1.Effects of Yigu capsule on the level of cbf ?-1 mRNA in bone of ovariectomized osteoporosis rats

AIM:To analyze the regulatory effect of Yigu capsule on core binding factor alpha 1 (cbf ?-1) gene expression in bone of ovariectomized osteoporosis (OP) rats. METHODS: Thirty-six 10-month old Sprague-Dawley female rats were randomized into three groups: sham-operated group, model group and drug group. After intervention by the corresponding methods, the femurs were collected, SYBR green Ⅰ fluorescence quantitative PCR technique was applied with the internal control of GAPDH according to the relative quantitative formula (2-Ct), the differentially expressed multiples between the model group or drug group and sham-operated group were calculated. RESULTS: Quantitative formula analysis showed that the level of cbf ?-1 gene expression in bone of model groups was decreased than that in sham-operated rats [compared with sham-operated group, P0.05). CONCLUSION: The results of this study show that cbf ?-1 gene expression in bone tissue of ovariectomized OP rat is decreased, and Yigu capsule increases the level of cbf ?-1 gene expression in bone of OP, indicating that Yigu capsule induces bone marrow mesenchymal stem cells into osteoblasts.
2.Relationship between enhanced CT examination and acute kidney injury in patients with acute pancreatitis

ObjectiveTo investigate the relationship between enhanced CT examination and acute kidney injury in patients with acute pancreatitis.MethodsThe clinical data of 162 acute pancreatitis patients were analyzed retrospectively.Acute kidney injury was diagnosed and graded by RIFLE criterion.The patients were divided into two groups according to enhanced CT examination:enhanced CT group and non-enhanced CT group.ResultsEnhanced CT group had 85 cases,and non-enhanced CT group had 77 cases.The increased creatinine level in enhanced CT group was higher than that in non-enhanced CT group [(59 ± 13 ) μ mol/L vs.(38 ± 11 ) μ mol/L],and there was significant difference between two groups (P <0.01 ).The incidence of acute kidney injury in enhanced CT group was higher than that in non-enhanced CT group [30.6% (26/85) vs.15.6% (12/77)],and there was significant difference between two groups (P <0.01 ).Enhanced CT examination increased the risk of acute kidney injury occurrence (OR =1.293,95% CI1.089-1.443).After adjusted by diabetes and APACHE Ⅱ score,enhanced CT examination was still anindependent risk factor of acute kidney injury (OR =1.282,95% CI 1.185-1.377 ).ConclusionsEnhanced CT examination is a potential risk factor of acute kidney injury in acute pancreatitis patients.The time of enhanced CT examination is worthy of further study.
3.The interventional evaluation and management of arteriovenous fistulae in patients with hepatic cancer

Objective To analyze the DSA manifestations of arteriovenous fistulae in hepatic cancer and to discuss the appropriate method for obstructing the fistulae in order to improve the therapeutic results for hepatic cancer associated with arteriovenous fistulae. Methods The imaging findings in 183 patients of hepatic cancer with urteriovenous fistulae, which were. selected from 637 patients with liver cancer, were retrospectively analyzed. For cases with mild arteriovenous fistulae (n = 89) the tumor-feeding arteries were embolized after iodized oil embolization procedure, while for cases with moderate and severe aneriovenous fistulae (n = 94) the abnormal shunts were obstructed before performing chemoembolization of the tumor in order to ensure that the effective infusion of the drug into the lesion and the sufficient deposit of iodized oil in the tumor could be obtained. Results The tumor-feeding arteries were successfully embolized in all 89 patients with mild arteriovenous fistulae. Of 94 cases with moderate and severe arteriovenous fistulae,excellent deposit of iodized oil was seen in 64 (68.1%), incomplete embolization in 13 (14.83%), poor oil deposit due to over-embolization in 11 (11.7%) and failure to block the fistulae as the fistulae were too large or too diffuse in 6 (6.4%). No serious complications occurred. Conclusion The arteriovenous fistulae accompanied with hepatic cancer can be effectively obstructed in most patients, with resultant clinical improvement of the patients' condition. This therapeutic technique is safe and reliable.
4.Effect of catalpol on activity of osteoblasts/osteoclasts and osteoblast ERα/βmRNA expression in osteoblast-osteoclast co-culture system

[ ABSTRACT ] AIM: To investigate the effect of catalpol on the activity of osteoblasts ( OB ) and osteoclasts ( OC) , and OB estrogen receptor ( ER) α/βmRNA expression in the OB-OC co-culture system.METHODS: OB and OC were isolated from the SD rats of 1 and 5 days old.In the OB-OC co-culture system, different concentrations of catalpol including low dosage (0.05 , 0.1, 0.5 and 1 mg/L), middle dosage (2, 5 and 10 mg/L), and high dosage (20, 50 and 100 mg/L) were added into the culture medium to detect the changes of OB proliferation by MTT assay.The catalpol at maximal dosage was added to OB section to detect the alkaline phosphatase ( ALP) activity of OB by pNPP method.The mRNA expression of ERα/βin the OB treated with catalpol in the co-culture system was detected by RT-PCR.The catalpol at maximal dosage was added to OC group to detect the activity of OC by microscopy and tartrate-resistantacid phosphatase ( TRAP) activity detection.RESULTS:In 0.05~2 mg/L catalpol groups, the proliferation of OB was significantly in-creased as compared with control group in the co-culture system, and it reached the maximum value when catalpol was at 0.05 mg/L, while in 5~100 mg/L catalpol groups, the proliferation of OB was not increased.The ALP activity of OB in 0.05 mg/L catalpol group was higher than that in control group.The catalpal at 0.05 mg/L promoted the mRNA expression of ERβin OB in the co-culture system, but did not increase the mRNA expression of ERαas compared with control group. Catalpol at 0.05 mg/L obviously inhibited the bone resorption and the TRAP activity in OC.CONCLUSION: Catalpol stimulates the proliferation and activity of OB, inhibits the bone resorption and activity of OC, and increases the mRNA ex-pression of ERβin OB in the OB-OC co-culture system, suggesting that high mRNA expression of ERβmay be the regula-tory pathway of catalpol in response to bone metabolism.
5.Therapeutic effect of carvedilol combined valsartan on chronic heart failure

