1.The clinical observation of the effects of maternal serum vitamin D levels on the developing nervous system in infants

Objective To investigate the effect of maternal serum vitamin D (VD)levels on the developing nervous system in in-fants.Methods A total of 1 012 pregnant women's serum VD levels of early pregnancy were given the right amount of VD supple-mentation according to the results.They were divided into 3 groups (normal,deficiency and insufficiency group)according to the third trimester VD levels;and their infants 6-8 months were tested on the developmental quotient (DQ),mental index (MI)and other neurological development index by the Developmental Screening Test for child under six(DST).Results There were up to 94.17% of pregnant women in early pregnancy had severe VD deficiency,the average levels was (16.21 ± 7.54)ng/mL;Although VD levels was increased after supplements,there were 46.25% of pregnant women still deficient in VD.The average DQ score of 1 020 infants was 92.67±16.45 and M1 score was 97.53±17.04,which accorded with normal distribution.The rate of the infants born by mothers who had the normal VD levels were superior to those with VD deficiency and insufficiency on the quotients of out-standing and smart,and there was a significant difference between them(P <0.01).But there was no statistical difference between the deficiency and insufficiency group (P >0.05).The level of VD was positively correlated with DQ and MI,and the correlation coefficients are 0.63 and 0.2(slightly correlated),respectively.Conclusion It should be pay more attention to monitoring the VD levels of the pregnant women during the pregnancy,and for lack should be promptly added.
2.Study on efficacy and accompanying toxic and side effects of volatile oil of Evodia Fructus based on stomach cold syndrome model.

OBJECTIVETo preliminarily study the effective dosage range and mechanism of the abirritation of volatile oil of Evodia Fructus on the stomach cold syndrome model in mice, and discuss the correlation between its accompanying toxicity and oxidative damage mechanism, in order to provide the experimental basis for explaining the efficacy-syndrome-toxicity correlation.
METHODThe stomach cold-syndrome model in mice was induced by the classic hot plate test by orally administrating with different doses of volatile oil of Evodia Fructus, in order to observe its abirritation and companying toxic and side effects and detect serum ALT, AST, PGE2, NO, NOS, MDA, SOD, GSH, GSH-Px, BUN, CR and hepatic ALT, AST. The companying toxic symptoms in mice were recorded in toxic reaction integral table.
RESULTVolatile oil of Evodia Fructus had an obvious analgesic effect at 30 min after the oral administration and reached the peak effect at 60 min, with certain "dose-effect" and "time-effect" relations, rises in serum and hepatic ALT and AST levels, serum PGE2, MDA, NO and NOS and hepatic indexes, decreases in SOD, GSH and GSH-Px and no notable change in BUN, CR levels and kidney weight/body ratio. Conclusion: The abirritation mechanism of volatile oil of Evodia Fructus was related to the inhibition of pain transmitter release, peroxidative damage and NO damage, which is accompanied by certain hepatotoxicity, mainly mainly oxidative damage, with a concurrent "dose-time-toxicity" relationship.

3.Study on efficacy accompanied by side effects of water extraction components of Evodiae Fructus based on syndrome model.

The range of effective dose and mechanism of abirritation about water extraction components of Evodiae Fructus on the stomach cold syndrome model in mice were preliminary studied. The method of stomach cold-syndrome model in mice was built, which were administrated with different doses water extraction components of Evodiae Fructus, observing abirritation and toxicity by the classical hot plate method, detecting the level of ALT, AST, PGE2, NO, NOS, MDA, SOD, GSH, GSH-Px, BUN, CR in serum and ALT, AST in hepatic tissue, and recording toxicity symptoms in mice according to the list of relevant toxicity reaction. The water extraction component of Evodiae Fructus has obvious analgesic action after administration 30 min, arriving peak effect after administration 60 min, showing certain "dose-time-toxicity" relationship. ALT and AST levels in mice serum and liver tissue enhanced; PGE2, MDA, NO, NOS enhanced in mice serum; SOD, GSH, GSH-Px reduced; the BUN, CR levels was no significant alteration; liver weight/ body weight enhanced; kidney weight/body weight was no significant alteration. The a irritation mechanism of volatile oil of Evodiae Fructus was connected with suppressing pain transmitters release, per oxidative damage mechanism and NO damage, which also induced hepatotoxicity and the mechanism of hepatotoxicity is main lyoxidative damage, showing certain "dose-time-toxicity" relationship in accordance to hepato-toxicity injury.

