1.The Complement Inhibition of Ursolic Acid and Tetrandrine on Tumor Related Signaling Pathways and Their Synergistic Antitumor Proliferation Effects

[Objective]This study was aimed to research the synergistic antitumor proliferation effects and their best proportion of ursolic acid(UA) and tetrandrine(Tet), a pair of compounds isolated from Chinese herbs which showed complement inhibition on the multiple signal pathways. [Methods] The reporter assays on tumor-related signal pathways for MAPK/ERK, MAPK/JNK, NF-κB, Wnt, Notch, Cell Cycle, Myc/Max and Hypoxia were used to study the effect of five different Chinese herbal compounds on tumor proliferation,it was concluded cepharanthine(Cep), Tet, 18α-glycyrrhetinic acid(18α-Gly), UA and luteolin(Lut). MTT assay and crystal violet staining were used to study the antiproliferative effect of 15 different compounds for the tumor cells of MDA-MB-231,SW480,MG63,PC3,DU145,HCT116,143B and MDA-MB-468, which is consisted with Cep, Tet, 18α-Gly, UA and Lut for the 15 different compounds. Coefficient of drug interaction(CDI) method was used to detect the synergistic effect of the two compounds. Combination of index(CI) and isobologram method was used to screen the best ratio of compounds in their antiproliferative effects. [Results] The signal pathway reporter assay showed that UA and Tet could complementarily inhibit tumor-related signaling pathways. And the results also showed that UA and Tet could induce synergetic anti-tumor cell proliferation in vitro. Furthermore, the optimal ratio of UA and Tet was 9:1 by using isobologram and CI method. [Conclusion] UA and Tet can be inhibited and complemented by 8 tumor-related signaling pathways, and we used MTT assay and crystal violet staining or other methods to confirm the synergistic antitumor proliferation effects, furthermore, the optimal proportion for UA and Tet were screened, and it provided a new insight to develop new anticancer formula in research.
2.Effect of Hyperbaric Oxygen on Cerebral Arteriosclerosis Vertigo

Objective To observe the clinical efficacy of hyperbaric oxygen on cerebral arteriosclerosis vertigo. Methods 100 patients were divided into treatment group (n=54) and control group (n=46). The treatment group accepted medicine and hyperbaric oxygen, while the control group accepted medicine only. The incidence of improvement and the measures of transcranial Doppler (TCD) were observed after treatment. Results The rate of improvement was 92.6% in the treatment group and 78.3% in the control group (P<0.05). The abnormal rates of TCD was 33.3% in the treatment group and 56.5% in the control group (P<0.05). Conclusion Hyperbaric oxygen therapy is effective on cerebral arteriosclerosis vertigo.
3.Determination of Ferulic Acid in Bogu Pill by HPLC

Objective To establish a method for the content determination of ferulic acid in Bogu Pill(BP). Methods Ferulic acid was obtained by ultrasonic extraction at room temperature and detected by HPLC. HPLC was performed with ODS chromatographic column, methanol-acetonitrile-1%acetic acid(15: 15: 70) as mobile phase and detection wavelength at 323 nm. Results The linearity of ferulic acid was in the range of 0.56 ?g~11.20 ?g/mL(r=0.9996), and the average recovery was 100.26%, RSD=1.48%. Conclusion This method was efficient and can be used for the quality control of BP.
4.Analysis on causes of death among children under 5 years old in a class 3A hospital during 2007-2015

Objective To investigate and analyze the death situation of children aged under 5 year old in a class 3A hospital during 2007-2015 to provide a basis for formulating corresponding interventional measures to reduce the disease mortality of children.Methods The medical records of children death in this hospital during 2007 2015 were retrospectively analyzed.Results The mortality rate of children under 5 years old in this hospital during 2007-2015 showed a decreasing trend year by year.There was the statistically significant difference in the mortality rates among different years,between male and female (3.58 %vs.2.10%) and between rural family registers and non-rural family registers(P<0.05);the neonatal mortality rate was highest (50.45 %);the top 5 places of death causes were premature delivery or low birth weight newborns,respiratory system diseases,nervous system diseases,congenital diseases and accidents.Conclusion The key for reducing the children mortality rate is to strengthen the perinatal health care,strengthen prenatal examinations and pregnant examinations,prevent the occurrence of respiratory system disease,pay attention to the influence of various factors on children health and prevention and treatment of accidents.
5.Analysis of paper publishing conditions by postgraduate background staff in an upper first-class hospital in Guangxi Province during 2010-2014

