1.The physiological effects of resveratrol and its potential application in high altitude medicine.

Resveratrol, as a natural polyphenolic compound, has a wide range of beneficial effects, which includes anti-tumor, cardiovascular protection, anti-oxidant and estrogen-like effects, and so on. Its various physiological properties are closely related to the therapeutic principle for prevention and treatment of high altitude hypoxia injury. Resveratrol may play an important role in relieving or curing high altitude diseases, especially high altitude polycythemia(HAPC). However, the literature about study and application of resveratrol in plateau medicine field is rarely reported up to now. In this review, we summarized the physiological effects of resveratrol, discussed the possible main principle of resveratrol for HAPC therapy, and looked forward to resveratrol's perspective or potential application in high altitude medicine.

2.Association of Neuropeptide S Receptor 1 Gene Polymorphisms with Inflammatory Bowel Disease

Background:Studies showed that neuropeptide S receptor 1(NPSR1)gene polymorphisms were associated with the susceptibility of inflammatory bowel disease( IBD)in Europe’s populations,however,there is no study on the relevance in Chinese population. Aims:To investigate the association of NPSR1 gene polymorphisms with IBD in Chinese Han population. Methods:A total of 457 IBD patients[355 cases of ulcerative colitis(UC)and 102 cases of Crohn’s disease ( CD)]from Zhongnan Hospital of Wuhan University and 500 healthy controls were recruited. Genotyping of 2 single nucleotide polymorphisms(SNPs)of NPSR1,rs323922(C→G mutation)and rs740347(G→C mutation)was performed by using PCR and sequencing techniques. Results:Differences of the frequencies of genotypes and alleles for rs323922 and rs740347 between UC,CD patients and controls didn’t reach statistical significance(P >0. 05). Genotype-phenotype analysis showed that there were some impact of genotypes for rs323922 and rs740347 on clinical phenotypes of IBD:( i) For rs323922,mutant CG genotype was correlated with male CD patients(OR:0. 441,95% CI:0. 230-0. 844)and CD involving the colon(OR:0. 425,95% CI:0. 199-0. 911),and might be a protective factor.(ii)For rs740347,mutant CC genotype was correlated with early onset CD patients( <16 years old)(OR:15. 019,95% CI:2. 634-86. 470)and mutant C allele was correlated with CD involving the colon(OR:2. 142,95% CI:1. 709-4. 294),both were risk factors. Conclusions:Polymorphisms of NPSR1 rs323922 and rs740347 are not contributors of IBD susceptibility in Chinese Han population,but might be correlated with some clinical phenotypes of IBD.
4.Aging performances for resisting low-temperature of three dental Yttria-stabilized zirconia ceramic core materials.

BACKGROUNDThe low-temperature resistance aging performance of Yttria-stabilized tetragonal zirconia polycrystal (Y-TZP) is the key effective factor that influences the long-term success rate of prosthesis. The objective of this study was to test and compare the aging performances for resisting low temperature of Lava Frame, Cercon Smart, and Upcera Yttria-stabilized zirconia core materials, via analyzing the micro and the crystal phases of the materials, and measure the three-point bending strength and the fracture toughness.
METHODSThe three zirconia green bodies were prepared as 60 test samples for three-point bending strength and as 60 test samples for fracture toughness. The test samples for three-point bending strength and fracture toughness were assigned to five groups and were treated respectively for 0, 5, 10, 15, and 20 hours to observe the micro and the crystal phases of the test samples. Then the three-point bending strength and fracture toughness were tested by X-ray diffraction (XRD).
RESULTSThe m phase content of Lava Frame was raised from 7.70% to 13.01%; the m phase content of Cercon Smart was raised from 4.95% to 8.53%; and Lava Frame is raised from 10.84% to 35.18%. The three-point bending strengths of the three zirconia core materials were higher than 1100 MPa and the fracture toughness was higher than 3 MPa·m(1/2). The three-point bending strength and the fracture toughness of Upcra zirconia decreased the most, followed by Lava Frame, and then by Cercon Smart.
CONCLUSIONThe aging resistance sequences of the three zirconia core materials are, from strong to weak, Cercon Smart, Lava Frame, and Upcera.

6.Mutation screening of MYH7 gene in a chinese pedigree with familial hypertrophic cardiomyopathy

Objective To identify the disease-causing gene mutation and investigate the genotypephenotype correlation in a Chinese pedigree with familial hypertrophic cardiomyopathy.Methods In this study we collected a large multigenerational Chinese family with FHCM.Total genome DNA was extracted from 67 subjects' peripheral leucocytes.The exons and boundary introns of MYH7 gene was amplified by PCR and directly sequenced by ABI PRISM 3700 DNA sequencer.Then,the mutation was examined.Results Fourteen family members had hypertrophic cardiomyopathy,including 4 deceased 2 of whom died from sudden death at young age.Analysis by echocardiography showed all the 10 living affected individuals have a maximal leftventricular-wall thickness of at least 13 mm.Three single nucleotide polymorphisms (SNP) which had been reported in NCBI SNP database,were found mutated.No mutation co-seperated with the disease was identified.Conclusion FHCM of this family was not caused by MYH7 mutation.Other genes should be screened to further identify the disease-causing gene mutation in hypertrophic cardiomyopathy.

9.A rat model of high altitude polycythemia rapidly established by hypobaric hypoxia exposure.

OBJECTIVETo investigate the effects of simple hypobaric hypoxia on parameters of hematology and blood rheology in order to establish a rat model of simulated high altitude polycythemia (HAPC) for the study of pathophysiologic mechanisms and medical prevention and treatment of HAPC.
METHODSForty-eight male Wistar rats were randomly divided into three normal control groups and three hypoxia model groups. Normal control group rats were bred in normoxia conditions, and hypoxia group rats were subjected to hypoxic exposure for 8 hours per day at simulated 5 500 m high altitude in a hypobaric chamber. After hypoxic exposure for 2, 4, 12 weeks, one group of normal control and hypoxia model rats were killed and blood was collected, respectively. Then parameters of erythrocyte and blood rheology were examined.
RESULTSMucous membrane of hypoxia model rats showed obviously cyanosis after 2 weeks hypoxic exposure. Hemoglobin concentration of hypoxia model rats were beyond 210 g/L after 2 weeks, 4 weeks and 12 weeks hypoxia exposure and significantly increased than that of normal control rats respectively. Besides, RBC counts, hematocrit, whole blood viscosity, erythrocyte aggregation index of hypoxia model rats were all notably higher than those of normal control rats respectively.
CONCLUSIONA rat model of high altitude polycythemia can be rapidly established by hypobaric hypoxia exposure at simulated 5 500 m high altitude for 8 hours daily.
