2.DNA POLYMORPHISM OF YERSINIA ENTEROCOLITICA

To investigate the inherent characteristics of the pathogenic Yersinia enterocolitica. Polymerase chain reaction (PCR),analysis of DNA sequence,randomly amplified polymorphic DNA (RAPD) were used to study the pathogenic and nonpathogenic 10 strains of Yersinia enterocolitica. The Pathogenic Yersinia enterocolitica of serotyps 0∶3?0∶9 and 0∶5,27 were confirmed containing pathogenic gene of adherent invasion locus(ail) and their DNA fingerprints were much more similar,and it was also found that the serotype 0∶22 contained ail gene whose fingerprints was partly similar with the pathogenic ones in molecular structure. The pathogenic Yersinia enterocolitica strains were similar in the inherent and polymorphic. The serotype 0∶22 may have relativly evaluated association with pathogenic Yersinia enterocolitica, which is possible that it got potentially pathogenicity.
3.Ambulatory urodynamics monitoring system in diagnosis of female stress urinary incontinence

Objective To compare the diagnostic accuracy of conventional urodynamic (CUD) and ambulatory urodynamic monitoring (AUM) in diagnosis of female stress urinary incontinence (SUI).Methods Forty women with SUI were prospectively enrolled and performed urodynamic studies after the ICI-Q-SF questionnaire.According to clinical symptoms,patients were divided into three groups:mild,moderate and severe group.Half patients in each groups performed CUD exam,and the other half of patients performed AUM exam.And two micturition cycles were recorded during AUM.Results There were no significant differrences in age,pregnant production times and ICI-Q-SF score between two groups.Three patients with SUI symptoms had negative findings in AUM group and 15 patients in CUD group (P<0.05).Among women with SUI,1 1 patients had positive findings in AUM group and 2 patients in CUD group (P< 0.05).Conclusion AUM can provide objective evidence for the majority of patients with SUI than CUD.
4.Linkage disequilibrium and mutation rate analysis of sixteen X-STR loci.

OBJECTIVE:
To assess the patterns of linkage disequilibrium (LD) of 16 STR loci on X chromo- some and investigate the genetic stability.
METHODS:
Genomic DNA samples extracted from blood stains from 500 unrelated individuals and 885 lineage members from Eastern Chinese Han population were genotyped through multiplex amplification using IDtyperX-16 kit by our independent research followed by capillary electrophoresis. LD was assessed by PowerMarker v3.25 software and mutation rate of every locus was analyzed.
RESULTS:
LD were not found at the 16 X-STR loci. Allele mutations were observed at 10 loci. Among them, mutation rates of DXS6809 and DXS7132 were both up to 0.0048.
CONCLUSION
When the 16 X-STR loci included in IDtyperX-16 kit were used for parentage testing, product princi- ples can be applied to calculate the likelihood, but mutation should be taken into consideration in the case that the genotypes do not meet the genetic law (especially at DXS6809 and DXS7132).

7.Electromyographic analysis of upper limbs and trunk during elbow extension activities in C_5 and C_6 SCI patients

Objective To explore how the spared muscles in upper limbs and trunk compensated for elbow extension movements in patients with SCI at the level of C_5 and C_6.Methods Fifteen patients with SCI at the level of C_5 and C_6 and fifteen healthy subjects were divided into an SCI group and a control group respectively.The surface EMG recorded from the anterior dehoid,middle deltoid,posterior deltoid,triceps braehii muscle,biceps braehii muscle,upper peetoralis,serratus anterior and latissimus dorsi during four elbow extension activities including grasp- ing cup,reaching for light-switch,wheelchair ambulation and upper limb weight-bearing.The relative EMG intensi- ties of muscles were expressed as a percentage of maximal voluntary contraction.Results Several muscles partici- pated in the activity simultaneously when both groups of subjects performed every motor task.But the primary muscles involved in the movements were different between the two groups.Furthermore,the SCI group tended to employ more muscles than the controls to perform a activity.The compensatory strategies used in various motor tasks by the spared muscles were different.Conclusion The motorueuron recruitment patterns used by the C_(5~6)SCI patients in per- forming elbow extension activities could be different from those by the healthy subjects.The patients accomplished performing different motor tasks by changing their compensatory strategies.
8.Determination of six chlorogenic acids in lonicerae flos extract by quantitative analysis multi-components with single marker.

This study is to determine six chlorogenic acids (chlorogenic acid, 5-O-caffeoylquinic acid, 4-O-caffeoylquinic acid, 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid and 4,5-di-O-caffeoylquinic acid) by quantitative analysis of multi-components with a single marker (QAMS). Chlorogeinc acid was used as internal reference to calculate the relative correction factors (RCF) of five compounds. Then the ruggedness of relative correction factors was tested on different instruments and columns. Meanwhile, a total of 4 batches of Lonicerae Flos and 20 batches of Lonicerae Flos extract with five different processing procedures were analyzed by external standard method (ESM) and QAMS, respectively. The ruggedness of relative correction factors was good. And the analytical results calculated by ESM and QAMS showed no difference. The quantitative method established was suitable for the quality evaluation of Lonicerae Flos extract.

9.Effect of Methyl-CpG binding domain protein 2 (MBD2) on AMD-like lesions in ApoE-deficient mice.

The role of methyl-CpG binding domain protein 2 (MBD2) in an ApoE-deficient mouse model of age-related macular degeneration (AMD) was investigated. Eight-week-old Mbd2/ApoE double deficient (Mbd2(-/-) ApoE(-/-)) mice (n=12, 24 eyes, experimental group) and MBD2 (wt) ApoE(-/-) mice (n=12, 24 eyes, control group) were fed on Western-type diet for 4 months. The mice were sacrificed, and total serum cholesterol levels were analyzed and Bruch's membrane (BM) of the eyes was removed for ultrastructural observation by transmission electron microscopy. Moreover, intercellular adhesion molecule 1 (ICAM-1) immunoreactivities were evaluated by fluorescence microscopy in sections of the eyes in both groups for further understanding the function mechanism of MBD2. There was no significant difference in the total serum cholesterol levels between control group and experimental group (P>0.05). Transmission electron microscopy revealed that AMD-like lesions, various vacuoles accumulated on BM, notable outer collagenous layer deposits and dilated basal infoldings of retinal pigment epithelium (RPE) were seen in both groups, and the BM in control group was significantly thickened as compared with experimental group (P<0.05). Fluorescence micrographs exhibited the expression of ICAM-1 in choroid was higher in control group than in experimental group. We are led to conclude that MBD2 gene knockout may lead to accumulation of more deposits on the BM and influence the pathogenesis of AMD via triggering endothelial activation and inflammatory response in choroid, improving microcirculation, and reducing lipid deposition so as to inhibit the development of AMD-like lesions. Our study helps to provide a new therapeutic approach for the clinical treatment of AMD.
