BACKGROUND: Intestinal mucosa was damaged by ischemia and hypoxia during severe burn, and injury and infection of oxygen-derived free radicals during reperfusion.OBJECTIVE: To study on the basis of sub-cell level whether danshen can improve respiratory function of mitochondrion of epithelial cell in intestinal mucosa and decrease the production of oxygen-derived free radicals so as to protect intestinal mucosa of burnt rats.DESIGN: Randomized controlled animal study with single sample.SETTING: Department of Burn, the Third Hospital of Chinese PLA.MATERIALS: The experiment was conducted at the Surgery Institute of the Third Hospital of Chinese PLA from December 2001 to February 2002.Totally 96 SD rats of clean grade were randomly divided into normal control group (n=24), burning model group (n=36) and danshen treatment group (n=36). 1 mL danshen parenteral solution had 1.5 g raw materials.METHODS: ① Rats in the burning model group and danshen treatment group were used to establish grade Ⅲ burning models with 20% scald of total body surface. ② Rats in the normal control group were not injured.③ After modeling, 1 mL/kg danshen parenteral solution was slowing injected into rats in the danshen treatment group through femoral vein, but saline was injected slowly into rats in the normal control group and burning model group. ④ Twelve rats from each of the burning mgdel group and the danshen treatment group were sacrificed at 1, 2 and 6 hours after modeling, and 8 rats in the normal control group were sacrificed at relevant time points respectively. ⑤ Samples of small intestine were collected to measure cytochrome aa3, cytochrome C, level of energy charge and activity of superoxide dismutase of mitochondrion of epithelial cells in intestinal mucosa.MAIN OUTCOME MEASURES: Cytochrome aa3, cytochrome C, level of energy charge and activity of superoxide dismutase of mitochondrion of epithelial cells in intestinal mucosa 1, 2 and 6 hours after modeling.RESULTS: Totally 96 SD rats entered the final analysis. ① Level of cytochrome aa3 of mitochondrion of epithelial cells in intestinal mucosa at various time points after modeling: Levels of cytochrome aa3 did not changed obviously in the burning model group and danshen treatment group 2 hours after modeling as compared with that in the normal control group (P＞ 0.05). Six hours after burning, levels in the burning model group were obviously lower than those in the normal control group (P ＜ 0.05), but those in the danshen treatment group were obviously higher than those in the burning model group [(3.16±0.13), (2.5640.15) μkat/g, P ＜ 0.05]. ②Measurements of cytochrome C, level of energy charge and activity of superoxide dismutase of mitochondrion of epithelial cells in intestinal mucosa at various time points after modeling: As compared with those in the normal control group, measurements were decreased obviously in the burning model group 1, 2 and 6 hours after modeling (P ＜ 0.05 or 0.01), but those in the danshen treatment group were obviously higher than those in the burning model group (P ＜ 0.05 or 0.01).CONCLUSION: Danshen can raise the levels of cytochrome aa3, cytochrome C, energy production and SOD, and also reduce the production of oxygen-derived free radicals so as to improve respiratory function of mitochondria of epithelial cells in intestinal mucosa of rats.

Objective To investigate the expression and significance of osteopontin (OPN) and monocyte chemoattractant protein-1 (MCP-1) in lipid -induced rat renal injury. Method Wistar rats were randomly divided into two groups: normal control rats (C group) and cholesterol fed rats (H group). Twelve weeks later, the 24h total urine protein amount, creatinine clearance rate and the levels of total serum cholesterol, LDL- cholesterol, HDL- cholesterol and triglycerides were measured. Kidney pathological changes were observed by HE and PAS statning. Immunohistochemitry were used to detect the expression of OPN, MCP-1 and ED1. Results Total serum cholesterol level, LDL-cholesterol level and 24h total urine protein amount in H group rats were higher than those in C group rats, and there were no significant difference between two groups in HDL-cholesterol and triglycerides level. Compared with C group rats, the expression of OPN and MCP-1 increased in cortical tubular epithelium of H group rats, and the number of ED1 + cells increased in tubulointerstitium as well. Either OPN or MCP-1 expression was positive correlation with the number of ED1 + cells. Conclusion The expression of OPN and MCP-1 in tubular epithelium increased in hypercholesterolemia rats, which might mediate M? infiltration into tubulointerstitium and lead to interstitial fibrosis.

This study primarily compares the characteristics and the application range of some kinds of bioactive ceramics and biodegradable ceramics, and introduces their latest development in degradability and strength et al. Among these materials, calcium polyphosphate is a new material as bone tissue engineering scaffold. With a controllable degradation rate and the required strength, it has been paid much attention.

Objective To explore the differences in the expression of inhibin(INH)receptors and activin (ACT)receptors in the follicular/luteinic phase in normal human ovaries and their relationship with female endocrine hormone levels.Methods Real time PCR and immunohistochemistry were used to determine the expression of inhibin receptors(INHR)genes,activin receptors(ACTR)genes.Serum estradiol(E2),follicle stimulating hormone(FSH),luteinizing hormone(LH),INHB,ACTA levels were determined by a solid quantitative sandwich enzyme immunoassay technique(Sandwich ELISA)in 21 women during follicular phase and another 21 women during luteinic phase,the correlations between each gene and each hormone were analyzed.Results(1)ACT type Ⅰ and Ⅱ receptors genes(ACTR Ⅰ A,ACTR Ⅰ B,ACTRⅡA,ACTR Ⅱ B)and INH receptor β-glycan genes were expressed higher in the follicular phase than in the luteinic phase:ACTR Ⅰ A(0.50±0.17 vs 0.36±0.18;P＜0.05),ACTR Ⅰ B(0.050±0.019 vs0.036±0.020;P＜0.05),ACTRⅡ A(0.10±0.04 vs 0.07±0.04;P＜0.05),ACTR Ⅱ B(0.28±0.10vs 0.19±0.11;P＜0.05),β-glycan(0.26±0.10 vs 0.17±0.09;P＜0.01).(2)The intensities of ACTR I A,ACTR Ⅱ A,β-glycan immunostaining in human normal ovaries in the follicular phase were significantly stronger compared to those in luteinic phase.In the follicular phase β-glycan expression was positively correlated with serum E2,FSH,LH,INHB levels.The correlation coefficient was 0.53(P＜0.05).0.74(P＜0.01),0.85(P＜0.01)and 0.76(P＜0.01)respectively.Conclusion In normal human ovary in the follicular phase INH and ACT bind their receptors and down-regulate or up-regulate FSH,thus influencing the follicular development.

