1.The migratory and invasive effects of CNE line after ALA photodynamic therapy

ObjectiveTo observe and detect the change of invasion and migration about CNE cell line after 5-ALA Photodynamic therapy.MethodsThe relation between the inhibition rate and 5-ALA concentration & energy density of lasers was detected by MTT.Scratch wound assay and matrigel artificial basement membrane invasion assay were perforned after photodynamic therapy with different photosensitizer concentration and energy density of lasers.ResultsThe results showed that photodynamic therapy can inhibit the growth of CNE cells.The migration of CNE cell was inhibited with appropriate photosensitizer concentration and energy density of laser after photodynamic therapy.It exhibited a linear relation between cell migration and the energy density, when the photosensitizer concentration was a maximum effective concentration of 1 mmol/L (P <0.05). The invasion of CNE cell was also inhibited after photodynamic therapy,there is a relationship between the change of invasion and 5-ALA concentration & energy density of lasers.It which exhibited a linear relation between cell invasion and the energy density in the same photosensitizer concentration (r >0.8, P <0.0001). ConclusionThe migration and invasion of CNE cells are both inhibited after photodynamic therapy. The change of migration and invasion is related to 5-ALA concentration & energy density of laser.
2.Diffusion and distribution of imported Oncomelania snails in non-endemic areas of schistosomiasis

Objecitve To observe the diffusion and distribution of imported Oncomdania snails in non-endemic areas of schistosomiasis.so as to provide scientific basis for developing the scheme of snail surveillance.Methods Oncomelania snails from the msrshland of Dongting Lake were imported in a waste land in March,2008,Qingxin County,which belonged to non-endemic areas of schistosomiasis.and their diffusion and distribution condition were observed from May to December in the same year.Resuits A total of 1 600 Oncomelania snails were imported.The occurrence rate of frame with snails in sunny days and cloudy rainy days were22.88%and 29.24%,respectively,and there was a statistically significant difference between them(χ~2=4.113,P=0.043).Those in waste Land and the sides/bottoms of the ditches were 9.38%and 35.99%,respectively,and the difference between them also showed a statistical significance(χ~2=67.246,P<0.001).The average density of living snails in sunny days and cloudy rainy days were 0.79 and 1.11 No./0.1 m~2,respectively,and those in waste land and the sides/bottoms of the ditches were0.11 and 1.34 No./0.1 m~2.The furthest diffusion distance of snailswas 4.5 m.Conclusions The detective opportunity of imported snails is relatively high in cloudy rainy days,which mainly distribute at the sides/bottoms of ditches and diffuse along the ditches.

5.Relationship Between Expression of Apoptosis-modulating Proteins and Multidrug Resistance in K562/VCR Cells

Purpose To explore the Relationship between expression of apoptosis-modulating proteins amdmultidrug resistance in K562/VCR cells. Methods Irnmunocytochemical methol and western blot wereused to analyze the expression of apoptosis-modulating proteins (Bcl - 2, Bcl-XL, Bax, Bak ) in multidrugresistant cell line K562/VCR and drugsensitive cell line K562. Results The positive cell rates ofapoptosis-suppressing protein Bcl-2 and Bcl-XL in K562/VCR were (40.0 ± 8.0) % and (60.0 ± 10.0) % .While the rates in K562 were (1.0 ± 0.3) % and (20.0 ± 4.0) %. There was significant difference in thepositive cell rates of Bcl - 2 and Bcl - XL between K562/VCR and K562 ( n = 3, P < 0.05 ). It was alsofound there was no significant difference in expression of Bax between K562/VCR and K562. Furthemore,Bak was not expressed in both K562/VCR and K562 or the expression was very low. Conclusions Wesuggest that Bcl-2 and Bcl-XL play important roles in multidrug resistance in K562/VCR, while Bax and Bakmight not be important.
6.Influence of Electro-acupuncture on E-Selectin and Resistin Protein Expression in Hippocampus of Rats with Chronic Stress Depression

