1.Study on the antibacterial activity of four kinds of nano-hydroxyapatite composites against Enterococcus faecalis.

OBJECTIVEThis study aims to compare and determine a kind of nano-hydroxyapatite composite material with good antibacterial efficacy on Enterococcusfaecalis (E. faecalis) in vitro.
METHODSWe investigated the antimicrobial activity of four kinds of nano-hydroxyapatite composites, namely, silver/hydroxyapatite composite nanoparticles (Ag/nHA), yttrium/hydroxyapatite composite nanoparticles (Yi/nHA), cerium/hydroxyapatite composite nanoparticles (Ce/nHA), and hydroxyapatite nanoparticles (nHA), against E. faecalis in vitro using the agar diffusion and broth dilution method by measuring the growth inhibition zone and the minimum inhibitory concentration (MIC), respectively.
RESULTSThe agar diffusion test results showed that Ag/nHA displayed an obvious growth inhibition zone, whereas Yi/nHA, Ce/nHA, and nHA showed no influence on E. faecalis. The MIC value of Ag/nHA was 1.0 g.L-1, and the three other materials had no effect on E.faecalis even at the high concentration of 32.0 g.L-1.
CONCLUSIONAg/nHA display a potential antimicrobial efficacy to planktonic E.faecalis. Whereas, the three other kinds of nano-hydroxyapatite composites (Yi/nHA, Ce/nHA, nHA) show no influence.

Objective To observe the effect of Shenfu Injection (SI)on adriamycin(ADR)-induced calcium overload in myocardial cells of rats.Methods SD rats were randomized into normal group, saline group, ADR group, ADR+saline group, low-, middle-and high-dosage SI groups (treated with SI 0.3, 0.6 and 1.2gkg-1d-1). Except the normal and saline groups, the rats in other groups were given ADR injection. Meanwhile,SI groups received treatment of SI for two weeks. Three weeks after modeling, the single myocardial cells were isolated. The free Ca2+ concentration was detected by ionic imaging method, and the ionic current through L-type calcium channel in cellular membrane was observed by patch clamp technique.Results The free Ca2+ concentration, the mean open probability of L-type calcium channel in cellular membrane and the mean ionic current through L-type calcium channel were increased in ADR group (P0.05).Conclusion The mechanism of SI counteracting the ADR-induced calcium overload in myocardial cells may be related with preventing external calcium from entering the cells,and with inhibiting the release of internal calcium.
4.Effects of Buyang Huanwu Decoction on L-type Calcium Channels of Cortical Neurons in Rats with Global Cerebral Ischemia and Reperfusion at Different Time Points

【Objective】 The effect of pretreatment with Buyang Huanwu Decoction(BHD) on L-type calcium channels of cortical neurons in rats with global cerebral ischemia was observed,thus to explore the molecular therapeutic mechanism of BHD for cerebral ischemia as well as the pathogenic mechanism of Ca2+ message in ischemic injury of neurons.【Methods】SD rats were randomized into pseudo-operation group,model groups and BHD groups.And the model groups and BHD groups were divided into six groups according to the time points after reperfusion(2nd,12th,24th,48th and 72nd hour after reperfusion respectively).BHD groups received 0.64g/kg BHD by gastric infusion,bid,for 5 continuous days.Except the rats in pseudo-operation groups,the rats in other groups were induced global cerebral ischemia by modified Pulsinelli's four-vessel occlusion.Cerebral cortical neurons in rats were isolated promptly in 2nd,12th,24th,48th and 72nd hour after reperfusion.The recorded single-channel current was amplified by the EPC-9 patch-clamping amplifier,and then input into the computer by Pulse+Pulsefit.The analytical software TAC was used to detect the opening time and opening probability of the L-type Ca2+ channels in rats at different time points after reperfusion.【Results】In the model groups,the opening time of the L-type Ca2+ channels was prolonged after reperfusion as compared with that in the pseudo-operation group,and the opening probability of the L-type Ca2+ channels arrived at the peaks in 2nd and 24th hour after reperfusion.In BHD groups,the opening time in 72nd hour after reperfusion was decreased(P

