1.Vascular endothelial growth gactor and its receptor expression in mandibular fracture healing

Objective:To study the expression of vascular endothelial growth factor(VEGF) and its receptor Flt1 in the process of mandibular fracture healing.Methods:The fracture models were made in the middle of the left mandible of 25 rabbits,then the specimens from the fracture site were taken 48 hours and 1,3,5 and 8 weeks after operation respectively. The expression of VEGF and Flt1 was detected by immunohistochemistry and in situ hybridization.Results:VEGF mRNA, VEGF and Flt1 were detected in many kinds of cells in the fracture site from 48 hours to 8 weeks after fracture, and high expression of VEGF and Flt1 were detected from 1 to 3 weeks after facture.Conclusions:VEGF and Flt1 are expressed through the process of fracture healing, VEGF may play important role in the process of blood vessel rebuilding after fracture.
2.Effects of lactated Ringer's solution on ischemic myocardium during optical coherence tomography

The examinations of optical coherence tomography (OCT) images were obtained in 136 patients. Lactated Ringer's solution or physiological saline was used randomly as perfusate. Their electrocardiogram and cardiac enzymes were acquired to compare the safety and image quality of two perfusates.The incidence of ventricular arrhythmia and rise of cardiac enzymes were lower in the lactated Ringer's solution group than those in the physiological saline group.Yet the image quality of OCT had no difference.The use of lactated Ringer's solution as perfusate instead of physiological saline in OCT examination may protect the ischemic myocardium and reduce the incidence of complications.

Objective:To investigate the effect of human p27~(kip1) gene on the proliferation of human tongue squamous cell carcinoma(TSCC) Tca8113 cells.Methods:Human p27~(kip1) gene was cloned from normal tissue and reconstructed into eukaryotic expression vector pcDNA3, the recombinant plasmid was transfected into Tca8113 cells with liposome.The integration and expression of exogenous objective gene in the target cells were examined by PCR and flowcytometry,the growth of transfected cells and non-transfected cells was investigated by MTT assay and flowcytometry.Results:p27~(kip1) cDNA was successfully cloned and reconstructed into vector pcDNA3.The gene was trasfected into Tca8113 cells and stable transfection was achieved by screening with G418.After p27~ kip1 transfection the cell proliferation was inhibited,proliferation index decreased(P

BACKGROUND:Tougu Xiaotong capsule is the clinical prescription for the treatment of osteoarthritis in Fujian University of Traditional Chinese Medicine, and the previous studies mainly focus on effect to cartilage.
OBJECTIVE:To observe the effect of Tougu Xiaotong capsule on the proliferation and differentiation of osteoblasts as wel as the expressions of bone remodeling correlated factors.
METHODS:Rat osteoblast-like cel line ROS17/2.8 cel s were incubated with Tougu Xiaotong capsule. The ROS17/2.8 cel s were divided into blank control group and Tougu Xiaotong capsule groups with different
concentrations. The cel proliferation was determined by methylthiazolyldiphenyl-tetrazolium bromide assay. Osteoblast differentiation biomarkers alkaline phosphatase activity, osteocalcin and bone mineralized nodules were measured with colorimetry, enzyme-linked immunosorbent assay and alizarin red staining, respectively. The real-time PCR and enzyme-linked immunosorbent assay were used to detect the expressions of bone remodeling factors osteoprotegerin/receptor activator of nuclear factorκB ligand.
RESULTS AND CONCLUSION:Compared with the control group, the Tougu Xiaotong capsule with the
concentration of 0.25-2 g/L could significantly promote the ROS17/2.8 cel proliferation (P<0.05), up-regulate the alkaline phosphatase activity, osteocalcin expression level and mineralized nodules area, and increase the
percentage of bone remodeling factors osteoprotegerin/receptor activator of nuclear factorκB ligand (P<0.05). The mechanism of Tougu Xiaotong capsule protecting osteoarthritis may partly result from the regulation of
proliferation and differentiation of osteoblasts and bone remodeling.
5.Prediction of hemorrhagic transformation after reperfusion therapy in patients with acute ischemic stroke

Reperfusion therapy, including intravenous thrombolysis and endovascular therapy, is an effective treatment for acute ischemic stroke. As a common complication after reperfusion therapy, hemorrhagic transformation is closely associated with the poor outcomes of patients. Early evaluation of clinical features, imaging and blood biomarkers can predict the risk of hemorrhagic transformation, so as to better guide the reperfusion treatment in patients with acute ischemic stroke.
6.Can CD44+/CD24- Tumor Cells Be Used to Determine the Extent of Breast Cancer Invasion Following Neoadjuvant Chemotherapy?.

