2.Relationship of TGF-β and IL-4R gene polymorphisms with risk of classical Hodgkin lymphoma.

OBJECTIVEThis study was aimed to analyze the relationship between single nucleotide polymorphisms of transforming growth factor-β1 G-800A and C-509T, interleukin-4 receptor V75I and susceptibility of CHL in adults.
METHODSPolymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was applied to analyze the expressed alleles of the selected SNP loca. The relationship between genomic polymorphisms of TGF-β1 and IL-4R and susceptibility of CHL were coupled with clinical data.
RESULTSTGF-β1G-800A and TGF-β1C-509T had obvious linkage equilibrium (D' = 0.879, r(2) = 0.83, P = 0.020). GT haplotype distribution frequencies in mixed cellularity Hodgkin lymphoma cases and control group were of 53.1% and 34.2%, respectively, with statistically significant (OR = 2.35, P = 0.000); distribution frequencies of mutant gene T/T in disease and control groups were of 38.8% and 15.3%, respectively, also with statistically significant (OR = 3.654, P = 0.000); frequencies of nodular sclerosis CHL patients with IL-4R V75I mutant gene A/A in disease and control groups were of 19.2% and 41.75%, respectively, also with statistically significant (OR = 3.156, P = 0.000).
CONCLUSIONSingle nucleotide polymorphisms of TGF-β1 G-800A, C-509T and IL-4R V75I has a significant correlation with Chinese susceptibility to classical Hodgkin lymphoma.

3.Expression of kinase insert domain-containing receptor in prostate adenocarcinoma.

OBJECTIVETo investigate the correlation between the expression of the kinase insert domain-containing receptor (KDR) and the histological grade of prostate adenocarcinoma.
METHODSForty-eight samples of prostate adenocarcinoma tissues and 20 samples of benign prostatic hypertrophy (BPH) tissues were studied by LsAB immunohistochemical staining.
RESULTSThe positive expression rate of KDR was 73% in prostate adenocarcinoma and 30% in BPH. The expression of KDR was stronger in prostate adenocarcinoma, and there was no relationship between staining intensity and the histological grade of carcinoma.
CONCLUSIONKDR, expressed more highly in prostate adenocarcinoma, promises to be a new target in the treatment of prostate adenocarcinoma.

4.Catwalk-assisted behavioral analysis of rat models of middle cerebral artery occlusion

Objective To investigate the feasibility of Catwalk-assisted system in analyzing the behavioral features of rat models of middle cerebral artery occlusion(MCAO).Methods Thirty male SD rats were randomly assigned to experimental group and control group.Suture-occluded method was used to establish the focal cerebral ischemia/reperfusion models in the experimental group.Traditional modified neurological severity scale(mNSS),vibrissae-evoked forelimb placing test and cylinder test were employed to analyze the behavioral characteristics of the SD rats 1,3,7,14 and 28 dafter establishment of rat models.Besides that,Catwalk-assisted gait analysis system was employed at the same time points.Results Catwalk-assisted gait analysis results suggest rats from experimental group had smaller intensity,print area and slower walk speed than rats from control group.Besides that,interlimb coordination also changed on the 3rd d of operation and existed until the 28th d of operation.Rotate step patterns like Ra(1.5%)and Rb(2.3%)were detected in experimental group but never seen in control group.Parameters as cadence,stance,swing and other regularity index had no significant changes between the 2 groups.Conclusion Catwalk analysis system,by analyzing such movement parameters of MCAO rats as intensity,speed,print area,step pattern and interlimb coordination,can commendably reflect the damage of brain ischemia and its changes crossing time,which can comprehensively analyze many behavioral features of the models.

6.Biological appraisal of human bone marrow mesenchymal stem cells during ex-vivo expansion.