BACKGROUND:Systemic lupus erythematosus (SLE) is classified into four types, and the major treatment is to tonify kidney and nourishyin, clear blood stasis and toxin by the traditional Chinese medicine (TCM). Even though, there are stil many patients with poor efficacy. Mesenchymal stem cels have the capacity of multiple differentiation, hematopoietic support and immune regulation, thus having been used for the treatment of refractory, recurrent SLE and achieving good effects. OBJECTIVE:To investigate the therapeutic effect of umbilical cord-derived mesenchymal stem cel transplantation on SLE patients with different patterns of syndromes. METHODS: Twenty-one SLE patients were clustered to four syndrome types of TCM, including heat-toxin,yin deficiency of liver and kidney,yang deficiency of spleen and kidney, andqi stagnation and blood stasis. The changes in clinical and laboratory indicators were analyzed statisticaly before and after cel transplantation. RESULTS AND CONCLUSION:The level of 24-hour proteinuria and SLE disease activity index scores in SLE patients were significantly decreased at 1, 3, 6 months after cel transplantation (P < 0.01). Umbilical cord-derived mesenchymal stem cel transplantation could significantly reduce the 24-hour proteinuria in SLE patients withyin deficiency of liver and kidney at 1, 3 and 6 months (P < 0.01), while slightly reduce the 24-hour proteinuria in SLE patients with heat-toxin andqi stagnation and blood stasis at 1, 3 months (P < 0.05) as wel as in SLE patients withyang deficiency of spleen and kidney at 1 month (P < 0.05). Additionaly, umbilical cord-derived mesenchymal stem cel transplantation could increase the serum albumin levels in al the SLE patients (P < 0.01), although the changes in patients with heat-toxin were moderate (P < 0.05). Al the SLE patients of four types had an increasing trend of their platelet counting after cel transplantation, but there was no statistical difference before and after cel transplantation. Taken together, umbilical cord-derived mesenchymal stem cel transplantation is effective for treatment of SLE, but has different therapeutic efficacy on SLE patients with different syndrome types of TCM.

Objective To explore the method for repair of total avulsion of 2~4 fingers and to find the best procedure. Methods The wrap-round flap with part toenail combind with the dorsal pedical flap to repair soft tissue defect of the dorsal of finger and part of the palmar of finger, the second simepulp flap to repair soft tissue defect of the palmar of finger,transfer of two tissues with one pedicle to repair total avulsion of 2~4 fingers. Results All of 9 composite transplants in 6 cases were survived,the received areas were primary healing in 8 finger,1 case repaired by the bilateral wrap-round flaps and second simepulp flaps for index and middle finger,the distal phalanx of middle finger necrosis because of dislocation of DIP,3 months later,injury healed after the necrosis of phalanx excised,but the nail and the nail bed were dead.Except 1 case which the grafted skin necrotized in the donor area of the big toe,the rest had primary healing.All cases were followed up 6~14 months,the activity of joints were: MP 0-70,PIP 0-40.The repaired pulps had excellent appearance,the nail was fine,1 finger's mail was atrophy and 1 finger's nail was defect.The donor area had good fountion without pain and edema,the grafted skin had normal color and the activity of joints were nomal. Conclusion The wrap-round flap with part toenail combined with the second simepulp flap is a good approach in treatment of the skin avulsion injury in the finger.

This study is to investigate the protein and mRNA expressions of pro-inflammatory and anti-inflammatory cytokines in U937 foam cells and effects of Ginkgo biloba extract (GbE) on the cytokines.U937 cells were cultured with different concentrations of GbE (0.1,1,and 10 μg·L-1),and stimulated by 100 mg·L-1 oxidized low density lipoprotein (ox-LDL) for 24 h.The expressions of interleukin-1β (IL-1β),tumor necrosis factor-α (TNF-α) and interleukin-10 (IL-10) in culture solution were detected by enzyme-linked immunosorbant assay (ELISA) and reverse transcriptase polymerase chain reaction (RT-PCR).The results showed that incubated with 100 mg·L-1 ox-LDL for 24 h,the U937 cells became foam cells,the protein or mRNA expressions of IL-1β,TNF-α,IL-10,and its receptor IL-10R in U937 foam cells were higher markedly than those in normal U937 cells.When the cells were pretreated with GbE (0.1,1,and 10 μg·L-1),the increases of IL-1β and TNF-α in U937 foam cells were remarkably inhibited,but IL-10 expression increased greatly.Especially when cells were pretreated with 10 μg·L-1 GbE,the protein and mRNA expressions of IL-1β and TNF-α were markedly lower than those in U937 foam cells.The protein expression of IL-10 and mRNA expressions of IL-10 and its receptor IL-10R were markedly higher than those in U937 foam cells.GbE inhibited production of pro-inflammatory cytokines IL-1β and TNF-α,but up-regulated the production of anti-inflammatory cytokine IL-10 and its receptor IL-10R in U937 foam cells,which might be related with its anti-atherosclerotic actions.
