BACKGROUND:It is necessary to dynamicaly monitor the survival, recognition and migration of neural stem cels after implantation.
OBJECTIVE:Toin vitro label fetal rat neural stem cels using MRI technology so as to provide applied evidence of neural stem cels in nervous system repair.
METHODS:Fetal rat neural stem cels were isolated, cultured and labeled folowed by identification and cel viability detection. A rat model of cerebral ischemia-reperfusion injury was established. Fetal neural stem cels labeled by superparamagnetic iron oxide particlesin vitro were transplanted into the left brain of model rats, and unlabeled fetal rat neural stem cels transplanted into the right brain. Prussian blue staining was used to observe the colonization and migration of implanted neural stem cels. MRI tracing was employed to monitor the signal changes of neural stem cels dynamicaly afterin vivo transplantation.
RESULTS AND CONCLUSION:Over 95% fetal rat neural stem cels were labeled successfuly by superparamagnetic iron oxide particles, and under electron microscope, there were iron particles in labeled neural stem cels, which were concentrated in the lysosome and endosome. MRI results showed that the labeled neural stem cels had a changing trend of low signals. No difference was found in the cel viability between labeled and unlabeled cels, but T2WI and T2*WI signals were reduced in labeled neural stem cels. These findings confirm that superparamagnetic iron oxide-labeled fetal rat neural stem cels can highly express, and MRI tracing can be used forin vivo monitoring of neural stem cels.

Objective To study the pollution levels, characteristics, causes and related adverse effects of indoor volatile organic compounds(VOCs) on human health after decoration. Methods The concentration of indoor air formaldehyde and TVOC were detected, meanwhile the adverse health effects were investigated by questionnaires. Result The results showed that the houses in which formaldehyde concentration exceeded the national indoor air quality standard occupied 47.1%, and three months later, the rate still was 40%,the highest level was 6.56 times of the standard. While the rate of exceed the national standard of TVOC was 76% within one month and 0% three months later respectively. As far as the concerned subjective syndromes, only the complaint of disgusted smell was significantly different between those who lived in newly decorated rooms and those who lived in the houses which taken as the control. Conclusion Now the pollution level of VOCs caused by decoration is some lower than before, but the pollution of formaldehyde was still pervasive and serious. The causes of pollution lay in artificial board materials used for furniture and decoration. Moreover, the complaints of the discomfort of eyes and upper respiratory tract will increase as people moved in the newly decorated rooms.

The study aimed to establish a population pharmacokinetic/pharmacodynamic (PPK/PD) model of warfarin. PCR-RFLP technique was used to genotype the CYP2C9 and VKORC1 polymorphisms of 73 patients. RP-HPLC-UV method was used to determine the 190 plasma concentrations of warfarin. Application of NONMEM, the clinical information and 263 international normalized ratio (INR) monitoring data were used to investigate the effect of genetic, physiological, pathological factors, other medication on clearance and anticoagulant response. The final model of warfarin PPK/PD was described as follows: CL = θCL · (WT/60)θWT · θCYP · eηCL (if CYP2C9*1/*1, θCYP = 1; if *1/*3, θCYP = 0.708); EC50 = θEC50 · θVKOR · eηEC50 (if VKORC1- 1639AA, θVKOR = 1; if GA, θVKOR = 2.01; V = θV; K(E0) = θK(E0); Emax = θEmax; E0 = θE0 · eηE0. Among them, the body weight (WT), CYP2C9 and VKORC1 genotype had conspicuous effect on warfarin PK/PD parameters. The goodness diagnosis, Bootstrap, NPDE verification showed that the final model was stable, effective and predictable. It may provide a reference for opitimizing the dose regimen of warfarin.

Objective To investigate the clinical value of the gated blood pool imaging phase analysis method in the evaluation of left ventricular mechanical synchronization in patients with chronic heart failure. Methods A total of 169 patients with chronic heart failure were enrolled in our study , using gated blood pool imaging phase analysis method to obtain left ventricular phase angle width ( PHB) and left ventricular phase angle standard deviation ( PSD) as evaluating left ventricular mechanical synchrony index; using tissue Doppler imaging (TDI) measurement of the standard deviation of systolic peak time(Ts-SD) of each segment by using the current prevailing 12 non-apical segments analysis method as evaluating left ventricular mechanical synchrony index, and parameters derived from both methods were compared. Results LVPHB was highly correlated with Ts-SD (r = 0. 83 ,P = 0. 000 ) . LVPSD was modestly correlated with Ts-SD ( r - 0. 69, P = 0. 000) . The ejection fraction measured by echocardiography was (42.93 ± 14. 89) % ,which was significantly higher than that measured by ERNA (39. 76 ± 17. 89)% (P <0. 01). Conclusions The evaluation of left ventricular mechanical synchrony in patients with chronic heart failure by the gated blood pool imaging can provide similar information with TDI, which can simultaneously measure two ventricular functions and get more accurate measurement of ejection fraction. Cardiac resynchronization therapy patients can be identified by combining two kinds of approaches, and cardiac resynchronization therapy responders could be improved as well. More patients with heart failure can benefit from cardiac resynchronization therapy therapy.

BACKGROUND: Cerebrovascular disease often causes dysfunction of the brain nerve, and nerve cel apoptosis is the important factor of cerebral nerve dysfunction. The excessive expression of c-fos can block the transduction of intracelular signal so that producing some apoptosis-promoting factors, which involve in nerve cel apoptosis process after ischemia injury of brain. Bcl-2 is an inhibited factor. It might to be the key to treat ischemic cerebrovascular disease by inhibiting or reducing the apoptosis of nerve cels after ischemia injury. OBJECTIVE: To investigate the therapeutic effect and mechanism of the Total Flavone of Hawthorn Leaf on chronic cerebral ischemia rats. METHODS: A total of 72 healthy male Sprague-Dawley rats were randomly divided into sham surgery group, model group, Total Flavone of Hawthorn Leaf group and ginkgo leaf group. Permanent bilateral carotid artery ligation was used to prepare chronic cerebral ischemia model in the model group, Total Flavone of Hawthorn Leaf group and ginkgo leaf group. Total Flavone of Hawthorn Leaf group and ginkgo leaf group respectively received 140 mg/kg Total Flavone of Hawthorn Leaf and 12.3 mg/kg ginkgo leaf intragastricaly for 36 days from 36 days after model induction. Model group and sham surgery group received 3.5 mL/kg physiological saline intragastricaly. RESULTS AND CONCLUSION: Compared with the model group, the expression of c-fos protein significantly deceased in the Total Flavone of Hawthorn Leaf group (P < 0.01), Bcl-2 expression levels significantly increased (P < 0.01), and Ca2+ content decreased (P < 0.05). Moreover, no significant difference in above indexes was detected between Total Flavone of Hawthorn Leaf group and ginkgo leaf group (P> 0.05). These data indicated that the protective effect of Total Flavone of Hawthorn Leaf on chronic cerebral ischemia was associated with its inhibition of neuronal apoptosis. Its mechanism of anti-apoptosis might be associated with up-regulating expression of Bcl-2, down-regulating expression of c-fos and decreasing Ca2+ content in brain.
