Objective To evaluate the feasibility and efficacy of optical coherence tomography (OCT) in interventional diagnosis and treatment of borderline coronary artery disease. Methods Sixteen 40%-70% angiographically stenotic lesions from 15 patients were enrolled in the current study. Intravascular OCT was applied to each lesion after informed consent was obtained to evaluate stenosis percentage, size of lipid core, thickness of fibrous cap, existence of plaque rupture, and presence of thrombosis or calcification. Therapeutic strategies were made according to OCT outcomes as well as clinical symptoms and electrocardiographic changes. Stent coverage, apposition, and tissue prolapse between stent struts were determined in the patients undertaken percutaneous coronary intervention(PCI). Results Qualitative OCT images were obtained in 14 lesions. Ten lesions were greater than 50% stenosis, with large lipid core and fibrous cap thinner than 65 ?m, and plaque ruptures were noted in 2 of the lesions. PCI were performed in these 10 lesions. Two lesions underwent predilatation and significant intima tearings and dissections were detected with OCT. Repeated OCT after stent implantation showed complete coverage in all 10 lesions. Focal incomplete apposition were noted in 2 lesions. Significant tissue prolapse occurred in 3 lesions and segmental incomplete deployment of stent in 2 lesions. The remaining 4 lesions had small lipid cores, thick fibrous caps but with no plaque ruptures, thus PCI was not performed. Conclusion Intracoronary OCT is an effective tool to determine features of coronary lesions. It plays an important role in diagnosis of vulnerable lesions, strategy-making in treating borderline coronary artery disease and evaluation of immediate result of PCI.

Background and purpose:DOC-2 serves as one of the tumor suppressing genes in human ovarian cancer and plays a role in the process of cell growth and differentiation. This study was to investigate the role of DOC-2 in the TGF? signal pathway and verification of the interaction between DOC-2 and TGF?Ⅲ receptor. Methods:The bait vector was constructed by inserting the PID domain of DOC-2(nDOC-2)into yeast express vector pGBKT7.pGBKT7-nDOC2 was transformed into the yeast AH109 and confirmed to be expressed. After the human foetus brain cDNA library had been transformed, the positive clones was screened by both nutrition defect medium and X-?-gal.The putative positive clones were sequenced and analyzed to get the DOC-2 interactive proteins.Furthermore, after the DOC-2 cDNA and TGF?Ⅲ receptor cDNA had been co-transfected into the human ovarian cancer cell line HO-8910 together, the interaction between DOC-2 and TGF?Ⅲ receptor was investigated by immunoprecipitation and Western blot.Results:21 putative positive clones were picked after being screened and sequenced.Three of them were identified as Homo sapiens partial mRNA for betaglycan(TBR Ⅲ gene),homo sapiens protocadherin gamma subfamily C3(PCDHGC3)and APLP1(amyloid beta precursor-like protein 1).The analysis by immunoprecipitation and Western blot showed that the interaction between DOC-2 and TGF?Ⅲ receptor could form protein complex. Conclusions:The three encoding proteins might participate in the DOC-2 signal pathway.DOC-2 might play as an essential role in the TGF?signal pathway by interacting with TGF?Ⅲ receptor.

Objective To explore the expression change of Toll Like Receptor 4 (TLR4) in lung ischemia-reperfusion injury in rabbits and the influence of clemastine fumarate on it. Methods Fifty rabbits were divided into five groups randomly (n=10): Sham (N+S), reperfusion for 2 hours (N+I1/R2), reperfusion for 4 hours (N+I1/R4), clemastine fumarate + reperfusion for 2 hours (F+I1/R2) and clemastine fumarate+reperfusion for 4 hours (F+I1/R4). The ischemia time in each group was 1 hour and normal saline was given respectively in groups of N+S , N+I1/R2 and N+I1/R4. Western blotting , RT-PCR and immunofluorescence were used to detect the expression of TLR4 in lung tissue , and the changes of ultrastructure in ischemia-reperfusion lung tissue were observed by electron microscope. Result The expression of TLR4 was elevated obviously in ischemia-reperfusion lung tissue (P<0.05), and there was positive correlation between the increased TLR4 level and reperfusion time (P<0.05), the swelled and thick-ridge mitochondria were observed in type II alveolar epithelial cells after LIRI (P<0.05); but clemastine fumarate inhibited the expression of TLR4 in lung tissue significantly caused by LIRI (P<0.05). And the mitochondria injury was reduced in the groups of clemastine fumarate. Conclusion TLR4 expression is elevated in lung tissue after LIRI; clemastine fumarate inhibits the expression of TLR4 caused by LIRI and protects the lung tissue from LIRI in rabbits.

Hearts from eight adult dogs were perfused through both coronary arteries with methylmethacrylate. Small specimens cut out from the left ventricular wall were eroded in 50% HCL for 5-7 days. The replicas were studied under the SEM (S-450). The main results were outlined as follows: The subendocardial capillary plexus composed of varied shapes and orientation of meshes was present. The capillary network of the myocardial layers was consisted of many parallel capillaries and between them a variety of models of anastomoses, such as "H" "K" and "8" modes etc, were observed. In our findings, it was demonstrated that the arteriole and venule were distributed between the myocardial bundles and their direction was often perpendicular to the myocardial bundles. The postcapillary venules were poured into venule by the "ginger or turnip root" arrangement. The replicas of the Thebesian veins were proved in our specimens. The A-V shunting was not observed.
