2.The heterogeneity of anti-GBM autoantibodies in sera from patients with anti-GBM disease and its clinical correlation

Objective:To investigate the heterogeneity of epitopes recognized by anti-GBM autoantibodies in sera from a large cohort of Chinese patients with anti-GBM disease and its clinical significance.Methods: The present study included 108 patients with anti-GBM disease who were diagnosed in our hospital, between Jan 1991 and May 2009, with complete clinical and renal pathological data. Sera or plasma exchange of the patients were used to incubate with cryostat section of normal human renal tissue for indirect immunofluorescence (IIF) assay. The cryostat sections of normal renal tissue were pre-treated by 6 mol/L urea to unmask cryptic epitopes, and untreated cryostat sections were used to detect natural exposed epitopes. The sera were diluted from 1:2 to 1:512 to determine titers of anti-GBM autoantibodies Patients with anti-GBM autoantibodies against cryptic or exposed epitopes were further stratified;their clinical and pathological associations were analyzed. Results: Sera from all the 108 patients could recognize cryptic epitopes on normal renal tissue ( urea treated section). IIF showed IgG linear staining along GBM. However, sera from 56/108 patients (group A) could also recognize exposed epitopes on normal renal tissue (untreated section) ; sera from the rest 52/108 patients (group B) could not recognize exposed epitopes. In urea treated condition, the average titer of anti-GBM autoantibodies from sera of patients in group A was significantly higher than that in group B (P<0.01) , ANCA-positive patients in group A were significant less than that in group B (P<0.01) . There was no significant difference between the two groups in regard to other clinical data (including serum creatinine) and renal histopathologic data. Conclusion: Anti-GBM autoantibodies from some patients with anti-GBM disease could recognize natural exposed epitopes, however, their anti-GBM titer for cryptic epitopes was higher than that of those recognizing cryptic epitopes only and the prevalence of serum ANCA was significantly less.
3.Distribution regarding tendency on personality disorder among college students in Shijiazhuang city

Objective To survey the prevalence of tendency on tendency of personality disorder among college students.Methods By means of stratified cluster sampling,498 students from 6 colleges in Shijiazhuang city and 204 students from 3 colleges in Beijing were studied through 'personality diagnostic questionnaire- revised UPDI'.Results The incidence rates on dependent personality (2.81%),histrionic personality (2.41% ) and borderline personality (2.21% ) were higher than obsessive- compulsive personality (0.40%) and schizoid personality (0.60%).The prevalence of personality disorder tendency was related to sex,major and years in college,blood type as well as their origins(from urban or rural).The overall incidence of personality disorder was 28.31% while the incidence rates of personality deviation and serious personality disorder tendency were 17.07% and 11.24% respectively.The incidence in males was higher than that in females.There appeared differences in dissociative personality,avoidant personality,paranoid personality,obsessive-compulsive personality,histrionic personality and narcissistic personality on people with different blood types.The scores of the city students were higher than that of the students from the rural areas regarding paranoid personality,dependent personality and narcissistic personality.Differences were also noticed between freshmen and students from other levels in the incidence rates on the tendency of avoidant personality disorder.Conclusion There were different incidence rates on the tendency of personality disorder among college students that related to sex,level in college and the origins where they were from(urban or rural).
4.Effects of polydatin on learning and memory and Cdk5 kinase activity in the hippocampus of rats with chronic alcoholism.

OBJECTIVETo observe the effects of polydatin on learning and memory and cyclin-dependent kinase 5 (Cdk5) kinase activity in the hippocampus of rats with chronic alcoholism.
METHODSForty rats were randomly divided into 4 groups: control group, chronic alcoholism group, low and high polydatin group. The rat chronic alcoholism model was established by ethanol 3.0 g/(kg · d) (intragastric administration). The abstinence scoring was used to evaluate the rats withdrawal symptoms; cognitive function was measured by Morris water maze experiment; Cdk5 protein expression in the hippocampus was detected by immunofluorescence; Cdk5 kinase activity in the hippocampus was detected by liquid scintillation counting method.
RESULTSThe abstinence score, escape latency, Cdk5 kinase activity in chronic alcoholism group rats were significantly higher than those of control group (P < 0.05). The abstinence score, escape latency in high polydatin group rats were significantly lower than those of chronic alcoholism group (P < 0.05); Cdk5 kinase activity in high and low polydatin group rats was significantly lower than that of chronic alcoholism group( P < 0.05); immunofluorescence showed that the Cdk5 positive cells of chronic alcoholism group were significantly increased compared with control group (P < 0.05), and the Cdk5 positive cells of polydatin groups were significantly decreased compared with chronic alcoholism group ( P < 0.05).
CONCLUSIONPolydatin-reduced the chronic alcoholism damage may interrelate with regulation of Cdk5 kinase activity.

5.Effects of progesterone on learning and memory and P2X7 receptor expression in the hippocampus after global cerebral ischemia/ reperfusion injury in rats.

