2.Effects of Neonatal Handling and Enriched Environment Stimulation on Brain Damage in Neonatal Rats Following Hypoglycemia

Objective To explore the effects of the neonatal handling and enriched environmental stimulation on brain damage in neonatal rats with hypoglycemia.Methods Thirty-six neonatal rats were randomly divided into the normal group,hypoglycemia intervention group and the hypoglycemia non-intervention group.Those rats in hypoglycemia intervention and hypoglycemia non-intervention groups were weaned for 12 h,then the blood sugar of both groups were monitored.After neonatal rat models with hyperglycemia were prepared,the rats in hypoglycemia intervention group received the neonatal handling for 14 d and then were kept in an enriched stimulation environment for another 14 d.Rats in normal group and hypoglycemia non-intervention group were fed in the routine way.Neonatal handling was done when the rats were born for 24 h.The rat was rubbed with the brush from head to tail softly.Rats in the hypoglycemia non-intervention group was not handled.The enriched environment stimulation was used after 15 d when the rats were born.Rats in the hypoglycemia intervention group was put into the enriched environment for 1 h per day until 28 d when the rats were born,and rats in the hypoglycemia non-intervention group was put into the normal environment.Then the body weight was scaled at 0 d,7 d,14 d,21 d and 28 d when the rats were born.Space learning and memory were tested with Morris earter at their third month's age.After that,changes of pathology was observed in their occipital cortex.Results The weight increase,the ability of space learning,memory and the number of survival pyramid neurons of occipital cortex in normal group were better than those in hypoglycemia intervention and hypoglycemia non-intervention group(Pa
3.Effect of nitric oxide on rostral ventrolateral medulla modulating cardiac sympathetic afferent reflex in rats with chronic heart failure

BACKGROUND: Nitric oxide in the central nervous system is involved in controlling the sympathetic outflow. The authors' recent data show that the reduction of nitric oxide in the rostral ventrolateral medulla (RVLM)enhanced the cardiac sympathetic afferent reflex (CSAR) evoked by stimulating the cardiac sympathetic afferent nerves in rats with chronic heart failure (CHF).OBJECTIVE: To further investigate the effect of nitric oxide in the RVLM on modulating the CSAR evoked by epicardial chemical stimulation in rats with CHF.DESIGN: Randomized controlled experiment.SETTING: Department of Physiology, Nanjing Medical University, and Department of Cellular and Integrative Physiology, University of Nebraska College of Medicine.MATERIALS: This study was carried out in the Department of Physiology, Nanjing Medical University from July 2003 to May 2004. A total of 52male Sprague-Dawley rats weighing 360-420 g were used, and were randomly divided into chronic heart failure group and control group with 23 in each group.METHODS: The rats were carried out either sham surgery or the left coronary artery ligation. Six to eight weeks later, all rats were anesthetized with α-chloralose and urethane and baroreceptor denervated and vagotomized. The CSAR was evoked by epicardial application of bradykinin (BK, 0.04 μg and 0.4 μg in 2.0 μL) to mimic the effect of chemical stimulation on the heart in the CHF state. The renal sympathetic nerve activity (RSNA), mean arterial pressure (MAP) and heart rate (HR) were recorded at baseline and during elicitation of the CSAR. 4.Clinical study of color Doppler ultrasound for early diagnosis of acute cholecystitis infection