This study was aimed to investigate the characteristics of human bone marrow mesenchymal stem cells during ex-vivo expansion, MSCs were isolated from human bone marrow. At each passage, the characteristics of proliferation kinetics, osteogenic and adipogenic differentiation potential were analyzed, and cell morphology, surface markers were investigated as well. The karyotype analysis was done in different passage cells. The infection HIV, HCV, HBV and TP were detected by ELISA. Mycoplasma contamination in vitro was detected by PCR method. HLA-SBT was used to reanalyze the results of HLA antigens and alleles. STR genetic loci were detected by PCR in the MSC1, MSC2, MSC3 and MSC4. The results indicated that the proliferative ability and osteogenic potential decreased with the increase of passage number during culture expansion. The multiple differentiation potential of MSCs was maintained during their life span. Karyotype analysis showed that MSCs from 4 groups before passage 8 were normal. The expression of CD29, CD44, CD105, CD166 and CD73 were positive. The expression of CD14, CD34, CD45, CD80, CD86 were all negative. SBT was used to identify HLA-A, B, Cw, DRB1, DRPB1, DQ alleles in the MSC1, MSC2, MSC3, MSC4. The genetype of STR in the MSC1, MSC2, MSC3, MSC4 was different. MSC 3 was examined by TP-ELISA to confirm the infectious disease of TP. MSC2 was contaminated by mycoplasma at passage 5. It is concluded that culture expansion causes MSCs to gradually lose their stem cell properties. During ex-vivo expansion of MSCs, the osteogenic differentiation potential is decreased. MSCs before passage 8 can be a valuable subject for basic research and clinical application.

7.Efficacy of erlotinib after the failure of gefitinib in patients with metastasis of non-small cell lung cancer with unknown EGFR mutation status.

OBJECTIVETo evaluate the efficacy of erlotinib in patients with metastasis of non-small cell lung cancer who had benefits from initial gefitinib treatment but finally demonstrated resistance, especially in those of unknown EGFR mutation status, and to compare the efficacy of erlotinib between patients who received erlotinib immediately after gefitinib failure and those who received chemotherapy before erlotinib.
METHODSForty Chinese patients who had been treated with erlotinib (150 mg daily) after gefitinib (250 mg daily) failure were evaluated retrospectively. All of these patients had achieved gefitinib treatment for at least three months with response of partial remission or stable disease. Among them, 16 patients shifted to erlotinib immediately after progression (Group G-E), and the other 24 patients inserted chemotherapy between gefitinib and erlotinib (Group G-C-E).
RESULTSIn the whole group, the disease control rate (DCR) of erlotinib was 52.5% (21/40) while the objective response rate (RR) was only 10.0% (4/40). The RR of the group G-E was 6.2% and the group G-C-E was 12.5%, and the DCR was 56.2% and 50.0% in the two groups, respectively, both without significant differences (P = 0.638 and P = 0.755). There was no correlation between the efficacy of erlotinib and that of initial gefitinib in both group G-E and group G-C-E (P = 0.365 and P = 0.658). The median progression-free survival (PFS) and overall survival (OS) for the erlotinib treatment were 3.0 and 12.0 months in the 40 patients. Statistically no significant difference was observed in PFS (4 months in the group G-E and 2 months in the group G-C-E, P = 0.768) and OS (12 months in both Groups, P = 0.510).
CONCLUSIONSErlotinib can be considered either immediately after gefitinib failure or following the insertion of chemotherapy after gefitinib failure in progressive non-small cell lung cancer patients who initially benefited from gefitinib.

Objective Circulating tumor cells (CTCs) have potential value in the clinical application of various tumors. This study was to investigate the role of CTCs and their chemokine receptor CCR9 in the invasion and metastasis of non-small cell lung cancer (NSCLC). Methods From May 2018 to June 2019, a total of 62 patients with NSCLC in the clinical oncology center of The People's Hospital of Guangxi Zhuang Autonomous Region were enrolled in this study. The CanpatrolTM CTC technique was used to detected the expressions of CTCs and CCR9 in CTCs in peripheral blood of patients. Furthermore, the relationships between expression levels of CTCs, CCR9 and clinical, pathological characteristics of NSCLC patients were analyzed. Risk factor monitoring and comprehensive intervention should strongly be advocated.
10.Purification of newborn rat Schwann cells by multiple digestions with low-concentration trypsin for differential cell detachment

Objective To describe a simple and efficient method to obtain large quantities of higllly purified Schwann cells(SCs).Methods SCs were isolated from the sciatic and brachial nerves of 3-to 5-day-old newborn SD rats by collagenase digestion.The isolated SCs were plated at the density of3x105/mL for primary culture,several rounds oftrypsin digestion were performed within 72 h to purify SCs. Results Compared with the purification using 1.25 g/L trypsin digestion in serial differential detachment procedures, Our protocol allowed easier and more efficient separation of the SCs from the fibroblasts.Immunocytochemical staining showed that the purity of the SCs exceeded 95%.Conclusion The purification protocol of the SCs we established can be easily carried out and yields well reproducible results to obtain large quantities ofhighly purified SCs for transplantation studies.