4.The effect of neural stem cell transplantation on the hindlimb motor function and caspase-3 expression of motor cortex in spinal cord transected rats

Objective To observe the effect of neural stem cell (NSC)transplantation on the hindlimb motor function and caspase-3 expression of motor cortex (MC) in spinal cord transected (SCT) rats. Methods S prague-Dawley (SD) rats were randomly divided into sham operated group, operation group( T9 transection), and subacute NSC transplantation group. The MC of each group( n= 8 )was harvested 7 days post operation (dpo), then western blot was employed to detect the level of caspase-3( β-actin was used as control ). The left 5 animals of each group were subjected to BBB score evaluation at 16th week,then the animals were sacrificed and the MC was harvested and performed immunostain by using caspase-3 rabbit antibody. Results Following NSC transplanta tion, BBB scores( ( 7.58 ± 0. 99 ) ) increased significantly than seen in the SCT animals( ( 5.16 ± 1.19) ). The expressional level of caspase-3 at 7day post operation was( (0.89 + 0.12)) in MC of SCT rats,while it decreased significantly to( (0.76 + 0.11 ) ) in NSC transplantation rats(P<0.05). The immunoreactive stain of caspase-3 was seen in the cytoplasm of pyramidal neuron in the cortex. Conclusion NSC engraft can downregulate the expression of caspase-3, corresponding to a significant improvement in hindlimb motor function. These findings indicate that NSC transplantation probably regulate the expression of apoptosis genes in MC to promote neurological function recovery in rats subjected SCT.
5.Comparison of curative effect of two treatment methods in children with bronchial asthma

Objective To investigate curative effect in children with bronchial asthma by two treatment methods. Methods 100 children with bronchial asthma,according to the different treatment were divided into treatment group 50 cases and control group(50 cases). The first seconds vital capacity(FEV1),peak expiratory flow (PEF) ,vital capacity 25% of the instantaneous velocity (V75), vital capacity 50% of the instantaneous velocity (V50) ,vital capacity 75% of the instantaneous velocity(V25) and their percentage the percentage of predicted value were observed. Results Total effective of 43 cases(86.0%) in treatment group after treatment 4 weeks were higher than that in control group of 28 cases (56. 5 %) (x~2 = 3. 987, P < 0.05); The percentage of FEV1 and PEF in two groups after treatment 12 weeks were significantly higher than that before treatment(x~2 = 4. 01,4. 21,4. 31,4. 08, all P < 0.05), but it between the two groups was not statistically significant (x~2 = 2. 31,2. 41, all P > 0.05); The pulmonary function of V75 、V50、V25 in two groups after treatment 12 weeks reached projected value of more than 90% ;The pulmonary function of V75 、V50、V25 after treatment compared with those before treatment were improved significantly (x~2 =4.285,4.234,4.311,4.278,all P<0.05). Conclusion The curative effect of bronchial asthma were confirmed in two methods,but treatment group were better than the control group.

7.Breeding Selenium-Enriched Yeast by Protoplast Mutagenesis.

This paper studied on breeding selenium-enriched yeast by protoplast mutagenesis. A strain which the content of selenium is the highest is selected from thirteen strains yeast. The optimum conditions to form protoplast are lysed by 1 g/100 mL lywallzyme for 120 min, the formation and regeneration being 95.2% and 21.8% respectively. By mutating breed a strain of A1 which the content of selenium is 821 mg/kg and the amount of dry cell of 0.84 g/100 mL is obtained.
8.Prevention of Post-operative Adhesion with Early Electro-acupuncture Treatment

ObjectiveTo investigate the effect of early electro-acupuncture treatment on prevention of post-operative adhesions.Methods40 rabbits were randomly divided into 5 groups: normal control group, model group 1, model group 2, early electro-acupuncture group and later electro-acupuncture group with 8 animals in each group. Muscle adhesion model was established by operation. The early electro-acupuncture group was received the treatment from the 11th to 20th day after the operation; later electro-acupuncture group was from the 21st to 30th day after the operation. The weight, gait and range of motion (ROM) of animals were observed and analyzed.ResultsThe standing, walking and ROM of animals of the early electro-acupuncture group improved; but only few gait indexes improved and ROM not changed in the later electro-acupuncture group.ConclusionThe early electro-acupuncture treatment can prevent the post-operative adhesion of muscles, but not having effect on formed adhesion.
9.NOK Activates STAT3 Signaling by a JAK2-Dependent Mechanism

Novel oneogene with kinase-domain (NOK) can activate multiple mitogenic signaling pathways including the janus kinases(JAK) and signal transducer and activator of transcription proteins (STAT). It was showed that NOK specifically and physicallyinteracted with STAT3 in human embryo kidney 293T (HEK293T) cells. In addition, NOK could directly interact with most of the STAT3 subdomains except coiled-coil and C-terminal domains. Removing ectodomain and transmembrane domain of NOK markedly enhanced its intermolecular interaction with STAT3. Also, NOK could co-immtmoprecipitate with JAK2. in vivo. Importantly, co-expression of NOK and JAK2 produced a synergistic effect on NOK-mediated STAT3 activation, while inactivating the kinase domain of JAK2 completely prevented this synergistic effect. Overall, the results indicated that NOK might complex with both STAT3 and JAK2 and activate STAT3 signaling by a JAK2-dependent mechanism.