Objective To investigate the clinical value of color Doppler ultrasound in the early diagnosis of acute cholecystitis infection ,in order to improve the diagnosis of acute cholecystitis infection .Methods 36 cases with acute cholecystitis infection were selected as the observation group , and during the same period the healthy people were randomly selected as the control group .The color Doppler performance of ultrasound was observed .The hepatic artery blood flow velocity and the maximum peak of the cystic artery (Vmax) and resistance index(RI) were meas-ured.ROC curve was observed to evaluate the specificity of color Doppler ultrasound for the disease .Results The incidence rates of weak bile sound transmission ,gallbladder wall thickness ,gallbladder enlargement ,ultrasound Mur-phy positive in the control group were 25.0%,16.7%,22.2%,8.3%,which in the observation group were 72.2%, 47.2%,100.0%,94.4%,the differences were statistically significant (χ2 =4.571,4.571,4.589,5.689,all P<0.05).The differences in hepatic artery and the cystic artery Vmax and RI were statistically significant between the two groups(P<0.05).ROC curve comparison showed that the correct rate of gallbladder enlargement in the observa -tion group was the highest (90.7%),the specificity was 90.7%,sensitivity was 90.6%,and other indicators of ultra-sound had no significant difference (χ2 =0.893,P>0.05).Conclusion Color Doppler ultrasound can show the form of acute infectious cholecystitis ,gallbladder wall degree of bile through sound etc .,it can improve the diagnostic value of the disease.
5.Adipose tissue engineering construction using adipose-derived stem cells and scaffolds in breast reconstruction

BACKGROUND:The development of adipose tissue engineering has solved a series of problems and brought new ideas for breast reconstruction. OBJECTIVE:To summarize the main biological characteristics of scaffolds, seed cells and adipose grafts for adipose tissue engineering construction, as well as their application prospects in breast reconstruction. METHODS:With the key words of "scaffold, mesenchymal stem cells, adipose tissue engineering, breast reconstruction, vascularization" in Chinese and English, respectively, a retrieval of CNKI and PubMed was performed for relevant articles published from 1999 to 2017. After initial screening, the eligible articles were further detailed, analyzed and summarized. RESULTS AND CONCLUSION:Scaffold materials with low immunogenicity, especially the decellularized adipose tissue, have a unique advantage in adipogenic induction that can be used to support the regeneration of human organs and tissues. Adipose-derived stem cells can also be used as the priority for adipose tissue engineering construction, which can promote tissue development and regeneration in vivo. However, much less is known about the use of adipose tissue engineering in breast reconstruction and the implementation of long-term stable growth of the adipose tissues. Further investigations are indispensable on how to realize breast reconstruction using tissue-engineered adipose so as to construct and maintain a stable structure entity throughout the entire patient's life. Therefore, strict selection of seed cells, scaffolds and cytokines is considered pivotal in the construction of cell microenvironment.
6.Relationship between regulation effect of salvia miltiorrhiza on AQP2 in kidney and promoting blood circulation and diuresis.

Partial nature of "promoting blood circulation and dieresis" of Salvia Miltiorrhizain was initially demonstrated by investigating the regulation effect of AQP2 expression in kidney of trauma blood stasis model rats with the Salvia Miltiorrhizain so as to provide guidance for its clinical deployment of administration. Random allocation was taken to averagely divide 30 SD rats into two groups: 10 rats in normal group and 20 rats in blood stasis syndrome group. Trauma blood stasis rat model was established by quantitatively beating. Then the rat model group was divided into model group and salvia group. After 7 days of treatment, the rat kidney AQP2 expression was detected, the content of urine AQP2 was compared and the damaged local muscle and kidney pathological changes were observed by immunohistochemical method and western blot method. Compared with that of the normal group, rats in model group had inflammatory cells infiltration, blood stasis and edema of the injured local muscles and up-regulated AQP2 expression, decreasing urinary output, and kidney tissues blood stasis and edema (P < 0.05). On the other hand, compared with that of the model group, those parameters of rats in salvia group were all decreasing except urine output (P < 0.05). Such result indicated that Salvia Miltiorrhiza can reduce trauma blood stasis rat content of urine AQP2 and down-regulated AQP2 expression in kidney tissue, so as to reduce the reabsorption of water by renal tubular and increase urine output. The promoting blood circulation effect of Salvia Miltiorrhizain can alleviate the degree of the damaged tissue edema and encourage urine drainage. This therapy is closely related to the effect of regulating AQP2 in kidney by salvia, so the purpose of this study by verifying "promoting blood circulation and diuresis" as the mechanism for the regulation effect of the salvia on AQP2 expression.

