4. Drying kinetics and mathematical modeling of Erzhi Pills during hot-air drying process

Objective: To study the change rule of water content in the drying process of hot air drying of thin layer drying characteristic and model research, and to provide reference for improving the quality of Erzhi Pills (EP). Methods: The moisture ratio, drying rate of dry basis, and drying rate curves along with the change of drying time and relationship between dry basis moisture content and drying rate of EP were studied using a hot air dryer at five levels of drying air temperature in the range of 50-90 ℃. The model was fitted and verified by the empirical model Henderson & Pabis model, Newton model, Page model, Logarithmic model, two term exponential model, Wang & Singh model, Midilli et al. model, etc. Meanwhile, based on Fick's diffusion law, the effective diffusion coefficients of water (Deff) and activation energy value in EP were calculated. Results: The drying curves of EP show that the moisture ratio and drying rate of EP were closely related to the temperature of drying medium, and the moisture of the material decreases with the prolonged drying time. It could be seen from the drying rate curve that the drying rate increased with the increase of hot air temperature, and the migration of moisture was accelerated. By comparing the correlation coefficient (R2), chi-square (χ2) and standard error (RMSE), we could see that the mean value of R2 of model number 7 was the maximum, the χ2 and RMSE mean were the smallest (0.996 86, 2.43 × 10−4, and 1.93 × 10−4), respectively. The results showed that the model number 7 could describe and predict the drying process of the pills. The experimental data had the effective diffusion coefficient (Deff) value of 8.6 × 10−11—3.13 × 10−10 m2/s, and the activation energy (Ea) was 30.97 kJ/mol. Conclusion: The hot air drying process of EP can be modeled and verified. The research provides a new way to explore the drying characteristics and quality of pills.
5.Cloning and Alignment of the Partial Mannanase Gene of Bacillus spp

By functional plates,16 strains which can produce?-mannana-se were isolated frnm 28 Bacillus spp.Using a pair of degenerated primers,the conserved fragments of?-mannanase gene from the selected strains were amplified by PCR.The obtained nucleotide fragments were sequenced and compared with the homologous?-mannanase genes in GenBank and a phylogenetic tree was generated.Comparing to the genes coding?-mannanase published,the cloned nucleotide fragments show the highest sequence identity between 62% and 98%.The genes coding fnr?-mannanase of Bacillus circulus have low identity while the?-mannanase genes of Bacillus subtilis and other Bacillus spp. have high identity.
6.Effects of dioscornin tablet containing serum on NF-kappaB p65, STAT3, and VEGF mRNA expressions in rats' synovial cell strain RSC-364 induced by IL-17 and TNF-alpha.

OBJECTIVETo observe the effects of Dioscornin Tablet (DT) containing serum on nuclear factor of kappa B (NF-kappaB) p65, signal transducer and activator of transcription 3 (STAT3), and vascular endothelial growth factor (VEGF) mRNA expressions in rats' synovial cell strain 364 (RSC-364) induced by interleukin-17 (IL-17) and tumor necrosis factor-alpha (TNF-alpha), and to investigate the underlying mechanisms for DT to inhibit angiogenesis of rheumatoid arthritis (RA).
METHODSIn this experiment, the vehicle control group, the cell model group, the DT containing serum group, and the positive control group (Tripterygium containing serum) were set up. The DT containing serum and the Tripterygium containing serum were prepared. The RA cell model was established by IL-17 combined TNF-alpha induced injury in RSC-364. The RA cells were intervened by DT containing serum and Tripterygium containing serum respectively. The DNA binding activity of NF-kappaB p65 was detected using TransAM NF-kappaB p65. The expression of STAT3 was observed using Western blot. The VEGF mRNA expressions were detected by real-time quantitative PCR.
RESULTSCompared with the vehicle control group, the NF-kappaB p65 activity, the expressions of STAT3 and VEGF mRNA increased significantly in RSC-364 induced by IL-17 +TNF-alpha (P < 0.01, P < 0.05). Compared with the model group, the NF-kappaB p65 activity, the expressions of STAT3 and VEGF mRNA decreased significantly in the DT containing serum group and the positive control group (P < 0.01, P < 0.05). There was no statistical difference between the two groups (P > 0.05).
CONCLUSIONDT inhibited the VEGF mRNA expression through inhibiting the NF-kappaB p65 activity and the STAT3 protein expression in the Janus kinase (JAK)-signal transducer and activating transcription factor pathway, thus inhibiting the angiogenesis of RA.

7.Comparative analysis of seven marine biological source of mineral drugs.

The marine biological source of mineral drugs recorded in Chinese Pharmacopoeia (2010 version) mainly including pearl, nacre, clam shell, common oyster shell, ark shell, cuttle bone, and sea-ear shell are widely used in clinical. Calcium carbonate and a small amount of protein are the main components in this type of drugs. In this paper, a systematical and comparable study were carried out by determination of calcium carbonate by EDTA titration method, the crystal of calcium carbonate by X-Ray powder diffraction and the total amino acids (TAAs) of the hydrolyzed samples by ultraviolet spectrophotometry method. As a result, the crystal structure is calcite for common oyster shell, mixture of calcite and aragonite for nacre and sea-ear shell, aragonite for the other drugs. The content of calcium carbonate ranged from 86% to 96%. Cuttle bone has the highest amount of TAAs among the seven drugs which reached 1.7% while clam shell has the lowest content of 0.16% on average. In conclusion, an effective method was developed for the quality control of marine mineral drugs by comprehensive analysis of calcium carbonate and TAAs in the seven marine mineral drugs.