Objective Based on the analysis of papers published by postgraduate background staff from an upper first-class hospital in Guangxi during 2010-2014,claimed that the medical scientific research administrators should pay more attention and give more support to such staff.Methods Literature metrology method was used to describe and analyze the hospital staff,postgraduate staff resource composition and published papers from the annual growth rate,per capita,constituent ratio,quantitative analysis of core authors group.Results In the latest five years,the hospital postgraduate staff had published 169 papers,143 papers were published in statistical source journals or SCI;the amount of paper publication increased annually,the average of per author published paper was 0.39;but the quality still has a lot space for improvement.Conclusions Although the amount of paper publication by postgraduate background staff increased annually,the average paper number published per author showed the features of great fluctuation,the quality of those papers should be improved.The core authors were formed,but the quantity of this author group needs to be increased,as well as the scientific quality of those core authors.
6.Expressions of interleukin-25 and interleukin-33 in peripheral blood of patients with chronic urticaria and their significance

Objective To detect the expression levels of interleukin (IL)-25 and IL-33 in peripheral blood of patients with chronic urticaria.Methods Ninety-three patients with chronic urticaria were included in this study along with 30 healthy individuals.All the patients were treated with loratadine for four weeks.Blood samples were collected from the healthy controls and patients before and after the four-week treatment.Enzyme-linked immunosorbent assay (ELISA) was performed to detect the serum levels of IL-25 and IL-33.The relationship between the expression levels of the two cytokines and urticaria severity was analyzed.Results The serum levels of IL-25 and IL-33 in the patients before treatment were significantly higher than those in the healthy controls (IL-25,139.86 ± 28.48 vs.114.41 ± 34.00 ng/L,P < 0.01; IL-33,91.95 ± 21.88 vs.79.80 ± 30.72 ng/L,P < 0.05),and positively correlated with the severity of urticaria (r =0.38,0.42,respectively,both P < 0.01).After four weeks of treatment,clinical improvement was observed in 81.72% of these patients with a significant decrease in the serum IL-25 level (116.48 ± 21.94 vs.139.86 ± 28.48 ng/L,P < 0.01),but no obvious changes in the serum IL-33 level (90.88 ± 20.62 vs.91.95 ± 21.88 ng/L,P > 0.05) compared with those before treatment.Conclusions The expressions of IL-25 and IL-33 are elevated in peripheral blood of patients with chronic urticaria,and positively correlated with the severity of urticaria.
7.Value of quantitatively analyzing tissue perfusion parameters of contrast enhanced ultrasound in the evaluation of antiangiogenic therapeutics effect in a mouse hepatoma model

Objective To evaluate the value of quantitatively analyzing tissue perfusion parameters of contrast-enhanced ultrasound in the evaluation of antiangiogenic therapeutics effect. Methods Kun-min mouse were subcutaneously implanted with H22 cells. Ten mouse were treated with thalidomide (200 mg/kg once daily) by intraperitoneal injection over 7 days,starting at day 2 post-implantation. Ten control mouse were treated with an equivalent volume of 0.5 CMC. Contrast-enhanced gray-scale ultrasound was performed on day 8 after bolus injection of SonoVue (0.1 ml) and the imaging was recorded on cine.Regions of interest within tumour were analyzed off-line with the software of SonoLiver to determine the area under the curve (AUC), maximum intensity (Imax), perfusion index (PI), mean transit time (mTT),time to peak (TTP) and quality of fit (QOF). Immediately after imaging, minces were euthanized and tumour tissue removed for fixation in a 10% formalin solution. Microvascular density (MVD) was measured after anti-CD34 staining. Results The body weight and the tumor volume of treated tumors were significant different (lower) than that of control tumors (P <0.05). Treatment with thalidomide resulted into a significant decrease in AUC,PI and Imax in comparison with control tumors ( P <0.05). There were no significant difference in mTT and TTP between control and treated tumours ( P >0.05). Treated tumours were associated with a significantly lower MVD as compared with control tumours( P <0.05). ConclusionsQuantitatively analyzing tissue perfusion parameters of contrast enhanced ultrasound shows promise for monitoring tumor response to antiangiogenic therapy.
8.Role of noncoding RNA in vascular injury, remodeling and aging