Objective To observe the effects of electro-acupuncture on the E-selectin and resistin protein expression in the hippocampus of rats of chronic stress depression;To explore the role and mechanism of electro-acupuncture in the micro-environment of brain with depression. Methods 40 Fourty rats were randomly divided into four groups:blank group, model group, model+EA group, and model+fluoxetine group by using chronic stress combined with solitary raising methods, 10 rats in each group. Electro-acupuncture intervention was used 1 hour before the modeling. Electro-acupuncture was given at points Baihui (Du 20) and Yintang (Extra) for twenty minutes (2 Hz, 0.6 mA) every day. Fluoxetine was given at the volume of 5 mL/kg, dosage of 10 mg/kg. E-selectin and resistin protein expression in hippocampus were determined by biotin label-based antibody array. Results Compared with the blank group, the protein expression of E-selectin and resistin in the model group increased (fold change=1.23, 1.22). Compared with the model group, E-selectin and resistin expression decreased (fold change=0.65, 0.62;fold change=0.76, 0.65). Conclusion Electro-acupuncture intervention could down-regulate E-selectin and resistin expression in the hippocampus of chronic stress depression model.
7.Exploration of the interactive mechanism between graduates quality and teaching reform in Chinese medicine colleges

Through the theoretical analysis of teaching reform and quality management in tradi-tional Chinese medicine col-leges, this paper puts forward the necessity of improving the quality management and optimizing the reform of education and teaching. Meanwhile, this paper further expounds the comprehen-sive, guiding, operable, qualitative and quantitative principles of teaching reform and quality management in traditional Chinese medicine colleges. Based on the above analysis, the paper puts forward and prac-tices the reform countermeasures to improve the interactive mechanism between the quality of graduates and the reform of teaching in traditional Chinese medicine colleges. The countermeasures include establishing an effective quality tracking feedback mechanism for graduates, actively carrying out the monitoring and evalu-ation of teaching quality, and constructing the measurement and evaluation system of teaching quality of traditional Chinese medicine education. This paper combines the needs of the current society for the culti-vation of talents of traditional Chinese medicine and the training quality standards, so as to promote the development of traditional Chinese medicine.
8.Analysis on Neisseria gonorrhoeae infection status and related factors in outpatients of sexually transmitted diseases in Shenzhen

Objective:To understand the prevalence of Neisseria gonorrhoeae (NG) infection and related factors in outpatients of sexually transmitted disease (STD) in Shenzhen and provide scientific evidence for targeted intervention. Methods:A cross-sectional study was conducted in STD outpatients who were aged 18-49 years, had had sexual intercourse, had not received antibiotics in the last 2 weeks and sought medical cares in 22 medical institutions in 6 districts of Shenzhen from 15 April, 2018 to 16 May, 2018, their basic information and urine samples were collected, and NG in urine sample was detected by nucleic acid amplification test. Logistic regression analysis was conducted to identify the related factors for NG infection.Results:The information collection and sample collection were completed for 8 324 STD outpatients, among these STD outpatients, 196 were NG positive, with a positive rate of 2.4% (196/8 324). The positive rate of NG in men (5.8%, 148/2 567) was higher than that in women (0.8%, 47/5 742) ( χ2=189.43, P<0.05). Aged 24 years or below ( OR=2.11, 95% CI: 1.44-3.09), single/divorced/widowed ( OR=1.98, 95% CI:1.38-2.84), having casual sex in the last 3 months ( OR=1.77, 95% CI:1.29-2.43) were the related factors for NG infection. Conclusions:We found that NG infection rate was high in STD outpatients in Shenzhen. The infection rates in men and in women differed obviously. It is necessary to conduct gonorrhea screening and intervention in STD outpatients with high-risk behaviors, such as extramarital and casual sex behaviors, and standardize the treatment to reduce the incidence and spread of gonorrhea and other STDs.
9.A survey of epidemic status of principal human parasites in Guangdong Province in 2015

