17.18) and the total GPS score was (61.71±9.50). Perfectionism was negatively correlated with procrastination behavior ( P<0.01), perfectionism was positively correlated with the pursuit of success ( P<0.01), perfectionism was negatively correlated with avoidance failure ( P<0.01), procrastination behavior was negatively correlated with the pursuit of success ( P<0.01) and procrastination behavior was positively correlated with avoidance failure ( P<0.01). Achievement motivation played a partial mediating role between perfectionism and procrastination behavior, with the mediating effect accounting for 47.89% of the total effect. Conclusions:The procrastination behavior and perfectionism of hematology nurses are both in the medium level. Both the pursuit of success and avoidance of failure have partial mediating effects between perfectionism and procrastination. From the perspective of behavioral motivation, strengthening the education and guidance of hematology nurses can avoid procrastination behavior, which has certain guiding significance for the career planning of hematology nurses.
7.A case of systemic amyloidosis beginning with purpura.

Primary systemic amyloidosis is a relatively rare disease, caused when abnormal extracellular deposition of fibrillary protein builds up in a variety of target organs, such as heart, kidneys, lungs liver, and so forth. The symptoms of the disease are usually vague, while many kinds of auxiliary or laboratory examinations especially pathologic biopsy can provide a clue for the diagnosis. Here we described a case who had purpura-like lesions in the initial stage, followed by progressive malfunctions in the kidneys, the heart, the lungs, as well as the liver. The final diagnosis was primary systemic amyloidosis determined by skin pathologic biopsy. And the disease led to a fatal outcome within three months after the diagnosis.

8.Effects of Yikunning on the apoptotic rate and expression of caspase-3 in rat ovaries during perimenopausal period.

OBJECTIVETo investigate the effects of Yikunning (compound of Chinese traditional Medicine, YKN) on the apoptotic rate and expression of caspase-3 in rat ovaries during perimenopausal period.
METHODSThirty female Wistar rats during perimenopausal period were selected by unforced aging. Then the rats were divided into 3 groups randomly: YKN group, livial control group and aged control group. Ten young female rats were selected as young control group. Intragastric administrations were conducted for 4 weeks once daily continuously. The apoptotic rate in rat ovaries were detected by TUNEL. The expression of caspase-3 mRNA and protein in rat ovaries were detected by RT-PCR and Western blot, respectively.
RESULTSThe apoptotic rate in rat ovaries in YKN group was lower than that in aged control group, which showed difference between them (P < 0.01). The levels of caspase-3 mRNA and protein in rat ovaries in YKN group were lower than those in aged control group, which showed differences among them (P < 0.01).
CONCLUSIONYKN can decrease the apoptotic rate and down-regulate the expression of caspase-3 mRNA and protein in rat ovaries of during perimenopausal period. It may be one of the molecular mechanisms of YKN postponed the ovarian failure and cured perimenopausal syndrome.

10.Healthcare-associated infection and pathogen spectrum in neonatal inten-sive care unit:a 5-year single-center study

Objective To understand the spectrum and changes of pathogens causing healthcare-associated infec-tion(HAI)in neonatal intensive care unit(NICU).Methods Clinical medical records of neonates with HAI in a hospital from January 2018 to December 2022 were collected,spectrum of pathogens causing HAI were and analyzed retrospectively.Results A total of 7 597 hospitalized neonates were investigated,and 240 of whom had 263 cases of HAI,with an HAI incidence of 3.16%and healthcare-associated case infection incidence of 3.46%.96 cases(36.50%)were bloodstream infection,70(26.62%)were respiratory system infection,and 57(21.67%)were in-fection without clear sites.A total of 170 pathogens were detected from specimens,78(45.88%)of which were Gram-positive bacteria,with Staphylococcus spp.accounting for the highest proportion,78(45.88%)were Gram-negative bacteria,mainly Klebsiella pneumoniae,and 14(8.24%)were fungi.The detection rate of Gram-negative bacteria showed an upward trend from 2018 to 2022(P<0.01).Conclusion The majority of HAI in NICU is bloodstream infection.In recent years,the detection rate of Gram-negative bacteria has been increasing year by year,and it is necessary to streng-then the prevention and control of HAI in clinical practice.
