1.Determination of Residual Solvents in Faeces Bombycis Extract by GC

Objective:To establish a method for the determination of solvent residues in faeces bombycis extract by GC .Meth-ods: A GC-headspace injection method was adopted to detect the residual organic solvents in the extract of faeces bombycis .A DB-5MS (30 m ×0.25 mm ×0.25 μm) quartz capillary column was used as the chromatographic column;nitrogen was used as the carrier gas at a flow rate of 0.6 ml· min-1;the injector temperature was 200℃;the detector was a flame ionization detector( FID) with the temperature of 250℃;the temperature program included two phases:the initial temperature was set at 40℃for 10 min, and then risen to 200℃at a rate of 5℃· min-1 and maintained for 5 min;the equilibrium temperature of headspace was 95℃;the equilibrium time was 30 min;the capillary temperature was 110℃;the injection ring temperature was 125℃.Results:The linear range of acetone , 2, 3-dimethylpentane, 3-methylhexane, heptane, 2,2-dimethylhexane, p-xylene, m-xylene, O-xylene and 2,4,6-collidine was 101-3 034μg· ml-1 , 100-2 995 μg· ml-1 , 107-3 197 μg· ml-1 , 101-3 019 μg· ml-1 , 99-2 962 μg· ml-1 , 45-1 358 μg· ml-1 , 44-1 325μg· ml-1, 47-1 411 μg· ml-1 and 104-3 130 μg· ml-1, respectively, and the average r was all above 0.992.The recovery of the method met the requirement , and the blank solvent showed no interference .Eight samples were all accordance with the requirements after the examination .Conclusion:The headspace injection method for the determination of residual organic solvents in the extract of faeces bombycis is simple and reliable .
2.Observation of Pharmacodynamics in Patients with Continuous Infusion of Domestic Atracurium under Total Intravenous Anesthesia

OBJECTIVE:To discuss the feasibility and average infusion velocity of domestic atracurium by continuous in-fusion and to determine and validate its clinical valence.METHODS:80patients undergoing selective operation were treated with domestic atracurium0.5mg/kg to facilitate intubation under the flaccidity monitoring(TOF),the continuous infusion velocity was regulated during operation to keep the level of T 1 at1%~10%of the control value,the infusion was stopped20min before the termination of operation.RESULTS:The average infusion rate was(5.1?0.4)(?g?kg)/min,the drug effect starting time and duration of action of initial dose were(203?47)s and(41.5?4.5)min respectively;Significant correlations were found between the initial time of T 1 and PTC during the TOF reaction-free period and between TR value and DBS value at extubation time(P

BACKGROUNO:Some experiments have proved that some traditional Chinese medicines (TCM) have good biocompatibility when they touch and interact with human body tissues, body fluid or blood.Therefore,the physicochemical properties and functions of biomaterials can be improved by compounding it with some TCM.OBJECTIVE:To evaluate the biocompatibility of gelatin-bletilla porous material carrying TCM.DESIGN:Single sample experiment.SETTING:Department of Acupuncture & Moxibustion and Orthopaedics.Hubei College of Traditional Chinese Medicine. MATERIALS:This experiment was carried out in the Department of Orthopaedics,Tongji Medical College,Huazhong University of Science and Technology between February and April 2005.Twelve mice,male and female in half,weighing 18-24 g, and 6 Japanese big-eared rabbits,weighing 2.8 to 3.0 kg,were chosen for this experiment.They were raised at (18±1)℃ in separate cages.METHODS: Spongy gelatin-bletilla porous materials carrying extract of Coptidis and Radix Salviac Militiorrhizae was prepared by freeze-drying method.The acute systemic toxicity test:The 12 mice were divided into experimental group and control group. Mice in the two groups were intraperitoneally injected with leaching liquor and normal saline,respectively (the dose was 50 mL/Kg),then the general state and body mass of mice were recorded at the 4th,24th,48th and 72nd hours after injection, respectively.The porous materials were implanted in dorsal muscle of rabbits.Two rabbits were sacrificed at the 1st, 2nd and 6th weeks after operation seperately,and tissue reaction of biomaterial in vivo was assessed by gross observation and histological examination. MAIN OUTCOME MEASURES: Gross observation and histological examination of tissue reaction of biomaterials in vivo.RESULTS: Twelve mice and six big-eared rabbits were involved in final analysis.The acute systemic toxicity test:Mice of two groups had good status,normal activity and appetite,stable respiration:and no abdominal irritation,exhaustion, cyanosis or death was found.Body mass of mice of two groups presented tendency of increase at the 24th,48th and 72nd hours after injection,and there were no significant differences between two groups (P>0.05).The implantation test:①Gross observation:The samples were tightly bounded with muscular fibrous tissues at postoperative 1 week.They were connected with muscular fibrous tissues and could not be easily separated at postoperative 4 weeks.Gelatin-bletilla porous materials carrying TCM could hardly been seen at postoperative 8 weeks.②Histological observation:There were a lot of infiammatory cells and mesenchymal stem cells around the samples at 1 week after the implantation of gelatin-bletilla porous materials carrying TCM; Some materials were degraded and infiammatory cell infiltration was reduced around the implanted materials at 4 weeks; The inflammatory cells were further reduced and the materials were absorbed and replaced by muscular tissues at 12 weeks.CONCLUSION:Gelatin-bletilla porous materials carrying TCM is safe and reliable for application in wound healing,skin and muscle tendon tissue engineering.
4.Effects of isoflurane anesthesia on cognitive function and expression of TSPO in brain tissues of aged rats

