2.Effect and mechanism of liraglutide on proliferation and apoptosis of human pancreatic cancer cells

Objective To investigate the effects and mechanism of glucagon-like peptide-1 ( GLP-1 ) receptor agonist liraglutide on the proliferation and apoptosis of human pancreatic cancer cells. Methods The human pancreatic cancer cell line MIA PaCa-2 was incubated for 24 h with liraglutide at various concentrations (10-1 000 nmol/ L), or with 100 nmol/ L liraglutide for various durations (0-72 h). Cell proliferation was determined by Cell Counting Kit-8 (CCK-8) analysis. RT-PCR and Western blot were used to detect the mRNA and protein expression levels of related genes. Results GLP-1 receptor was expressed in the MIA PaCa-2 cells. Liraglutide suppressed cell proliferation, up-regulated the expression levels of pro-apoptotic protein Bax and down-regulated the expression levels of anti-apoptotic protein Bcl2 in human pancreatic cancer cells in a dose- and time-dependent manner. Meanwhile, liraglutide down-regulated the expression levels of insulin receptor (INSR) and insulin-like growth factor-1 receptor (IGF-1R), and the phosphorylation levels of their downstream signaling proteins Erk1 / 2 and Akt, in a dose- and time-dependent way. Conclusion Liraglutide inhibits proliferation and promotes apoptosis of human pancreatic cancer cells; the process may be mediated via suppressing the expression of INSR and IGF-1R and inhibiting activation of the downstream MEK/ Erk1 / 2 and PI3k/ Akt pathways.
3.Application effects of “hands-off” delivery method in puerpera with vaginal delivery

Objective To explore clinical application effects of hands-off method among puerperas delivered through vagina. Methods To utilize retrospective study design, a total of 1 040 puerperas, who could not received natural labour from July to November 2013, were selected as observation group by convenient sampling method and underwent hands-off method to deliver children, while 1 005 prerperas were taken deliver technical protection of perineum from July to November 2010 as control group. We compared puerperas′delivery conditions and injury of perineum, and observed and analyzed the influencing factors of puerpera′s episiotomy and perineal laceration. Results No incidences of Ⅲ and Ⅳ degree perineal laceration and obstetric anal sphincter injury happened. The rate of puerpera′s episiotomy in the observation group was 25. 38% compared with 13.93% in the control group (P <0.01). The Ⅱ degree of perineal laceration was 48. 85% in the observation group lower than 82. 09% in the control group (P<0. 01). Comparing injury of perineum between two groups, the medium injury of observation group (Ⅱdegree of perineal laceration and episiotomy, 74. 23%) was lower than 96. 02% puerperas of control group (P <0. 01). The rate of vaginal wall hematoma in the observation group was 0. 77% lower than 4. 68% in the control group (P<0. 01). The length of second stage of labor and length of perineum wound suture were shorter than those of the control group (P<0. 05);the amount of bleeding 2 h after labor in two groups had no statistical significance (P>0. 05). Regression analysis showed that, maternal age, analgesia, newborn weight were the influence factors of lateral episiotomy (P <0. 05);maternal age, weeks of gestation, analgesia, newborn weight were the influence factors of perineal laceration (P<0. 05). Conclusions The hands-off method to deliver child can elevate the perineal laceration degree of perineum, reduce the length of the second stage labor, and shorten the length of perineal wound suture.
4.Investigation on the career motivations and professional self-concept of nurses in nurses of gynecology and obstetric department in a three-level general hospital

Objective To explore the difference of the professional self-concept in nursing staff of gynecology and obstetric department with the different career motivations in a three-level general hospital.Methods A total of 97 nurses were selected and investigated with professional self-concept scale and career motivation questionnaire.Results Among 97 nurses,44 nurses voluntarily chose the nursing job and their score of professional self-concept and job satisfaction was ( 3.04 ± 0.36),(2.91 ± 0.53 ),respectively.Fifty-three nurses passivity chose the nursing job by external factors and their score of professional self-concept and job satisfaction was(2.89 ± 0.29),(2.49 ± 0.53 ),respectively,and the difference between the two group was statistically significant (t =2.295,3.868,respectively ; P < 0.05 ).Scores of two groups except job satisfaction score in 53 nurses group were more than 2.5 and 2.5 was a level boundary value.Conclusions Nurses with different career motivation have different professional self-concept.It is important for nursing educator to pay attention on cultivating nurse students' professional interest and career motivation.
5.Effect of telephone follow-up on breastfeeding intention of unipara during the puerperium

