1.Influence of Nbn gene on the development of granular cells in hippocampus of neonatal mouse

Objective To investigate the influence of neuro-specific DNA repair gene Nbn on the development of granular cells in hippocampus of monatal mouse. Methods The hippocampus development of postnatal mouse was evaluated by comparing the morphological differences of granular cells in hippocampus between the Nbn gene knockout mice (Nbn-CNS-Del) and the controls (Nbn-CNS-Ctr). Serial sections, immunohistochemistry and stereology methods were used to quantitatively analyze the development of granular cells in hippocampus of mice on the postnatal day 7, 14 and 21 (P7d, P14d and P21d). Results In Nbn-CNS-Ctr group, the granular layer in hippocampus manifested "C" in shape, the section area of granular layer was increased, and so was the cell body of granular cells in hippocampus during P7d to P21d; the inner arm granular cells in granular layer of hippocampus emerged on P7d, and most obvious changes occurred during P7d to P14d. While in Nbn-CNS-Del group, the granular layer in "C" shape were seen to form in most of the specimens during P7d to P21d, though they were sparsely distributed. 3.Imaging Diagnosis of the Bone Infarct

Objective To evaluate the imaging characteristics of the bone infarct . Methods 13 cases of the bone infarct proved by clinical manifestation and surgical pathology were collected,there were 5 males and 8 females. All patients had X-ray films, 4 were imagedwith CT scanning , 7 were imaged with MRI , and 2 underwent all three techniques . The bone infarct were classified into the early,themiddle and later stages.X-ray , CT and MRI findings of bone infarct ,were analysed characteristics . Results Partial osteoporosin and punctate calcifications could be seen on X-ray and CT , in the early and middle stages , which presented as iso-/low-signal on T_1WIand iso-/high-signal on T_2WI in the center of the bone infarct with a tortuous low-signal on T_1WI and high-signal on T_2WI zonearound.In the later stages,the bone infarct was characterize by irregular sclerosis and tortuous calcification on X-ray and CT,and by low-signal on both T_1WI and T_2WI . Conclusion MRI is the most effective way to find the bone infarct in the early stages , X-ray is helpful in diagnosing the bone infarct in the middle and later stages , CT is more sensitive than X-ray . X-ray ,CT and MRI should be combined in diagnosing the bone infarct .
4.Effect of Endotoxin Tolerance on Cytokine

Objective To further determine the inflammation?associated cytokine changes during endotoxin(LPS)tolerance. Methods Kun?ming mice were divided into groups. The endotoxin tolerance mouse model was established with repeated injection of LPS. Serum levels of tumor ne?crosis factor?α(TNF?α),interleukin?10(IL?10)and IL?17 were determined by ELISA assay. Results The serum concentrations of TNF?α,IL?10 and IL?17 were 2 261.49±1 090.28 pg/mL,7 780.42±2 320.04 pg/mL,and 27.28±13.76 pg/mL in the LPS control group,which were increased significantly compared to the normal healthy control group. The serum concentrations of TNF?α,IL?10 and IL?17 were 1 174.19±450.91 pg/mL, 1 293.26±520.85 pg/mL,and 22.26±11.44 pg/mL in the endotoxin tolerance group. Levels of TNF?αand IL?17 were further decreased by dexameth?asone treatment during LPS tolerance inducing,while the IL?10 level did not change significantly. Conclusion Compared to the status of endotoxin inflammation,levels of TNF?αand IL?10 decreased significantly,while levels of IL?17 were not significantly changed in the status of endotoxin toler?ance. Dexamethasone could further suppress TNF?αand IL?17 levels during endotoxin tolerance inducing.
5.Ocular vestibular evoked myogenic potential elicited by different types of stimulus among normal young Chinese people.

OBJECTIVE:
To observe waveform differences among ocular vestibular evoked myogenic potentials (ACS-oVEMP) elicited by different types of air conducted sound in normal young Chinese subjects.
METHOD:
Twenty-two adult volunteers (44 ears) were recruited as research subjects including 12 males and 10 females aged between 18 and 35. Five hundred Hz tone burst, 125 Hz tone burst and click were employed as stimulus for conventional ACS-oVEMP examinations in bilateral ears of each subject. The response rate, threshold, n I latency, p I latency, n I-p I latency interval, amplitude and inter-aural asymmetry were recorded and compared among groups.
RESULT:
The response rate was 100% (44/44) in 500 Hz tone burst group, 40.9% (18/44) in 125 Hz tone burst group and 29.6% (12/44) in click group There were statistical significant differences between the 500 Hz tone burst group and the other groups (P < 0.05). Waveform amplitude was the largest in the 500 Hz tone burst group (7.3 +/- 5.5) muV than that in the 125 Hz tone burst group (3.8 +/- 1.5) muV and that in the click group (5.8 +/- 1.3) muV, which had statistically significant differences between the former and the later two groups (P < 0.05). Other waveform parameters presented similar differeces.
CONCLUSION
ACS-oVEMP waveforms could be largely affected by different stimulus. 500 Hz tone burst was the best stimulus with highest response rate, biggest amplitude and therefore most easily recognizable waveform in ACS-oVEMP examination.

