2.PTEN mRNA expression in proliferative LECs of the rabbit

· AIM: To observe the effects of PTEN and the tumor suppressor gene in proliferative LECs of the rabbit.·METHODS:Forty-two white rabbits were randomly divided into test group (36 rabbits) and control group (6rabbits). The transparent lenses of treated rabbits were operated with extra capsular cortex extraction, and the controls were kept untouched. The rabbits were sacririced at 1d, 3d, 1wk, 2wK, 1mo and 2mo after surgery.Immunohistochemistry was applied to detect proliferative cellular nuclei antigen (PCNA) as well as hybridization in situ and reverse-transcriptase polymerase chain reactions (RT-PCR) were applied to detect phosphatase as well as tensin homology deleted on chromosome 10(PTEN) mRNA in lens equartor.·RESULTS:PCNA expression increased significantly to a high level at 1wk, began to reduce 2wk later, and recovered to the normal level 1 or 2mo after the surgery.PTEN mRNA expressed positively in normal rabbit LECs.The relative PTEN mRNA contents reduced greatly at 1dafter operation and remained low level at 3d. It began to increase slightly at 1wk, kept rising at 2wk and regained the normal expression after 1 or 2mo. There was inverse correlation between the PTEN mRNA and PCNA expression.·CONCLUSION: PTEN mRNA expresses positively in normal rabbit LECs plasma. PTEN participates in LECs proliferation and correlates with lens proliferative conditions.
4.Evaluation of the Vertebral Artery Anatomic Course with Multi-slice Spiral CT and Its Clinical Application

Objective To evaluate the value and the significance of 16-slice spiral computed tomography angiography(MSCTA) in identifying the vertebral artery course variation.Methods MSCTA data in 22 patients with anomalous path of vertebral artery were retrospectively analyzed,13 of them had undergone DSA examinations,and 5 of them had also undergone MRI examinations.Multi-planar reconstruction(MPR),curved MPR,thin-slab maximum intensity projection(MIP),volume rendering(VR) or slab VR(cut-plane VR) were used to assess the anomalous path of the vertebral artery and its relationship with the adjacent large vessels.Virtual endoscopy(VE) was used to evaluate the orifice of the vertebral arteries and its relationship with the aortic arch and the ostia of the adjacent large vessels.Different views of vertebral artery were analyzed on DSA.MSCTA and DSA images were analyzed in blinded fashion for the accuracy of anomalous vertebral artery path and origin detection.Results were compared in a secondary consensus evaluation.Results The anomalous path of the vertebral artery in all 22 patients were correctly displayed on MSCTA,7 of them had anomalous origin of vertebral artery,the anomalous of ostia showed clearly.The vertebral arteriae with course variation were all going up at the front of the foramen transversarium of C6,then bended into the C5,C4 or C3 foramen transversarium.DSA could only be detected the anomalous origin in 13 patients undergone DAS.5 patients indirectly showed having course variation of vertebral artery by MRI.Conclusion MSCTA is superior to DSA in showing the anomalous path and orifice of vertebral artery,it should be considered as a non-invasive imaging tool for these diseases.
5.Applied microanatomy of cervical nerve and its related structure

Objective To study the course of different cervical nerve segments and their related intervertebral foramen’s size so as to increase safety and decrease complications in microinvasive surgery for cervical syndrome. Methods Fifteen human cervical specimens (30 sides) were anatomically observed for the course, branches and distribution of cervical nerves. Results The diameter of intervertebral foramen ranges from 0.54 to 0.65 cm, and increases gradually from top to bottom. The vertical diameter and anteroposterior diameter between C4, C5 and C6 vertebrae are smaller, and those between C3 and C7 are larger, but the diameter line of nerve root in intervertebral foramen from up to down gradually increases. The anteversion angle between nerve root and spine cord on horizontal plane is from 15? to 19?, within a small variation, while the declination angle on coronal plane gradually decreases from C3 to C7. There exist plenty of anastomosis branches among the cervical dorsal rami. Conclusion In the range of 0.6 cm around articular process at the entrance of intervertebral foramen, it is the narrowest part, the removal of which may alleviate the pressure on nerve root and benefit spinal stability. Because of the general existence of anastomosis branches of cervical nerve, the symptoms of cervical syndrome are not completely consistent to innervation. The intervertebral foramen between C4, C5, C6 is relatively small and the diameter line of nerve root is comparatively large, so the nerve root at C4, C5, C6 is most likely to be pressed.
6.Validity of panoramic radiographs for measurements of mandibular asymmetry

Objective To evaluate the reliability of the panoramic radiographs for diagnosing mandibular asymmetry.Methods A total of 25 cases with no asymmetry in chief complaint or in appearance were selected for symmetry group.Pa and PAC from 16 cases of facial asymmetry were selected for asymmetric group.Marker points were fixed,a reference line and the reference plane drawn,and then relevant measurement items determined.The repeatability of the fixed position was observed.The difference of all measurement items between the left and right on Pa in symmetry group and asymmetric group were compared respectively and the asymmetric rate of corresponding measuredvalues on two sides between two groups were observed too.Correlation test was used to evaluate the measured-values on Pa and PAC in asymmetric group.Results Dispersion became smaller with the help of reference line and reference plane.And the dispersion was larger when Me was used as a marker other than MF.Most measured-values in symmetry group on Pa and all measured-values in asymmetric group on Pa showed statistically significant difference (P< 0.05).The mean difference of CP-GoP distance representing the vertical measured-value was 4.73 mm (P<0.01); and the mean difference of GoP-MF distance representing the horizontal measured-value was 5.77 mm (P<0.01).The horizontal measurements on PA and PAC showed closer correlation (r=0.78~0.85,P<0.01) while the vertical was poorer (r=0.64,P<0.01).Conclusions This study suggests that marking assisted with reference line and reference plane could get a more repeatable fixed-point on Pa.MF point could be used as a fixed-point for measuring mandibular asymmetry on Pa and the Pa measurement can be used to evaluate mandibular asymmetry.
7.Micro -incision phacoemulsification with silicone oil removal for eyes with silicone oil tamponade combined cataract after vitrectomy