Objective To explore the effect of telephone follow-up on breasffeeding intention of unipara during the postpartum period.Methods The quasi-experimental method was used and 100 recruited unipara were divided into the control group ( n =50 ) and the experimental group ( n = 50 ).A continuing telephone follow-up was provided by community nurses to the experimental group 1 week,2 weeks,and 4 weeks after delivery,and provided psychological nursing and solution to the difficult of breasffeeding. Unipara breasffeeding intentions were collected when their discharge and 6 weeks after delivery when they received postpartum cheek up in clinics.Then,unipara breastfeeding intentions,attitude,perception behaviour control,and subjective standard were observed.Results During hospital,the average score of breastfeeding intention among 100 unipara was (4.58 ± 0.46 ),the average score of perception behaviour control was ( 3.31 ± 0.46 ),the average score of attitude was (4.29 ± 0.43 ),the average score of subjective standard was ( 3.84 ± 0.42 ),and there was no significantly difference in breastfeeding intention and others between two groups (P >0.05).Six weeks after delivery,the average score of breastfeeding intention,perception behaviour control,subjective standard,attitude in turn was (4.45 ±0.51),(3.24 ±0.59),(3.91 ±0.46),(4.18 ±0.54) ; breastfeeding intention and others between two groups,there was no significantly differences( P > 0.05).The breastfeeding intention in intervention group was maintain higher after six weeks,and no difference was detected when compared with that during hospital (P>0.05),while the average score of attitude in control group was decreased from (4.32 ±0.46) to (4.13 ±0.59),and the difference was statistically significant (P<0.05).Conclusions The telephone follow up provided by community nurses can increase the intention of new mothers' breastfeeding intention by continuing consulting,encouragement and supports.
6.Effects of WeChat-based transitional care on the cognition of pelvic floor rehabilitation in postpartum women

Objective To explore the effect of WeChat-based transitional care on the cognition of pelvic floor rehabilitation in postpartum women, so as to provide a reference for clinical generalization in the future. Methods By convenience sampling, we selected puerpera with childbirth and postpartum reexamination of a class Ⅲ grade A of Beijing from September to October 2016 in the control group (n=47); we selected puerpera with childbirth from December 2016 to January 2017 in the WeChat intervention group (n=47). Puerpera of two groups were all treated with conventional health education in the next day after delivery. Beyond that, puerpera of WeChat intervention group adopted WeChat public platform management. All of them were investigated on cognition of pelvic floor rehabilitation in the next day and 6 to 8 week after delivery. Results The percentage of very unclear cognition of pelvic floor rehabilitation were 38.3% and 6.4% in the control group and the WeChat intervention group respectively 6 to 8 week after delivery with a significant difference (χ2=-2.82,P<0.05). The accuracy of "Childbirth is relevant to pelvic floor muscle injury" and "Incontinentia urinae will show after pelvic floor muscle injury" were all 100% in the WeChat intervention group and 87.2%, 83.0% respectively in the control group with significant differences (P< 0.05). Conclusions WeChat-based transitional care can deepen the cognition of puerpera on the pelvic floor.
7.New approaches to developing high-quality maternal and infant nursing care

Starting from the innovation and diversification of maternal and infant nursing service models, it discusses the challenges faced by China's maternal and infant nursing services in the new era as well as the new approaches to developing high-quality maternal and infant nursing care. These include enhancing the physical and mental health of pregnant and postpartum women and promoting the transformation and application of evidence-based evidence in maternal and infant nursing practice, among others.
8.The Relationship Study between Expressions of P2X5 Receptor and Deficiency-cold Syndrome/Deficiency-heat Syndrome at Various Ambient Temperatures.

