1.Scientific practice of Chengjiang Acupuncture School in the Republic of China.

Under the influence of Chinese medicine scientization, Chengjiang Acupuncture School, which was originated in the Republic of China (1912 - 1949), has also undergone a series of scientific practice. The Chengjiang School established scientific research, education and medical organizations such as the Research Society of China Acupuncture-Moxibustion, China Acupuncture-Moxibustion School, Training School of China Acupuncture-Moxibustion and Sanatorium of Acupuncture-Moxibustion. Zhenjiu Zazhi (Journal of Acupuncture-Moxibustion) was established, and monographs and textbooks such as Zhongguo Zhenjiu Zhiliaoxue (Therapeutics of Chinese Acupuncture-Moxibustion) and Zhongguo Zhenjiuxue Jiangyi (Lectures on Chinese Acupuncture-Moxibustion) were published. Meanwhile, based on clinical practice, large members of acupuncture personnel with scientific literacy emerged. Therefore, it is held that the scientization of Chengjiang School has performed a profound influence on the development of modern acupuncture-moxibustion.

2.Construction of plant expression vectors with PMI gene as selection marker and their utilization in transformation of Salvia miltiorrhiza f. alba.

OBJECTIVETo construct plant expression pCAMBIA1301-PMI by substituting PMI for hygromycin resistance gene in pCAMBIA1301 and obtain transgenic Salvia miltiorrhiza f. alba using PMI-mannose selection system.
METHODThe 6-phosphomannose isomerase gene (PMI) of Escherichia coli was amplified by PCR. Sequence analysis showed that it shared 100% amino acids identities with the sequences of PMI genes isolates reported in the NCBI. Based on pCAMBIA1305, the plant expression pCAMBIA1305-PMI was constructed successfully by substituting PMI for hygromycin resistance gene in pCAMBIA1305. pCAMBIA1305-PMI was transformed into Agrobacterium tumefaciens LBA4404, and then the leaves of S. miltiorrhiza f. alba were inoculated in LBA4404 with pCAMBIA1305-PMI.
RESULTPlant expression pCAMBIA1301-PMI was successfully constructed and the leaves of S. miltiorrhiza f. alba inoculated in LBA4404 with pCAMBIA1305-PMI were selected on medium supplemented with a combination of 20 g x L(-1) mannose and 10 g x L(-1) sucrose as a carbon source. The transformation efficiency rate was 23.7%.
CONCLUSIONGenetic transformation was confirmed by PCR, indicating that a new method for obtaining transgenic S. miltiorrhiza f. alba plants was developed using PMI-mannose selection system.

3.Effects of photodynamic therapy on the ultrastructure of glioma cells.

OBJECTIVETo study the change in ultrastructure of C6 glioma cells after photodynamic therapy (PDT), to compare morphological differences in necrosis and apoptosis before and after PDT treatment, and to evaluate the effect of photodynamic therapy on the blood brain tumor barrier (BTB) of C6 glioma.
METHODSThe model was produced by transplanting C6 glioma cells cultured in vitro using Peterson method into the caudate nuclei of Wister rats. The experiment group received PDT for two weeks after the operation. The sub-cellular structure, blood-brain-barrier (BBB) and BTB in both groups were observed under electron microscope.
RESULTSApoptosis in different phases and necrosis could be observed in some C6 glioma cells. Swelling occurred on the ultrastructure of cellular organs such as mitochondria and endoplasmic reticulum in most of the cells. Damage to the BTB, reduction of the number of cellular organs in endothelial cells of the capillary blood vessels, stretch of the tight junction, and enlargement of the gaps between endothelial cells were also seen in the experiment group. Meanwhile, limited impact on the normal sub-cellular structures and BBB was observed.
CONCLUSIONPDT could induce apoptosis and necrosis of C6 glioma cells due to the damage to the ultrastructure of mitochondria and endoplasmic reticulum. The weakened function of C6 glioma BTB initiated by PDT makes it possible to perform a combined therapy of PDT and chemotherapy for glioma.

4.Discussion on scientific-school characteristics of chengjiang acupuncture school.

To find out the factors of scientific schools in Chengjiang acupuncture school according to its characteristics and demands on the basis of connotations of the genres, schools, scientific schools. The results show that the Chengjiang acupuncture school is a typical scientific school, including followed elements: school leaders, plenty of famous group members, unified system research pattern and school spirit that is commonly followed.

