OBJECTIVE:To investigate the effects of pravastatin on serum levels of CRP and TNF-? in patients with diabetic nephropathy and study its possible mechanism in the treatment of diabetic nephropathy. METHODS:28 patients with diabetic nephropathy were treated with provastatin plus routine therapy (trial group),another 28 patients with diabetic nephropathy were treated with routine therapy alone (control group). The macroscopic characteristics, tissue, caterpillar body wall and powder of Ophiocordyceps xuefengensis in different batch numbers were observed and researched by the macroscopic and microscopic identification methods. The result shows that the morphology, size, abdominal annulations of caterpillar, etc. of 0. xuefengensis are the macroscopic identification characteristics, the caterpillar body surface mycelium, body wall sculpture and crochets on abdominal legs are the microscopic identification characteristics. These characters are stable and regular discriminant features, which are proved to be the identification basis of O. xuefengensis. In addition, The characters such as crochets on abdominal legs arrange in two parallel ellipse rings, the inner crochets are long strip, and the external toes are unciform, are specific.

7.Bladder saline perfusion before catheter removal in TURP patients

Objective To evaluate the clinical significance of bladder saline perfusion before catheter removal in TURP patients. Methods From 2009 to 2011,140 patients received TURP were enrolled in this study.Patients were divided into perfusion group (70 cases with bladder saline perfusion before catheter removal) and control group (70 cases without perfusion). Results Comparing with the control group (33.1 ± 5.4) min,the time waiting for urination was shorter in perfusion group ( 3.7 ± 0.2 ) min ( P <0.05 ).The recovering time to normal urination was shorter in perfusion group (7.7 ± 1.2 ) d than in control group (11.7 ± 1.3) d (P < 0.05 ) as well.In the first urine after catheter removal and first urine on the next day morning,white blood cell count of 2 groups (4.5 ± 0.1 ) vs ( 6.9 ± 3.5 ) ; ( 3.7 ± 0.2 ) vs (4.3 ±0.5) had significant differences ( P < 0.05 ). Conclusion Bladder saline perfusion before catheter removal in TURP patients is simple and effective for the restoration of normal voiding.
8.MTT assay for detecting 5-fluorouracil chemosensitivity of human breast carcinoma cell line.

OBJECTIVETo assess the feasibility of MTT colorimetric assay for testing the in vitro chemosensitivity of breast cancer cells to 5 fluorouracil (5-Fu).
METHODSThe chemosensitivity of human breast carcinoma cell lines MCF-7 and MDA-MB-435S to 5-Fu at different concentrations was evaluated with MTT assay.
RESULTS5-FU treatment resulted in dose-dependent growth inhibition of the breast cancer cells with both low and high metastatic capacities.
CONCLUSIONSMTT assay may help select appropriate chemotherapeutic agents and provides evidence for individualized chemotherapy for breast cancer.

9.Application of temperature sensitive yeast models with definite target in the screening of potential human Pin1 inhibitors.

This study is to explore new lead compounds by inhibition of Pin1 for anticancer therapy using temperature sensitive mutants. As Pin1 is conserved from yeast to human, we established a high-throughput screening method for Pin1 inhibitors, which employed yeast assay. This method led to the identification of one potent hits, 8-11. In vitro, 8-11 inhibited purified Pin1 enzyme activity with IC50 of (10.40 +/- 1.68) micromol x L(-1), induced G1 phase arrest and apoptosis, showed inhibitory effects on a series of cancer cell proliferation, reduced Cyclin D1 expression, was defined as reciprocally matched for protein-ligand complex in virtual docking analysis and reduced cell migration ability. In vivo, we could observe reduction of tumor volume after treatment with 8-11 in xenograft mice compared with vehicle DMSO treatment. Altogether, these results provide for the first time the involvement of 8-11 in the anticancer activity against Pin1.

10.Experimental studies and clinical application of quickly prefabricated random skin flap.

OBJECTIVETo enlarge the ratio of length to width of a prefabricated random skin flap in a short period, in order to meet a special clinical need.
METHODSA white rat and a white mini-pig was chosen for an animal model for the experiment. They were divided into the experimental group and the control. The prefabricated flap was formed as the planned design. In the experimental group, the flaps were treated with a desired pressure in certain intervals at the planned part of the flap. We estimated and inspected the digitized perfusion of flap microcirculation, and made overcong and wide random skin flap survive within 24 hours, and this technique was finally used for the clinical wound treatment.
RESULTSAs expected, the survival area of the flap in the experimental group was significantly larger than the control. This technique was successfully used to repair the defects of the special sites in plastic and reconstructive surgery.
CONCLUSIONThe method of quickly prefabricated random flap could enlarge the ratio of length to width of the flap and put the flap in shape in short time. With the digitized estimating and inspecting, this method could obviously improve the survival area of the random flap and could also be very useful for the clinical treatment.
