1.Effects of renal ischemia on microRNAs and VEGF-NOTCH signal moleculars in mice

Objective To investigate the expression changes of microRNAs and VEGF-NOTCH in renal ischemic injury in mice, and to explore the potential mechanism associated with renal angiogenesis.Method Male Balb/c mice were subjected to a standard renal ischemia to induce acute kidney injury (AKI) after 45 min of bilateral renal artery clamping. Following 4 h, 24 h of reperfusion or sham operation, kindey tissues were collected and subjected to detect the expression changes of microRNAs which relatived with angiogenesis and VEGF, Flk-1, Notch1 mRNA by Quantitative Real-time RT-PCR. Flk-1 protein was detected by Western blotting analysis at 24 h and 72 h following Ischemia/Reperfusion(I/R) injury. The expression of CD31 was examined in tissue sections by immunohistochemistry staining, and the microvessels in ischemic region of each group were counted. Results miRNA-210 and miRNA-92a expression increased significantly, with prominent changes at 4 h and 24 h after reperfusion( P < 0.05 ). VEGF and Flk-1 mRNA expression and Flk-1 protein were increased in renal I/R compared with control group respectively (P<0.05 ).Immunohistochemistry staining results of CD31 showed a significant increase of microvessels in renal ischemic region. Conclusion This study first reported the changes in miRNAs expression in response to kidney I/R in mouse. our results implied that miRNAs may be involved in targeting VEGF-Notch pathway signaling to regulate angiogenesis after renal I/R injury. It provided novel insights into the angiogenesis mechanism of renal ischemic injury.
2.Model TJ-IV Computer-Assisted Vectorcardiogram Analysis System

uring last 16 years we have successfully developed the computer-assisted vectorcardiogram analysis systems: model TJ-Ⅰ, TJ-Ⅱ, and TJ-Ⅲ, but some technical problems remained unresolved, such as the recognition accuracy for vectorcardiograms, measurement of the parameters of complicated QRS waves, the ratio of T loop length to width, and the area of spatial vectors etc. A new system, model TJ-Ⅳ was designed to resolve these technical problems. The system was equipped with a 586 computer with a CPU of 120 MHz. Special new low-noise amplifier was employed and C language was used for programming. Three graph recognition techniques were used to enhance the accuracy of VCG recognition. 32 orthogonal electrocardiograms and vectorcardiograms were displayed and printed, and 566 parameters of vectorcardiograms were calculated. Our results with 150 cases showed that the system had high accuracy of graph recognition, and parameter calculation and the results were essentially consistent with those of manipulative methods. We were led to concluded when compared with TJ-Ⅲ system, the new version has higher accuracy of processing and measurement for vectorcardiograms, is able to process more vectorcardiographic parameters, with higher processing speed.

4.Diagnostic value of endoscopic ultrasonography in staging of rectal cancer

Objective To evaluate the diagnostic value of endoscopic ultrasonography (EUS) in staging of rectal cancer (RC).Methods From January 2015 to January 2017,the clinical data of 204 patients with RC and received EUS and surgery were retrospectively analyzed.Patients were divided into surgery alone group (155 cases) and preoperative neoadjuvant chemoradiation therapy (CRT) plus surgery group (49 cases).The preoperative staging by EUS and postoperative pathological staging of two groups were compared.Kappa test was performed for statistical analysis.Results Compared with postoperative pathologic diagnosis,the accuracy rate of EUS in the evaluation of invasion depth of RC in surgery alone group was 81.9% (127/155),and the accuracy rates in the diagnosis of Tis,T1,T2,T3 and T4 were 3/4,11/13,82.1%(32/39),91.1%(41/45) and 74.1%(40/54),respectively,with a good consistency (kappa=0.751,P<0.01).However,the accuracy rate of EUS in the invasion depth of RC in CRT plus surgery group was 34.7% (17/49),and the accuracy rates in the diagnosis of T2,T3 and T4 were 1/13,2/7 and 14/16,respectively,with a poor consistency (kappa =0.107,P=0.850).Compared with postoperative pathologic diagnosis,the diagnostic accuracy rate of EUS in evaluating regional lymph node metastasis in surgery alone group was 70.3% (109/155),and the accuracies in the diagnosis of cases with or without regional lymph node metastasis were 40.7% (24/59) and 88.5% (85/96),respectively,with a poor consistency (kappa=0.317,P<0.01).The diagnostic accuracy rate of EUS in evaluating regional lymph node metastasis of preoperative CRT plus surgery group was 51.0% (25/49),and the accuracies in the diagnosis of cases with or without regional lymph node metastasis were 5/11 and 52.6% (20/38),respectively,with a poor consistency (kappa =0.014,P =0.911).Conclusions EUS can accurately evaluate the depth of tumor invasion and lymph node metastasis in preoperative staging of RC,which may be helpful for determining clinical treatment strategy.However,for patients received CRT treatment,EUS has a limited value in diagnosing and staging the tumor.
5.Trans-theoretical model based intervention affecting on physical activity promotion for patients with ;hypertension

Objective To evaluate the effectiveness of trans-theoretical model ( TTM ) based intervention for patients with hypertension to promote physical activity. Methods A total of 118 patients were selected and randomly divided into experimental group (n=56) and control group (n=62) by random number table. The patients of the experimental group received the TTM-based intervention while the patients of the control group received the conventional intervention. The results of two groups were compared. Results The score of behavior changing stage and motor behavior of different follow-up time in two groups had significant differences (F=46. 37,50. 44;P <0. 01). The systolic pressure was (124. 93 ± 10. 29) mmHg, diastolic pressure (78.32 ±8.42) mmHg in the experimental group that were lower than (133. 24 ± 11. 87), (83. 02 ± 7.97)mmHginthecontrolgroup(t= -4.05, -3.11; P<0.01).Conclusions TTMbasedinterventionis effective for patients with hypertension to promote physical activity.
6.Effect of 3D laparoscopic hepatectomy in glissonian approach for hepatocellular carcinoma on the cellular immune function

