1.Research progress of targeting PD-1/PD-L1 in immunotherapy for non-small-cell lung cancer

Lung cancer is the leading cause of cancer-related death in the world.PD-1 is a member of the CD28 family expressed on the surface of a variety of immune cells.PD-L1, one of PD-1 ligands, plays a pivotal role in the ability of tumor cells to evade the host immune system.A recent study showed that signaling via mutant EGFR in lung tumor cells upregulates directly tumor PD-L1 expression.In this review, the clinical significance of the PD-1/PD-L1 pathway in immunotherapy for non-small-cell lung cancer (NSCLC) and the relevance to targeted therapy were summarized.
2.Advances of programmed death ligand 1 and its inhibitor in non-small cell lung cancer

Programmed death-ligand 1 (PD-L1 ),which is highly expressed in NSCLC,can be divided into two categories:membrane PD-L1 and soluble PD-L1 .PD-L1 participates in tumor immune escape through combining with its receptor.PD-L1 immune checkpoint inhibitors have entered the phase Ⅰ studies and showed a good application prospect.It is also found that the sensitivity of PD-L1 immune checkpoint inhibitors is strongly associated with the expression of PD-L1 in tumor.Therefore,PD-L1 can be used as a biomarker to pre-dict its curative effect.
3.Change of Cytochrome Oxidase Expression in Type Ⅱ Alveolar Epithelial Cell of Rats Exposed to Hyperoxia

Objective To observe the expression of mitochondrial encoding cytochrome oxidase subunitⅠ(COXⅠ) and subunitⅡ(COXⅡ) in type Ⅱalveolar epithelial cell(AEC Ⅱ) of rats exposed to hyperoxia and explore the role of COXⅠand COXⅡ in hyperoxia-induced lung injury.Methods AECⅡ were gained by primary culture from 19-days fetal rats lung.After purified,AECⅡ were randomly assigned to hyperoxia group and air group.Hyperoxia group was flushed the flake with 950 mL/L O2 and 50 mL/L CO2 at 3 L/min for 10 min,then sealed.Both groups were in CO2 culture chamber(37 ℃,50 mL/L CO2).After 6,12 and 24 hours of exposure,AECⅡ were harvested and extracted for total RNA.COXⅠand COXⅡ mRNA were detected by reverse transcription polymerase chain reaction(RT-PCR).The results were analyzed by SPSS 12.0 softwore.Results Compared with air group,COXⅠmRNA expression in hyperoxia group increased significantly at 6 hours(t=3.832 P=0.019) and 12 hours(t=10.431 P=0),respectively,then decreased to the equivalent level in 24 hours(t=0.360 P=0.731).Compared with air group,COXⅡmRNA expression in hyperoxia group increased significantly at 6 hours(t=2.795 P=0.035),then decreased to the equivalent level at 12 hours(t=0.892 P=0.40) and 24 hours(t=2.018 P=0.071).Conclusions Exposure of hyperoxia up-regulate the expressions of COXⅠmRNA and COXⅡmRNA in AECⅡ,which may be a protective mechanism of hyperoxia-induced lung injury.
4.Application value of monoenergetic-plus imaging in the diagnosis of early gastric cancer

Objective To explore the value of dual-source CT monoenergetic-plus imaging in the diagnosis of early gastric cancer. Methods Imaging data of 2 6 cases with pathologically proven early gastric cancer who underwent dual-energy scanning were analyzed retrospectively.Monoenergetic-plus images with energy levels at 40 keV,50 keV,60 keV and 70 keV were reconstructed by using an advanced monoenergetic software (syngo.via.VA30A,Dual energy Mono+).Contrast-to-noise ratio (CNR)of the lesion,image noise,overall image quality score and lesion conspicuity score were com pared between different monoenergetic-plus images and conventional polychromatic images at 1 2 0 kVp.Results The CNRs (3 .2 4 ± 0 .7 6 ,2 .9 4 ± 0 .9 2 ,2 .4 7 ± 0 .7 9 ,2 .3 7 ± 0.98 and 1.22±0.40 respectively)and image noises [(32.7±1.82)HU,(25.6±2.61)HU,(20.30±2.1 7)HU,(1 7.03±2.1 5)HU and (18.15±2.12)HU respectively]among 40 keV,50 keV,60 keV,70 keV and 120 kVp groups all had statistical differences (F =24.654 and 227.05,P <0.01).The CNR in 50 keV group was higher than that in 60 keV,70 keV and 120 kVp groups,but similar to that in 40 keV group (P >0.05).The SD value in 50 keV group was lower than that in 40 keV group,but higher than that in other three groups (P <0.05).The overall image quality score (3.08±0.27,3.65±0.49,3.31±0.47,3.23±0.43 and 3.1 5±0.37,respectively) and lesion conspicuity score (4.15±0.67,3.92±0.48,3.27 ±0.45,3.08±0.27 and 3.04±0.20)among the five groups all had statistical differences (Z =25.343 and 70.426,P <0.05).The overall image quality score of 50 keV group was higher than that of the other four groups (P <0.05).Lesion conspicuity score in 50 keV group was higher than that in 60 keV,70 keV and 120 kVp groups and similar to that in 40 keV group (P =0.1 52).Conclusion The 50 keV monoenergetic-plus images can increase the CNR value of early gastric cancer while maintaining the overall image quality.
5.Preparation and Characterization of Monoclonal Antibody Specific to PfCP-2.9 Chimeric Protein of Plasmodium falciparum

