2. Evaluation on taste-masking effect of Andrographis Herba by electronic tongue

Objective: To evaluate the feasibility of taste-masking by electronic (E)-tongue and provide a reference for the drug taste-masking study. Methods: Andrographis Herba was used as the carrier of bitter Chinese materia medica (CMM), sodium acetate (NaAc), sodium cyclamate (SC), adenosine-5′-monophosphatedisodiumalt (AMP), and 2, 4-dihydroxyhenzoic acid (DA) as the taste-masking agents. The data obtained by the E-tongue were analyzed through principal component analysis (PCA), and Euclidean distance in the data space of different dimensions was calculated. Regarding the distance of bitterness (D) as the evaluation index, then whether the data analyzed by the E-tongue and volunteers were the same or were not investigated. Results: The evaluation results of the taste-masking agents by E-tongue were similar to the taste by volunteers except NaAc, suggesting that some conditions should be required to evaluate the effect of CMM taste-masking. Conclusion: This study shows that E-tongue could be used for evaluating the effect of CMM taste-masking under certain conditions. To evaluate the gustation information by E-tongue, we should use the professional knowledge of pharmacy, chemical and other related disciplines to analyze and improve this evaluation method and model gradually.
4.Effect of Yisheng herbal injection on chronic allograft nephropathy in rat

Objective To investigate the preventive and therapeutic effectiveness of Yisheng herbal injection against chronic allograft nephropathy in rats.Methods Renal transplantation was performed from SD to Wistar closed colony strain rats. Before the transplantation, the kidney was preserved in 0- 40 ℃ heparin sodium chloride solution for one h. The rats were divided into 4 groups: Group A only received CsA 10 mg/kg every day; Groups B, C and D received low dose of Yisheng+CsA, high dose of Yisheng+CsA and CsA+MMF, respectively. Blood creatinine, pathological changes of the renal grafts and the expression of transforming growth factor ?1 in transplanted renal tissues were observed 4, 8 and 12 weeks after operation.Results After the transplantation, the serum creatinine level in group A was significantly higher than in groups B, C and D ( P
5.Hemodynamic effects of open-loop and closed-loop target controlled infusion of propofol and fentanyl for general anesthesia induction

Objective To observe the hemodynamic effects of open-loop target controlled infusion (OLTCI) and closed-loop target controlled infusion (CLTCI) of propofol and fentanyl for general anesthesia induction. Methods Twenty-four female patients with ASA grade I-II who were performed thyroidectomy were randomly allocated into two groups: OLTCI group and CLTCI group(n=12). In OLTCI group, anesthesia induction and maintenance were performed with propofol and fentanyl at the target concentrations of 3 ?g/mL and 3 ng/mL, respectively. In CLTCI group, double CLTCI were performed. Titrations of propofol and fentanyl were guided with bispectral index (BIS) and product of systolic pressure and heart rate (HR). Initiative concentrations of this closed-loop system were 3 ?g/mL and 3 ng/mL, step-up or step-down concentrations were 0.5 ?g/mL and 0.5 ng/mL, and the highest concentrations were 6 ?g/mL and 5 ng/mL, respectively. HR, mean arterial pressure (MAP), HR variability, BIS value and the dosages of propofol and fentanyl in various time of the two groups were recorded. 6.EFFECT OF GUIDED RELAXATION METHOD WITH MUSIC ON PAIN SENSITIVITY IN AC-UPUNCTURE

Objective To detect the effect of Guided Relaxation Method with Music on pain sensitivity and hemodynamics in acupuncture.Methods 70 volunteers aged 18 to 22, all in good condition, were randomized to study group or control group (each 35).In the control group, volunteers were only treated with acupuncture in a routine way while volunteers in the study group were treated with acupuncture using Guided Relaxation Method with Music .Comparing the change of heart rate and blood pressure before and after the experiment, and the pain sensitivity between these two groups.Results By comparing the two groups, it showed that there were no significant differences in heart rate, diastolic blood pressure and systolic blood pressure level(p>0.05).Numerical rat-ing scale scores in study group was much lower (p<0.001), and systolic blood pressure level after the experiment decreased more significantly than that in the control group (p<0.05).However, no significant differences can be seen in heart rate and diastolic blood pressure level after the experiment (p>0.05).Conclusion Guided Relaxation Method with Music could help to alleviate the feeling of pain, and make acupuncture therapy more acceptable.
7. Masking Efficiency and Regularity of Bitterness Suppressants to Berberine Hydrochloride Based on Tongue Taste and Electronic Tongue Taste

