1.Methylation status and mRNA expression of caspase-8 in human hepatocellular carcinoma cells resistant to tumor necrosis factor-related apoptosis-inducing ligand

Objective To study the association of methylation status of C5 of the cytosine in the CpG di-nucleotide of caspase-8 promoter and expression of caspase-8 mRNA with the resistance to tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) in human hepatocellular carcinoma cell lines,and to evaluate the effect of demethylation agent 5-Aza-2′-deoxycytidine(5-Aza-CdR) on the resistance to TRAIL of human hepatocellular carcinoma cell lines.Methods Methylation status of caspase-8 promoter was measured with methylation-specific PCR method(MSP).Expression of caspase-8 mRNA was detected with RT-PCR.Apoptosis induced by TRAIL was observed by Acridine Orange/Ethidium Bromide(AO/EB) staining.Results Unmethylated status of caspase-8 promoter was found in both HepG2 and SMMC 7721 hepatocellular carcinoma cells.5-Aza-CdR neither up-regulated caspase-8 mRNA expression nor increased the sensitivity of hepatocellular carcinoma cells to TRAIL.Conclusion caspase-8 promoter methylation status and caspase-8 mRNA expression are not related to the resistance to TRAIL.5-Aza-CdR can not increase the sensitivity of hepatocellular carcinoma cells to TRAIL.
2.Research on healthcare management of pregnant women in transient population in Beijing

Objective To establish the healthcare management mode for pregnant women adapting to the characteristics of transient population which can safeguard the maternal and infant health or reduce the maternal mortality rate and prenatal mortality rate. Methods Five suburban communities of Chaoyang District, Haidian District and Fengtai District of Beijing were chosen as intervention group, which received systemic healthcare management of pregnant women in transient population, another five suburban communities were selected as control group. Results Both maternal mortality rate and prenatal mortality rate in intervention group were lower than those in control group. To evaluate the effect of systemic healthcare for pregnant women, an average of four antenatal clinic visits were completed in intervention group and eight visits in control group. There was no statistical difference in occurrence of maternal complications, newborn weight and occurrence rate of neonatal asphyxia between two groups. The expense of hospital vaginal delivery was no more than 1000 yuan in intervention group and more than 1000 yuan in control group, while the outcomes of mothers and neonates in two groups had no significant difference. Conclusion The systemic healthcare and community management mode for pregnant women in transient population can safeguard the maternal and infant health with scientific, practical, and replica-ble significances.
4.EUS in preoperative TNM staging of esophageal carcinoma

Objective To study preoperatively on TNM staging of esophageal carcinoma by endo-scopes ultrasonography ( EUS). Methods Sixty-one patients with esophageal carcinoma were preoperatively staged by EUS. The results were compared with the postoperative histopathological staging according to the new (1997) TNM classification. Results Clinical staging of T subsets by EUS was reliable with an overall accuracy rate of 86. 9% , while that of N subsets was relatively more difficult with an overall accuracy rate of 52. 5% ; sensitivity and specificity of regional lymph nodal metastases were 88. 9% and 23. 5% respectively. Conclusion EUS is relatively an accurate measure in assessing the depth of tumor infiltration, whereas further efforts are needed to improve the accuracy in N staging. EUS will be helpful in choice of the appropriate therapeutic procedure and predicting the possibility of surgical resection.
5.The clinical significance of prostate specific antigen density value in the differential diagnosis of prostate carcinoma and benign prostate hyperplasia

Objective To investigate the clinical significance of prostate specific antigen density (PSAD)value in the differential diagnosis from malignant and benign prostate disease .Methods 56 patients who enrolled in this hospital during April 2008 to No-vember 2012 were reviewed ,all of them were suspected as prostate carcinoma with free total PSA value at 4-10 ng/mL .Prostate volume were determined via transrectal ultrasound ,while serum PSA was determined by chemiluminescence method ,and all patients were finally pathologically diagnosed via tissue biopsy or prostatectomy .21 cases were proved to be prostate carcinoma (prostate carcinoma group) ,and the other 35 cases were proved to be benign prostate hyperplasia(BPH) .And the statistical significances of tPSA ,prostate volume and PSAD values were compared between the two groups .Results Compared with BPH group ,serum level of tPSA was insignificant ,while the values of prostate volume and PSAD were significantly lower in the BPH group than in prostate carciboma group .Considering the mean prostate volume(39 .27 mL) as the reference point ,40 mL was set as the cut-off value of prostate volume .Therefore ,there were 80 .95% prostate carcinoma group patients whose prostate volume were less than 40 mL , and 68 .57% benign prostate hyperplasia group patients whose prostate volume were larger than 40 mL .Considering the PSAD val-ue(0 .147) as the reference point ,0 .15 was set as the cut-off value of PSAD .Therefore ,the sensitivity ,specificity ,positive predic-tive value and negative predictive value among patients with prostate volume less than 40 mL ,the datas were respectively as 80 .95% ,72 .73% ,85% and 61 .54% .While ,among patients with prostate volume larger than 40 mL ,were respectively as 75% , 66 .67% ,27 .27% and 94 .11% .Conclusion PSAD was of certain value in the differential diagnosis of prostate carcinoma in PSA gray zone cases .If PSAD=0 .15 was consider as the cut-off point ,the sensitivity and specificity of differential diagnosis for prostate carcinoma decreased as the prostate volume increase .However ,the influencing factors are still to be elucidated .
6.Empirical study and treatment analysis of corrosive gastritis by toilet cleaner