AlM: To observe the clinical effect of micro-incision phacoemulsification combined with silicone oil removal from pars plana in eyes with silicone oil tamponade after vitrectomy.
METHODS:Forty-three patients (43 eyes) with cataract and silicone oiltamponade after vitrectomy underwent micro-coaxial phacoemulsification combined with silicone oil removal from pars plana between February 2013 and November 2013. lntraocular lens was implanted at stage l in all cases. The best corrected visual acuity was observed at 3mo after operation, and the complications were recorded.
RESULTS: The best corrected visual acuity in 39 cases (91%) was improved at 3mo after operation, and the best outcome was 0. 6. There were still 4 cases (9%) without improvement of visual acuity. There was no incidence of complications such as posterior capsular rupture, residual silicone oil, corneal endothelial decompensation, hyphema, vitreous hemorrhage, intraocular lens dislocation, secondary glaucoma and ora serrata mutilation after operation in all patients.
CONCLUSlON: Micro - invision phacoemulsification combined with silicone oil removal from pars plana is an effective and safe method for eyes with cataract and silicone oil tamponade after vitrectomy.
8.Design, synthesis and antidepressive activity of duloxetine derivatives

In this paper, duloxetine was chosen as the lead compound. The pharmacophores with 5-HT(1A) antagonism activity were used to replace the naphthyl of duloxetine. A series of duloxetine derivatives had been designed and synthesized and whose structures were confirmed with elemental analysis, MS and H NMR. All synthesized compounds were tested by tail suspension test and forced swimming test in vivo. The test results revealed that most of the compounds have shown better activity than duloxetine at the same dosage. Some of them are worth to be studied further.
9.Antioxidation of Picroside Ⅱ on Cerebral Ischemic Injury in Rats and Its Optimum Dosage and Time Window

Objective To optimize the therapeutic dose and time window of picroside Ⅱ in treating cerebral ischemic injury in rats by orthogonal test. Methods The forebrain ischemia models were established by bilateral common carotid artery occlusion (BCCAO) method. The successful models were randomly grouped according to orthogonal experimental design and treated by injecting picroside Ⅱintraperitoneally at different ischemic time with different doses. The concentrations of MDA, NO and H2O2 in serum and brain tissue were respectively determined by thiobarbituric acid assay, nitratase reductase assay and chemiluminescence immunoassay. Results The optimized composition of the therapeutic dose and time window of picroside Ⅱ in cerebral ischemic injury were ischemia 1.5 h with 10 mg/kg, 1.5 h with 20 mg/kg and 1.5 h with 10 mg/kg body weight according to the expressions of MDA, NO and H2O2 in serum, and ischemia 1.5 h with 10 mg/kg, 1.5 h with 20 mg/kg and 1.5 h with 20 mg/kg body weight according to the expressions of MDA, NO and H2O2 in brain tissue. Conclusion On the basis of the principle of lowest therapeutic dose with longest time window, the optimized composition of the therapeutic dose and time window in cerebral ischemic injury is injecting picroside Ⅱ intraperitoneally with 10-20 mg/kg body weight at ischemia 1.5 h.
10.Antidepressant-like effects of ammoxetine in mice of behavioral despair model and monoamine neurotransmitter depletion model

OBJECTIVE To study the antidepressant effects of ammoxetine(AMX)and the underlying mechanisms. METHODS Two behavioral despair models,the tail suspension test (TST) and the forced swimming test(FST),were used to evaluate the antidepressant-like effects of AMX 2.5-20 mg · kg-1 following oral administration. Monoamine neurotransmitter p-chloro-phenylalanine(p-CPA)andα-methyl-p-tyrosine(AMPT) depletion models in mice were used to investigate the effects of AMX on levels of 5-serotomin(5-HT)and norepinephrine(NE)in the brain. RESULLTS The results of behavioral study showed that compared with normal control group,AMX(10 and 20 mg · kg-1)reduced the immobility time of mice by 51.4% and 80.7% in the TST(P<0.05,P<0.01) or by 48.0% and 66.2% in the FST (P<0.05),respectively. Locomotion activity test indicated that AMX did not increase or decrease the movement distance of mice,demonstrating that AMX had no excitatory or inhibitory actions on the central nervous system. Moreover,AMX(5,10 and 20 mg·kg-1)exerted antidepressant effects in the p-CPA induced 5-HT depletion model and AMPT induced NE depletion model,as evidenced by the significantly reduced immobility time,ie,63.9%,93.4%,90.5% and 61.9%,77.2%,100% reduction in the TST (P<0.01),respectively,and AMX at the dose of 20 mg·kg-1 significantly increased the concentrations of 5-HT and NE by 144.7% and 57.2% in the mouse brain(P<0.05) ,respectively. CONCLUSION AMX has strong antidepressant-like effects in behavioral despair models and monoamine neurotransmitter depletion models in mice,which is involved in the increased levels of 5-HT and NE in the brain.