Objective To analyze the epidemic status of principal human parasites, so as to provide scientific evidence for making prevention countermeasures for Guangdong Province in the future. Methods In 2015, a survey was performed according to the scheme of "The 3rd National Survey of Principal Human Parasites"as well as the incidence of parasites disease in Guangdong Province,the residents at 48 investiation sites in counties were surveyed. The survey of the soil-transmitted nematodes (Ascariasis, Ancylostoma, Trichuris trichiura and Enterobius vermicularis) and intestinal protozoa were performed based on the ecological regions and stratified by economic and geographic situation.The survey of food borne Clonorchis sinensis was performed along with the soil-transmitted nematodes in rural areas, and it was performed by the sample method of random cluster in cities and towns.The residents in each investigation site as the object, the ovum of the soil-transmitted nematodes, Clonorchis sinensis and other helminths were examed by the modified Kato-Katz method, test tube filter paper was used to identify Hookwormspecies, and the intestinal protozoa was checked by direct smear method. The transparent tape anal swabs method for children aged 3 - 6 years to check Enterobius vermicularis. Results Totally 12 401 residents of 48 survey sites from 22 counties were surveyed, and the total infection rate of intestinal parasites was 8.29%(1 028/12 401). The infection rate of soil-transmitted nematodes was 3.39% (420/12 401),in which the infection rate of Ascariasis, Ancylostoma, Trichuris trichiura and Enterobius vermicularis were 0.52% (64/12 401), 1.89%(234/12 401),0.46%(57/12 401),and 0.52%(65/12 401),respectively.The infection rate of Clonorchis sinensis was 4.90%(608/12 401). Nine hundred and sixty-seven children were tested for eggs of Enterobius vermicularis with the infection rate of 12.41% (120/967). The number of hook larva culture was 153, among them, 140 were hookworm larvae of America and no duodenal hookworm larvae and other nematode species were found.Totally 9 309 residents were tested for intestinal protozoa infection,and the infection rate was 0.31%(29/9 309). Conclusion In Guangdong Province, the infection rate of soil-transmitted nematodes is decreasing while the infection rate of Clonorchis sinensis is still high, and the control work of parasites still should be strengthened especially for food borne parasitic diseases.
10.Animal experimental study of biliary fibrosis in end to end anastomosis after transection of the common bile duct

Objective:To explore the mechanism of biliary fibrosis after end to end anastomosis of bile duct.Methods:12 Neijiang miniature pigs (6-8 months old, weight 30-40 kg) were divided into experimental group ( n=6) and control group ( n=6). The model of end to end anastomosis after transection of the common bile duct was established in experimental group. Control group was a sham operation group, and only T tube was placed. After 9 months, Masson staining, fluorescence quantitative PCR and immunohistochemistry were used to analyze the expressing changes of pro-fibrotic factor transforming growth factor-β1 (TGF-β1), connective tissue growth factor (CTGF) and epithelial-mesenchymal transition (EMT) markers, including cytokeratin-19 (CK19), E-cadherin (E-Cad) and fibroblast specific protein-1 (S100A4), α-smooth muscle actin (α-SMA) and collagen components Collagen I (COL-1), collagen III (COL-3) and fibronectin (FN) in the anastomotic bile duct tissues. Results:Masson staining showed that the submucosal collagen fibers increased significantly in the experimental group. Compared with the control group, the mRNA expression of TGF-β1 [(3.482±0.313) vs. (1.000±0.102), t=18.43, P<0.001], CTGF [(2.160±0.287) vs. (1.000±0.103), t=9.32, P<0.001] were increased, the difference was statistically significant. Compared with control group, the mRNA and protein expression of CK19 and E-Cad were decreased in the experimental group, while the mRNA and protein expression of S100A4 and α-SMA were increased in the experimental group (all P<0.01). Conclusion:It was feasible in the short term to perform an end-to-end anastomosis after transection of the common bile duct, but there was an obvious fibrosis in the anastomotic bile duct tissue at later time.
