1.Sequencing VP4, VP7, NSP1, NSP4 genes of human rotavirus strain G1P8

Background: Rotavirus is the main cause of acute viral gastroenteritis in children under 5 years old. The virus leads to over 600000 children deaths a year in the world, 80% of which occur in the developing countries. In Viet Nam, 50%-70% the children\u2019s hospitalizations for acute diarrhea were resulted from rotavirus infection. Objective: To sequence nucleotides and amino acids of VP4, VP7, NSP1, and NSP4 genes of 5 passages of human rotavirus strain G1P8. Materials and method: A study was conducted in rotavirus sample of 5 passages of human rotavirus strain G1P8: B17A3; B17.3; B17.3 pp32vero15; B17.3 pp36TKP2; B17.3 pp43.7vero in Centre for Disease Control and Prevention, Atlanta, United State. Methods: using NucliSen Kit for detection of ARN; RT-PCR; sequencing genes by ABI 3100 machine. Results and Conclusion: Sequencing nucleotides and amino acids of VP4, VP7, NSP1, and NSP4 genes of 5 passages of human rotavirus strain G1P8 showed that: the number of nucleotide mutations ofVP4, VP7, NSP4 genes occurring among the passages were 3 (at nucleotit 175, 419, 790), 1 (at nucleotit 644), 3 (at nucleotit 134, 254, 482), respectively. All these mutations resulted in changes in amino acid composition. No mutation was found in NSP1 gene.

3.Research progress in receptors involved in rotavirus infection.

Rotaviruses, which are recognized as one of the major etiological agents among infants and young children with diarrhea, consist of three concentric layers of protein capsid with the enclosed double-stranded RNA genome. Rotaviruses infect host cells mainly by identifying the specific receptors on cell surfaces and binding to them. Therefore, receptors are important factors for viruses infecting cells. So far, there have been many receptors found to be involved in rotavirus infection, including sialic acid, integrin, Toll-like receptor, and blood group antigen. This article provides an overview of receptors involved in rotavirus infection.

4.Advances in Correlation between Rotaivrus and Histo-blood Group Antigens.

Group A rotaviruses (RVs) are major pathogens associated with acute gastroenteritis in young children and animals worldwide. VP4 is responsible for interaction with the host and viral attachment. Recent study showed that the distal portion of rotavirus (RV) VP4 spike protein (VP8*) is implicated in binding to human histo-blood group antigens (HBGAs), which is new cellular receptors on rotavirus, Published in Nature and Journal of Virology in 2012. The paper describes advances in correlation between rotaivrus and HBGAs, summarizes the main achievements has gotten, Clarify the significance of study on Rotaivrus and HBGAs.

5.Detection of bovine rotavirus G10P11 in a diary farm in Daqing, China.

Group A rotavirus are the most frequently detected viral agent associated with the acute diarrhea in calves. In order to investigate the situation of rotavirus strains circulating in diary farms, a total of 117 fecal specimens were collected from diarrhea calves under 4 weeks-age on Yinluo diary farm in Daqing region in China from 2008 to 2009. Ten specimens were detected to be positive by a Rotavirus Group A Diagnostic Kit, which confirmed that the rotavirus was important viral agent associated with diarrhea in this diary farm. Based on the new classification system, G10P[11] genotype was determined in rotavirus positive samples. Sequence and Phylogenetic analysis indicated DQ-75 strain was introduced into our country with imported bovine.

7.The genetic stability of recombinant adenovirus expressing human rotavirus VP6 gene which used Ad41 as vector.

OBJECTIVETo investigate the genetic stability of non-replicating recombinant adenovirus which used Ad41 as vector and could express VP6 gene of group A rotavirus during continous passage, in order to develop the vaccine of rotavirus.
METHODSThe recombinant adenovirus rvAd41-VP6 (o) was prepared by our laboratory early, it then was continuously propagated on 293TE7 cells for 14 passages. After that samples of the infected cells were collected at every 2 passages for the detection of the integration of the VP6 gene by PCR, and the expression of the target protein was detected by Western Blot analysis.
RESULTSAnalysis by PCR revealed that, there was stable integration of specific VP6 gene in the rvAd41-VP6 (o), Western Blot analysis confirmed that rvAd41-VP6 (o) could stably expressed the group-specific antigen structural protein VP6 (o), and it had preferable genetic stability.
CONCLUSIONThe recombinant adenovirus rvAd41-VP6 (o) which could stably express the VP6 (o) gene had favorable biological property in vitro, and it has provided a basis for further research of animal immunization.

Rotavirus is the leading causal agent of severe acute gastroenteritis in children aged <5 years. A specific pharmacologic agent for the treatment of rotavirus-infected children is lacking. In China, only the Luo Tewei oral vaccine (Lanzhou Institute of Biological Products, Shanghai, China), which is produced from Lanzhou lamb rotavirus vaccine (LLR), is available. Studies have hypothesized that the genotype of LLR is G10P[12], To identify the genotype of LLR by reverse transcription-polymerase chain reaction, we showed that the VP7 and VP4 genotypes of LLR were G10 and P[15], respectively, based on sequencing, alignment and phylogenetic analyses. In conclusion, we identified the genotype of rotavirus strain LLR to be G10P[15].