PURPOSE: To investigate the distribution of CD44+/CD24- cells in breast cancers in relation to tumor size before and after the administration of neoadjuvant chemotherapy. METHODS: CD44+/CD24- tumor cells obtained from breast cancer specimens were characterized in vivo and in vitro using tumor formation assays and mammosphere generation assays, respectively. The distribution of CD44+/CD24- tumor cells in 78 breast cancer specimens following administration of neoadjuvant chemotherapy was also evaluated using immunofluorescence assays, and this distribution was compared with the extent of tumor invasion predicted by Response Evaluation Criteria in Solid Tumours (RECIST). RESULTS: In 27/78 cases, complete remission (CR) was identified using RECIST. However, 18 of these CR cases were associated with a scattered distribution of tumor stem cells in the outline of the original tumor prior to neoadjuvant chemotherapy. After neoadjuvant chemotherapy, 24 cases involved cancer cells that were confined to the tumor outline, and 21 cases had tumor cells or tumor stem cells overlapping the tumor outline. In addition, there were 6 patients who were insensitive to chemotherapy, and in these cases, both cancer cells and stem cells were detected outside the contours of the tumor volume imaged prior to chemotherapy. CONCLUSION: CD44+/CD24- tumor cells may be an additional parameter to evaluate when determining the extent of breast cancer invasion.

8.Effects of Electroacupuncture at Shenting and Baihui on Ability of Learning and Memory, and LIM-domain Containing Protein Kinase 1 Phosphorylation in Rats with Cerebral Ischemia-reperfusion

Objective To investigate the effect of electroacupuncture (EA) at Shenting (DU24) and Baihui (DU20) acupoints on hippocampal synaptic plasticity and learning and memory function, and its possible mechanism in rats with cerebral ischemia-reperfusion. Methods Thirty-two male adult Sprague-Dawley rats were randomly divided into sham group (n=8), model group (n=8), electroacupuncture (EA) group (n=8) and non-acupoint group (n=8). The model group, EA group and non-acupoint group were performed with left middle cerebral artery occlusion (MCAO). The EA group received electroacupuncture at Shenting and Baihui, while the non-acupoint group received electroacupuncture at two fixed non-acupoints from bilateral ribs, for fourteen days. Then they were tested with Morris Water Maze test, and their synaptic structure of hippocampal neurons was observed with transmission electron microscope. The level of total LIM-domain containing protein kinase 1 (T-LIMK1) and LIM-domain containing protein kinase 1 phosphorylation (p-LIMK1) were detected with Western blotting. Results Compared with the sham group, the latency increased (t>6.789, P<0.01) and the frequence crossing platform decreased (t= 8.695, P<0.001) in the model group, while the number of synapse in hippocampal neurons decreased, and the level of T-LIMK1 (t=7.568, P< 0.01) and p-LIMK1 (t=8.874, P<0.001) decreased. Compared with the model group, the latency decreased (t>4.938, P<0.01) and the frequence crossing platform increased (t=- 7.891, P<0.001) in the EA group, while the number of synapses increased, and the level of T-LIMK1 (t=-6.473, P<0.01) and p-LIMK1 (t=-6.579, P<0.01) increased. There was no significant difference between the the non-acupoint group and the model group in all the indices (P>0.05). Conclusion Electroacupuncture at Shenting and Baihui can improve the ability of learning and memory in rats with cerebral ischemia-reperfusion, which may relate with the increase of LIMK1 phosphorylation and hippo-campal synaptic plasticity.