Objective To evaluate the effects of isoflurane anesthesia on the cognitive function and expression of translocator protein 18 kDa ( TSPO) in the brain tissues of aged rats. Methods Twelve pathogen?free male Sprage?Dawley rats, aged 20 months, weighing 500-550 g, were randomly divided into 2 groups ( n=6 each) using a random number table: control group ( group C) and isoflurane anesthesia group ( group I) . The rats inhaled 2% isoflurane in 100% O2 for 4 h in group I, or 100% O2 for 4 h in group C. The rats underwent Morris water maze test at 24 h after anethesia. The escape latency and frequency of crossing the original platform were recorded. Then the rats were sacrificed, and the hippocampus and cerebral cortex were isolated for determination of the expression of TSPO and Iba1 by Western blot and quantitative real?time reverse transcriptase?polymerase chain reaction. Results Compared with group C, the escape latency was significantly prolonged, the frequency of crossing the original platform was decreased, and the expression of TSPO and Iba1 protein and mRNA in the hippocampus and cerebral cortex was up?regulated in group I ( P<0. 05 ) . Conclusion Isoflurane anesthesia results in decreased cognitive function through up?regulating the expression of TSPO in the brain tissues of aged rats.

Existing antidepressants exhibit delayed onset of action,which can decrease the compliance of the patients and enhance the risk of suicide.How to produce early-onset antidepressants with higher efficacy and lower adverse reactions has become a crucial point in the research of antidepressants.It has been demonstrated that selective 5-HT1Aantagonist,?2 antagonist and 5-HT2Aantagonist can accelerate the response of classic antidepressants.Furthermore,5-HT/NE dual reuptake inhibitor and 5-HT/NE/DA triple reuptake inhibitor can also produce early onset of action.Here,several reasons for the delayed onset of action and the progress in the development of early-onset antidepressants are reviewed.

The safety of Chuangyuling (CYL) dressing-a multifunctional medicine carrying biomaterial was evaluated in order to provide foundation for the application of CYL as material used in the wound healing. The traditional Chinese medicine (TCM) extract solution was compounded with scaffolds (gelatin and Bletilla hyacinthine gum), and then frozen and dried to form spongy and porous material CYL. According to the standard of biological evaluation of medical devices that was instituted by the ministry of health of China, the biological evaluation of CYL dressing was conducted. The results showed that all the contents of biological evaluation test consisting of acute toxicity, skin irritation, sensitization and cytotoxicity met the requirement of standards. It was concluded that the biomaterial carrying TCM (CYL dressing) is safe for application of wound healing.