Non-coding RNAs ( ncRNA ), including ribosomal RNA( rRNA), transfer RNA( tRNA), MicroRNA ( miRNA), long noncoding RNA(lncRNA) and small nucleolar RNA(snoR-NA), are a class of RNA that have multiple functions and are not translated to proteins. MicroRNA and lncRNA are involved in the injury, remodeling and aging of blood vessels, and it is necessary to understand the regulatory roles of MicroRNA and lncRNA in these processes. It is reported that MicroRNA and lncRNA are not only participated in the regulation of oxidative response, inflammation, cell proliferation and migration, and phenotype transition, they are also involved in the regulation of gene expression by conducting different mechanisms, including transcriptional regulation, post-transcriptional modification and chromatin remodeling. These aspects of regulation by MicroRNA and lncRNA are related to cardiovascular diseases, such as ath-erosclerosis, hypertension, myocardial infarction, stroke, pul-monary hypertension and diabetes, and thus provide a new way for genetic diagnosis and therapy of cardiovascular diseases.
9.Loratadine in combination with desloratadine for the treatment of chronic spontaneous urticaria in children:a clinical observation

Objective To evaluate clinical efficacy and safety of loratadine combined with desloratadine in the treatment of chronic spontaneous urticaria(CSU)in children. Methods A total of 177 children with CSU were enrolled into this study, and randomly and equally divided into 3 groups:combination group treated with an age?based dose of desloratadine tablet every morning and a weight?based dose of loratadine tablet before sleep every night for consecutive 28 days, loratadine group treated with a half tablet of placebo(starch tablet)every morning and oral loratadine tablet before sleep every night for consecutive 28 days, and desloratadine group treated with a half tablet of placebo (starch tablet) every morning and oral desloratadine tablet before sleep every night for 28 consecutive days. Possible adverse reactions were observed and recorded after the start of treatment, and therapeutic effects were evaluated at the end of treatment. Results A total of 166 patients completed the trial, including 55 in the combination group, 56 in the loratadine group and 55 in the desloratadine group. After 28?day treatment, the total response rate was significantly higher in the combination group(90.9%, 50/55)than in the loratadine group (71.4%[40/56],χ2=6.865, P<0.05)and desloratadine group(74.5%[41/55],χ2=5.153, P<0.05). No significant difference in the incidence of adverse reactions was observed among the combination group (10.9%[6/55]), loratadine group (8.9%[5/56]) and desloratadine group (9.1%[5/55], P > 0.05). Conclusion Combination of loratadine and desloratadine was superior to loratadine or desloratadine alone in the treatment of childhood CSU, and there was no significant difference in the incidence of adverse reactions among the 3 treatment groups.
10.An epidemiological study on insomnia in permanent residents over the age of 18 years old in YiBin City

Objective To describe the prevalence and risk factors of insomnia among adults in YiBin City. Meth?ods Using multi-stage stratified cluster sampling, 12000 subjects over 18 years old were identified in YiBin City. The subjects were screened using the insomnia survey self-edited and risk factors of insomnia were identified through logistic analysis. Results A total of 11227 subjects completed the survey in which 1671 people (14.9%, 95%CI:14.2%~15.5%) were diagnosed insomnia using ICD-10. Among them, 9.4% (157 patients) had seeked for professional help. The multi?variate logistic analysis showed that low level of life quality(OR=2.42,P<0.01), jobless(OR=1.87,P<0.01), over 60 years old(OR=1.69,P<0.01), fewer years of education(OR=1.41,P<0.01), female(OR=1.41,P<0.01)and lower annu?al per capita income(OR=1.15,P=0.01)were risk factors in insomnia people. Conclusion The incidence of insomnia is high in YiBin city. Sleep quality in high-risk group needs more attention and corresponding measures should be taken in order to increase the rate of asking for help in insomnia people.