Objective To compare the effects of 3D laparoscopic hepatectomy (3D-LH) and open hepatectomy(OH)in glissonian access for hepatocellular carcinoma(HCC)on postoperative immunosuppression by analyzing cellular immune function. Methods During January 2014 to December 2017,sixty patients randomly scheduled to undergo hepatectomy were divided into 3D-LH group and OH group,with 30 patients in each group. All patients' peripheral blood was sampled to measure cell-mediated-immunologic markers(CD4+,CD8+T cell) before operation and in the mornings of the 1st,7thday after surgery. The effects of different operative methods on cellular immune function in patients organism were investigated. Results The decreased level in 3D-LH group was less than that in OH group in postoperative 1stday. The improvement in the 3D-LH group was more faster than that in the OH group on postoperative 7thday. There was statistical significance between the two groups(P<0.05), respectively. Conclusion When compared to open hepatectomy ,3D laparoscopy liver resection in glissonian approach for HCC,less effects in cellular immune function.
7. Detecting hexamethylene diamine in workplace air by high performance liquid chromatography

OBJECTIVE: To establish a method for detecting hexamethylene diamine in workplace air by high performance liquid chromatography( HPLC).METHODS: Hexamethylene diamine in the workplace air was collected by silica gel tube,and each was added with a concentration of 0.05 mol/L sodium bicarbonate solution,each with 0.60 mL of dansyl chloride solution( a mass concentration of 240.00 mg/L),and the volume was adjusted to 5.00 mL with acetonitrile,heating for 40.00 min in bath water,acetonitrile:water( 75:25,V/V) as the mobile phase,quantitated by the standard curve method,using HPLC for determinationstandard.RESULTS: The linear range of hexamethylene diamine was 0.040 0-6.000 0 mg/L,the correlation coefficient was 0.999 3,and the detection limit was 0.003 8 mg/L,and the minimum detection concentration was 0.002 5 mg/m~3( calculated by sample volume of 3.0 L); The within-run relative standard deviation( RSD) was 2.1%-3.0%,and the between-run RSD was 2.9%-3.6%.The average desorption efficiency of the method was 91.4%-94.1%.The sampling efficiency was 98.5%-99.6%.CONCLUSION: The method is simple,rapid,sensitive,accurate and suitable for the detecting hexamethylene diamine concentration in workplace air.
8.A Chromosome-level Genome Assembly of Wild Castor Provides New Insights into Its Adaptive Evolution in Tropical Desert

Wild castor grows in the high-altitude tropical desert of the African Plateau,a region known for high ultraviolet radiation,strong light,and extremely dry condition.To investigate the potential genetic basis of adaptation to both highland and tropical deserts,we generated a chromosome-level genome sequence assembly of the wild castor accession WT05,with a genome size of 316 Mb,a scaffold N50 of 31.93 Mb,and a contig N50 of 8.96 Mb,respectively.Compared with cultivated castor and other Euphorbiaceae species,the wild castor exhibits positive selection and gene family expansion for genes involved in DNA repair,photosynthesis,and abiotic stress responses.Genetic variations associated with positive selection were identified in several key genes,such as LIG1,DDB2,and RECGI,involved in nucleotide excision repair.Moreover,a study of genomic diversity among wild and cultivated accessions revealed genomic regions containing selection signatures associated with the adaptation to extreme environments.The identification of the genes and alleles with selection signatures provides insights into the genetic mechanisms under-lying the adaptation of wild castor to the high-altitude tropical desert and would facilitate direct improvement of modern castor varieties.
9.Characteristics and management mechanism of imported malaria from overseas during the malaria elimination phase in Wuhan City

Objective To analyze the characteristics of imported malaria epidemic from overseas in Wuhan, to explore the management mechanism of on-site cases, and to accumulate experience for the treatment of imported malaria in large cities after malaria elimination. Methods The epidemiological data on imported malaria from abroad during the period of malaria elimination (2010-2019) in Wuhan were collected. The gender, age and severe illness-related factors of the cases were analyzed. Based on the characteristics of the epidemic and the current situation of prevention and control, the content and experience of the “Municipal-District 24-7” case mechanism were discussed. Results The medical resources in Wuhan were the best in the central region, resulting in a large number of imported malaria cases, with a total of 474 cases reported from 2010 to 2019 (40.79% of the total number of cases in Hubei Province), including 359 cases of falciparum malaria, 36 severe cases and one death (the death rate was 0.28%). The patients were mainly young and middle-aged men aged 20 to 49 years old (97.26%). There were many referral cases (40.30%), and there was no seasonal clustering of cases reported. The undiagnosed proportion at the first visit was 44.85%, and the time of attack-diagnosis was 4 days or more in 61.00% of cases. The occurrence of severe cases was related to unconfirmed diagnosis at the first visit (χ2=35.46, P<0.001) and attack-diagnosis time (Z=-6.49, P<0.001). Conclusion Imported malaria occurs frequently in Wuhan, mainly falciparum malaria. However, “Municipal-District 24-7” case mechanism has effectively curbed the occurrence of severe and death cases and provided valuable experience for case management in similar cities in China.