Objective To study the gastrointestinal tract damage induced by acid detergent toilet cleaner and treatment methods.Method Acute toxicity of toilet cleaner was studied using big white rabbits.Subacute and chronic toxicity and treatment methods were studied by hospital patients.Results After falling 15 ml toilet cleaner into stomach through mouth in 2 rabbits,both died of gastric perforation.After one patient misapplied with 50 ml to//et cleaner,gastric corrosive ulcer was found by gastroscopy after treatment 7 d,gastric constrictive inflammation was found by upper gastrointestinal barium meal at 14 d after treatment,and gastric scarring was found by gastroscopy after treatment 51 d.Conchusions Acid detergent toilet cleaner mainly harm gastric mucosa,gastric perforation is easily induced in acute stage,gastric ulcer induced in subacute stage,and scarring induced in chronic stage.Taking orally pure milk,inhibiting gasuic acid secretion and haemostasis are mainly treatment methods.Examination of gastroscopy and upper gastrintesinal barium meal may be carried out in subaeute and chronic stages.
7.Hepatocarcinoma HepG2 cell apoptosis promoted by hTERT RNAi through cytochrome C dependent mitochondrial pathway

Objective To probe the mechanism of hepatocarcinoma cell apoptosis promoted by human telomerase reverse transcriptase (hTERT) RNA interference (RNAi) by mitochondrial pathway in vitro. Methods Westernblot was employed to detect the intracellular expressions of caspase-9, hTERT, Bcl-2 and Bax, and mitochondrial and cytoplastic cytochrome C (cyt C) in HepG2 cells transfected by pSliencer 3.1-H1 neo-shTERT (small hairpin RNA hTERT). Result In the HepG2 cells transfected by pSliencer 3.1-H1 neo-shTERT, expressions of hTERT, Bcl-2 and mitochondrial cyt C were significantly down-regulated, while Bax and cytoplastic cyt C were obviously up-regulated, and active caspase-9 was found in addition to procaspase-9 compared with that in negative control cells and untransfected cells. Conculsion hTERT RNAi may suppress hTERT expression to result in reduction of Bcl-2 and increase of Bax, and then induce hepatocarcinoma HepG2 cell apoptosis by mitochondrial pathway subsequent to cyt C release from mitochondria to cytoplast.
8.Screening of the Stable Expressing HPV18 E5 Protein Cell Line and its Influence on Cell Proliferation and the Cell Cycle.

We wished to screen the cell line that stably expresses the HPV18E5 protein, and to ascertain the influence of HPV18E5 protein on cell proliferation and the cell cycle. The HPV18E5 gene was amplified by the polymerase chain reaction. Then, the His-tag pSecTag-HPV18E5 eukaryotic expression vector was constructed by digestion ligation and connection. The recombinant plasmid was transfected into Balb/c3T3 cells with lipofectamine, and positive cell lines were screened by a culture medium containing bleomycin. HPV18E5 expression in cells was confirmed by western blotting and immuno-enzymatic methods. The influence of HPV18E5 on cell proliferation and the cell cycle were detected by Cell Counting Kit-8 and flow cytometry, respectively. The pSecTag-HPV18E5 eukaryotic expression vector was constructed. After 21-day selection in a culture medium containing 400 μg/mL bleomycin, stably expressing HPV18E5 protein cells were harvested. Compared with control groups, cell proliferation in HPV18E5 stably expressed cells was obviously increased, as was the S phase in the cell cycle. Our results suggested that HPV18E5 influences cell proliferation and the cell cycle. Our study has laid the foundation of the biologic properties of HPV18E5 protein, which will aid further studies on the mechanism of action of carcinogenesis.

9.Enhanced TRAIL antitumor activity on human gastric cancer cells induced by 5-Aza-CdR is involved in up-regulation of caspase-8

Objective To study the effects of demethylation agent 5-Aza-2′-deoxycytidine (5-Aza-CdR) on the antitumor activity of tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) against gastric carcinoma. Methods Expression of caspase-8 mRNA was examined by RT-PCR. Antitumor activity of TRAIL protein was measured by MTT method. Results The inhibition rates of treatment with 200 ng/ml TRAIL for 72 h on gastric cell lines SGc 7901, Kato 3, and AGS were 9.83%, 11.94%, and 4.04%. After treatment with 5-Aza-CdR, the inhibition rates of 200 ng/ml TRAIL on gastric cell lines increased to 38.98%, 52.42%, and 30.72%. Before exposure to 5-Aza-CdR, expression of caspase-8 mRNA was low and an increased expression of the caspase-8 was found in the three gastric cancer cells after treatment with 5-Aza-CdR. Conclusion Treatment with 5-Aza-CdR can increase TRAIL antitumor activity on human gastric cancer and its mechanisms might be involved in the up-regulation of caspase-8 gene.
10.Trabeculectomy with scleral tunnel treating refractory glaucoma

Objective To investigate the clinical effecl of trabeculectomy with scleral tunnel in the treatment of refractory glaucoma. Design Prospective, randomized and comparative clinical study. Participants 87 patients (98 eyes) with refractory glaucoma. Methods The patients were randomly assigned to receiving trabeculectomy with or without scleral tunnel. The tunnel group (50 eyes) underwent trabeculectomy with an additional deep scleral tunnel of 5.0mm?1.5mm beneath the superficial scleral flap. The control group (48 eyes) underwent conventional trabeculectomy. The average follow-up period was 6 to 12 months posloperatively. Main Outcome Measures Visual acuity, intraocular pressures (IOP), filtering blebs, operative and postoperative complications. Results (1) No significant differences in visual acuity were found between two groups. (2) The postoperative IOPs were significantly lower than the preoperative IOPs in both groups, while the IOPs on the 7th day after the surgery between the two groups were not different significantly. The average postoperative IOP at the 6th month in the tunnel group was 14.34?3.95 mmHg and 19.57?7.76 mmHg in the control group, which were different significantly (P