7.Comparative Study of Propofol and Isoflurane on Inflammatory Cytokines in Patients with Intracranial Neoplasm

Objective To explore the neuroprotective mechanism of propofol by comparing the influence of propofol and isoflurane on inflammatory cytokines ( TNF-α、IL-1、ICAM-1 ) in patients with intracranial tumors. Methods One hundred and sixty-eight patients with intracranial neoplasm were randomly divided into two groups:the propofol ( Group P) and isoflurane (Group I),84 cases in each. Patients were given with propofol (3-6 μg·mL-1) by plasma target-controlled infusion or with continuously inhaled isoflurane ( 1%-2%) , respectively. The serum levels of TNF-α, IL-1 and ICAM-1 were detected before anesthesia and at 0,24,and 48 h after operation. Results The serum levels of TNF-α,IL-1 and ICAM-1 were significantly increased after operation as compared to baseline in both groups. The serum level of TNF-α was(69. 11±8. 95) and (76. 26±11.28) μg·mL-1,IL-1 was(21.57±3.19) and (29.58±4.38) ng·L-1,and ICAM-1 was (1.63±0.24)and (1.94±0.29) g·L-1 at 24 h post operation in Group P and Group I,respectively. These inflammatory cytokine levels were significantly higher in group I compared to group P at 24 and 48 h after operation (P<0. 05 or P<0. 01). Conclusion The target-controlled infusion of propofol brings about lower level of inflammatory reaction than isoflurane inhalation in patients with intracranial neoplasm,which may attribute to the mechanism of brain protection against injury.
8.Cloning of the Coding Region and 3′Non-coding Region of Calmodulin 2 in Guinea Pig

Objective To clone the coding region and 3′non?coding region of calmodulin 2(CaM2)in guinea pig,to provide the genetic informa?tion for studying the gene function of Calmodulin 2. Methods Total RNA was extracted from heart tissue of guinea pig,the coding region and 3′non?coding region of CaM2 were amplified by RT?PCR and 3′?RACE PCR methods,and the recombinant plasmid was constructed by inserting cDNA of the coding region and 3′non?coding region of CaM2 into the cloning vector by genetic engineering technology followed by DNA sequencing and se?quence analysis. Results The cloned coding region of CaM2 was 450 bp,and the 3′non?coding region of CaM2 was 660 bp. The amino acid se?quences of the coding region of CaM2 was consistent with those of other CaM subtypes,and the 3′non?coding region of CaM2 had low homology with those of other subtypes. Conclusion The cloning of CaM2 coding region and 3′non?coding region in guinea pig was the foundation for further study on the gene function of CaM2 and its role in related diseases.
10.Data analysis of hearing of 719 recruited students from teenager flying school and discussion of relative standards

Objective To analyze the data on hearing of recruited students from a teenager aviation school according to both the standards of PLA and those of the US Armed Forces , to learn about their differences in hearing standards in medi-cal selection, and to raise suggestions on establishing hearing standards for medical selection of flying cadets from teenager aviation schools .Methods The data collected on the first hearing test about hearing of students from Hebei Teenager Aviation School were collected .The validity of the data was determined according to the medical selection standards both of PLA and of the US Armed Forces respectively .The cases of qualified or unqualified recruits at high frequency and middle -low frequency were counted respectively and analyzed statistically .Results The unqualified recruits determined by PLA′s standards tended to be unqualified too when determined by the US Armed Forces standards .During the screening of high frequency hearing and the general hearing condition , the proportion of unqualified cases determined by the US selection standards was higher than that determined by PLA′s selection standard .In the screening of middle-low frequency hearing , no statistically significant difference was found in the proportion of unqualified cases when the two different selection standards were used.Conclusion The hearing standard for medical selection of flying cadets at high frequency in the US Armed Forces is stricter than that in the PLA .Meanwhile, the hearing standard at middle-low frequency in the US Armed Forces is different from that in the PLA , though there are no statistically significant differences in the proportion of unqualified cases.The hearing standards of the US Armed Forces are recommended in recruiting flying cadets for teenager flying schools .