Objective: To explore therapeutic effect of carvedilol combined valsartan on chronic heart failure (CHF), and its influence on cardiac function and level of high sensitive C reactive protein (hsCRP).Methods: A total of 78 CHF patients, who were treated in our hospital from Jul 2011 to Jul 2015, were selected.According to therapeutic method, they were divided into valsartan group (n=38, received valsartan based on routine treatment) and combined treatment group (n=40, received carvedilol based on valsartan group).Therapeutic effect, cardiac function, serum hsCRP level and quality of life (QOL) score before and after treatment were observed and compared between two groups.Results: Total effective rate of combined treatment group was significantly higher than that of valsartan group (92.50% vs.71.05%), P=0.022.Compared with before treatment, there were significant reductions in heart rate, cardiothoracic ratio (C/T), left ventricular end-diastolic dimension (LVEDd) and serum hsCRP level, and significant rise in left ventricular ejection fraction (LVEF), left ventricular fractional shortening (LVFS), emotional index, health index and life satisfaction score in both groups after treatment except C/T of valsartan group, P<0.01 all;compared with valsartan group after treatment, there were significant reductions in heart rate [(124.39±10.07) beats/min vs.(101.18±8.72) beats/min], C/T [(0.63±0.15) vs.(0.53±0.12)], LVEDd [(43.32±4.02)mm vs.(40.38±3.76)mm] and serum hsCRP level [(32.12±3.99) pg/ml vs.(10.29±2.08) pg/ml], and significant rise in LVEF [(45.59±9.99)% vs.(59.97±10.02)%], LVFS [(24.58±5.67)% vs.(31.89±6.68)%], scores of emotional index [(70.20±10.02) scores vs.(86.58±12.28) scores], health index [(73.36±8.08) scores vs.(85.58±11.01) scores] and life satisfaction [(74.34±8.39) scores vs.(88.12±13.35) scores] in combined treatment group,P<0.05 or <0.01.Conclusion: Carvedilol combined valsartan possesses significant therapeutic effect on chronic heart failure.It can significantly reduce hsCRP level, improve cardiac function and quality of life in these patients, which is worth extending.
6.Application of periodic reporting system in clinical teaching

Objective To explore the effect of periodic reporting system applied in clini-cal teaching.Method 62 nursing students of grade 2006 and 2007 practising in our hospital were selected and each group leaders reported their practical information to the teaching regula-tor every other month.Result If nursing students rehearsed the operational procedures and the items and communication skills that the teachers asked them to pay attention to,nursing stu-dents would be more self-confident in practice. Meanwhile,about 70% teachers didn’t introduce the working environment or clinical nursing knowledge to nursing students.Instead,they made them run errands.Conclusion Periodic reporting system was conducive to nursing students’suc-cessful completion of the internship program,and conducive to teaching regulators to understand nursing students’practical demands and the overall level of clinical teaching,which increased the overall level of clinical teaching and nursing students’satisfaction of teaching.
7.Unilateral Low - frequency Sensorineural Hearing Loss (40 Cases Analysis)

Forty cases of unilateral low- frequency sensorineural hearing loss(LFSNHL) were analysed. The symptoms included tinnitus(87.5% ), deafness(75% ), vertigo(35%) and ear fullness(25% ). Audiome-try showed LFSNHL in all cases. The threshold difference between low frequencies and high frequencies was 26. 32 dB. The cause of LFSNHL, the relation between tinnitus, LFSNHL and endolymphatic hydrops were discussed. We hope that the diagnosis of LFSNHL should be standardized.
8.Study on polymorphism of human eighth complement component(C8A)by using allele specific amplification method

For the purpose of detecting the polymorphism of the human eighth complement component(C8A),an allele specific amplification method were established for detecting two alleles of C8A based on a nucleotide exchange of the C8? cDNA sequence.100 DNA blood samples from unrelated individuals of Han population in Chengdu were typed by this method.The result of comparison with the data of protein typing of the same samples using SDS PAGE method proved that the described method is efficient,correct and reliable for C8A genotypes.This methtod is valuable for further application in population genetic study and forensic science practice.

Objective To evaluate the in vivo efficacy of modified platelet(PLT) additive solution(PAS-ⅢM) with trehalose as a substitute for plasma for the storage of platelet concentrates at low temperature(10℃).Methods Rabbit platelet concentrates were collected and stored in different media and temperature conditions,plasma(22℃),70%PAS-ⅢM/30% plasma(10℃) and plasma/trehalose(-85℃).Platelets in plasma(22℃) on storage day 3,in 70%PAS-ⅢM/30% plasma(10℃) on storage day 3,7,9,and in plasma/trehalose(-85℃) thawed on day 20 were transfused to rabbit thrombocytopenia model.The in vivo recoveries and survivals of the three preserved platelets and fresh platelets were measured and evaluated after transfusion into rabit model of thrombocytopenia.Results The survival of platelets stored in 70% PAS-ⅢM/30% plasma(10℃) on day 9 was lower than that of fresh platelets(P0.05).Recovery and survival of frozen platelets were significant lower than those of the other groups(P