5.Microsurgical resection of large acoustic neuroma through the retrosigmoid approach: a clinical analysis

Objective To discuss the surgical skills in microsurgical resection of large acoustic neurornas via the retrosigmoid approach for reducing the neural damage and complications of such surgeries. Methods Twenty-one cases of large acoustic neuroma treated with microsurgery via the retrosigmoid approach were retrospectively analyzed in view of the surgical approach, microsurgical techniques, intraoperative accidents, and management of the postoperative complications. Results The tumors were totally removed in 18 cases and subtotally in 3 cases. The facial nerve function was well preserved in 17 cases (80.9%), and the acoustic nerve was preserved in 5 cases (23.8%). Intraoperative cardiac arrest occurred in 1 case, and 4 patients developed short-term postoperative complications, but no death occurred in these cases. Conclusion Microsurgical resection through the retrosigmoid approach is safe for removal of large acoustic neuroma, and proficient microsurgical skills and thorough understanding of the microanatomy are crucial for total tumor resection and reduction of the nerve injuries and surgical complications.
6.Sequence analysis of VP7 gene from rotavirus field strain from Guangzhou, China.

OBJECTIVETo compare the difference of VP7 fragment at the nucleotide level between rotavirus Guangzhou field strain R97-196 and rotavirus Beijing field strain or prototype strains.
METHODSThe VP7 fragment amplified from Guangzhou field strain R97-196 by RT-PCR was cloned into the T-A cloning vector pUCm-T and sequenced.
RESULTSThe VP7 fragment from Guangzhou field strain R97-196 was 1,062 bp in length and contained two open reading frames which is consistent with that reported in the literature. The sequence analysis revealed that the cDNA from R97-196 shared higher nucleotide and amino acid identities with Beijing field strain T73 (98% and %, respectively) than with serotype 2-4 (G types) rotavirus (from 74% to 77% and 73% to 81%, respectively). The divergence of amino acid sequences is mainly within the nine divergence regions. The phylogenetic analysis revealed that the VP7 fragment from R97-196 was far away from rotavirus serotype G1 strain Wa.
CONCLUSIONSThe VP7 gene fragment from rotavirus Guangzhou field strain R97-196 belongs to rotavirus serotype G1. And variation of rotavirus VP7 gene fragment seems to be a geographic matter.

The purpose of this investigation is to improve ethanol production and decrease acetate formation in Saccharomyces cerevisiae strain YS2-?adh2.The strain YS2-?adh2 with deleted alcohol dehydrogenase Ⅱ(adh2) gene was isolated in our lab with higher ethanol production than that of the strain YS2.The ace-taldehyde dehydrogenase Ⅵ(ald6) gene encoded a cytosolic acetaldehyde dehydrogenase,a key enzyme of the pyruvate dehydrogenase(PDH) bypass,transfers acetaldehyde to acetate.To disrupt ald6 gene of the strain YS2-?adh2,ald6 gene targeting cassettes were synthesized by long flanking homology PCR(LFH-PCR) and then were transformed into YS2-?adh2 mutants by LiAc/SS Carrier DNA/PEG method.Positive transformants were selected with G418 and further confirmed by PCR.Once correctly integrated into the genome,the selective marker was rescued by transforming the plasmid pSH65 into the positive transformants and inducing the Cre expression with a Cre/loxP-mediated marker removal procedure.We named the ald6 gene knocked-out strain as YS2-?adh2-?ald6 which has a 12.5% higher ethanol production and a 18% lower acetate formation compared to the strain YS2.
9.The effect of Tenascin-R and anti-EGF-L antiserum on the cortical neurons in vitro in rats

Objective To study the effect of the EGF-L functional region containing human TN-R protein in prokaryocytes and anti-EGF-L serum on the cortical neuronsin vitro in rats. Methods The anti-EGF-L serum was obtained from the rabbits immunized with the protein of EGF-L, then combine with TN-R coat petri dish to prepare various culture substrate.The effect on adhere,migrate and neurite outgrowth of neuron by differ culture substrate was observed. Results High concentration of antiserum against protein of EGF-Lwas successfully obtained from the rabbits,and it has no effect on the neuron when coated on petri dish as culture substrate with PLL in vitro,but can add the adhesive to the neuron,partly neutralize the inhibitory on the neuron neurite of TN-R.The antiserum can also allow the neuron with its neurite to migrate to the area coated with TN-R. Conclusion The anti-EGF-L serum can weaken the inhibitory on the neuron of TN-R in vitro,the function in vivo need further research,
10.Introduction on the academic origin of Chengjiang acupuncture school.

The academic life, thoughts and contributions of over ten representatives of Chengjiang acupuncture school were investigated with the methodology of literature comparative analysis to abstract the academic origin of the school. And the result shows that based on the traditional theory of Chinese medicine, influenced by the ideological trend of scientization, focused on clinical practice and enlightened by Japanese acupuncture in a certain degree, Chengjiang acupuncture school is an open and inclusive Chinese medicine school which does not only emphasize on carrying on of the theoretical system of Chinese medicine, but also actively adjust itself to the development of the society.