7.Comparison of two prediction models for mortality ofacute paraquat poisoning

Objective:
To compare the effects of Cox proportional hazard regression model (Cox model) and extreme gradient boosting model ( XGBoost model ) on the prediction of the mortality of acute paraquat poisoning (APP).
Methods:
The APP cases admitted to Qingdao Eighth People's Hospital and Shandong Provincial Hospital from January 1st of 2018 to December 1st of 2020 was recruited and divided into a training group and a verification group by a random number table. The Cox model and XGBoost model were established to select the predictors for APP mortality. Receiver operating characteristic ( ROC ) curve was drawn to analyze the predictive power of the two models, and the calibration was evaluated using Hosmer-Lemeshow test.
Results:
Totally 150 APP cases were recruited. There were 75 cases each in the training group and in the verification group, with 52 and 55 cases died respectively, accounting for 69.33% and 73.33%. The Cox model showed that paraquat intake, the time from taking poison to seeing a doctor, the time for the first perfusion, the time for the first vomiting, aspartate aminotransferase, alanine aminotransferase, serum creatinine, blood urea nitrogen, white blood cell, lactic acid, creatine kinase isoenzymes, glucose, serum calcium and serum potassium were the predictors of APP mortality ( all P<0.05 ). The XGboost model showed that the predictive power of the factors in a descending order were the time from taking poison to seeing a doctor, the time for the first vomiting, the time for the first perfusion, lactic acid, white blood cell, paraquat intake, serum creatinine, serum potassium, serum calcium, creatine kinase isoenzymes, glucose, aspartate aminotransferase, blood urea nitrogen and alanine aminotransferase. The area under curve ( AUC ) of the XGBoost model for predicting was 0.972, which was greater than 0.921 of the Cox model ( P<0.05 ). The predicted results of the Cox model and XGBoost model were consistent with the actual situation ( P>0.05 ).
Conclusion
The Cox model and XGBoost model are consistent in predicting the mortality of APP, but the latter is better.

9.Fatty acid participates in up-regulation of diabetes on function and expression of CYP1A2

Aim To investigate the mechanism of dia-betes changing the hepatic CYP1 A2 through in vitro cell culture study.Methods The function of CYP1 A2 in HepG2 and Fa2N-4 cells were evaluated by determi-ning the level of phenacetin metabolism,and the mR-NA expression of CYP1 A2 in cells was detected by real time PCR.HepG2 cells were co-cultured with serum of diabetic rats(type 1 and type 2)and normal rats,then the CYP1 A2 function in cells were evaluated.Then, the HepG2 and Fa2N-4 cells were co-cultured with a series of concentrations of saturated (including palmitic acid and stearic acid)and unsaturated fatty acids(in-cluding oleic acid and linoleic acid)for 48 h,and the function and expression of CYP1 A2 in the cells were compared.Results It was found that the activities of CYP1 A2 were higher in cells incubated with diabetic serum of both type.All high concentration of fatty acids could increase the function and expression of CYP1 A2 in both HepG2 and Fa2N-4 cells.Conclusion It is speculated that the abnormal level of fatty acids under diabetic state might be part of the reasons why diabetes change the hepatic CYP1 A2,which provides the basis for future study.
10.Correlation analysis among quality of life, knowledge, beliefs, behavioral and pulmonary function outcomes in stable-stage patients with chronic obsturctive pulmonary disease

Objective To explore the relationship between quality of life and knowledge,beliefs,behavioral,pulmonary function outcomes in stable-stage patients with chronic obsturctive pulmonary disease (COPD).Methods Using the general data questionnaire,COPD assessment test (CAT),knowledge,attitude and behavior questionnaire to investigate 93 stable-stage COPD patients in urban and rural areas,and measure the level of forced expiratory volume in one second (FEV1%).Results CAT total score was (22.56 ± 6.40) points which was in severe level;knowledge,beliefs,behavior total score was (41.94 ± 8.20) points,and FEV1% was (59.81 ± 7.64) %.The CAT was negatively correlated with knowledge,beliefs,behavior total score and two dimensions of beliefs,behavior and FEV1%,r values were-0.262,-0.288,-0.217,-0.256 respectively,P < 0.05.Conclusions The COPD patients in stable-stage have a high level of CAT.Targeted invention should be combined with the status of the knowledge,beliefs and behavior to improve the level of air flow,and quality of life of patients.
