1.SYNERGISTIC EFFECT OF HYPERTHERMIA ON PHOTOCARCINORIN PHOTODYNAMlC THERAPY

In this paper, we describe a synergistic effect of hyperthermia on photocarcino-rin photodynamic therapy. The results suggest that the synergistic action of hyperthermia and photodynamic reaction was markedy better than that of hyperthermia alone plus photocarcinorin alone. with hyperthermia treatment (42℃, 30 min) alone, the fraction of cells inactivated was 7%, while with photodynamic effect alone(1J/cm2) that was 30% (?5%).when the same dosage of irradiation was applied after a rise in temperature, the fraction of cells inactivated increased to 80% (?5%) and when temperature was raised to the same amount after irradiation the fraction of cells inactivated increased to 92%. It can be seen that hyperthermia enhances, the response of tumor cells to photocarcinorin photodynamic effect in a synergistic way.
2.A PRELIMINARY REPORT OF THE EXPERIMENTAL STUDY ON THE EFFECT OF HPD AND LASER ON HUMAN HEPATOCARCINOMA CELL LINE

The present article reports the observation of the effect of HPD and laser on human hepatocarcinoma cells.The experimental result shows that HPD (50?g/ml) does not influence the growth of the hepatocarcinoma cell line,and laser alone for as long as 20 minutes does not bring about any changes of the shape of cell. Cyto-logical damage occurrs when the HPD culture medium is irradiated with lasers.A remarkable killing effect on the human hepatocarcinoma cell line is shown along with increasing HPD concentration and laser power.In order to distinguish the degree of cell damage, we suggested that changes of cell shape could be classified into three degrees.It was discovered in the experiment that most of cultured cells developed degeneration and necrosis after being treated with HPD plus laser but still there were some islands of cells alive.Authors considered that this deficiency might be unevenness in the distribution of laser action to account for the relapse of some treated cases and accordingly suggested that in the HPD treatment plus laser, the order of distribution of the irradiation should be well plotted in order to obtain better therapeutic effect.
3.EXPERIMENTAL STUDIES ON THE CYTOTOXICITY OF HAEMATOPORPHYRIN DERIVATIVE AND LIGHT IN DIFFERENT WAVE-BANDS ON HUMAN TUMOR CELL LINE

Department of Pathological Anatomy of Second Military Medical College Photosensitive reaction of HPD and its cytotoxic action were studied, focussing on the spectral effect of the irradiating light. A human hepatocarcinoma cell line(SMMC-7721)with HPD was used. Killing effects on tumor cells after irradiation of light in three different wave-bands were observed, both qualitatively and quantitatively.Results were compared with those of Kinsey and Matsushima.
4.Pathological Observations of 31 Cases of Sudden Death due to Acute Pancreatitis

The pathological changcs of pancreases, livers, kidneys, lungs and hearts in 31 cases of sudden fatal pancreatitis arc reported. It is pointed that those pathological changes may be related trypsin, especially elastinasc and phosphohpasc A,. The clinical features,of sudden fatal pancreatitis and the mechanisms why those patients died suddenly are also discussed.
5.THE RELATIONSHIPS BETWEEN PATHOLOGICAL TYPES AND INFILTRATING DISTANCES OF GASTRIC CANCER

In this paper are discussed the relationships between the infiltrating distances and the pathological types of gastric cancer by measuring the excised specimens of middle and dvanced stages in 50 cases.It is confirmed that the infiltrating distances vary according to the pathohistolog-ical types. The tubular adenocarcinoma may be divided into two subtypes: the large tubular adenocarcinoma and the small tubular adenocarcinoma. In all pathohistological types of gastric cancer the undifferentiated carcinoma, "signet-ring" cell carcinoma and tubular adenocarcinoma infiltrated farther, and the papillary adenocarcinoma and the small tubular adenocarcinoma into a less.The infiltraing type and gastric sinus carcinoma have tendency to infiltrate farther which can be observed with naked eye.These results may offered some assistance in the surgical treatment of gastric cancer.
6.Morphologic Study of Transplanted Human Hepatocaranoma in Nude Mice after Photodynamic Therapy with Sulfonated Aluninum Pathalocyanine

The morphologic changes of human hepatocaranoma transplanted in nude mice afler photodynamic therapy (PDT) with sulfonated aluminum phthalocyanine (ALSPQ were studied The cancer cells after PDT with ALSPC showad vacuolar degeneration and separation from each other. The endothelium of blood vessel was damaged and hemorrhage occured Under electron microscope the endoplasmic reticulum enlarged, mitochondria swelled, perinudear space dilated, and chromosome aggregated and condensed. The membrane of nuclei and cells broke off and tumor cells died at last The endothdial cells had the same changes mentioned above. This result suggests that ALSPC has photodynamic effect on the human hepatocellular carcinoma tranaplanted in nude mice. And the membrane system of cells is probably the main target.
7.Influence of photodynamic therapy of endogenous ALA-induced PP Ⅸ on tumor morphological pathology

To evaluate the practical photodynamic therapy (PTD) efficiency on tumor of en-dogenous ALA-induced PP Ⅸ. Methods: Morphoraglcal alterations were observed after PDT 0f protopor-phyrin Ⅸ (PP Ⅸ) by application of endogenous 6-aminolaevulinic acid(ALA) in Kunming mice bearing H22hepatic carcinoma. Results: After PDT, the tumor cells showed degeneration, deformation and nuclearnecrosis and the tumor tissues were defused with hemorrhage and necrosis. The ultrastructural pathologyindicated that mitochondria in tumor cells were injured and destroyed. C0nclusi0n: The pathological alter-ations suggest that PDT of ALA on H22 hepatic carcinoma was conducted by the destruction of mitochon-dria.
8.Anti-melanoma action of small molecular peptides derived from Brucea javanica (L.) Merr.globulin in vitro

Objective:The morbidity of malignant melanoma keeps increasing annually.It has high risks of metastasis,drug resistance,and poor prognosis in clinics.Moreover,the available medicines used commonly,such as dacarbazine,temozolomide,the v-Raf murine sarcoma viral oncogene homolog B1(BRAF) inhibitor vemurafenib,and the programmed cell death protein 1 inhibitor pembrolizumab,have some limitations at some extent.Therefore,a more effective therapeutic strategy is still urgently necessary.Methods:In this study,Brucea javanica(L.)Merr.globulins were hydrolyzed with pepsin,then ultra-filtrated to collect small molecular peptides (≤3 kDa).The peptides were then analyzed by anti-proliferative assay,cell-cycle distribution,apoptosis assay,and in vitro wound-scratch assay.Finally,western blotting was conducted to elucidate the underlying anti-melanoma mechanism.Results:The small molecular peptide from B.javanica significantly inhibited malignant melanoma cell proliferation with the IC50 of 2.72 μg/mL for 72 h.Further analysis indicated that B.javanica peptides arrested cell cycle at the S and G2/M phases and induced apoptosis by upregulating p21,p53,Bax,caspase-3,and cleaved PARP while downregulating Bcl-2 expression.The inhibitory migration effects were also confirmed by wound-healing assay.Conclusion:The small molecular biopeptides from B.javanica may be a promising bioactive agent candidate for melanoma treatment.