Objective To prepare and characterize monoclonal antibody against a malaria vaccine candidate, PfCP-2.9 chimeric protein of Plasmodium falciparum. Methods BALB/c mice were immunized with PfCP-2.9,and the spleen cells were used for fusion with SP2/0 cells. The monoclonal antibodies were analyzed by ELISA,Western blotting as well as growth inhibition assay. Result A monoclonal antibody was obtained. It interacted with the PfCP-2.9 recombinant protein by ELISA and Western blotting. The interaction of the monoclonal antibody with the protein was reduction-sensitive,indicating that the antibody recognized a conformational epitope. Moreover,the antibody also recognized the cultured parasites of P.falciparum by indirect immunofluorescent antibody test(IFA). When tested by growth inhibition assay,the antibody significantly inhibited parasite growth in vitro of 56% inhibition rate at the antibody concentration of 0.3 mg/ml. Conclusion A monoclonal antibody against PfCP-2.9 malaria vaccine candidate has been obtained,which recognizes a conformational epitope of the protein and natural protein.
6.A road to new healthy countryside:Policy design and initial effects of China's healthy rural re-construction

Healthy rural reconstruction is a widely explored intervention implemented by China Rural Health Devel-opment Project with the support of the World Bank and UK Department for International Development. By using initial pro-ject baseline data survey, mid-term evaluation and supervision, terminal evaluation of acquired qualitative and quantitative data,this paper analyzes the intervention policy design and initial effects of China's healthy rural reconstruction. With transforming the living environment in rural areas as the starting point, the project has relatively achieved good results, by using different policy measures such as improving the environment, creating a social environment conducive to villagers' health, expanding and providing high-quality health services, foster healthy behaviors and lifestyles, etc. The main results include the expansion of the project to cover 745 administrative villages, the environment of healthy villa-ges is much better than other neighboring villages, villagers' health knowledge has been greatly increased, lifestyle be-haviors of villagers have improved, the attitudes and participation rate of residents in cultural and sports activities have improved, etc. This paper suggests implementing the healthy rural reconstruction nationwide. It is very important to in-tegrate the concept of healthy village throughout the connotation of all kinds of new rural reconstruction, so the concept of health is deeply rooted in rural areas and imperceptibly change people's living behaviors and habits.
7.Management and control of hypertension and diabetes in rural areas of China: Based on the practice of China Rural Health Project

With the support of World Bank ( WB ) and UK Department for International Development ( DFID) , China Rural Health Project ( hereinafter referred asHealth XI Project) has successfully covered 40 coun-ties in 8 provinces. With the establishment of community diagnosis and health records as the entry point, hyperten-sion, diabetes and other major chronic diseases as the starting point, and focus on the needs of healthy people, high-risk groups and patients, the project mainly adopts health education and promotion, health management, disease management and other measures to explore the establishment of a new model of rural chronic disease management. By analyzing the monitoring data of chronic diseases in the project zones, this study found that the number of registered and managed patients with hypertension and diabetes increased significantly, from 397,113 and 136,326 in 2009 to 1,500,252 and 388,846 in 2013, respectively. The management rate also increased from 60. 8% and 32. 2% in 2009 to 92. 2% and 88. 8% in 2013, respectively. The results of the 5th National Health Service Survey show that, the control rates for self-reported hypertension and diabetes ((53. 8% and 50. 2%, respectively) in the project zones were significantly higher than the national average in rural areas ( 54 . 9% and 38 . 3%, respectively ) . This paper suggests that, with focus on training, health education, health promotion, health management and disease manage-ment as the core mainline, the chronic disease management model has effectively improved the chronic disease service capabilities in rural areas,. The comprehensive and integrated chronic disease interventions implemented by the pro-ject in the rural areas is practical, and it has value of popularization and application.
8.Development of Teachers Ability and Teaching Bases for Rehabilitation Therapy

10.Experimental study of biomimetic nanoparticle probe containing hematoporphyrin monomethyl ether and its imaging and sonodynamic therapy for breast cancer

Objective:To develop a biomimetic nanoparticle probe of hematoporphyrin monomethyl ether (HMME) coated with breast cancer cell membrane, to observe its ability to target homologous breast cancer cells in vitro, and to investigate its effect of enhanced photoacoustic imaging and sonodynamic therapy (SDT) for breast cancer in vitro.Methods:The cell membrane of breast cancer 4T1 was extracted by chemical cleavage and repeated freezing and thawing. Then the HMME-coated polylactic acid-glycolic acid copolymer biomimetic nanoparticle was prepared by double emulsification and extrusion. The basic characteristics of nanoparticles were detected. The target ability of nanoparticles to homologous breast cancer cells and the enhancement of photoacoustic imaging were observed in vitro. Singlet oxygen sensor green (SOSG) was used to verify the reactive oxygen species (ROS) production of nanoparticles, and its SDT effect on breast cancer cells was evaluated by CCK8 cytotoxicity assay.Results:The size of the prepared CHP-NPs was uniform, the morphology was spherical "core-shell structure" , the particle size was (275.23±8.25)nm, and the surface potential was (-18.43±0.45)mV. It was observed that CHP-NPs could target homologous 4T1 cells under laser confocal microscopy. In vitro photoacoustic imaging experiments show that the photoacoustic signal of nanoparticles increases with the increase of its concentration. According to SOSG probe detection, CHP-NPs could produce ROS under ultrasonic irradiation.When CHP-NPs was incubated with 4T1 cells alone and no ultrasonic irradiation was used, the cell survival rate was not significantly affected. When the concentration was 0.6 mg/ml, the cell survival rate was still 95%. After ultrasonic irradiation, CCK8 experiment showed that the CHP-NPs had a significant SDT effect on breast cancer cells.Conclusions:The biomimetic nanomolecular probe of breast cancer cell membrane is successfully prepared. The probe has good ability to target homologous tumor, and can significantly enhance tumor photoacoustic imaging and SDT effect.