Objective To study effects of baicalin(Bai) on AngⅡ in diabetic nephropathy(DN) rats and to explore its preventive and therapeutic effect on treating DN from the non-hemodynamic mechanism of AngⅡ.Methods Nineteen specific pathogen-free male rats were randomly divided into three groups:normal control group(5 rats),diabetic nephropathy model control group(7 rats),baicalin treated group(7 rats).DN rat model was established by one-dose intraperitoneal injection of streptozotocin.Seven weeks after the modeling,rats in baicalin group recieved intraperitoneal injection of baicalin solution(40 mg/kg),rats in DN model group received intraperitoneal injection of normal saline(1 mL for each rats),and the normal control group did not receive any intraperitoneal injection.After treatment for 6 weeks,orbital blood from all rats was collected to determine plasma AngⅡ by radioimmunoassay.Unilateral kidney was processed by 4 % paraformaldehyde fixation and then was used for the determination of TGF-? by immunohistochemical assay.The other unilateral kidney was got out to prepare for renal cortex homogenate,and then was used for the determination of renal tissue AngⅡ by radioimmuno assay.Results The quantity and degree of TGF-? expression in baicalin group was decreased,and there was statistical significance between baicalin group and the model group(P
6.Expression and diagnostic value of 34βE12, Galectin-3 and HBME-1 in thyroid nodules

[ABSTRACT]OBJECTIVETo study the expressions of 34βE12, Galectin-3 and HBME-1 in thyroid nodules, and to explore its diagnostic value for papillary thyroid carcinoma (PTC).METHODSEn VisionTM immunohistochemical technique was used to detect the expression of 34βE12, Galectin-3 and HBME-1 in 352 thyroid lesions. The correlation between the expressions of the 3 protein markers and clinicopathological characteristics was evaluated. The receiver operating characteristic area under the curve (ROC-AUC) and their index for diagnosis evaluation were also calculated.RESULTSThe positive rates of 34βE12, Galectin-3 and HBME-1 in 246 PTC lesions were significantly higher than those in benign nodules (P<0.001). There was no relationship between the expression of the 3 protein markers and clinicopathological characteristics (eg. gender, age, numbers of lesions, tumor size, capsular invasion, lymph node metastasis, TNM staging). The ROC-AUC of 34βE12, Galectin-3 and HBME-1 for diagnosis of PTC was 0.936, 0.915 and 0.898 respectively. The sensitivity of 34βE12, Galectin-3 and HBME-1 for diagnosis of PTC was 94.3%, 95.5% and 91.1% respectively, while the specificity was 81.1%, 71.7% and 83.0% respectively, and the diagnostic accuracy rate was 90.3%, 88.4% and 88.6% respectively.CONCLUSION The expressions of 34βE12, Galectin-3 and HBME-1 are statistically different between PTC and benign lesions, but no associations are found with clinicopathological characteristics, indicating the three protein markers have important diagnostic value for PTC.

8.Clinical significance of detection for mutated BRAF gene in thyroid nodules based on amplification refractory mutation system

OBJECTIVE To explore the feasibility of detection for mutated BRAF V600E gene based on amplification refractory mutation system(ARMS),and to evaluate its clinical significance of BRAF V600E gene mutation in thyroid nodules.METHODS The method of ARMS was used to detect BRAF V600E mutation status in 179 patients with PTC and 115 patients with benign lesions.The diagnosis index of BRAF V600E mutation status for identifying the nature of the thyroid nodule was calculated.The potential correlation between BRAF V600E mutation and PTC clinicpathological characteristics was also analyzed.RESULTS Detection of BRAF V600E mutation status in thyroid lesions based on ARMS was feasible and believable.The positive rate of mutated BRAF V600E gene in PTC was 82.68%,whereas the rate in benign lesions was only 1.74%,indicating statistical differences between the two groups(x2=183.568,P<0.01).The diagnostic sensitivity of BRAF V600E mutation was 82.68%,specificity was 98.26%,accuracy was 88.76%,and Youden index was 0.8094.There was no associations between the BRAF V600E mutation status and PTC clinicpathological characteristics(eg.gender,age,tumor size,numbers of lesions,bilateral lesions,extrathyroidal extension and lymph node metastasis).CONCLUSION Detection of BRAF V600E mutation based on ARMS has higher sensitivity and specificity in distinguishing PTC from benign lesions,indicating BRAF V600E gene is an ideal marker of PTC for clinical early diagnosis.