OBJECTIVETo detect the expression of the peripheral blood P2X5 receptor at various ambient temperatures, and to explore its relationship with deficiency-cold syndrome and deficiency-heat syndrome.
METHODSSubjects were selected by questionnaire and expert diagnosis, and assigned to the normal control group, the deficiency-cold syndrome group, and the deficiency-heat syndrome group, 20 in each group. 5 mL venous blood was collected at room temperature (25 °C) and cold temperature (-4-5 °C) respectively. Then the expression of P2X5 receptor was relatively quantified by real-time fluorescence quantitative PCR, and compared at room temperature and cold temperature respectively.
RESULTSThe expression of P2X5 receptor in deficiency-cold syndrome and deficiency-heat syndrome groups was lower than that in the normal control group at room temperature (P < 0.05). It decreased more at cold temperature in the deficiency-cold syndrome group than in the normal control group (P < 0.01) as well as in the deficiency-heat syndrome group (P < 0.05). The expression of P2X5 receptor showed no difference in all groups at two different temperatures (P > 0.05).
CONCLUSIONSThe expression of P2X5 receptor was different in different syndrome groups at various ambient temperatures. Ambient temperatures had insignificant effect on the expression of P2X5 receptor of the population with the same syndrome.

9.Effect of various ambient temperatures on activities of mitochondrial complex II in patients of deficiency-cold syndrome and deficiency-heat syndrome.

OBJECTIVETo explore activity laws of mitochondrial complex II in patients of deficiency-cold syndrome (DCS) and deficiency-heat syndrome (DHS) under various ambient temperatures.
METHODSSubjects were recruited by questionnaire and expert diagnosis from grade 1 - 3 undergraduates at Henan College of Traditional Chinese Medicine in November 2012, and assigned to a normal control group, the DCS group, and the DHS group, 20 in each group. Their venous blood samples were collected at two different temperature conditions. Activities of mitochondrial complex II were measured by spectrophotometry.
RESULTS(1) Comparison of mitochondrial complex It under various ambient temperatures: Compared with room temperature in the same group, activity values were all increased in the normal control group at cold temperature with significant difference (P <0.05), but there was no significant difference in the DCS group and the DHS group (P >0. 05). Compared with the normal control group, activity values of complex H were reduced in the DCS group at cold and room temperatures with significant difference (P <0.05). Compared with the DCS group, activity values of complex It were increased in the DHS group with significant difference (P <0. 05). (2) Changes of adjustment rates: Compared with room temperature, the adjustment rate all rose at cold temperature in the normal control group and the DHS group with significant difference (P <0.05), but with no significant difference found in the DCS group (P >0. 05). Compared with the normal control group at the same temperature, the adjustment rate in the DHS group and the DCS group was all reduced at cold and room temperatures with significant difference (P <0. 05). There were no significant difference in the adjustment rate between the DHS group and the DCS group (P > 0. 05).
CONCLUSIONSEnvironment temperature can affect the activity of mitochondrial complex II with different influence degrees on different syndrome types of people, but its change trend are basically identical.

10.Prognostic influencing factors of acute myocardial infarction patients performed emergency percutaneous coronary intervention supported by IABP

Objective To investigate the effect of prognostic factors and nursing care in of acute myocardial infarction(AMI)patients performed emergency percutaneous coronary intervention supported by IABP.Methods Twenty seven AMI patients treated with emergency PCI,supporting by intra-aortic balloon pump(IABP).We analyzed retrospectively elinical characters and situation of interventional treatment of the patients.Results Among 27 cases.there were 12 cases who were recovered,4 suffered congestive heart failure repeatedly.and ll died during the hospitalization.There was no difference in age,sex,the history of hypertension,diabetes and smoking,the site of infarction and ejection fraction of left ventficular.The prognosis was different in the patients with grade of Killip in cardiac function and no flow post PCI(P<0.05).Conclusions The grade of Killip and no flow post PCI were the effective factors for prognosis in Acute Myocardial patients with emergency PCI.