OBJECTIVETo explore the effects of progesterone (PROG) on learning and memory and P2X7 receptor expression in the hippocampus after global cerebral ischemia/reperfusion (I/R) injury in rats.
METHODSForty-eight male SD rats were randomly divided into four groups (n = 12) that were normal group, sham-operated group, I/R group and I/R+ PROG group. The global cerebral I/R injury models were established by improved Pulsinelli's four vessel occlusion, the learning and memory were evaluated by Y-maze, the expression of P2X7 receptor protein in the hippocampus were detected by the immunofluorescence, the activity of superoxide dismutase (SOD) in the hippocampus were detected with hydroxylamine oxidation method, the content of malondialdehyde (MDA) in the hippocampus were detected with pen-thiobarbituric acid method.
RESULTSThere was no significant difference of the learning and memory and positive expression cells of P2X7 receptor protein and the activity of SOD and the content of MDA in the hippocampus between normal group and sham-operated group. Compared with sham-operated group, the learning and memory of I/R group were significantly decreased (P < 0.01), Compared with I/R group, the learning and memory of I/R + PROG group were significantly increased (P < 0.05). Compared with sham-operated group, the positive expression cells of P2X7 receptor protein in the hippocampus of I/R group was significantly increased (P < 0.01), Compared with I/R group, the positive expression cells of P2X7 receptor protein in the hippocampus of I/R + PROG group was significantly decreased (P < 0.01). Compared with sham-operated group, the activity of SOD in the hippocampus of of I/R group was significantly decreased (P < 0.01), and the content of MDA in the hippocampus of I/R group was significantly increased (P < 0.01). Compared with I/R group, the activity of SOD in the hippocampus of I/R+ PROG group was significantly increased (P < 0.05), and the content of MDA in the hippocampus of I/R + PROG group was significantly decreased (P < 0.01).
CONCLUSIONPROG could improve the learning and memory ability following global cerebral ischemia/reperfusion injury in rats, the protective mechanism might be associated with down-regulating the expression of P2X7 receptor protein and attenuating oxygen-derived free radicals in the hippocampus.

6.Functional difference of malate-aspartate shuttle system in liver between plateau zokor (Myospalax baileyi) and plateau pika (Ochotona curzoniae).

To explore the adaptive mechanisms of plateau zokor (Myospalax baileyi) to the enduring digging activity in the hypoxic environment and of plateau pika (Ochotona curzoniae) to the sprint running activity, the functional differences of malate-aspartate shuttle system (MA) in liver of plateau zokor and plateau pika were studied. The ratio of liver weight to body weight, the parameters of mitochondria in hepatocyte and the contents of lactic acid in serum were measured; the open reading frame of cytoplasmic malate dehydrogenase (MDH1), mitochondrial malate dehydrogenase (MDH2), and the partial sequence of aspartate glutamate carrier (AGC) and oxoglutarate malate carrier (OMC) genes were cloned and sequenced; MDH1, MDH2, AGC and OMC mRNA levels were determined by real-time PCR; the specific activities of MDH1 and MDH2 in liver of plateau zokor and plateau pika were measured using enzymatic methods. The results showed that, (1) the ratio of liver weight to body weight, the number and the specific surface of mitochondria in hepatocyte of plateau zokor were markedly higher than those of plateau pika (P < 0.01 or P < 0.05), but the content of lactic acid in serum of plateau pika was significantly higher than that of plateau zokor (P < 0.01); (2) MDH1 and MDH2 mRNA levels as well as their enzymatic activities in liver of plateau zokor were significantly higher than those of plateau pika (P < 0.01 or 0.05), AGC mRNA level of the zokor was significantly higher than that of the pika (P < 0.01), while no difference was found at OMC mRNA level between them (P > 0.05); (3) mRNA level and enzymatic activity of MDH1 was significantly lower than those of MDH2 in the pika liver (P < 0.01), MDH1 mRNA level of plateau zokor was markedly higher than that of MDH2 (P < 0.01), but the activities had no difference between MDH1 and MDH2 in liver of the zokor (P > 0.05). These results indicate that the plateau zokor obtains ATP in the enduring digging activity by enhancing the function of MA, while plateau pika gets glycogen for their sprint running activity by increasing the process of gluconeogenesis. As a result, plateau pika converts the lactic acid quickly produced in their skeletal muscle by anaerobic glycolysis and reduces dependence on the oxygen.

7.Study on effect of different compatibility of saponins contained in banxia xiexin tang on intestinal bacterium metabolism.