OBJECTIVE: To explore the masking efficiency and regularity of bitterness suppressants (BS) to a representative bitterness substance berberine hydrochloride (BBR). METHODS: Some quantitative evaluation methods of BS efficiency were established based on relevant literature and our previous research. Efficiency of respectively added into eight kinds of BS such as acesulfame, on bitter degree 3 BBR solution (0.05 mg•mL-1) were evaluated using the established methods. Bitterness masking regularity of BS on BBR solution were explained by respectively establishing the relationships between tongue tasted bitter decreased value (ΔI), electronic tongue tasted bitter decreased value (ΔIe) with BS concentration (ρ). RESULTS: ①Five tongue taste bitter masking efficiency (calibrated masking rate, bitterness revised standard half effect index and so on), two electronic tongue bitter masking efficiency(first taste reduce and aftertaste reduce) were established. ②Seven kinds of bitterness suppressants efficacy indicators can be used to compare the bitterness suppressants efficacy of BS to BBR solution, and the maximum bitterness reduction of 8 kinds of BS to BBR solution is above 1.8. ③ΔI-ρ Weibull regularity models of eight BS and nine ΔIe-ρ models of six BS were brlilt.HP-β-CD, glycine and crystalline fructose had good response to two sensor, stevioside, soluble soybean polysaccharide and acesulfame had a good response to one sensor and xanthan gum and aspartame had no response to any sensor. CONCLUSION: Seven evaluation indexes and methods are built based on human tongue and electronic tongue taste method. Max standard capability index of BS is suitable for the development of bitterness suppressants in taste method and two indicators of electronic tongue are also can be used as indicative indicators. Masking regularity of BS to BBR solution is exposed which could provide reference for the development and application of more BS.
8. Detection of epidermal growth factor receptor mutations in small specimens of non-small cell lung cancer by amplification refractory mutation system

Objective: To explore the feasibility of the application of small specimens as alternatives of gross specimens to detect EGFR (epidermal growth factor receptor) mutation in NSCLC (non-small cell lung cancer) by ARMS (amplification refractory mutation system). Methods: Biopsy specimens from 181 cases of NSCLC were collected, in which 157 were small specimens (including specimens acquired through CT-guided percutaneous lung biopsy, endobronchial ultrasound-guided transbronchial needle aspiration, lymph node biopsy, bronchoscopic biopsy and aspiration of pleural effusion) and 24 were gross specimens. QIAGEN DNA extraction kit was used to extract DNAs from NSCLC specimens. Then AmoyDx EGFR Mutation Test Kit - a highly sensitive real-time PCR-based test was used to detect EGFR mutations. The detection rates of EGFR mutations in small specimens and gross specimens were compared by Chi-square test or Fisher's exact test. Results: The total EGFR mutation detection rate of all biopsy specimens was 39.8% (72/181). The detection rates of small specimens and gross specimens were 38.9% and 45.8%, respectively (P = 0.515). Furthermore, EGFR mutation frequencies were significantly higher in non-smokers (P = 0.033) and in patients with adenocarcinoma (P < 0.001). Conclusion: A relatively high detection rate of EGFR mutation in NSCLC small specimens can be obtained through ARMS. For patients with advanced NSCLC whose gross specimens cannot be easily obtained, the alternative small specimens can be used in clinical EGFR mutation detection. Copyright © 2012 by TUMOR.
9.Effect of ceramide on GSTA1 in Caco-2 cells.

This study is to investigate the effects of ceramide on GSTA1 expression in Caco-2 cells. After being exposed to ceramide for a fixed time, GSTA1 protein expression was detected by Western blotting analysis; GSTA1 mRNA expression was detected by real time PCR; dual luciferase assay was used to analyze GSTA1 transcriptional activity and GSTA1 activity was determined toward androstanedione (AD) as substrate. The data showed that ceramide can significantly induce the expression of protein and GSTA1 mRNA, and increase transcriptional activity and enzyme activity of GSTA1. The results demonstrated that ceramide may increase resistance to chemotherapeutics in Caco-2 cells by up-regulating the expression of GSTA1.
10.Comparative study of external dissection & internal ligation versus Milligan-Morgan haemorrhoidectomy for mixed hemorrhoids

Objective To study the efficacy of external dissection & internal ligation for beating mixed hemorrhoids, and compare with Milligan-Morgan. Methods One hundred and twenty-six patients with mixed hemorrhoids were divided into two groups: treatment group(66 cases)and controlled group(60cases).In treatment group used external dissection & internal ligation for treating mixed hemorrhoids, but in eontrolled group used Milligan-Morgan. Then the differences between both groups in healing time of wound surface, as well in the statuses of bleeding, pain, edema, anus stenosis, and anus overflow liquid were observed. Results Averaged healing time in treatment group was significantly shorter than that in controlled group, (8.2±2.6)days vs (17.4±3.8)days, P<0.01.The anus stenosis and anus overflow liquid were significantly less in treatment group than those in controlled group, P<0.01.Conclusion External dissection & internal ligation of hemorrhoids for treating mixed hemorrhoids decreases anal transitional zone(ATZ)disorganization, shorten healing time, and significantly decreases crissum scar,protectes anus function effectively.