6.Monitoring the migration of bone marrow derived mesenchymal stem cells to intracranial glioma by sodium iodide sympoter

Objective To construct a recombinant lentiviral expression vector containing NIS and EGFP gene,and to explore the feasibility of NIS gene for monitoring the bone marrow derived mesenchymal stem cells (BMSCs) migration to the intracranial glioma.Methods The NIS and EGFP gene fragments were subcloned into lentiviral vector pLVX-puro,then packaged and amplified in HEK293T cells to obtain recombinant lentivirus pLVX-CMV-NIS-EGFP.pLVX-CMV-0-EGFP was constructed as control.BMSCs were isolated,cultured,and transfected by lentivirus.The antibiotic-resistant transfected BMSCs (BMSCs-NIS-EGFP and BMSCs-EGFP) were selected.The expression of NIS gene was examined by Western blot.Functional NIS activity was confirmed by the uptake of 125I and the inhibition effect of NaClO4.The nude mice intracranial glioma models were established.MicroSPECT was performed at 24 h post BMSCs-NIS-EGFP injection via the tail vein.Results pLVX-CMV-NIS-EGFP and pLVX-CMV-0-EGFP were successfully constructed and packaged.BMSCs were successfully isolated and cultured.Stable cell lines BMSCs-NIS-EGFP and BMSCs-EGFP were constructed after lentivirus transfection and puromycin selection.The expression of NIS gene was detected by Western blot in BMSCs-NIS-EGFP,but not in BMSCs-EGFP.BMSCs-NIS-EGFP showed significantly more uptake of 125I (nearly 10 times than the uptake in BMSCs-EGFP) and the uptake could be significantly inhibited by NaClO4.The nude mice intracranial glioma models were successfully established and the BMSCs-NIS-EGFP in glioma foci could be visualized by microSPECT imaging at 24 h post injection.Conclusions A recombinant lentivirus containing NIS gene could be successfully constructed for monitoring BMSCs migration towards intracranial glioma.It might provide evidence on the research of BMSCs and NIS gene mediated therapy for glioma.
7.Imaging Diagnosis of Nerve-Root-type Cervical Spondylosis and the Correlative Study on the Nerve Root Disturbance

Objective To explore the value of imaging diagnosis of nerve-root-type cervical spondylosis(NRTCS)and to study the relative factors to the nerve root disturbance.Methods 188 cases of NRTCS clinically proved and with complete imaging data were retrospectively analyzed.The imaging characteristics of CR,CT and MRI of NRTCS were compared and the relative factors to the nerve root disturbance were analyzed in combination with the clinical characteristics.Results ①CR films in six-position could show the whole expression of the cervical spine.The anterior-posterior and functional position CR showed the physiological curvature change in 122(64.9%),narrowing of intervertebral space in 96(51.2%)and vertebral-body unstability or slide in 49(26.1%).The oblique CR showed the intervertebral-foramen lessening or narrowing in(26.1%)and Luschka-or intervertebral-joint osteophyte in 82(43.2%);②CT scans showed Luschka-or intervertebral-joint osteophyte in 76(40.4%),disc herniation and calcification of cartilage nodus in 80(42.6%)and hypertrophy or calcification of ligamenlum flavum in 66(39.8%);③MRI showed intervertebral-disk degeneration and protrusion in 105(55.8%),hypertrophy or calcification of ligamentum flavum in 51(27.1%),the nerve root and spinal cord compressed in 69(36.7%)and edema or degeneration of spinal cord in 23(12.2%).Conclusion The nerve root disturbance is closely correlated with the imaging appearances in NRTCS.It is of importance in early diagnosis and treatment for NRTCS to analyse synthetically the imaging data and clinical characteristics.
8.Imaging Features of Charcot’s Joint

Objective To analyze X-ray ,CT and MRI findings of Charcot’s joint.Methods 24 cases of Charcot’s joint were collected,there were 11 males and 13 females.24 cases were examined with radiography,9 with CT ,5 with MRI,and 2 were examined with all three techniques.X-ray,CT and MRI imaging findings of Charcot’s joint were analysed.Results There were two types of Charcot’s joint,hypertrophic(12 cases) and atrophic(12 cases).X-ray and CT findings of hypertrophic type were hyperostotic osteosclerosis with osteophytes,periarticular soft tissue swelling and ossification.While the atrophic type appeared as extensive bone resorption and disappearance,periarticular debris and soft tissue swelling,articular dislocation.These imaging findings of Charcot’s joint also could be showed by MRI.The joint capsule thickened periarticular soft tissue markedly enhanced after Gd-DTPA was administrated with definite medial margin.Conclusion Charcot’s joint is of typical imaging appearances.
9.Effects of Celecoxib on Proliferation and Apoptosis of Human Endometrial Adenocarcinoma Cell Line HEC-1B

Objective To study the effects of cyclooxygenase-2(COX-2) selective inhibitor celecoxib on proliferation and apoptosis of human endometrial adenocarcinoma HEC-1B cell line. Methods Methabenzthiazuron(MTT) assay was used to examine the effects of different concentrations of celecoxib on proliferation of HEC-1B cells.When HEC-1B cells were treated with different concentrations of celecoxib for 24 h,cell cycle and apoptosis were detected by flow cytometry.The expression of COX-2 mRNA was analyzed by RT-PCR. Results MTT results indicated that celecoxib could inhibit HEC-1B cell proliferation in a time-and dose-dependent manner.When HEC-1B cells were treated with different concentrations of celecoxib for 24 h,flow cytometry results showed that the cell percent of(G_(0)/G_(1)) phase increased,S and G_(2)/M phase decreased,and cell apoptosis rate also increased,which was significantly different from that of the control group(P