OBJECTIVETo compare the effect of intestinal flora on the metabolism of the Banxia Xiexin Tang (BXT) full prescription group, the sweet-nourishing group and saponins contained in single ingredients ginseng and liquorices.
METHODThe anaerobic incubation technology for intestinal flora in vitro was adopted to incubate the BXT full prescription group, the sweet-nourishing group and extracting solution of the single ingredients, under anaerobic conditions at 37 degrees C. Samples of different incubating time points were collected. The high-speed separation and content determination of various prototypes and metabolites were conducted with LC-MS/MS method, and then their degradation rate K was calculated to observe the difference and characteristics in metabolism of different compatible groups.
RESULTIntestinal flora could transform saponins into their metabolites. Having comparing spss one factor variance, we learned the difference in saponin metabolites of different compatible groups. As for the degradation rate of glycyrrhizic acid, the sweet-nourishing group > the full prescription group > the single prescription group (P < 0.05). Rb1 degraded the most slowly in the full prescription group. As for the degradation rate of Re, the single prescription group > the sweet-nourishing group > the full prescription group (P < 0.05).
CONCLUSIONThe sweet-nourishing group and the sweet-nourishing group have different effect in inducing or inhibiting intestinal flora. The single prescription group shows in inhibition in metabolites of Rb1 and Rg1. Glycyrrhizic acid metabolites are promoted by glycyrrhetinic acid, which facilitates the efficacy of drug absorption. The compatibility of compounds has no impact on metabolites of Rb1 and Rg3.

8.Association between risk factors during maternal pregnancy and the neonatal period and childhood bronchial asthma.

OBJECTIVETo study the association of the risk factors during maternal pregnancy and the neonatal period with childhood bronchial asthma.
METHODSA total of 306 children with asthma (asthma group) and 250 healthy children (control group) were enrolled. Their clinical data during the neonatal period and the maternal data during pregnancy were retrospectively studied.
RESULTSThe univariate analysis showed that there were significant differences in the rates of maternal use of antibiotics during pregnancy, use of antibiotics and probiotics during the neonatal period, preterm birth, cesarean section, low birth weight, and breast feeding (>6 months) between the asthma and control groups (P<0.05). The multivariate logistic regression analysis showed that use of antibiotics during pregnancy (OR=3.908, 95%CI: 1.277-11.962), use of antibiotics during neonatal period (OR=24.154, 95%CI: 7.864-74.183), preterm birth (OR=8.535, 95%CI: 2.733-26.652), and cesarean section (OR=4.588, 95%CI: 2.887-7.291) were independent risk factors for childhood asthma. The use of probiotics during the neonatal period (OR=0.014, 95%CI: 0.004-0.046) and breast feeding (>6 months) (OR=0.161, 95%CI: 0.103-0.253) were protective factors for childhood asthma.
CONCLUSIONSThe early prevention of childhood asthma can be improved by reducing the use of antibiotics during pregnancy, reducing cesarean section, avoiding abuse of antibiotics during the neonatal period, trying breast feeding and taking probiotics in early stage.

9.Application of endovascular balloon dilatation in removing long-term indwelling incarcerated hemodialysis catheter: preliminary results in 4 cases

Objective To evaluate the safety and effectiveness of endovascular balloon dilatation in removing incarcerated tunnel cuffed catheter (TCC) . Methods The clinical data and the imaging materials of 4 hemodialysis patients with incarcerated TCC, who received endovascular balloon dilatation at Taizhou Hospital of Zhejiang Province, China, during the period from January 2017 to March 2018, were retrospectively analyzed. After the treatment, the patient's vital signs were monitored, and the procedure-related complications such as hemopneumothorax, subcutaneous hematoma, arrhythmia, etc. were documented. Results The mean age of the 4 patients was (73.3±6.4) years. The average indwelling time of TCC was 5.5 years (4-8 years) . Successful treatment of incarcerated TCC was achieved in all 4 patients. In one patient, both 5-mm and 6-mm balloons were used to simultaneously dilate the dual cavities of TCC.Temporary arrhythmia occurred in one patient during operation. No serious postoperative complications occurred. Conclusion For the remove of long-term indwelling incarcerated TCC, endovascular balloon dilatation is minimally-invasive, safe and effective, although more researches are needed to further confirm its safety and reliability.
10.In vitro and in vivo assesemet of sodium norcantharidin lipid microsphere.

AIMTo prepare lipid microsphere of sodium norcantharidin (NCTD) and then study their characters and pharmacokinetic behavior.
METHODSDynamic Light Scattering, HPLC and retrodialysis technique were used to determine the in vitro characters of the NCTD loaded lipid microsphere (LM), such as the particle size, xi-potential, content, incorporation ratio, release profile and changes after dilute. And the plasma concentration was determined by HPLC-MS, compared with NCTD aqueous solution at the same time.
RESULTSEvery property showed that the LM was preferable. The average diameter was about 200 nm. The xi-potential was - 38 mV. The content was close to 100%. And the incorporation ratio exceeded 80%. After i. v. administration of single dose, the pharmacodynamic parameter of LM AUC was 111.28 microg x mL(-1) x h(-1). The data of plasma concentrations showed that the NCTD LM was of two compartment. There was no obvious difference between in vivo parameters of LM and reference solution.
CONCLUSIONThe NCTD LM was eligible and the character of it in vivo was not changed.
